Abstract

In the body, 2-methoxyethanol compound may be converted to MAA. MAA is a strong oxidant and may cause oxidation stress in spermatozoa. Oxidation stress is a disturbance on phosphorilation that increases ROS concentration, and it produces lipid peroxide in spermatozoa membrane resulted in high MDA concentration. One of indicator of spermatozoa membrane disturbances is a lack of spermatozoa membrane integrity. The main purpose of this research was to determine the relationship between MDA concentration in sperm and membrane integrity of spermatozoa in rats. The animal of treated groups (n = 40) were divide into 8 groups of 5 each. The rats were given subcutaneous injection with 0,2 ml of 200 mg/kg/day for 1 day (P1), 3 days (P2), 6 days/week (P3), and 12 days/two weeks (P4), respectively the control group was injected with physiological saline of the same volume. The concentration of MDA was measured by spectrophotometer and observing membrane integrity used HOS method to watch the spermatozoa response on hypoosmotic condition. The results of the research indicated that 2-ME caused the increasing in sperm MDA concentration and the decrease of spermatozoa membrane integrity. There was negative correlations between MDA concentration and spermatozoa membrane integrity.

Abstract

The research was conducted to evaluate the effect of amphetamine as pshycostimulan drug on ultrastructure spermatogenic cells of male rats (Rattus norvegicus L.). Ultrastructure spermatogenic cells described from six male rats of Wistar strain. They were divided in to 2 groups (control and treatment group). Four mg/kg/days amphetamine were given sub-cutaninjection. In control to injection with physiologic saline. The last of the treatment the rat were killed after 100 days the amphetamine injection. The testis were released into fixative solution. Ultrastructure morphologic analysis of the testis were evaluate of quantitative to assess the effect of amphetamine on spermatogenic cells. Observation of spermatogenic cells (Spermatogonia, Spermatocyte, and Spermatyd cells) were done under the electrone microscope. The result of this experiment showed different morphologically from the amphetamine- treated group and their respective control. In the treatment groups, Spermatogonia, Spermtocyte, and spermayd cells less endoplasmic reticulum than normal spermatogenic cells (control). More vacuolization in membrane line of Spermatogoium cells and in cytoplasm of Spermatocyte, and Spermatic cells than normal spermatogenic cells (control).

Abstract

Immunomodulators are substances acting as stimulants or suppressants against the immune system, either to specific or non-specific activity. The non-specific through the production of leukocytes as the first response in fighting against an attacking antigen. Meanwhile, the specific, hrough recognition of antigens by lymphocytes during reattack. This research aimed to determine the immunomodulatory activity of the extracts combination nanoparticles of Allium sativum, Curcuma mangga, and Acorus calamus on the leukocyte profile of mice. This study used 5 treatments and 6 repetitions. The treatment groups were K- (untreated group), P1 (extracts combination nanoparticle dose of 25 mg/kg), P2 (extracts combination nanoparticle dose of 50 mg / kg), P3 (subur kandungan herbal medicine dose of 75 mg / kg), and P4 (Clomiphene citrate dose of 0.9 mg / kgBW). The parameters used included the total number of leukocytes and their differential value. The data that met the parametric assumptions, such as normally distributed and homogeneous were examined using the One Way ANOVA test, and when there was a significant difference, it was processed with the Duncan assessment. While those that did not meet the assumptions were evaluated using a non-parametric analysis. The statistical results showed that administration of extracts combination nanoparticles of A. sativum, C. mangga, and A. calamus at doses of 25 mg/kg and 50 mg/kg were able to suppress the inflammatory reaction by decreasing the total number of leukocytes. However, the differential leukocyte count was able to maintained or modulate immune system, indicating by the percentage of neutrophil, basophil, and eosinophil in the normal range. At a dose of 50 mg/kg, decreased the percentage of lymphocytes, while for monocytes, all dosage ranges were able to increase their number.

Abstract

Diabetes Mellitus condition (DM) correlates with the damage that occur in pancreas especially in Langerhans islets, one of the damage indicators is diameter changes of Langerhans islets that getting smaller compare with the normal sizes. The use of natural ingredients to control diabetes mellitus can be done by infusing sweet beauty watermelon albedo which can be found abundant in traditional market. Albedo layer of watermelon contain antioxidant compounds that can help to regenerate Langerhans islets of pancreas. This study purpose was to know the effect of giving watermelon’s infusion albedo to diameter of Langerhans islets in diabetes mellitus rats (Rattus norvegicus), and analyze the potential process occurred based on the result as information source for other research. This study used 25 male rats (Rattus norvegicus) as animal model with 5 groups treatment which were normal group, positive group as control (DM without treatment), A group (DM+1000 mg/kgBB watermelon’s infusions albedo), B group (DM+1500 mg/kgBB watermelon’s infusions albedo), and C group (DM+2000 mg/kgBB watermelon’s infusions albedo). After all the treatments, the rats were anesthetized and dissected and then their pancreas was taken. The pancreas was processed into histological slides by fixing the pancreas in a fixative solution (10% formalin) then dehydrated in a series of graded ethanol and embedded in paraffin wax, cut at a thickness of 4 µm, and finally stained with hematoxylin-eosin dye for microscopic observation, according to the stages Histopathological analysis of the gonads was examined using standard methods. Data was collected by measuring the size of Langerhans islets with microscope Olympus CX33  and connected to the raster image application.  Data results from this study indicate that watermelon’s infusion albedo had effect to increase the diameter of Langerhans islets of rat with diabetes mellitus

Abstract

Diabetes is closely related to immune response problems when it occurs chronically. Pegagan (Centella asiatica) is a medicinal plant with active compounds. Madecassoside is beneficial in treating diabetes, and nanoparticle technology is expected to enhance the medicinal potential and availability of pegagan compounds. The aim of this study was to determine the effect of chitosan-coated pegagan nanoparticles on the cytokine profile of chronic diabetic mice, which included CD4+TNF-α+, CD8+TNF-α+, CD4+IFN-γ+, CD8+IFN-γ+ and IL-6+. An experimental study with a randomized complete block design (CRD) consisting of six treatments with seven replicates was conducted. The groups were: healthy mice as negative control; diabetic mice treated with distilled water as positive control and diabetic mice treated with nanoparticle coated with chitosan (NPC) 20 mg/kg, 30 mg/kg, 40 mg/kg, and metformin 130 mg/kgBW. The data were tested using one-way analysis of variance (ANOVA) with a significance level of 5% and continued with the Duncan’s multiple range test. The results showed that pegagan NPC could significantly reduce the relative number of CD4+TNF-α+, CD8+TNF-α+, CD4+IFN-γ+ and CD8+IFN-γ+ and IL-6 in the dose of 20 mg/kg, 30 mg/kg and 40 mg/kg (p<0.05). The treatment dose of 20 mg/kg reduced CD4+TNF-α+, CD8+TNF-α+, CD4+IFN-γ+, CD8+IFN-γ+ to the levels of healthy mice and a dose of 30 mg/kg could reduce IL-6 as in healthy mice. These findings suggest that chitosan-coated pegagan nanoparticles are a promising therapy for diabetes, as they have the potential to modulate the immune response associated with chronic diabetes.

Abstract

Breast cancer can perform metastasis to distant organs and cause more than 90% of malignancy-related deaths. The anti-metastasis potency of nanodiamond-conjugated annonacin and squamocin against MCF-7 cells is currently studied. First, IC50 determination of both free annonacin and squamocin to evaluate their potency as cytotoxic agents. Upon getting the IC50 value, both compounds are conjugated into nanodiamonds. Drug loading efficiencies of nanodiamond-conjugated annonacin and squamocin are 88.9% and 89.1%, respectively. Meanwhile, the ND-annonacin and ND-squamocin complex size is 150-300 nm based on SEM imaging. Subsequently, cell viability assessment of MCF-7 was performed with six cohort designs, namely, K (control cell), AN (annonacin), SQ (squamocin), NDAN (nanodiamond-conjugated annonacin), and NDSQ (nanodiamond-conjugated squamocin). Both IC50 and cell viability are assessed by MTT assay after 24 h incubation. All cohorts also underwent gene expression analysis subject to the metastasis markers CTNND1 (catenin delta 1), NOTCH4, and C-JUN. Here, the IC50 of both free annonacin (4.52 µg/ml) and squamocin (10.03 µg/ml) are more than IC50 of potent anticancer (< 4 µg/ml) for pure compounds. However, nanodiamond conjugation to both compounds can decrease cell viability better than free compounds. Compared to K, nanodiamond-conjugated annonacin and squamocin significantly decreases cell viability after 24 h incubation. Bioinformatics analysis confirmed significant pro-metastasis (C-JUN and NOTCH4) upregulation and anti-metastasis (CTNND1) downregulation in tumors compared to normal. Recent findings demonstrated that nanodiamond-conjugated annonacin can significantly upregulate CTNND1 and significantly downregulate C-JUN and NOTCH4. Even so, nanodiamond-conjugated squamocin upregulate CTNND1 but not significantly and significantly downregulate C-JUN and NOTCH4.

Abstract

Anggota Familia Sciaenidae mencakup sekitar 70 genera dan 270 spesies di seluruh dunia. Penelitian tentang hubungan kekerabatan fenetik anggota Familia Sciaenidae belum banyak diinformasikan. Hal ini dapat dijadikan sebagai data dasar anggota Familia Sciaenidae yang didaratkan di TPI Asemdoyong, Pemalang. Tujuan penelitian ini untuk mengetahui jumlah spesies, hubungan kekerabatan fenetik dan kunci identifikasi sederhana anggota Familia Sciaenidae yang didaratkan di TPI Asemdoyong Pemalang. Metode yang digunakan adalah survey, pengambilan sampel secara purposive random sampling sebanyak 3 kali. Variabel yang diamati adalah hubungan kekerabatan fenetik anggota Familia Sciaenidae. Parameter yang dihitung adalah jumlah spesies dan nilai koefisien similaritas. Hubungan kekerabatan dianalisis menggunakan program NTSYS versi. Hasil penelitian ini ditemukan lima spesies dari Familia Sciaenidae yaitu Johnius amblycephalus, Johnius carouna, Johnius heterolepis, Pennahia anea, Otolithes ruber. Kekerabatan fenetik terdekat adalah J.heterolepis dan J. carouna dengan nilai koefisien similaritas 0,811. Kekerabatan fenetik terjauh adalah J.amblycephlaus dan P.anea serta O.ruber dengan nilai koefisien similaritas 0,556.

Abstract

Acaudina rosettis is one of the commercial sea cucumber species that is widely consumed on the east coast of Surabaya, East Java, Indonesia. Although its abundance in nature has begun to show signs of decreasing, studies on this species are still very limited. This study aimed to examine indicators that can be used to determine the sex of A. rosettis individuals. Acaudina rosettis samples were collected from Madura strait in the mid of Hijri month for three months. The sea cucumber samples were then dissected. Body wall without viscera and gonads was weighed. The gonads were then characterized macroscopically and microscopically, prepared into histological slides with hematoxylin-eosin staining. The results showed that A. rosettis samples could be classified into three sex groups based on the type of gamete cells observed in the tubules; male (spermatogenic), female (oogenic), and undetermined sex (no gamete cells found). Gonads had three colors; yellow, pink, and orange and were in five stages of reproductive phase; gametogenesis (G), mature (M), spawning (S), and post-spawning (PS). Male samples had yellow and orange gonads, female samples were pink and orange, and all undetermined sex were orange. The G gonads were yellow and orange, M were yellow and pink, while S and PS were orange. Body wall and gonad weight at the same reproductive phase did not show any difference between males and females. Based on the results, gonad color can be used as an indicator of sex determination, but only in G and M reproductive phases.

Abstract

This study aimed to determine the effect of arak bali on the quality of spermatozoa include morphology, motility, viability, membrane integrity of spermatozoa rat (Rattus norvegicus). The study was conducted in two phases: the first phase of the deployment of questionnaires conducted in five districts in Bali to determine the type and frequency of arak bali consumption and phase II made arak bali giving treatment in experimental animals. This study used 24 male rats (170-200 grams), divided into four groups: one control and three treatments (by arak bali containing 40% alcohol as much as 0.1 and 0.5 mL and 0.1 mL much alcohol synthesis, for 45 days. the results showed that of the five districts in Bali, most people consume arak bali commercial and most of the frequency of consumption of the week more than one bottle (350 mL). the provision of arak bali in experimental animals, degrade the quality (morphology, motility, viability, membrane integrity), the greater the volume given declining spermatozoa quality.

Abstract

Polytyrene nanoplastics (NPs) (<1 µm) have high toxicity when entered and accumulated in cells. NPs accumulation causes oxidation stress, thus increasing reactive oxygen species (ROS) production, resulting in necrosis or apoptosis, as well as affecting endogenous antioxidant activity, such as superoxide dismutase (SOD) and catalase (CAT). Cinnamomum burmanni plant contains flavonoids, cinnamaldehyde, phen, and olic acid, potentially exogenous antioxidants. The study aims to analyze the potential of C. burmanni leaf extract for SOD and CAT levels, sperm quality, epithellia tubulus thickness, tubulus seminiferus diameter, and number of spermatogenic Rattus norvegicus cells exposed to NPs. Twenty-five male R. novergicus are divided into five groups (n = 5): two controls (without and with NPs) and three groups (combination of NPs and variations in extract concentrations of 100, 200, and 400 mg/kg. The SOD and CAT levels were measured with an ELISA kit. The histology was observed by counting the spermatogenic cells, measuring the epithellia thickness, and tubulus seminiferus diameter. The sperm motility, viability, and sperm count were observed to determine the sperm quality. Leaf extract of C. burmanni treatment with different concentrations not significantly increased SOD levels but significantly decreased the levels of CAT (P<0.05). The addition of C. burmanni leaf extract significantly increased the spermatogenic cell count, epithelia tubulous thickness, sperm viability, and sperm count (P<0.05), while sperm motility and tubulous seminiferous diameter not significantly increased. According to the results, C. burmanni leaf extract has antioxidant potential against the toxic effects of NPS.
