Abstract

The microbial community in Kawah Hujan, Kamojang, West Java, Indonesia, was analyzed using 16S-rRNA-gene-sequencing combining with denaturing-gradient-gel electrophoresis (DGGE) technique. Two different cell lysis methods, enzymatic-based, and physical treatment-based DNA extraction, were used to isolate chromosomal DNA for 16S rDNA gene-fragment amplification. The DGGE profiles showed some differences in banding pattern that were obtained from both cell lysis methods. The DNA sequence analysis of the individual DGGE bands revealed that most of the band sequences obtained by physical treatment were close to 16S rRNA gene fragments from the bacterial domain, while most of band sequences performed by enzymatic method had high homology with 16S rRNA gene fragments from archaeal domain. Further analysis of the sequences from both methods performed by comparisons with the Ribosomal Database Project showed that some of DGGE sequences from Kawah Hujan consisted unique 16S rDNA sequences.

Abstract

Telah dilakukan analisis kualitatif dan kuantitatif asam lemak tak jenuh Omega-3 dan Omega-6pada minyak ikan patin (Pangasius pangasius) dengan menggunakan metode kromatografi gas. Kadar Omega-3yang diperoleh dari minyak ikan patin yaitu berkisar antara 1,16-12,44(%W/W) dan kadar Omega-6 berkisaranantara 12,278-15,961(%W/W). Dari kadar Omega-3 dan dari kadar Omega-6 yang diperoleh dari penelitianini, ikan patin dapat digunakan sebagai sumber Omega-3 dan Omega-6 alternatif. Dalam penelitian ini jugadilakukan karakterisasi minyak ikan patin yang meliputi kadar minyak, bilangan asam, bilangan penyabunan,dan bilangan peroksida dengan metode Badan Standarisasi Nasional. Dari hasil penelitian pada ikan patindengan berat 650-879 gram diperoleh kadar minyak rata-rata 3,827(%W/W), bilangan asam berkisar antara3,667-19,521 mgKOH/gr, bilangan penyabunan berkisar antara91,319-192,656 mg KOH/gr, dan bilangan peroksida berkisar antara 0,778-17,78 mek/kg.

Abstract

Ogan ethnic population in Indralaya, Ogan Ilir District, South Sumatra was known to have used several types of plants, there are Swietenia mahagoni, Averrhoa carambola, Syzygium samarangense, Musa acuminata, Nymphaea rubra, Syzygium polyanthum, and Andrographis paniculata for the treatment of hypertension. But, scientific information has not yet been found secondary metabolites that are responsible for providing these activities. The study began with the extraction of methanol from the seven types of plants using methanol as a solvent. Each thick extract was tested for its activity using the 1,1-diphenyl-1-picryllhydrazyl (DPPH) method. The extract which showed the highest activity (methanol extract) was subsequently carried out in vitro antihypertension test using the Angiotencyn Converting Enzyme (ACE) method. Antioxidant test results showed that the methanol extract from S. samarangense stem bark had the highest antioxidant activity with IC50 was 83.06 μg/mL. Antihypertension test of methanol extract from S. samarangense stem bark obtained IC50 ACE was 61.56 μg/mL. Based on the IC50 value, it shows that S. samarangense stem bark has potential as a source of antioxidant compounds as well as a source of antihypertension compounds. Keywords: Syzygium samarangense, stem bark, antioxidant, antihypertension

Abstract

Pelatihan pemanfaatan limbah organik dari sisa rumah tangga, pekarangan dan perkebunan menjadi bokashi sebagai media tanam berbagai jenis sayuran, buah-buahan dan tanaman telah dilaksanakan di Desa Tanjung Pering, Ogan Ilir. Kegiatan ini bertujuan memberikan wawasan dan keterampilan bagi peserta dalam memanfaatkan limbah menjadi pupuk yang bernilai ekonomi dalam menunjang sektor pertanian yang berkelanjutan. Kegiatan ini merupakan kerjasama Universitas Sriwijaya dengan Dinas Kehutanan Provinsi Sumatera Selatan (Seksi Penyuluhan dan Pemberdayaan Masyarakat dan Seksi Pengendalian Kebakaran Hutan dan Lahan). Kegiatan ini merupakan bagian dari Program Pengabdian kepada Masyarakat (PPM) oleh Unsri kepada desa binaan. Melalui kegiatan ini diharapkan masyarakat mulai memanfaatkan limbah di lingkungan masyarakat menjadi produk yang bernilai ekonomi.

Abstract

Ogan ethnic community in South Sumatera has been used a decoction of the stem bark of Syzygium samarangense L (locally named “jambu air” in Indonesia) for hypertension medicine. Hypertension is a degenerative disease caused by free radical activity in the body. This study aims to scientifically prove the use of a decoction of the stem bark of S. samarangense so that its use by the Ogan ethnic community can be justified. The research began with fractionation of the stem bark by gradient solvents, and each fraction was tested for antioxidant activity. Isolation of antioxidant compounds from active fractions was carried out by chromatographic techniques. Antioxidant tests on fractions and pure compounds were carried out by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. Based on spectroscopic data analysis and by comparing with literature, the pure compounds were identified as quercetin and gallic acid. Both of these compounds have strong antioxidant activity with IC50 quercetin 11.16 and gallic acid 11.43 µg/mL. This research proves that the use of the stem bark of S. samarangense for a degenerative disease by the Ogan ethnic community contains the truth.

Abstract

Leaves of jambu air (Syzygium samarangense L.) has been used by local residents as medicine for various diseases caused by free radical agents in human’s body. This study aims to find a new source of antioxidants from medicinal plants and their endophytic fungi Syzygium samarangense leaves was fractionated by maceration method using gradient solvent i.e.  n-hexane, ethyl acetate, and methanol. The antioxidant activity of the leaf fractions was determined using 1,1diphenyl-2-picryl hydrazyl (DPPH) method. The chemical compound was isolated from active fraction by chromatographic techniques and their chemical structure was identified using spectroscopy techniques.  The endophytic fungi was isolated from S. samarangense leaves and continued to cultivation in potato-dextrose broth (PDB) medium for four weeks. The medium was extracted using ethyl acetate and determined its antioxidant activity. Fungi’s isolate with highest activity was analyzed to identify its molecular. Ethyl acetate fraction of S. samarangense leaves showed highest antioxidant activity. Spectroscopy analysis result concluded the isolated compound is 5,7-dihydroxy-6,8-dimethyl flavanone. Four endophytic fungi had been isolated form S. samarangense leaves. Antioxidant activity test showed that ethyl acetate extract of endophytic fungi BJA-1 has the highest value. Molecular identification of BJA-1 shows high homology with Lasiodiplodia venezuelensis strain CBS 129753.

Abstract

Tetracera indica Merr. in Musi Banyuasin, is one of the traditional medicine used by the community for the treatment of kidney stone disease and gout, but this claim is not recorded in the treatment of kidney stones and gout in Indonesia.  In this study, isolation of antioxidant compound from ethyl acetate extracts of supit (Tetracera indica) was done. The isolation was carried out through step gradient polarity extraction, and separated and purified by chromatography technique. The determination of the structure of the isolated compound was performed by spectroscopy method including UV, IR, and NMR 1D and 2D, and antoxidant activity was determined  by DPPH method.  An  active antioxidant compound was isolated from ethyl acetate extract in form of yellow solid (15 mg).  Based on spectroscopic analysis the isolated compound was 5,8-dihydroxy-7-methoxyflavone.  The compound showed strong antioxidant activity (IC50 8.25 μg/mL) higher than standard ascorbic acid (IC50 11.3 μg/mL).  This data concluded that efficacy of supit (Tetracera indica) for the treatment related to antioxidant activity (uric acid) is proven by the identification of one antioxidant compound of this plant.

Abstract

Flacourtia  rukam is a plant popular among people to treat hypertension, especially the Musi Banyuasin of south Sumatera, Indonesia. Isolation and identification of chemical compounds from F. rukam leaves and evaluation of their effects on antihypertensive activity have been conducted. Isolation of chemical compounds using chromatographic methods and identification using spectroscopic methods were compared with the reported data. The drug’s effects on antihypertension were determined using the  angiotensin converting enzyme (ACE) inhibitory method. Two compounds were first reported and isolated from the leaves of F. rukam and identified as apigenin (1) and lupeol (2). These compounds were demonstrated to be effective in treating antihypertension with IC50 656.51 ± 1.55 µg/mL for apigenin and 15.12 ± 0.72 µg/mL for lupeol. It can be concluded that  F. rukam leaves is a potential ACE inhibitor can be explored further as an effective antihypertensive agent.

Abstract

Desa Tebing Gerinting merupakan salah satu desa di Indralaya Selatan Kabupaten Ogan Ilir, Sumatera Selatan.  Sumber mata pencaharian penduduk sangat beragam mulai dari petani, peternak, berkebun, pedagang dan usaha rumah tangga membuat kemplang, dengan rata-rata tingkat perekonomian yang masih rendah. Usaha pembuatan kemplang juga mempunyai kendala dalam pemasarannya dengan banyaknya masyarakat yang membuka usaha tersebut. Untuk itu perlu diperkenalkan  alternatif lain sebagai peluang usaha pada masyarakat  Desa Tebing Gerinting. Pada kegiatan ini telah  diperkenalkan kepada masyarakat inovasi keripik berbahan baku kelapa aneka rasa yang renyah dan berkualitas  dengan nilai jual tinggi sebagai suatu peluang usaha.  Dari kegiatan yang dilakukan terlihat bahwa pengolahan kelapa menjadi keripik merupakan suatu pengetahuan baru bagi masyarakat Tebing Gerinting, sehingga terlihat rasa ingin tahu dan antusias peserta dalam kegiatan ini.  Masyarakat mengetahui bahwa pengolahan kelapa menjadi keripik juga sangat sederhana  dan bahkan memiliki nilai jual yang tinggi. Survei yang dilakukan menunjukkan masyarakat suka akan produk yang dibuat dengan persentase penilaian rasa biasa (15%), suka (65%), dan sangat suka (20%).  Kegiatan ini mendapatkan respon yang baik dan telah menambah pengetahuan masyarakat untuk mencoba alternatif lain peluang usaha untuk meningkatkan pendapatan masyarakat.

Abstract

Abstract: Colony morphology yeast isolates from tuak and its application in producing ethanol from sugarcane bagasse has been conducted. Yeast isolates used in this work were HT4, HT5, and HT20 obtained from Tuak. These isolates have cream to yellow in color, spherical to oval in shape, and grow as budding cells. Our goal is to find yeast isolates used as microbial agent in fermentation process producing ethanol from lignocellulosic biomass as raw material.  Based upon morphology showed that these isolates were Candida species.  Isolate HT5 then used to see its potent  as  agent in bioethanol production.  Fermentation of 5 g sugarcane bagasse substrate resulted 0.0008% ethanol.  This results indicated that isolate has a potent as microbial agent for fermentation, however the optimum condition of process is needed to furthermore study.    Keywords: Candida species, bioethanol, TuakAbstrak (Indonesian): Telah dilakukan penelitian terhadap morfologi koloni dari isolat khamir dari tuak dan aplikasinya dalam produksi etanol dari ampas tebu.  Isolat khamir yang digunakan adalah koloni HT4, HT5, dan HT20  yang telah didapat dari tuak.  Isolat-isolat ini memiliki bentuk warna krem hingga kuning dengan bentuk bulat hingga oval dan pertumbuhannya dengan membentuk tunas.  Tujuannya adalah untuk mendapatkan isolat ini akan digunakan sebagai agen mikroba pada proses fermentasi menghasilkan etanol dari biomasa lignoselulosa sebagai bahan bakunya.   Berdasarkan morfologi menunjukan bahwa isolat-isolat ini adalah spesies Candida.  Isolat HT5 selanjutnya digunakan untuk melihat potensinya sebagai agen dalam produksi bioetanol.  Fermentasi menggunakan 5 g subtrat ampas tebu menghasilkan etanol  sebesar 0.0008%.  Hasil ini mengindikasikan bahwa isolat memiliki potensi sebagai agen fermentasi menghasilkan etanol akan tetapi masih perlu diteliti lebih lanjut untuk kondisi optimum prosesnya.Kata kunci: spesies Candida, bioetanol. tuak
