Abstract

Seedless fruit is one of the criteria (necessary) to improve the quality of Simadu tangerine. The most effective method to obtain seedless triploid cultivars is hybridisation between tetraploid and diploid parents. Simadu tangerine is a diploid plant. Tetraploid Simadu tangerine can be obtained with doubling chromosome using colchicines.The aim of this research was to obtain tetraploid Simadu tangerine shoot which would serve as parent to produced seedless Simadu tangerine. Shoot-tips of Simadu tangerine without leaves were treated with colchicines at four different concentrations (0, 0.1, 0.2, and 0.3%) for 3 hours. The results showed that the high concentration of 0.3% reduced survival rate. The colchicine treatments reduced growth of shoot-tip of Simadu Tangerine.The leaves of colchicines treated shoots were thicker than control. Leaves from control (0% colchicine) and 0.1% colchicine treated shoots had 8.67 and 18.25 chloroplast per pair of guard cells. Compared to those of control, leaves with 0.1% colchicine had lower stomatal density, and larger stomatal size. It appeared that 0.1% colchicine treatment resulted in tetraploid Simadu Tangerine Shoot.Keywords:chloroplasts, doubling chromosomes, stomatal size, stomatal density

Abstract

ABSTRACTMandarin Citrus cv. garut is a of local citrus variety with several superiority, but not competitive to imported citrus. Quality improvement have been the subject of citrus breeding programme. The objective of this research was to determine lethal dose 50 (LD50) and putative mutans regenerant selected based on morphological markers. Callus was irradiated at doses of 0, 20, 30, 40, 50, 60, 70, 80, 90 and 100 Gy and regenerated through somatic embryogenesis. The result of radiosensitivity showed that lethal dose 50 (LD50) analyzed by Curve Expert 1.4 software was 75.31 Gy. Observation on callus showed variation on morphology and weight of callus. Callus growth was not inhibited at doses 0-50 Gy but at doses 60-100 Gy callus growth was inhibited. Gamma irradiation also affected the formations of somatic embryos. After six weeks on maturation medium, the highest number of embryo somatic was produced from doses of 20 and 100 Gy and following culture on germination medium, the highest number of planlet was produced from doses of 20 and 40 Gy. Abstract

The gene that regulates callose could be modified by CRISPR/Cas9 technology. This research aimed to insert the CRISPR/Cas9-CsCS gene into Ponkan orange genome using Agrobacterium tumefaciens. The explants were soaked in a bacterial suspension for 20 minutes and incubated for 2-3 days. In vitro acceleration growth was conducted with a two-factor completely randomized design. The first factor is the type of explant with three levels (nucellar embryo, zygotic embryo, cotyledon node), and the second factor is the type of media with 6 levels (VMW, MT, MSK0, MSK1, MSK2, MSK3). The results showed of all explant types, the highest plant height average and number of leaves were obtained in a media combination of MS + Kinetin 2 mg L-1, MT, and MS + Kinetin 3 mg L-1. The highest transformation efficiency was in the nucellar embryo explant, while the highest regeneration efficiency was in the zygotic embryo explant. The highest shoot tip grafting percentage was achieved in the cotyledon node explant at 100%. In the grafting phase, the putative transformants before and after artificial bacterial inoculation showed that Ponkan 606 and Ponkan 597 had the highest plant heights, respectively. The intensity of Huanglongbing attacks after bacterial inoculation showed that three genotypes did not show HLB symptoms in the 24th week of observation, namely genotypes 598, 606, and 607. This study concluded that gene transformation in citrus plants produced three genotypes that did not show HLB symptoms. Abstract

Plant regeneration post-genetic transformation play an important an role in genome editing activities that should be optimized via several factors such as the composition of the medium and the concentration of plant growth regulators. This study aimed to optimize the kinetin concentrations and medium compositions for shoot multiplication originating from cotyledon node explants of several local citrus cultivars. The cotyledonary nodes from three citrus cultivars (Batu 55, Siam Madu, and Proksi-1 Agrihorti) were incubated in MS medium with Morel and Wetmore vitamins (VMW) supplemented with several kinetin concentrations (0; 0.2; 0.4; 0.6; 0.8; and 1 mg/L). The best kinetin concentrations for number of shoots variable were then combined with MT medium. The results showed that kinetin concentration at 0.8 mg/L gave the best number of shoot in Batu 55 cultivar as well as 1 mg/L concentration in Siam Madu and Proksi-1 Agrihorti cultivars. The combination 1 mg/L kinetin with Murashige and Tucker (MT) medium showed the best number of shoots, percentage of shoot formation, number of leaves, number of nodes, and shoot length in this study. This medium composition could be further used for shoot multiplication in genetic transformation in those three citrus cultivars, including genome editing activities in development of new improved citrus varieties.

Abstract

Tetraploid citrus are important for interploidal hybridization to create triploid seedless citrus. Colchicine is the most commonly used as antimitotic agent to induce polyploid plants. Tetraploid induction by colchicine in Pummelo ‘Nambangan’ was conducted in vitro using different types of explants. The aim of this research was to induce tetraploid pummelo ‘Nambangan’ by colchicine treatment using germinated seed, shoot tip and cotyledonary node as explants. Tetraploid shoot induction was conducted by soaking germinated seeds, shoot tips and cotyledonary nodes in 0.1 % colchicine for 1, 3 and 5 hours. Regenerant shoots were grown on MS medium and their growth was observed after four weeks in culture. Ploidy level was determined using flow cytometry analysis. Stomata density, length and width of stomatal guard cell were also recorded. The results showed that shoot elongation was inhibited by colchicine treatment. Soaking of shoot tip explants in 0.1 % colchicine for 1 hour resulted in 66.66 % of putative tetraploid shoots. Compared to diploid shoots, tetraploids had lower stomata density but bigger in guard cell size.

Abstract

One of difficulties in mangoes hybridization is polyembryonic seeds. This phenomenon reduces the chance of recovering true hybrid seedlings. The identification of the zygotic embryo is difficult and the possible approach is by using molecular marker. The objective of this study was to evaluate the utilization of two different marker systems (dominant markers and SSR) to characterize the occurrence of zygotic or nucellar embryo in polyembryony mango cultivars Garifta Merah, Lalijiwo, Manalagi, Madu, Saigon Kuning, and Saigon Merah. The type of embryo was evaluated by comparing the exhibiting amplification patterns, if different from the mother plant considered as zygotic and identified as nucellar if they exhibited the same banding pattern as the mother plant. From both of this marker systems out of the 16 SSR and 16 evaluated dominant primers, nine primers of each systems amplified the largest number of allele and sharply defined band. Dendogram analysis showed that the evaluated dominant markers could distinguish the zygotic and nucellar embryo clearly compare to evaluated SSR primers. The number of zygotic embryos derived from SSR was 64% and dominant markers were 47%. Based on zygotic and nucellar size, number and position, indicating no relationship between the type of embryo and embryo size, number and the position.

Abstract

Tanaman telang dapat dimanfaatkan sebagai alternatif pakan ternak karena kandungan nutrisinya. Produktivitas tanaman telang dapat ditingkatkan dengan menginduksi tanaman poliploidi dengan kolkisin. Tanaman poliploidi mempunyai ukuran fenotipik yang besar daripada diploidnya yang berpengaruh terhadap produktivitas tanaman tersebut. Tujuan dari penelitian ini adalah mengetahui pengaruh jenis telang dan lama perendaman kolkisin terhadap kecambah, karakter fenotipik dan tingkat ploidi tanaman. Penelitian ini dilaksanakan di Balai Besar Pengujian Standar Instrumen Bioteknologi dan Sumber Daya Genetik Pertanian (BSIP Biogen). Penelitian ini menggunakan Rancangan Acak Lengkap dengan  duafaktor yaitu jenis telang (putih dan biru) dan lama perendaman kolkisin (0, 24, dan 48 jam. Data dianalisis secara statistik dengan ANAVA pada taraf nyata 5%. Telang biru dengan perendaman kolkisin selama 24 jam menghasilkan panjang daun, panjang stomata dan jumlah kloroplas tertinggi. Telang putih yang diberikan perlakuan perendaman kolkisin selama 24 jam menghasilkan tinggi kecambah dan lebar kotiledon yang paling tinggi dan perendaman kolkisin selama 48 jam menghasilkan karakter lebar daun, panjang dan lebar mahkota bunga terbesar. Telang biru dan putih dengan perlakuan perendaman kolkisin selama 24 dan 48 jam menghasilkan tanaman miksoploid (diploid dan tetraploid).

Abstract

Digital transformation presents new challenges for copyright protection in Indonesia. Law No. 28 of 2014 on Copyright is considered to be insufficiently responsive to the dynamics of digital infringement, such as illegal distribution, works based on modern technology, and weaknesses in the protection system and royalty distribution on digital platforms. This article discusses the urgency of updating the Copyright Law to make it more adaptive, including expanding the scope of protection in the digital realm, integrating security technologies such as Digital Rights Management (DRM) and watermarking, strengthening the responsibility of User-Generated Content (UGC) platforms, and aligning regulations with international standards. Key challenges include the difficulty of cross-border law enforcement, low public literacy, dualism in protection systems, and the lack of transparency in collective management organizations. Proposed solutions include inter-agency collaboration, improving public literacy, utilizing work protection technologies, and reforming the royalty distribution system. With comprehensive regulatory updates and cross-sector collaboration, copyright protection in Indonesia is expected to become more effective and competitive in the digital age.
