Abstract

Water pollution that occurred 2014 in Wangi river flow caused by many things, such as household waste, industrial waste disposal and the market as well as the erosion process. The sourcesed of river stream that comes from the river Prigen and empties into the river flow Bangil Pasuruan. Pollution in rivers Wangi had a negative effect on the environment. Streams that previously could be used by residents for everyday activities began to change in recent times. The activities necessary to manage liquid waste so that the effluent that discharged into the environment does not exceed effluent quality standards have been set. The activity will be successful if any commitment and control as well as the cooperation of all parties.

Abstract

Conservation and preservation of the waters environment of needed, in order to protect the flow of river water to downstream areas. Waters often contaminated and carried out the destruction by human activity itself. The threatens of existence of the flow of Kali Mewek. Kali Mewek of the flow needed some special handling related to the preservation river a worth, because of the cluster housing developmented. Activities that need to be done to efforts the preservation and protection is operation and maintenance river a worth; and the conservation, development water allocation, water quality control and ecotourism.

Abstract

Carp (Cyprinus carpio) is one of important freshwater cultivated fish species in Indonesia. Aeromonas hydrophila  is accounted as potential pathogenic bacteria in Carp, Hence the control for the infection of A. hydrophila is very important. In this study we investigated the effectiveness of crude extract of Dayak onions (Eleutherine palmifolia (L.) Merr) for the treatment of A. hydrophila infected fish. Furthermore, alteration of histophatology of several organs, gills, kidney, liver and muscle were also investigated. Crude Extract of Dayak Onion (Eleutherine palmifolia (L) Merr. (CEDO) with the doses of 50, 60, 70 and 80 ppm were used for treatment of A. hydrophila infected carp for 11 days. Prior to investigation of histopatology of gills, kidneys, hearts, muscle, the Carp was moved to fresh-non infected fish tank. The Result indicated that gill experienced Hyperplasia, Fusion, Necrosis. Kidney experienced Degeneration, Congestion, Necrosis. Liver experienced the vacuole degeneration, congestion, Melano macrophages, necrosis. Muscle experienced Edema and necrosis. But fish treated with 80 ppm treatment showed less damage  and tissue repair.Keywords : E. palmfolia (L.) Merr, A. hydrophila, C. carpio, Histopathology, Gills, Kidney, Liver,  Muscle

Abstract

Climate change impact such as an increased on the intensity and the frequency of rainfall in the oceans cause sea level rise which then impacts the water surface in the river. This condition has an impact on fishery cultivation in coastal areas. People in Sidoarjo regency are increasingly affected by the alteration of water surface in coastal areas. Sidoarjo region is divided into 5 sub-development areas of one of the 5 sub-regional development units (SSWP V) which are a coastal area with aquaculture fisheries aquaculture economic center. Physical parameter indicators including secondary data and spatial map (GIS) were conducted. The result showed that 4 villages with the highest physical vulnerability of Sidoarjo Regency to the impact of climate change including Kalanganyar Village, Kupang, Tambak Kalisogo and Permisan. Keywords: Coastal, Exposure, GIS, Sea level rise

Abstract

Protein ekstraseluler (ECP) bakteri memiliki kemampuan imunogenik dan dapat meningkatkan sistem imun tubuh inang. Komponen ECP diantaranya flagellin, protease dan metalloprotease mampu berdifusi pada sel inang untuk mengaktifkan respon imun. Tujuan penelitian adalah untuk mengetahui produksi protein ekstraseluler E. tarda dengan presipitasi ethanol pada konsentrasi berbeda sebagai potensi protein imunogenik secara in vitro. Metode penelitian yaitu kultur bakteri E. tarda, preparasi protein ekstraseluler, presipitasi protein, konsentrasi protein ditentukan denganspektrofotometer dan SDS-PAGE, uji in vitro protein ECP E. tarda, dan uji nilai RPS. Hasil protein ekstraseluler yang diperoleh dengan presipitasi ethanol tertinggi pada konsentrasi 90% sebanyak 5,69 mg/ml dan terendah pada konsentrasi 80% sebanyak 4,81 mg/ml. Hasil karakterisasi protein dengan SDS-SDSPAGE antara 30-60 kDa. Hasil uji in vitro vaksin E. tarda (presipitasi ethanol 100%) baik dari uji viabilitas maupun sterilitas menunjukkan tidak terdapat koloni yang tumbuh. Hasil nilai RPS pada ikan nila (Oreochromis niloticus) mencapai 100% dengan masa pemelihaaraan 9 hari pasca imunisasi. ECP bacteria have immunogenic abilities and can increase the host's immune system. ECP components which are flagellin, protease, metalloprotease can diffuse to host cell for the activation immune response. The study aimed to determine the production of extracellular protein E. tarda with ethanol precipitation at different concentrations as a potential immunogenic protein by in vitro test. The research methods were E. tarda bacterial culture, extracellular protein preparation, protein precipitation, protein concentration determined by spectrophotometer and SDS-PAGE, in vitro ECP E. tarda protein test, and RPS value test. The results of extracellular protein obtained by the highest ethanol precipitation at a concentration of 90% as much as 5.69 mg/ml and the lowest at 80% concentration as much as 4.81 mg/ml. The results of the characterization of proteins with SDS-SDS-PAGE between 30-60 kDa. The results of the in vitro test of E. tarda vaccine (100% ethanol precipitation) from both the viability and sterility tests showed that there were no growing colonies. The results of the RPS in tilapia (Oreochromis niloticus) reach 100% with a maintenance period of 9 days after immunization.

Abstract

The aim of study to determine the environmental quality, content and distribution of heavy metals in the waters of the Wangi River, Beji Sub-District, Pasuruan Regency in relation to the interests of environmental conservation around the river. Method of sampling doned by chosen, taking into account the condition of research area, current and depth of research area.The intended to see to what extent pollutant concentrations are spreading. Observation of water quality during research in different seasons of the rainy season (October 2016 to February 2017) and dry season (March to September 2017). Based on the results of environmental quality testing and heavy metals can be concluded that the environmental quality at the Wangi river flow is already at the threshold, so that the required waste prevention and environmental conservation activities. This should get great attention from the public in general and the local government in particular. The hope for the future is pollution of the Wangi River can be avoided and overcome.

Abstract

Immunostimulatory dose given to the fish to be precise, should not be less and should not be excessive. This is because the immunostimulatory properties which can only work at the optimal dose.Therefore,themethod fordetermining the dosage and administration techniques immunostimulatory should be studied in depth.Research on the determination of the dose and the best maceration time on the crude extract phenol Gracilaria sp. using LC50 is intended to determine how best dose and how long it's best used in the immersion process to extract.rangedose used is 1 g/l; 1,5 g/l; 2 g/l; 3 g/l; 4 g/l and 5 g/l.The results of these observations indicate that doses ≥ 3 g/l has a 10-hour LC50.While the dose ≤ 3 g/l has a 14-hour LC50.However, further research is continued in the time range for prolonged submersion namely 10, 12 and 14 hours.The results showed the soaking time 12 hours showed LC50 15 hours.It concluded that the dose that can be used to test immunostimulant is 1, 1.5 and 3 g/l with a soaking time of less than 12 hours.

Abstract

The study was conducted to determine total leucocyte and plasma protein of infected koi (Cyprinus carpio) after immersion in the extract of Gracillaria verrucosa. The experimental design of this research was Completely Randomized Design. Different doses of extract were used five treatments and three replicates. Data was analyzed by using ANOVA (one-way) there were significant different of total leucocyte. Total protein plasma at treatments of fish infected, showed protein of ben thinner. Doses immersion treatments at 19 ppm get significant effects of humoral and celluler system the infected of koi fish.

Abstract

Gracilaria sp. is one type of red algae that has many benefits. It is widely used as a cosmetic, gelatin and medicinal materials. . In the fisheries, especially in the field of aquaculture, utilization of algae has been quite widely used. One of them is the use of red algae as a madicine ingredient and immunostimulant material to minimze the use of antibiotics in fish. Research on determination of solvent extraction and extract duration done to know the best of solvent extraction and extract duration used as a reference to determine the time of the maceration when the Gracilaria sp. extraction process. The appropriate solvent expected to produce high yield (results of extraction). The type of solvent used is ethanol 80%, ethanol 96%, acetone and aquades. The range of time that is used for the maceration is 24, 48 and 72 hours. The results showed that the best type of solvent is ethanol 96% with long soaking for maceration during 48 hours. The resulting yield is 0.9 with a moisture content of 10%.
