Abstract

The Jaguar cichlid, Parachromis managuensis (Günther, 1867), is native to Central America, with introduc­tions reported from West Java and Central Java Provinces of Indonesia. On 7-8 January 2019, sixteen specimens of P. managuensis were collected from Karangkates, the largest hydropower reservoir in East Java Province of Indonesia. A description of the morphological characters of specimens is provided.

Abstract

Analisa hematology yang mengkaji aspek anatomi, fisiologi, dan patologi darah, merupakan tahapan yang biasa dilakukan dalam bidang kedokteran untuk mengetahui kondisi kesehatan pasien. Dalam bidang perikanan analisa hematology bisa diterapkan sebagai early detection system untuk mencegah terjadinya kematian masal dalam pembudidayaan ikan. Parameter yang penting untuk diketahui dalam analisa hematology adalah jumlah leukosit. Penghitungan manual yang biasa dilakukan selama ini sangat tergantung pada keahlian dan jam terbang laboran. Kelelahan mata, bentuk yang kecil dan jumlah yang banyak seringkali menjadi faktor yang menyebabkan tingkat akurasi penghitungan berkurang. Dalam penelitian ini penghitungan otomatis dilakukan dengan pendekatan image processing. Tingkat roundness, area dan perimeter digunakan untuk membedakan antara partikel sel darah dan bukan sel darah. Tahap preprocessing yang dilakukan untuk meningkatkan akurasi perhitungan antara lain dinamic contrast, filling dan opening. Algoritma watershed digunakan untuk memisahkan sel yang bertumpuk. Hasil pengujian terhadap 80 citra leukosit ikan menghasilkan tingkat akurasi sistem yang dibangun mencapai 96,84%Kata Kunci : pengolahan citra, leukosit, ikan roundness.

Abstract

The coastal area of Mayangan District is the center of the economic development of the city of Probolinggo. Mangrove forest management that lacks environmental standards such as fish and shrimp ponds and mangroves that are within the scope of the area, causes damage and decreases the quality and sustainability of coastal and marine resources. This Community Service intends to introduce intensive shrimp farming techniques in ponds that are environmentally friendly and sustainable as well as introduce the importance of mangrove ecosystems in coastal areas and in supporting the development of fish/shrimp cultivation business activities by socializing the vannamei shrimp cultivation model with "Ecogreen Aquaculture" technology. The forms of service activities that will be carried out include socializing the vannamei shrimp cultivation model , socializing the importance of mangrove ecosystems in improving the environment in supporting the development of aquaculture activities and trial activities of "Ecogreen Aquaculture" technology.

Abstract

Several studies of marine bioactive compounds have been carried out using seagrass. Enhalus acoroides is a type of seagrass that has bioactive compounds including alkaloids, flavonoids, tannins, steroids, and saponins that have potential as an antibacteria. The aim of study is to investigate the phytochemical compound contained in the crude extract of Enhalus acoroides and its antibacterial activities of Enhalus acoroides against Staphylococcus aureus. The seagrass was collected, washed, dried, grind and exposed to extraction by vacum rotary evaporator at temperature of 40°C and was analyzed for their phytoconstituents. Further, the crude extract was tested against pathogenic bacterial at different concentrations using disc diffusion method to determine the effect of bioactive compounds in E. acoroides to bacteria. In the present study, qualitative test of phytochemical from ethanol extract of Enhalus acoroides leaves contained phytochemical compound of alkaloids, flavonoids, tannins, steroids and saponins, while the extract of roots contained all of compounds except saponins. Based on research can be concluded, that Enhalus acoroides also has antibacterial activity against Staphylococcus aureus that showed by the presence of inhibition zone. Abstract

Pseudomonas fluorescens is a gram-negative pathogenic bacteria which is one of the problems in fish farming that can causes death in fish. The alternative to overcome this bacterial attack is by using of natural material such as dayak onion bulbs (Eleutherine palmifolia (L) Merr), which are known to have secondary metabolites. The purpose of this research was to determine the antibacterial activity against P. flourescens that contained in the dayak onion bulbs (E. palmifolia (L) Merr) ethanol fraction and the content of antibacterial compounds from the best dayak onion bulbs (E. palmifolia (L) Merr) ethanol fraction. The method used in this research was antibacterial test with MIC (Minimum Inhibitory Concentration) followed by disc test and FTIR (Fourier Transform Infrared Spectroscopy) to analysis secondary metabolite. Fractionation was conducted using Chloroform: Methanol (9:1 v/v). MIC test result showed that 100 ppm was the minimum concentration that can inhibit the growth of P. fluorescens. Disc test reported that fraction 4 was the best fraction, with the dominant compounds that contained in the bulbs was phenolic compound based on FTIR test. So, the fraction of dayak onion bulbs (E. palmifolia (L) Merr) can be used as potential leads to discover new drugs.

Abstract

Indonesian waters have six types of turtles that can live, spawn and breed. Sea turtle conservation becomes an important and urgent program to be done in order to protect and save sea turtle population in Indonesia. One of the factors that most affect the turtle population is the cause of degradation of pathogenic factors. Alas Purwo National Park, East Java, there is some communities that have activities turtle conservation. Conservation is done by securing and protecting turtle eggs. Turtle eggs that have hatched are released into the sea once it is ready. This study aims was to determine the type of bacteria and fungi that infect hatchlings and environmental factors that influence. This research is descriptive method to Hawksbill turtle (Eretmochelys imbricate) is by way of random sampling. Sampling of microbes in sea turtle was conducted using cotton swab method and then microbes was cultured and indentified in laboratory. The results showed The kind of parasites and microbes which were indentified in hatching and adult Hawksbill sea turtles were fungus with genus Aspergillus sp., Geotrichum sp., Fusarium sp., and Gliocladium sp. ; bacteria are Pseudomonas aeruginosa and Enterobacter cloaceae; and parasite is Chelonibia testudinaria barnacles.  The parameter average value of water in pond indicated 28.1 – 29.2°C for temperature, 32 - 34 ‰ for salinity, 7.78 – 8.2 for pH, and 3.86 – 4.21 mg/L for DO.

Abstract

Nile tilapia and Striped Catfish can be classified as freshwater fish species that have great cultivation potential. Inadequate cultivation management leads to the occurrence of disease attacks on fish. Hematological examination was applied to determine the level of fish health. This research was aimed to know the difference between erythrocyte cell and leucocyte cell count by using Hematology Analyzer Tool Software (computer based) and hand tally counter (human based), and to know the description of hematocrit component, hemoglobin and leucocytes differential on tilapia fish and Striped Catfish. The methods employed in this research were experimental method and descriptive method. The results showed that the calculation of erythrocyte cells and leucocyte cells by using Hematology Analyzer Tool Software was similar to the calculation by using hand tally counter. It was because the result of t value was smaller than t of the 5% table. Based on the description of the hematocrit component, hemoglobin and leucocyte differentials, it could be said that nile tilapia and Striped Catfish were within the range of abnormalities.

Abstract

The purpose of this study described the influence and the dose of guava leaf raw extract (P. guajava) as an immunostimulant towards liver histotapologi Patin (Pangasius sp.) Challenge tested A. hydrophila bacteria. This study uses a completely randomized design (CRD) with 4 treatments and 3 replications. The treatments used was of guava leaf extract doses A dose of 2% (2.4 g / 120 g), and 4% (4.8 g / 120 g), C 6% (7.2 g / 120 g) and K without a guava leaf extract and tested bacteria A. hydrophila challenge. Based on the results obtained in the study of liver histopathology Patin (Pangasius sp.), there was damage Inflammation and necrosis of the liver tissue in order from the most severe damage to the least obtained at treatment K (0%) 3,07, A ( 2%) 2.53, B (4%) of 2.33, and the best treatment is C (6%) with an average damage of 1.67. Necrosis of liver tissue damage in a sequence of severe damage to the least, namely the treatment of K (0%) 2,67, A (2%) 2.33, B (4%) 2.20, and C (6%) 1,53. Lowest average gained in treatment C (6%). The results showed that the dose can reduce the level of tissue damage that was at a dose of 6% (7.2 g / 120 g). The best dose is 6% (7.2 g / 120 g), which can reduce the level of damage indicate the level of tissue damage smaller percentage of damage 6-25%.

Abstract

The impact of use antibiotic can cause problems on aquaculture such as bacterial resistance and residues for consumer. Therefore, one of alternatives that can be used to inhibit or kill bacteria, one of which is the use of plankton Chaetoceros calcitrans. This research aims to know the influence of the granting C. calcitrans extract that can inhibit or kill bacteria and know the best dose for inhibit A. salmonicida bacteria. The experiment method  used randomize completely design consist of four treatment and two control. A treatment (0.3 ppm), B treatment (15.3 ppm), C treatment (30.3 ppm),  D treatment (45.3 ppm), positive control (add chlorampenicol 5ppm) and negative control (without extract). The result of research showed the highest average of clear zone diameter in D treatment 5.37 mm and the lowest average of clear zone diameter in A treatment 2.89 mm, with linear equation y = 3.2067 + 0.0509x and coefficients R2 = 0.9262. The relationship between C. calcitrans extract in inhibitng gwoth of A. salmonicida bacteria showed that the average diameter of the clear zone has increase in line with the addition of extracts. Form the research showed that the extract of C. calcitrans was bacteriostatic (inhibits the growth of bacteria) and best obtained at the treatment dosage of D with a dose of 45.3 ppm and included into the category of middle.
