Abstract

Edwardsiella tarda is a type of gram-negative intracellular pathogen that can cause mass demise in fish farming. The development of edwarsiellosis due to E.tarda infection causes significant economic losses for cultivators. Aloe vera is a perennial succulent plant like a cactus, drought withstand, and belongs to the Liliaceae family, of which there are more than 360 known species (Wijaya and Masfufatun, 2022). Aloe vera is one of the plants known to contain active ingredients that can inhibit the growth of bacteria. The results of phytochemical analysis of a crude extract of Aloe vera showed the presence of active compounds, specifically flavonoids, tannins, triterpenoids, and saponins. MIC analysis showed that the lowest absorbance value of Aloe vera was obtained at a concentration of 50 mg/L with a value of 0.316, indicating a decrease at each concentration. The concentration of 50 mg/L is close to the positive control which indicates that at a concentration of 50 mg/L, it can be the reference for the lowest concentration in determining the dose of Aloe vera extract which is known that the presence of antibacterial compounds in Aloe vera extract can inhibit the growth of E.tarda bacteria. The inhibition zone test was carried out with graded concentrations of 50 mg/L, 100 mg/L, 150 mg/L, 200 mg/L and 250 mg/L with an observation time of 24 hours post-incubation and 48 hours post-incubation. Inhibition zone analysis with the highest diameter was at 200 mg/L. SEM analysis by distributing a crude extract of Aloe vera affected E.tarda bacteria specifically, lysis occurring in bacterial cells

Abstract

Uji hematologi sangat bermanfaat sebagai alat diagnostik didalam menetapkan status kesehatan ikan. Salah satu aspek dari infeksi adalah terjadinya perubahan gambaran darah. Pemeriksaan darah dapat digunakan sebagai indikator keparahan suatu penyakit tertentu. Beberapa parameter yang dapat memperlihatkan perubahan patologi pada darah meliputi jumlah leukosit, deferensial leukosit, eritrosit. Dari hasil penelitian diketahui jumlah rata-rata sel darah merah dalam darah ikan botia sehat pada masing-masing perlakuan K-, K+, A, B, C, D berturut-turut adalah 2.470.000, 2.500.000, 2.560.000, 2.350.000, 2.440.000, dan 2.430.000. Sementara itu pada ikan botia yang terinfeksi bakteri A.hydrophila diketahui jumlah rata-rata eritrosit pada masing-masing perlakuan K-, K+, A, B, C, dan D berturut-turut adalah 2.420.0000, 1.770.000, 1.780.000, 1.890.000, 1.780.000, dan 1.870.000. jumlah rata-rata eritrosit ikan botia yang terinfeksi bakteri A.hydrophila, dan kemudian diobati dengan ciprofloxacin pada masing-masing perlakuan K-, K+,A, B, C, dan D berturut-turut adalah 2.420.000, 1.630.000, 2.090.000, 2.280.000, 2.500.000, dah 2.540.000. Sementara jumlah rata-rata sel darah putih dalam darah ikan botia yang sehat pada masing-masing perlakuan K-, K+, A, B, C, D berturut-turut adalah 97.000, 90.000, 88.000, 87.000, 93.000 dan 88.000. Sementara itu jumlah rata-rata sel darah putih pada ikan botia yang terinfeksi bakteri A.hydrophila, pada masing-masing perlakuan K-, K+, A, B, C, D berturut-turut adalah 97.000, 174.000, 174.000, 179.000, 185.000 dan 185.000. Sedangkan dari hasil penelitian terhadap ikan botia yang terinfeksi A.hydrophila dan kemudian diobati dengan ciprofloxacin diperoleh hasil perhitungan rata-rata leukosit pada masing-masing perlakuan berturut-turut adalah sebagai berikut  980.000, 189.000, 125.000, 137.000, 133.000, dan 136.000. Abstract

The viral nervous necrosis (VNN) is betanodavirus of the family Nodaviridae with acute infection and associated with high levels mortality up to 100% of numerous larval-stage marine and freshwater fish species. The danger signals of viral infection that are capable of activating APCs (Antigen Presenting Cells), furthermore produced molecules protein receptors such as interferon and heat shock proteins. Stimulating interferon type I (IFN I) induce several antiviral molecules, further binding the actin cytoskeleton to reach the site of infection. This study demonstrated a correlation between increased levels of Hsp70 (heat shock proteins) and actin filamentous (β-actin) within invasion wild-type isolate of ssRNA VNN from Asian Seabass (L. calcalifer) juvenile. Furthermore, expression of Hsp70 and actin as an indicator or biomarker of stressed states in fish. The reverse transcriptase polymerase chain reaction (RT-PCR) method used to finding expression Hsp70 and β-actin. Whereas nested RT-PCR used for VNN genes. The up-regulation of Hsp70 was observed on the brain tissue higher than eye tissue of sample positive infected VNN. Whereas different expression of β-actin, β-actin receptor expression tends to be stable in the organs of the brain and eye, both invaded VNN or normal, while on the eye, the ratio slightly increased based on the intensity of the band. The research shows that altered expression of heat shock protein 70 and beta-actin gene receptors in the target organs that response to invasion VNN at Asian seabass (L. calcalifer) juvenile. Alteration expression of heat shock protein 70 and beta-actin can be used as an indicator or biomarker of stress cells, especially from VNN invasion at fish. Abstract

Dayak onions (E. palmifolia (L) Merr) is a herb used as medicine and known to have antibacterial activity. This research aimed to evaluate antibacterial activity of Dayak onion (Eleutherine palmifolia (L) Merr) extract against fish pathogen Pseudomonas fluorescens. The method used in this study was extraction by maceration, UV-Vis spectrophotometry analysis on the extract and antibacterial activity test with Minimum Inhibitory Concentration (MIC) as well as disc diffusion test. The results of this study showed that the yield value was 8.87%. Based on UV-Vis analysis, it confirmed that extract of Dayak onion (Eleutherine palmifolia (L) Merr) contains predominantly flavonoids and phenols with its derivatives, that have antibacterial activity. The results of MIC and disc diffusion test showed potential antibacterial activity against P. fluorescens, as observed from the value of absorbance and inhibition zones that formed. Abstract

Koi herpesvirus (KHV) is a virus that usually infects carp and koi causing mass mortality, as well as economic and social loss. The results of varied test are often found in its development. Allegedly, there are mutation or genetic variation of KHV in Indonesia. Based on these problems, this research aims to understand the genetic variation and kindship of KHV isolate in the carp (Cyprinus carpio) in Lombok Island of West Nusa Tenggara Province based on thymidine kinase sequence. Water quality of fish pond was checked. KHV clinical symptom was checked visually in each fish sample, then sacrifices the organ (gill) for PCR analysis and DNA sequencing. The research results show that clinical symptoms that appear are pale fish gill and bleeding on the surface of the body. The temperature in the research site shows an optimum range for KHV development. The results electrophoresis of carp suspected with KHV shows five positives out of a total six samples. Genetically, KHV isolates from Lombok Island of West Nusa Tenggara Province is identical with the GenBank isolate code KHV-J/U/I and Indo_0C05WNT2.Keywords: Carp, Koi herpesvirus, Phylogenetic, Thymidine kinase.

Abstract

Phytoplankton can be used to estimate the potential for vannamei shrimp production. It can be used as a provider of nutrient sources and has an important role in improving water quality. This study aims to analyze the phytoplankton community structure on the productivity of intensive vannamei shrimp ponds. The research was conducted in Jatisari Village, Banyuwangi, East Java, Indonesia. Carried out parameters were the calculation of density, phytoplankton diversity index, culture performance (SR, FCR, ADG), and water quality (temperature, water transparency, salinity, pH, DO, NO2, NO3, PO4, NH4). The results showed that there were six classes and 33 genera from both ponds, Chlorophyceae (10 genera), Bacillariophyceae (8 genera), Cyanophyceae (9 genera), Dinophyceae (3 genera), Euglonophyceae (1 genus), Criptophyceae (1 genus). The index value of the two ponds shows moderate diversity, H 'pond 1 is 1.76, and pond 2 is 2.02. The two plots' cultivation performance was SR 92% and 80%, FCR 1.08 and 1.13, ADG 0.31 g.day-1 and 0.35 g.day-1, respectively. The physical and chemical parameters of the research showed a good enough value for the life of vannamei shrimp and phytoplankton.Keywords: Cultivation performance, Phytoplankton, Vannamei shrimp.

Abstract

Highlight Research:It has been found that many fish died with wounds from farmers in Blitar, Indonesia.The mucosal smear test from mucosal gills of C. carpio infected with Myxobolus.In the blood smear test of C. carpio, there was no myxobolus found in the blood.In gill organ testing using SEM that Myxobolus is found in gills (C. carpio).The shape of Myxobolus resembles an imperfect ball with a hollow in the middle. AbstractThe biggest problem that is often considered to be an obstacle to Common carp culture is the emergence of disease attacks. One type of disease that often attacks the seeds of Cyprinus carpio is Myxobolus (a systemic parasite that can cause harm to fish farming). The aim of this study was to determine the molecular expression through the smear test on C. carpio gills, to determine the image of the gill organs of C. carpio using the SEM test, and to determine the description of the spores of Myxobolus sp. Data were analyzed using descriptive methods. Descriptive method used was comparative descriptive comparing molecular expression in the test of gill mucosal smear of fish using a light microscope and gill organ testing using Scanning Electron Microscope (SEM) on C. carpio infected by Myxobolus sp. In this study, the results showed that in C. carpio infected with true Myxobolus found the presence of Myxobolus in the mucosal smear test and SEM test on gill tissue, but not found in the blood smear test.

Abstract

Ketahanan pangan merupakan program yang telah di canangkan oleh pemerintah. Dalam pelaksanaanya pembuatan wilayah dengan ketahanan dapat diintegrasikan dengan wilayah penopang ekonomi lainnya seperti daerah wisata sehingga akan didapat sentra Ekonomi yang mampu dijadikan sebagai sumber penghasilan warga setempat. Desa Kebontunggul merupakan desa yang ada di Kecamatan Gondang Kabupaten Mojokerto. Melalu program Matching Fund Kedaireka yang didanai oleh Kementerian Pendidikan, Kebudayaan, Riset, dan Teknologi Universitas Brawijaya mengadakan program hilirisasi Teknologi berupa pengembangan Kawasan Peternakan sapi perahyang diintegrasikan dengan Kawasan wisata yang ada di daerah setempat yaitu Lembah Mbendcirang yang sebelumnya sudah beroprasi. Tim Matching Fund Universitas Brawijaya mengadakan pemetaan wilayah di desa Kebontunggul terkait potensi lokasi sapi perah, sumberdaya paka, serta akses lokasi integrasi dengan lembah mbencirang. Selanjutnya dengan bantuan drone dan satelit Map Tim Martching Fund Universitas Brawijaya Melakukan pembuatan Materplan dimana dalam pembuatannya juga melibatkan masyarakat setempat sehingga sesuai dengan harapan masyarakat namun tetap memegang aspek Teknolologi yang dikembangkan oleh Universitas Brawiajaya. Selin itu juga dilakukan pelatihan budidaya sapi perah yang dilakukan oleh Tim Matching fund 2022 Universitas Brawijaya

Abstract

Cyprinus carpio is one type of freshwater fish. In Indonesia, carp outbreaks in carp since June 2002 have raised concerns, especially for fish farmers. One of the factors causing the outbreak of this disease is the bacteria Aeromonas salmonicida, which can cause fish to get sick and even die. Immunostimulants with natural ingredients that are environmentally friendly and can be used to increase the immune response of fish, one of which is maggot. The data analysis used in this study was a simple completely randomized design (CRD) with 6 treatments difference in the dose of maggot as an immunostimulant (0%, 25%, 50%, 75%, 100%) and three replications. The independent variable is the difference in the dose of maggot as an immunostimulant. The dependent variables include survival rate, total leucocytes, differential leukocytes, and water quality (pH, temperature, and DO). The FT-IR test results obtained that the best compound content in maggot flour is flavonoid compounds where these compounds can increase immune response. So, it is proven by phytochemical tests with positive maggot flour containing flavonoids, tannins, and alkaloids. In total erythrocytes, hematocrit, hemoglobin, total leukocytes, and differential leukocytes showed the best immune response in treatment C with a dose of 75% maggot.

Abstract

Peningkatan padat tebar adalah salah satu cara untuk meningkatkan kinerja produksi pada budidaya ikan cupang hias endemik serban malem (Betta rubra Perugia 1893). Kegiatan ini bertujuan mengevaluasi pengaruh padat tebar yang berbeda terhadap sintasan dan pertumbuhan ikan serban malem (Betta rubra Perugia 1893) hasil domestikasi. Kegiatan penelitian ini dilakukan di laboratorium sistem dan teknologi akuakultur, Program Studi Akuakultur, Universitas Teuku Umar. Penelitian ini dilakukan secara eksperimental dengan menggunakan metode rancangan acak lengkap. Penelitian padat tebar ini dilakukan dengan empat perlakuan dan tiga ulangan. Perlakuan padat tebar yang dilakukan adalah 1 ekor/liter (P1), 2 ekor/liter (P2), 3 ekor/liter (P3) dan 4 ekor/liter (P4). Pelaksanaan kegiatan penelitian selama 60 hari. Hasil proksimat pakan yang diberikan mengandung protein 31,27%, karbohidrat 44,63%, lemak 4,63%, serat 2,56%, kadar abu 13,03% dan kadar air 3,89%. Pemberian pakan dilakukan dengan frekuensi dua kali sehari yaitu pagi dan sore hari. Pengoleksian data penelitian dilakukan pada awal dan akhir penelitian. Analisis data menggunakan analisis varians dengan selang kepercayaan 95%. Hasil penelitian menunjukan bahwa padat tebar ikan yang berbeda berpengaruh nyata (P<0,05) pada sintasan dan pertumbuhan ikan serban malem (Betta rubra Perugia 1893) hasil doemstikasi. Padat tebar terbaik untuk sintasan dan pertumbuhan ikan serban malem adalah perlakuan 1 ekor/liter (P1).
