Abstract

Highlight Research:It has been found that many fish died with wounds from farmers in Blitar, Indonesia.The mucosal smear test from mucosal gills of C. carpio infected with Myxobolus.In the blood smear test of C. carpio, there was no myxobolus found in the blood.In gill organ testing using SEM that Myxobolus is found in gills (C. carpio).The shape of Myxobolus resembles an imperfect ball with a hollow in the middle. AbstractThe biggest problem that is often considered to be an obstacle to Common carp culture is the emergence of disease attacks. One type of disease that often attacks the seeds of Cyprinus carpio is Myxobolus (a systemic parasite that can cause harm to fish farming). The aim of this study was to determine the molecular expression through the smear test on C. carpio gills, to determine the image of the gill organs of C. carpio using the SEM test, and to determine the description of the spores of Myxobolus sp. Data were analyzed using descriptive methods. Descriptive method used was comparative descriptive comparing molecular expression in the test of gill mucosal smear of fish using a light microscope and gill organ testing using Scanning Electron Microscope (SEM) on C. carpio infected by Myxobolus sp. In this study, the results showed that in C. carpio infected with true Myxobolus found the presence of Myxobolus in the mucosal smear test and SEM test on gill tissue, but not found in the blood smear test.

Abstract

Ketahanan pangan merupakan program yang telah di canangkan oleh pemerintah. Dalam pelaksanaanya pembuatan wilayah dengan ketahanan dapat diintegrasikan dengan wilayah penopang ekonomi lainnya seperti daerah wisata sehingga akan didapat sentra Ekonomi yang mampu dijadikan sebagai sumber penghasilan warga setempat. Desa Kebontunggul merupakan desa yang ada di Kecamatan Gondang Kabupaten Mojokerto. Melalu program Matching Fund Kedaireka yang didanai oleh Kementerian Pendidikan, Kebudayaan, Riset, dan Teknologi Universitas Brawijaya mengadakan program hilirisasi Teknologi berupa pengembangan Kawasan Peternakan sapi perahyang diintegrasikan dengan Kawasan wisata yang ada di daerah setempat yaitu Lembah Mbendcirang yang sebelumnya sudah beroprasi. Tim Matching Fund Universitas Brawijaya mengadakan pemetaan wilayah di desa Kebontunggul terkait potensi lokasi sapi perah, sumberdaya paka, serta akses lokasi integrasi dengan lembah mbencirang. Selanjutnya dengan bantuan drone dan satelit Map Tim Martching Fund Universitas Brawijaya Melakukan pembuatan Materplan dimana dalam pembuatannya juga melibatkan masyarakat setempat sehingga sesuai dengan harapan masyarakat namun tetap memegang aspek Teknolologi yang dikembangkan oleh Universitas Brawiajaya. Selin itu juga dilakukan pelatihan budidaya sapi perah yang dilakukan oleh Tim Matching fund 2022 Universitas Brawijaya

Abstract

Cyprinus carpio is one type of freshwater fish. In Indonesia, carp outbreaks in carp since June 2002 have raised concerns, especially for fish farmers. One of the factors causing the outbreak of this disease is the bacteria Aeromonas salmonicida, which can cause fish to get sick and even die. Immunostimulants with natural ingredients that are environmentally friendly and can be used to increase the immune response of fish, one of which is maggot. The data analysis used in this study was a simple completely randomized design (CRD) with 6 treatments difference in the dose of maggot as an immunostimulant (0%, 25%, 50%, 75%, 100%) and three replications. The independent variable is the difference in the dose of maggot as an immunostimulant. The dependent variables include survival rate, total leucocytes, differential leukocytes, and water quality (pH, temperature, and DO). The FT-IR test results obtained that the best compound content in maggot flour is flavonoid compounds where these compounds can increase immune response. So, it is proven by phytochemical tests with positive maggot flour containing flavonoids, tannins, and alkaloids. In total erythrocytes, hematocrit, hemoglobin, total leukocytes, and differential leukocytes showed the best immune response in treatment C with a dose of 75% maggot.

Abstract

Peningkatan padat tebar adalah salah satu cara untuk meningkatkan kinerja produksi pada budidaya ikan cupang hias endemik serban malem (Betta rubra Perugia 1893). Kegiatan ini bertujuan mengevaluasi pengaruh padat tebar yang berbeda terhadap sintasan dan pertumbuhan ikan serban malem (Betta rubra Perugia 1893) hasil domestikasi. Kegiatan penelitian ini dilakukan di laboratorium sistem dan teknologi akuakultur, Program Studi Akuakultur, Universitas Teuku Umar. Penelitian ini dilakukan secara eksperimental dengan menggunakan metode rancangan acak lengkap. Penelitian padat tebar ini dilakukan dengan empat perlakuan dan tiga ulangan. Perlakuan padat tebar yang dilakukan adalah 1 ekor/liter (P1), 2 ekor/liter (P2), 3 ekor/liter (P3) dan 4 ekor/liter (P4). Pelaksanaan kegiatan penelitian selama 60 hari. Hasil proksimat pakan yang diberikan mengandung protein 31,27%, karbohidrat 44,63%, lemak 4,63%, serat 2,56%, kadar abu 13,03% dan kadar air 3,89%. Pemberian pakan dilakukan dengan frekuensi dua kali sehari yaitu pagi dan sore hari. Pengoleksian data penelitian dilakukan pada awal dan akhir penelitian. Analisis data menggunakan analisis varians dengan selang kepercayaan 95%. Hasil penelitian menunjukan bahwa padat tebar ikan yang berbeda berpengaruh nyata (P<0,05) pada sintasan dan pertumbuhan ikan serban malem (Betta rubra Perugia 1893) hasil doemstikasi. Padat tebar terbaik untuk sintasan dan pertumbuhan ikan serban malem adalah perlakuan 1 ekor/liter (P1).

Abstract

Latukan Village is one of the villages in Lamongan Regency with Independent Village status in the 2020 Building Village Index (IDM). Independent Village has the highest status in the IDM assessment scale published by the Ministry of Villages. Through the  Empowered Village Program, Latukan Village seeks to build a village icon by constructing the Latukan “Koi Village”. However, the complete design concept regarding the development of Latukan Village as a  Kampung Koi Tourism Village has not yet been formed optimally. Therefore, it is essential to prepare strategic development steps, which include elements of Koi Fish Cultivation, Post-harvest Management, Transportation, Marketing Management, the establishment of recreational and educational parks, as well as human resource development, in this case, the Koi Fish Cultivator Group and the Harvest Management Group. The solution is to create a development design for the Kampung Koi Tourism Village as part of the BUM Desa business unit. For this reason, the Brawijaya University Community Service team designed an activity to build a stronger and more sustainable Latukan Village Icon. Service activities consist of identifying the status of the problem and planning solutions, forming a koi fish cultivator group and making portable spawning ponds, strengthening the role of BUMDesa, village government, and cultivators through focus group discussions and comparative studies, and koi fish cultivation and breeding training. All activities have been conducted and compiled in scientific publications and mass media. In its implementation, this activity involved five students concentrating on fisheries cultivation and fishery product technology.

Abstract

Salah satu upaya pencegahan yang dapat dilakukan untuk mengatasi penyakit pada ikan yaitu menumbuhkan kekebalan seperti vaksinasi karena kekebalan tubuh yang meningkat pada ikan dapat mencegah infeksi penyakit. upaya telah dilakukan untuk mengembangkan program imunisasi yang efektif dengan menggunakan vaksin yang dibuat dari sel utuh bakteri yang diinaktivasi menggunakan formalin dan/atau pemanasan Tujuan penelitian ini untuk mengetahui hasil uji viabilitas dan hasil uji adhesi sel epitel usus menggunakan vaksin inaktif formalin dengan konsentrasi berbeda. Metode dalam penelitian ini adalah metode eksperimen dengan rancangan percobaan menggunakan rancangan acak lengkap (RAL) terdiri dari 3 perlakuan (A=2%, B=3%, dan C=4%) dan kontrol dengan 3 kali ulangan. Hasil penelitian diperoleh hasil kandidat vaksin inaktif bakteri A.hydrophila sebanyak 9 ml/tabung falcon. Infeksi bakteri A. hydrophila dengan kepadatan 109 cfu/ml sebanyak 0,1 ml dengan masa pemeliharaan 1 minggu ditandai terjadi pengerasan di bagian perut dan terdapat kerusakan pada sirip punggung pada hari ketujuh. Hasil uji viabilitas menunjukkan tidak terdapat pertumbuhan koloni bakteri A. hydrophila dibandingkan dengan kontrol. Hasil uji adhesi sel epitel usus pada kontrol dan perlakuan A, B, C diperoleh rata-rata 10.3, 0.73, 0.43, dan 0.23 bakteri/10 sel epitel. Analisis uji ANOVA diperoleh nilai uji F Hitung > F Tabel, artinya perlakuan memberikan pengaruh sangat berbeda signifikan antar semua perlakuan. Hasil analisa regresi diperoleh (R2) sebesar 0.66 bahwa hubungan keterkaitan antara vaksin inaktif konsentrasi formalin berbeda dengan jumlah bakteri A. hydrophila yang menempel pada sel epitel sebesar 66%.   One preventive effort to tackle diseases in fish is by boosting immunity, such as through vaccination, as increased immunity in fish can prevent disease infections. Efforts have been made to develop effective immunization programs using vaccines made from whole bacterial cells inactivated using formalin and/or heat. The objective of this study was to determine the results of viability tests and intestinal epithelial cell adhesion tests using formalin-inactivated vaccines with different concentrations. The method used in this study was an experimental approach with a completely randomized design (CRD) consisting of three treatments (A=2%, B=3%, and C=4%) and a control, each with three replications. The study results showed that the candidate inactivated vaccine of A. hydrophila bacteria was obtained at 9 ml/falcon tube. A. hydrophila bacterial infection with a density of 10⁹ CFU/ml at 0.1 ml and a maintenance period of one week was characterized by abdominal hardening and dorsal fin damage on the seventh day. The viability test results showed no bacterial colony growth of A. hydrophila compared to the control. The intestinal epithelial cell adhesion test results for the control and treatments A, B, and C showed average bacterial adhesion of 10.3, 0.73, 0.43, and 0.23 bacteria/10 epithelial cells, respectively. ANOVA test analysis showed that the calculated F-value was greater than the table F-value, indicating a highly significant effect among all treatments. Regression analysis resulted in an R² value of 0.66, suggesting that 66% of the variation in the adhesion of A. hydrophila bacteria to epithelial cells was associated with different formalin concentrations in the inactivated vaccine.

Abstract

Myxobolus koi is a type of parasite that infects many freshwater fish through myxospores, resulting in a disease called myxobolosis. This research aimed is to investigate the clinical symptoms that occur in the gills of infected Koi carp (C. carpio). The utilized methods in this study are molecular detection, histopathology, and Scanning Electron Microscopy (SEM). Koi carp samples that are suspected to be infected with Myxobolus koi show several clinical signs, such as swollen and pale-colored gills and gill covers (opercula) that do not close completely, as well as white spots that appear on the edge of the lamella. The results of this study showed that through histological observations, there were changes characterized by hyperplasia of the primary lamella cartilage that envelops parasitic cysts and the encapsulation process that surrounds the cysts by gill cartilage in infected fish;SEM showed oval and elongated spores of a size of ± 12 µm (800x). TheDNA template from gill tissues in clinically infected and uninfected fish was examined byPCR testing with primers ERB 1 (Forward) and ERB 10 (Reverse); the results of electrophoresis in infected fish were detected at 2000 bp. Abstract

Kratom (Mitragyna speciosa) is an indigenous tropical herbal plant to the Northern Malay Peninsula and Thailand. Empirically kratom leaves have several properties as herbal medicines. Currently, the treatment of diseases caused by bacteria that are resistant to antibiotics requires new compounds that have high potential. The material studied was kratom leaf extract. The solvent used for extraction is methanol. Phytochemical screening carried out includes the examination of alkaloids, flavonoids, steroids/terpenoids, phenols, tannins, and saponins. The test bacteria used was Aeromonas hydrophilla. The medium used is Nutrient Agar (NA). NaCl 0.9% as a bacterial suspension. Testing the antibacterial activity carried out with 10 (ten) variations of concentration namely 3%, 6%, 9%, 12%, 15%, 18%, 21%, 24%, 27% and 30 %. Fresh kratom leaf is collected and then wet sorted. The extraction process was using the method of maceration. This study aims to analyze the antibacterial activity of kratom leaf, measure the secondary compounds of kratom leaf extract, and measure the best concentration to kill Aeromonas hydrophilla. The extraction process obtained blackish-brown extract with 62.27 grams of extract (31.14%). Secondary metabolites of kratom leaf extract show positive results of alkaloid, saponins, tannins, phenolics, steroids, and triterpenoids. Antibacterial activity against Aeromonas hydrophila which is characterized by a kill zone around the paper disc with the best concentration that shows a wide killing zone was 24% of the extract.Keywords: Antibacterial, Extract, Kratom, Mitragynine, Phytochemical Screening.

Graphical Abstract     Highlight Research The GC-MS analysis of the asiatica leaves extracts identified 53 bioactive compounds. The crude extracts of asiatica showed antibacterial efficacy against fish pathogenic bacteria. 13-Hexyloxacyclotridec-10-en-2-one has the potential to be an inhibitor of DNA gyrase. Bioactive compounds derived from C. asiatica leaves extracts show potential as antibacterial agents.     Abstract Antimicrobial agents are crucial for managing bacterial infections in fish cultures. Centella asiatica is a medicinal plant recognised for its diverse bioactive compounds with important antibacterial properties. The present study aimed to investigate the antibacterial activity of C. asiatica leaves bioactive compounds on fish pathogenic bacteria using an in vitro and in silico approach. The maceration method was used to extract bioactive compounds from C. asiatica leaves and was identified using Gas Chromatography-Mass Spectrometry (GC-MS). In vitro analysis of antibacterial activity was evaluated using the minimum inhibitory concentration method. While in silico molecular docking is applied alongside assessing Lipinski's rules of five, as well as absorption, distribution, metabolism, excretion, and toxicity properties. The result of the GC-MS examination of the C. asiatica leaf extracts identified 53 bioactive compounds. In vitro studies showed antibacterial efficacy of leaf extracts against fish pathogenic bacteria (Streptococcus agalactiae, Bacillus subtilis, and Staphylococcus aureus) with minimum inhibitory concentration values of 12,5 mg/ml. In silico molecular docking analysis showed that several bioactive compounds have the potential to be DNA gyrase inhibitors. Compound 13-Hexyloxacyclotridec-10-en-2-one has the highest inhibition with binding energy of −7,4 Kcal/mol compared to ciprofloxacin as drug standard with a binding energy value −7,3 Kcal/mol. The following compound is gamma.-Muurolene (−6,7 Kcal/mol), Copaene (−6,6 Kcal/mol) and Humulene (−6,6 Kcal/mol). These results suggest that bioactive compounds of C. asiatica leaves extracts hold promise as potential antibacterial agents for treating fish pathogenic bacteria infections.

Abstract

Modern strategies incorporate preventive measures such as extracellular products, as soluble proteins with antigenic properties, as well as generating a broad immune response. The aim of research is to determine and analyze the effect of the extracellular product of E. tarda on enhancing the immunity of Nile tilapia (O. niloticus). The research design with treatment durations of d0, 3rd, 6th, 9th day, and control (A, B, C, D, K). ECP vaccination was injected intraperitoneally at a dose of 0.1 ml/fish. The parameters analyzed are the expression of IL-1, IL-6, and IFN-, phagocytic activity, leukocyte differential, total white blood cells, and relative percent survival. The results of phagocytosis activity were highest in treatment C at 84.18% 1.81. The highest leukocyte differential in treatment C was lymphocytes at 56% 2.00, monocytes at 28% 2.00, and 16% 0.00, with a total leukocyte count of 29 x 10^3 cells 2.64, and RPS values ranging from 83.33% to 100% over 9 days of post-vaccination maintenance. Then, post-vaccination, there was an increase in the expression levels of the cytokine genes IL-1 and IL-6, but there was no increase in the expression levels of the IFN- gene compared to the control in Nile tilapia.
