Abstract

Bukit Tiga Puluh National park (TNBT) of Riau Province is a nature conservation area with a high diversity of fauna, one of which is the butterfly. Butterfly species diversity is influenced by the vegetation in their habitat. The TNBT utilization zone has a habitat type with varied vegetation. There is no information about the species of butterflies in the area. This study aimed to identify morphological character of butterflies and vegetation based on morphological characters. The method used was exploration. The catching area of the butterfly was the Granite waterfall and the Bukit Lancang path of TNBT utilization zone. Samples made as insectarium and herbarium. The  identification of butterfly characters consisted of the head, thorax, abdomen, legs and wings. There were a number of different types of butterflies and plants between the Granite waterfall area (22 and 28 species) and the Bukit Lancang paths (5 and 9 species). The type of butterfly was dominated by the family Nymphalidae. This is due to the amount of vegetation and environmental conditions that are more favorable in the granite waterfall area. The key determinants of butterfly species are the characters of the wing. The results of this study can contribute to maintaining the TNBT butterflies in Riau Province and also as database information of butterflies species in Indonesia.

Abstract

Musa acuminata Colla is a unique genomic species which differs from other species in this genus. It has typical color of reddish-purple on its fruit. Musa acuminata is recorded as rare banana in Riau, particularly in Kampar District. This research aims to study the influence of several cutting types of corm of shrimp banana originated from Kampar District through in vitro culture. It also purposes to determine the best formation of M. acuminata shoot through various concentration treatments of BAP only and BAP combined with Kinetin. The study used randomized block design (RBD) by adding come BAP concentration (0, 4, 8 mg/l) and BAP combined with Kinetin (0 mg/l BAP+0.4 mg/l Kinetin, 4 mg/l BAP+0.4 mg/l Kinetin, and 8 mg/l BAP+ 0.4 mg/l Kinetin). The cutting type of banana corm involve no cut (whole) and half-to-half cutting in MS media, each with 5 replicates. The result showed that either addition of BAP and BAP combined with Kinetin gave the best percentage of live explants and shot formation up to 100%. Treatment of 8 mg/l BAP resulted in the highest shoot percentage up to 100%, the fastest shoots appearance at 34.00 days after planting and the longest shoot reached 2.83 cm with 2.33 shoots counted on half-to-half cutting type. This study also confirmed that the addition of plant regulation substances from cytokinin group gave good impact in the formation of shrimp banana shoots.

Abstract

Moss club (Lycopodiella cernua) is one of fern members distributed in Riau Province. The aimed of this study were to characterized the morphological characters and to analized its DNA sequence based on rbcl primer. Samples were collected from the field, documented and characterized the morphological charaters. DNA were isolated and amplified using PCR method and rbcl primer.  DNA sequnces were then analized using BLAST   Lycopodiella cernua collected from Riau Province was characterized by having creeping and erect rhizome that covered by microphyl, and terminal strobili. Rbcl fragmen of Lycopodiella cernua was sucssesfully amplified with 643 bp length. BLAST result confirmed the  rbcl fragmen of this club moss.

Abstract

Siam orange (Citrus nobilis Lour.) is a prominent agriculture commodity in Riau province. It tastes sweet, with fragrant aroma and thin skin. Its cultivation, however, needs improvement to gain better quality of product. One effort that is using the in vitro culture. The study aimed to determine the best IBA and NAA concentrations to trigger root formation on siam orange transplant and used Completely Randomized Design (CRD). The result showed that the rooting reached 100% on all treatments. The fastest rooting was observed in 2 mg/L IBA treatment (7.67 days after planting). The most root growth was obtained with combination of 0.5 mg/L NAA with 1.5 mg/L IBA (2.67 units). The longest root was obtained through the mixture of 1.5 mg/L NAA with 0.5 mg/L IBA (2.27 cm).

Abstract

Drendan (Lansium domesticum var. aqueum (Jack) Miq. is one of Indonesia's native tropical fruits. Drendan has high antioxidant activity and contains high nutrients, but has started to rarely be found in the field because its management and cultivation is rarely done due to its sour taste in vitro culture is a method or technique of plant propagation which can be an alternative to obtaining drendan seedlings in large quantities and in a short time vegetatively. The purpose of this study was to determine the effect of BAP on shoot induction from drendan seed explants and determine the best concentration of BAP on shoot growth of drendan seed explants in vitro. This study used a completely randomized design with BAP concentrations of 0, 1, 3, 5 and 7 mg / L BAP. The result of this research is the addition of BAP has been able to increase the percentage of live explants and the percentage of shoot formation. Giving BAP did not significantly affect the number of shoots and had a significant effect on shoot emergence time and shoot length. The addition of BAP resulted in slower shoot emergence time. Giving BAP was not able to stimulate shoot induction, but the addition of BAP could increase shoot length growth at a concentration of 5 mg / L BAP of 0.52 cm.

Abstract

Family Orchidaceae has about 800 genera which are already difficult to find and almost extinct, including the orchid genus Grammatophyllum. A species of rare orchids, Grammatophyllum stapeliiflorum, is hard to find in original habitat. The addition of Growmore, BAP and 15% coconut water is expected to increase the growth and development of the G. stapeliiflorum orchid protocorms. This study use a Completely Randomized Design, consisting of 6 treatments (control, 1 and 3 mg/l BAP, 1 and 3 g/l Growmore, and 150 ml/l coconut water) in MS media with 5 replicates with observation for 4 weeks after planting. The results showed did not differ markedly in the number of protocorms, number of shoots and number of browning protocorms, but differ markedly in the parameter protocorm color based on DMRT test. Treatment of 3 mg/l BAP gives the best results on the number of shoots 2,60 protocorms and number of browning protocormss at the least amount 0,80 protocorms. The number of protocorms most widely on the treatment of 1 g/l Growmore 14,40 protocorms. Protocorms color is best found in 3 g/l Growmore with green color. This research managed to multiply the number of protocorms and induces protocorms shoots from Grammatophyllum stapeliiflorum orchid.

Abstract

The an annual herbaceous tropical plant which is one of the species of the genus Tacca from the Dioscoreaceae family is Tacca chantrieri. T. chantrieri has a unique inflorescence morphology like that of a bat. The people of Southeast China and Thailand have used by T. chantrieri rhizome as traditional medicine because the methanol extract contains secondary metabolites such as diarylheptanoids, pseudofurostan, withanolide, taccalonolide, and saponins. To maintain its sustainability, it is necessary to propagate T. chantrieri by using in vitro culture techniques such as callus culture. The purpose of this study was to determine the response of T. chantrieri leaf callus formation and to determine the optimal concentration with various concentrations of 2,4-D and BAP in vitro. This study used a completely randomized design consisting of control treatments, 1 and 2 mg L-1 2,4-D and concentrations of 0.5; 1.0; 1.5; 2.0 2,4-D combined with 3 mg L-1 BAP. The observations were made for 60 days after planting. The results showed that the concentration of 1.5 mg L-1 2,4-D + 3 mg L-1 BAP affected the percentage of live explants and the percentage of callus formation by 100% respectively, and the time of callus appeared 18.75 days after planting.

Abstract

Duku (Lansium domesticum Corr.) adalah salah satu buah tropis bernilai ekonomis yang diminati oleh masyarakat dengan rasa manis dan bernilai gizi tinggi. Biji duku memiliki sifat poliembrioni yang akan menghasilkan tanaman lebih banyak dan seragam sesuai dengan induknya. Perbanyakan dengan teknik in vitro akan menghasilkan tanaman dalam jumlah yang banyak dalam waktu yang relatif singkat dibandingkan secara konvensional. Tujuan penelitian yaitu untuk mengetahui respon poliembrioni biji duku yang dibelah tiga secara membujur  dengan penambahan BAP (Benzyl Amino Purine)  secara in vitro. Penelitian ini menggunakan Rancangan Acak Lengkap dengan konsentrasi BAP = 0, 1, 3, 5 dan 7 mg/L BAP. Hasil penelitian menunjukkan bahwa waktu muncul tunas tercepat yaitu 2,20 MST pada konsentrasi 1 mg/L BAP. Jumlah tunas tertinggi pada perlakuan 3 mg/L BAP yaitu 2,00 tunas, namun perlakuan pemberian konsentrasi BAP belum mampu meningkatkan jumlah tunas.

Abstract

Gotu kola (Centella asiatica (L.) Urb.) is a medicinal plant that contains chemical compounds as triterpenoids and saponins.  The chemical compounds can be produced quickly using in vitro callus induction.  The callus is a very important source of planting material in regenerating new plants.  Therefore, by inducing callus the need for seedling in large quantities achievable in a short time.  This study aimed to determine the effect of the BAP single and combination of BAP and NAA, and determine the effective concentration of a BAP single and combination of BAP and NAA on callus induction of gotu kola leaf explant using in vitro method.  This study used Complete Randomized Design (CRD) which consisted of nine treatments (control, 1 mg/l BAP, 2 mg/l BAP, 1 mg/l BAP + 0,1 mg/l NAA, 2 mg/l BAP + 0,1 mg/l NAA, 1 mg/l BAP + 0,3 mg/l NAA, 2 mg/l BAP + 0,3 mg/l NAA, 1 mg/l BAP + 0,5 mg/l NAA, 2 mg/l BAP + 0,5 mg/l NAA) with five replication for each treatment. Data obtained from observation were analyzed descriptively.  The results of this research showed that a single BAP and combine of BAP and NAA were able provide a response in the form of colour, swelling and callus formation.  The best concentration that from the most optimal callus and the highest callus growth (+++) is the treatment of P5 ( 1 mg/l BAP + 0,3 mg/l NAA) which is equal to 100% with the caracteristics of green callus and callus covering the entire surface of explants.  All the callus produced was textured compact, while the colour of the callus was green and brown. Urb.) merupakan tanaman berkhasiat obat yang mengandung berbagai bahan aktif seperti triterpenoid dan saponin.  Bahan aktif tersebut dapat diproduksi dengan cepat menggunakan teknik induksi kalus secara in vitro.  Kalus merupakan sumber bahan tanam yang sangat penting dalam meregenerasi tanaman baru.  Oleh karena itu, dengan menginduksi kalus kebutuhan bibit dalam jumlah banyak dapat dicapai dengan waktu singkat.  Penelitian ini bertujuan untuk mengetahui pengaruh penambahan BAP tunggal dan kombinasi BAP dan NAA, dan menentukan konsentrasi terbaik dari penambahan BAP tunggal dan kombinasi BAP dan NAA terhadap pembentukan kalus dari eksplan daun pegagan secara in vitro. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) yang terdiri atas 9 (sembilan) perlakuan (kontrol, 1 mg/l BAP, 2 mg/l BAP, 1 mg/l BAP + 0,1 mg/l NAA, 2 mg/l BAP + 0,1 mg/l NAA, 1 mg/l BAP + 0,3 mg/l NAA, 2 mg/l BAP + 0,3 mg/l NAA, 1 mg/l BAP + 0,5 mg/l NAA, 2 mg/l BAP + 0,5 mg/l NAA) dan masing-masing diulang sebanyak 5 (lima) ulangan.  Data hasil pengamatan yang diperoleh dibahas secara deskriptif karena tidak semua ulangan menghasilkan kalus.  Hasil penelitian menunjukkan bahwa pemberian perlakuan BAP tunggal dan kombinasi penambahan BAP dan NAA mampu memberikan respons pada eksplan daun pegagan berupa perubahan warna, pembengkakan dan terbentuknya kalus.  Konsentrasi terbaik yang mampu membentuk kalus paling optimal dan pertumbuhan kalus paling tinggi (+++) yaitu perlakuan P5 ( 1 mg/l BAP + 0,3 mg/l NAA) yakni sebesar 100% dengan ciri-ciri kalus berwarna hijau dan kalus menutupi seluruh permukaan eksplan.  Semua kalus yang dihasilkan bertekstur kompak, sedangkan warna kalus yang dihasilkan hijau dan coklat. Abstract

Jeruk kasturi (Citrus microcarpa) adalah salah satu jenis buah yang termasuk ke  dalam genus Citrus, memiliki kandungan vitamin C, dan anti oksidan juga termasuk tinggi. Perkembangan secara generatif setelah berumur 5 tahun. Penelitian ini bertujuan untuk mengetahui efek dan menentukan konsentrasi BAP dan Kinetin terbaik secara tunggal atau kombinasi dari kedua regulator pertumbuhan untuk pembentukan tunas dari eksplan epikotil jeruk kasturi secara in vitro pada media Ms. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) terdiri dari 16 taraf perlakuan (kontrol, 0,5 mg/l BAP, 1 mg/l BAP, 1,5 mg/l BAP, 0,5 mg/l Kinetin, 1 mg/l Kinetin, 1,5 mg/l Kinetin, 0,5 mg/l BAP+ 0,5 mg/l Kinetin, 0,5 mg/l BAP + 1 mg/l Kinetin, 0,5 mg/l BAP + 1,5 mg/l Kinetin, 1 mg/l BAP + 0,5 mg/l Kinetin, 1 mg/l BAP + 1    mg/l Kinetin, 1  mg/l BAP + 1,5 mg/l Kinetin, 1,5 mg/l BAP + 0,5 mg/l Kinetin, 1,5 mg/l BAP + 1 mg/l Kinetin, 1,5 mg/l BAP + 1,5 mg/l Kinetin). Hasil penelitian ini menunjukkan bahwa pemberian perlakuan BAP dan Kinetin tunggal atau kombinasi mampu memberikan respons pada eksplan epikotil jeruk kasturi berupa waktu muncul tunas, jumlah tunas, jumlah daun dan panjang tunas. Konsentrasi terbaik yang mampu membentuk jumlah tunas paling banyak yaitu perlakuan P6 (1,5 mg/l Kinetin) sebesar 1,6 tunas.
