Abstract

This research aimed to examine the effect of baker’s yeast as immunostimulant on growth of nile tilapia and to find out a proper dose of baker’s yeast that promoted optimal growth of fish. Juveniles with an average weight of 31.83 g were obtained from stock available at Aquaculture Study Program of the Faculty of Fisheries and Marine Sciences.  This research was conducted at the experimental ponds located at the Faculty of Fisheries and Marine Sciences for three months. During the experiment, fish was fed with pellet supplemented with baker’s yeast as treatment for four weeks as much as 4%/bw/day, twice daily.  The doses of baker’s yeast consisted of 0, 10, 20, 30, and 40 g/kg pellet. Fish growth including absolute and relative growth was measured at two weeks interval.  Research results showed that  after two weeks of feeding, supplementation of baker’s yeast did not affect the growth of fish but after four weeks of feeding, supplementation of baker’s yeast significantly increased fish growth. Fish fed pellet supplemented with 10 g baker’s yeast displayed the highest growth. As conclusion, supplementation of baker’s yeast in fish food could enchance the growth of fish.   Keywords:  Oreochromis niloticus, immunostimulant, baker’s yeast, growth

Abstract

The aimed of the study was to isolate and select the probiotic Lactobacillus from carp intestine potential for use in aquaculture. Carp as a source of probiotics with a size of about 200 g was  taken from the Center for Fish Hatchery and Disease Control (BP3I), Tateli. After aseptic surgery, the intestine was separated and cleaned, weighed as much as 1 g, put in a mortar and crushed until smooth. The scoured results were put into a tube and added 9 ml NaCl to get a ratio of 1:10. The tube that already contains intestinal scouring was then centrifuged at a speed of 1000 RPM for 10 minutes. The centrifugation process was carried out 2 times to get the supernatant that has been cleared of debris. The supernatant was removed and diluted to obtain a concentration of 10-2  to 10-3. The solution was then spread on MRS (De Man Rogosa and Sharpe Agar) media and incubated at 28oC in the Incubator for 24-48 hours. The bacteria that grew in culture fmedia was reisolated several times to get a single pure colony.. The pure colonies were propagated in the same media to be further identified through gram staining, biochemical, proteolytic, amylolytic, cellulolytic, antibiotic resistance, antagonistic, pathogenicity tests. The results of the isolation obtained the characteristics of a colony of milky white color, round and convex with long cells, gram-positive and confirmed based on biochemical tests as Lactobaccilus sp. The bacterium also had proteolytic, amylolytic, cellulolytic, antagonistic, antibiotic-sensitive activity that were characterized by the formation of clear zones in the area around probiotic discs, and did not cause death either as a disease or as a result of poisoning. Thus it can be concluded that Lactobaccilus sp. isolated from carp intestines can be used as probiotics for cultivation.

Abstract

The aim of this research was to count the total leukocytes and phagocytosis activity of goldfish seeds treated with probiotics. The test fish used were goldfish seeds measuring 5-8 cm. 500 fish will be taken from the Tateli Fish Cultivation and Development Center (BP3I). This study used a completely randomized design (CRD) with 5 treatments, where each treatment had 3 replications. The treatments used were probiotic bacteria from the intestines of goldfish with different concentrations. The treatments tested consisted of: A: without the addition of probiotics, B: addition of probiotics 1x106 cfu / mL, C: addition of probiotics 1x107 cfu / mL, D: addition of probiotics 1x108 cfu / mL, E: addition of probiotics 1x109 cfu / mL. Each concentration of bacteria that has been prepared is then sprayed on the commercial feed evenly using a sprayer and then dried to room temperature. After drying the feed was coated with 2% egg yolk and dried again at room temperature. Fish are stocked in an aquarium measuring 60x40x40cm3 with a density of 20 fish / aquarium. Each aquarium is equipped with an aerator and a water pump for recirculation. The fish were then given feed that had been added with probiotics at the same dose and frequency of administration as in the acclimatization process. The treatment is given for 6 weeks. At the end of the treatment feeding, fish samples were taken from each experimental unit to measure immune parameters, namely total leukocytes and phagocytic activity. Based on the results of research using the probiotic Lactobacillus sp. which originated from the intestines of goldfish and added to the feed had a significant effect on increasing total leukocytes and phagocytosis activity and the optimal concenration was  1x107 - 1x108 cfu / mL with a duration of 6 weeks of administration

Abstract

The purpose of this study was to determine the feasibility of blood meal in various level inclusins in  promoting  the growth rate and nutrient utilization of O. niloticus.  The results showed that the absolute growth of tilapia in treatment 2 (with a combination of 20%) was higher than the other treatments. The daily growth rate of tilapia in treatment 2 (3.5% / day) was higher than other treatments. Likewise, with the feed change value, treatment 2 showed significantly different from other treatments. The combination of tuna fish meal waste and blood meal showed significantly different results from the results of statistical tests

Abstract

The purposed of research was to evaluate the ability of Andong leaf extract as natural antimicrobial against pathogenic bacteria, Aeromonas hydrophila. The plant was collected from yards at Kasuang Village, Tondano Disctrict of Minahasa Regency. After washing, the leaf was cut into small pieces, eased using a mortar and then extracted by maceration using alcohol 75% for 24 hours with ratio between simplicia and solvent was 1:1, 1:2, 1:4 and 1:8.  Antimicrobial activity of the extract was examined through disc diffusion method. The results showed that leaf extract of andong was able to inhibit the growth of bacteria  with strong and the sensitivity of the extract was categorized as strong.

Abstract

This study aimed to examine the effect of several types of natural substances in increasing the growth of tilapia optimally. The fish were taken from Board of Fisheries and Freshwater Aquaculture Tatelu (BPBAT), North Sulawesi Province. Fish were acclimatized for a week in ponds in the “mina padi” system, with a density of 25 fish/net. After acclimatization, the fish were fed pellet with the addition of ginger (Zingiber officinale), garlic (Allium sativum Linn), and yeast (Saccharomyces cerevisiae) for 30 days as much as 10% /fish body weight /day with the frequency of feeding twice a day, namely at 08.00 and 16.00. This study used a completely randomized design (CRD) with four treatments, namely treatment A: ginger flour 7.5 g/kg of feed, treatment B: garlic flour 15 g/kg of feed, treatment C: Yeast 5 g/kg of feed and treatment D: commercial feed (control), each had three replications. The weight of fish was scaled weekly. The results showed that the growth of fish fed with the addition of baker’s yeast was better than fish fed with the addition of ginger, garlic, and commercial feed, but statistically the growth of fish fed with baker’s yeast and ginger were not significantly different from each other.

Abstract

This study aimed to examine the effect of probiotic Effective Microorganism-4 (EM4) on the growth rate of the natural feed population of Daphnia magna and to determine the right dose to obtain the highest growth. This research was conducted at the Aquaculture Technology Laboratory, Faculty of Fisheries and Marine Sciences, Sam Ratulangi University, Manado. The time of the study was carried out from April to May 2021. The experimental design used was designed according to a 4 x 4 factorial experimental design in Completely Randomized (CRD). The first factor was the dose of EM4 with 3 levels, namely 0.75 mL, 1 mL and 1.25 mL with 1 control, then the second factor is the time difference (weeks) I, II, III, and IV. The results of the study using the Analysis of Variance (ANOVA) using the JMP application showed 1. The dose difference in the density of daphnia on the culture medium was not significantly affected by the difference between the culture medium and chicken manure (value 'Prob>F)' = 0.1411), 2. The difference in density of daphnia on culture medium was very significant by differences in culture time (Value 'Prob>F' = 0.0003), 3. Interaction, The effect of culture medium factors (EM-4) dose and chicken manure on the density of daphnia changed significantly at the time of the change in the level of the time factor (weeks) (Value 'Prob>F' = 0.0439). Further contrast tests showed that the density of daphnia at week-I was significantly different from the density of daphnia at week-II, week-III, and week-IV and the density of daphnia at week-II, week-III and week-IV were not significantly different.

Abstract

The purposes of research were to examine the effect of extract of Acalypha indica leaf on the growth of Nile tilapia and determine the dose of leaf extract that could enhance the growth of tilapia optimally. The fish used were Nile tilapia taken from the Tatelu Center for Freshwater Aquaculture with an average weight of 3.79±0.50 g. The medicinal plant A. indica was collected from around the Faculty of Fisheries and Marine Science complex and from the North Bolaang Mongondow area. After being washed, the leaves were dried and then ground to obtain leaf powder. Leaf flour was then extracted by maceration method with 95% alcohol solvent. After being concentrated, the extract was then used as a treatment with different concentrations, including A: 0 g extract/kg feed, B:10 g extract/kg feed, C:20 g extract/kg feed, D:30 g extract/kg feed and E: 40g extract/kg feed. The data collected consisted of absolute growth, daily growth and relative growth. The results showed that the extract of A. indica leaf added to the feed had a significantly effect on the growth of tilapia indicated by an increase in absolute growth, daily growth, and relative growth (p<0.05). The best fish growth was achieved in fish fed with feed with the addition of 20 g extract/kg feed. In conclusion, leaf extract of A. indica can increase the growth of Nile tilapia

Abstract

The purpose of this study was to identify the bacteria Aeromonas sp. in a tilapia (Oreochromis niloticus) culture pond in Matungkas Village, Dimembe District, North Minahasa Regency. Bacterial identification was carried out at the Laboratory of the Fish Quarantine Center for Quality and Safety Control of Fishery Products, Manado. The time of the study was carried out from February 2021 - June 2021. Ten samples of fish were selected from 3 existing ponds for this study with the criteria of fish that have clinical signs of disease infection such as pale body surface, red spots and ulcers. From 10 samples, 19 isolates were obtained to be identified. The bacterial method was carried out based on the results of gram tests and biochemical tests which included: Gram Tests, Motility Test, Oxidase Test, Oxidative Fermentative Test, Tolerance Test to incubation temperature, Tolerance Test to pH of the media, Tolerance Test to the concentration of NaCl in the media and rimler – shotts selective media test. Of the 19 isolates from 10 samples identified, there were 7 isolates that were positive for A. hydrophila, namely: N1.4.1, N2.1.1, N2.1.2, N2.2.1, N2.3.2, N3.3.1, and N3.3.2. Twelve other isolates were negative for A. hydrophila. The characters of A. hydrophila identified are creamy in colour, gram negative, oxidase positive, bacteria tolerance to temperature 4 °C positive, 37 °C positive, 50 °C negative, can move (motile), fermentative, pH tolerance test media pH 3 negative, pH 5 positive, pH 9 positive, pH 11 positive, tolerance test for NaCl concentration 0.5% positive, 3% positive, 5% negative, rimler – shotts positive. All sampling sites (ponds) in Matungkas Village were infected with bacteria.

Abstract

This study aimed to examine the effect of adding ciplukan (Physalis angulata) to feed on the growth of tilapia and to find a dose of ciplukan that was able to increase the growth of tilapia (Oreochromis niloticus). The test fish used were tilapia with an average size of 5-8 cm as many as 300 fish from the Center for Freshwater Aquaculture (BPBAT), Talelu. This research was conducted at the Aquaculture Technology Laboratory, Faculty of Fisheries and Marine Sciences, Sam Ratulangi University. This study used a completely randomized design (CRD) with 4 treatments, where each treatment had 3 replications so there were 15 experimental units, the treatment used was ciplukan flour with different concentrations consisting of: A: 0 g flour/kg feed, B : 10 g flour/ kg feed, C : 15 g flour/ kg feed, D : 20 g flour/ kg feed. The data collected consisted of fish weight to measure the fish growth. The results showed that the addition of ciplukan leaf extract in feed with different concentrations gave significantly different effects on the growth of tilapia with treatment B (10 g) showing the highest fish growth reaching 5.24 g, followed by treatment C (15 g) with the highest growth. reached 4.35 g. While the fish treated with D (20 g) only reached 3.16 g in weight where this value was not much different from the growth of fish in control A. Duncan's further test results showed that treatments B, C, D were not significantly different from each other, but these three treatments were very significantly different compared to fish that were not given ciplukan leaf extract. The study showed that ciplukan leaf extract added to feed gave significantly different effects on fish growth, with the best dose of 10 g extract/kg feed. Thus it can be concluded that ciplukan leaf extract can increase growth of Nile tilapia.
