Abstract

Drug storage is something that must be considered because it can affect the quality of the drug. Storage conditions must adapt to the stability of each drug. This study aims to determine the level of knowledge about the storage of syrup drug preparations in patients in all public health centers in Pekanbaru. The research method is analytic observational with cross sectional design. The research sample was the people who visited all health centers in Pekanbaru city which were selected by purposive sampling technique according to the inclusion and exclusion criteria and obtained a sample of 160 respondents. The instrument is a knowledge questionnaire that has been tested for validation and reliability. The knowledge questionnaire contained 14 valid questions (r=0.818). Presentation of data with bar charts and narrative text. The results of the study revealed that there were 66 respondents (41.25%) in the good knowledge category and 94 respondents (58.75%) in the sufficient knowledge category and none of the respondents had less knowledge in the category. From these results, it can be concluded that most people do not know well how to store syrup drugs, so it is necessary for the role of pharmacists to provide education to the public, especially regarding how to store drugs correctly

Abstract

Degenerative a disease that continues to grow and can affect a person's productivity and quality of life. Usually, people only check themselves after their symptoms, even though early detection is important. The aim of the service is to provide an overview and improve the status of the community both promotively, preventively, or curatively by providing counseling about degenerative diseases. The method used is to conduct a health examination and continued with the provision of counseling. The activity was attended by 42 people in the Car Free Day area of Pekanbaru city. The activity begins with a health check that is tailored to the symptoms felt and continues with counseling. Many factors cause degenerative diseases, such as genetics, age, physical activity, and diet. This activity can increase public insight and knowledge about degenerative diseases.

Abstract

The new pyrazolo-pyridine compounds derived from monoketone curcumin analogues (P4Cl and P2CL) were synthesized in two stages. The first stage was the synthesis of curcumin monoketone (Cs) analogue through the Claisen-Schmidt reaction in the heating method using reflux. The second step was the synthesis of P2CL and P4Cl through nucleophilic addition and intramolecular cyclization in the heating method using a Monowave 50 apparatus. The structures of the compounds P2CL and P4Cl were confirmed by spectroscopic analysis including UV-Vis, FT-IR, 1H-NMR and HRMS. Based on molecular docking studies, the compounds P2Cl and P4Cl have weak potential as COX-2 enzyme inhibitors with a binding free energy value of -4.02 kcal/mol and an RMSD of 1.19.

Abstract

Scaffold pirazol maupun piazolo-piridin telah dilaporkan memiliki potensi aktivitas biologis yang menarik dan terdapat pada berbagai struktur senyawa obat yang telah disetujui oleh food and drug administration (FDA). Pada penelitian ini, senyawa pirazolo-piridin tersubstitusi metoksi sebagai turunan dari analog kurkumin monokarbonil telah disintesis melalui dua tahap reaksi. Tahap pertama adalah sintesis analog kurkumin monokarbonil tersubsitusi metoksi melalui reaksi kondensasi Claisen-Schmidt dengan bantuan iradiasi microwave. Tahap kedua adalah sintesis senyawa pirazolo-piridin melalui reaksi adisi nukleofilik yang diikuti oleh reaksi siklisasi intramolekular dalam reaktor tertutup, monowave 50. Kemurnian produk hasil sintesis telah dipastikan melalui analisis HPLC. Struktur senyawa pyrazolo-piridin telah dikonfirmasi melalui analisis spektroskopi UV-Vis, FT-IR, dan 1H-NMR. Berdasarkan studi molecular docking, senyawa tersebut  tidak menunjukkan potensi aktivitas yang baik sebagai inhibitor enzim siklooksigenase-2 (COX-2), karena hanya memiliki nilai energi bebas pengikatan sebesar -5,98 kcal/mol. Selain itu, pirazolo piridin tersubstitusi metoksi juga tidak dapat membentuk satupun ikatan hidrogen dengan sisi aktif COX-2. Di sisi lain, celecoxib sebagai kontrol positif memiliki energi bebas pengikatan sebesar -11,97 kcal/mol dan dapat membentuk ikatan hidrogen dengan residu asam amino Gln178, Leu338, Arg499, dan Phe504 pada sisi aktif COX-2 (PDB ID: 3LN1). Pyrazol and pyrazolopyridine scaffolds have been reported to have many interesting biological activities and present in various structures of drug compounds that have been approved by the food and drug administration (FDA). In this study, a methoxy-substituted pyrazolo-pyridine compound as derivative of monocarbonyl curcumin analogs was synthesized in two steps of reaction. The first step was the synthesis of monocarbonyl curcumin analog through the Claisen-Schmidt condensation with the assist of microwave irradiation. The second step is the synthesis of pyrazolopyridine through nucleophilic addition followed by intramolecular cyclization in a closed-vessel reactor, monowave 50. The purity of synthesized product was confirmed by HPLC analysis, and the structure has been confirmed through UV-Vis, FT-IR, and 1H NMR spectroscopic analyses. Based on the molecular docking study, the pyrazolo-pyridine did not show good activity as cyclooxygenase-2 (COX-2) inhibitor, because it only has a binding free energy of -5.98 kcal/mol. In addition, methoxy-substituted pyrazolo-pyridin also can not form any hydrogen bonds with the active site of COX-2. On the other hand, celecoxib as a positive control has a binding free energy of -11.97 kcal/mol and can form hydrogen bonds with Gln178, Leu338, Arg499, and Phe504 amino acid residues on the active site of COX-2 (PDB ID: 3LN1).

Abstract

Dengue hemorrhagic fever (DHF) is a disease caused by the dengue virus (DENV). Dengue virus can enter the human body through the Aedes aegypti and Aedes albopictus mosquitoes. According to the Indonesian Ministry of Health, dengue hemorrhagic fever (DHF) is still a serious health problem in Indonesia. The type of dengue virus serotype most commonly found to cause infection in the human body is the DENV-2 serotype. This study aims to determine whether Ginger (Zingiber officinale) isolate compounds have potential as dengue DEN-2 NS2B/NS3 inhibitors. Samples used are compounds with IUPAC names (S)-5-hydroxy-1-(4-hydroxy-3-methoxyphenyl) octan-3-one (4-gingerol) and (S)-5-hydroxy-1-(3-methoxy-4-methylphenyl) decan-3-one. The method used is molecular docking and Pharmacophore using the MOE (Molecular Operating Environment) 2022.0901 software package. The results obtained based on the observed parameters of the two compounds isolated from ginger (Zingiber officinale) could be estimated as potential dengue DEN2 NS2B/NS3 inhibitors.

Abstract

Rationality of drug use is the rational use of drugs in which patients receive treatment according to clinical needs. Indragiri Hulu Regency has a total of cases of People with Mental Disorders (PMD) until December 2020, there were 553 cases with the highest number being schizophrenia as many as 393 cases. Antipsychotics were the first drug therapy to be effective in treating schizophrenia. This study aims to determine the rationality of the use of antipsychotics with rationality indicators referring to the World Health Organization which includes the right indication, right patient, right drug, right dose, and alert for side effects. The sample in this study were schizophrenic patients in 10 health centers in Indragiri Hulu Regency with the highest number of patients. This research is descriptive analytic done retrospectively. The results of this study are the right indication 100%, the right patient 100%, the right drug 98,77%, the right dose 61,72%, and the alert for side effects 71,60%.  Keywords: rationality, schizophrenia, antipsychotic

Abstract

Background: Pharmacy students consist of undergraduate and professional pharmacy students. They are candidates for future pharmacist health workers who require practical experience. Offline learning with hands-on practice methods in health facilities supports their professional skills. Objective: This study aimed to determine pharmacy students' knowledge and attitude toward implementing offline learning methods during the new normal era. Methods: This study involved pharmacy students from Indonesia who were asked to participate in an e-questionnaire about the vaccination program, COVID-19 health protocols, pharmacist competence, and attitude toward implementing offline learning. The students' scores were based on their knowledge and attitude. Statistical analysis was performed to compare the scores between the two groups, and a correlation test was conducted to assess the relationship between the students' knowledge and attitudes. Results: A total of 652 pharmacy students were divided into two groups, undergraduate and professional pharmacy students, in a 3:1 ratio. About 74.6% of undergraduate students and 78.5% of professional pharmacy students had good and moderate knowledge. The level of knowledge was not different between the two groups (p=0.602; p>α). Professional pharmacy students were more ready to engage in offline learning compared to undergraduate students (p=0.001; p<α). However, there was a relatively low correlation between knowledge and attitudes, with r = 0.079 (p=0.043; p<α). Conclusion: Professional pharmacy students are more ready for offline learning methods than undergraduate students, particularly in hands-on field practice.

Abstract

Pada penelitian ini, 2'-hidroksicalkon (C345) dan flavonol (F345) tersubstitusi trimetoksi telah disintesis menggunakan metode iradiasi microwave dan metode pengadukan. Struktur kedua produk telah dikarakterisasi melalui analisis spektroskopi UV-Vis, FT-IR, 1H NMR, 13C NMR, dan HRMS. Hasil analisis spektroskopi menunjukkan bahwa kedua produk memiliki struktur yang sesuai dengan struktur molekul target yang diharapkan. Selain itu, hasil sintesis juga menunjukkan bahwa metode iradiasi microwave terbukti dapat mempercepat waktu reaksi (dari 1,5-3,0 jam menjadi 3-6 menit) dan meningkatkan rendemen produk murni pada sintesis senyawa C345 (55,31 %) dan F345 (83,65 %). Selanjutnya, hasil docking menunjukkan bahwa kedua senyawa dapat membentuk ikatan hidrogen dengan beberapa residu penting pada sisi aktif dan dapat terikat pada kedua situs katalik MPro SARS-CoV-2 (PDB ID:6M2N), yaitu His41 dan Cys145 melalui interaksi hidrofobik dengan nilai energi bebas pengikatan yang lebih negatif (-8,95 dan -9,02 kcal/mol) dibandingkan dengan baicalein sebagai inhibitor pembanding. Hasil kajian in silico lainnya juga menunjukkan bahwa kedua senyawa memiliki profil farmakokinetik yang baik dan memiliki sifat kemiripan dengan obat berdasarkan aturan Lipinski, Ghose, Veber, Egan, dan Muegge. Selain itu, senyawa F345 juga diprediksi memiliki risiko toksisitas yang lebih kecil dibandingkan dengan baicalein. Synthesis, Structural Characterization, and In Silico Study of the Potential of 2'-Hydroxychalcone and Trimethoxy-Substituted Flavonols as Inhibitors of the Main Protease (MPro) of SARS-CoV-2. In this study, trimethoxy-substituted 2'-hydroxychalcone (C345) and flavonol (F345) were synthesized using microwave irradiation and stirring methods. The structure of the two products were characterized by spectroscopic analyses including UV-Vis, FT-IR, 1H NMR, 13C NMR, and HRMS. The result of spectroscopic analyses showed that the products had structures consistent with the expected target molecules. In addition, the synthesis results showed that the microwave irradiation method was proven to speed up the reaction time (from 1.5-3.0 hours to 3-6 minutes) and increased the yield of pure product in the synthesis of compounds C345 (55.31 %) and F345 (83.65 %). Furthermore, the docking result showed that the two compounds can form hydrogen bonds with several important residues on the active site and also can bind to catalytic dyad of the SARS-CoV-2 MPro (PDB ID:6M2N), namely His41 and Cys145 through hydrophobic interactions with a more negative binding free energy (-8.95 and -9.02 kcal/mol) than baicalein as a reference inhibitor. The result of silico studies also showed that the two compounds exhibited good pharmacokinetic profiles and drug-likeness properties based on Lipinski, Ghose, Veber, Egan, and Muegge rules. In addition, compound F345 was also predicted to has a smaller toxicity risk compared to baicalein.

Abstract

Berkurangnya stabilitas suatu obat dapat menyebabkan efek toksik dan efek yang tidak diinginkan lainnya dan salah satu penyebabnya yaitu obat sudah rusak dan melewati masa kadaluwarsa. Saat ini diharapkan agar semua orang memiliki pengetahuan tentang obat kadaluwarsa dan obat rusak dan juga bagaimana penanganan yang harus dilakukan saat ditemukan adanya obat kadaluwarsa dan obat yang sudah rusak. Tujuan dari kegiatan ini untuk melihat pengaruh pemberian media leaflet tentang waspada obat kadaluwarsa dan/atau rusak serta memberikan pengetahuan seputar obat kadaluwarsa dan/atau rusak di SMAN 10 Pekanbaru. Kegiatan pengabdian masyarakat ini dilakukan dengan sistem diskusi dan demonstrasi, dimulai dari pengisian lembar Kuisioner (pre-test), pembagian leaflet, edukasi secara lisan dan diakhiri dengan pengisian lembar Kuisioner (post-test) terkait pemahaman siswa terhadap informasi yang telah disampaikan. Hasil akan dievaluasi berdasarkan skala guttman. Hasil pre-test dan post-test dimana jumlah siswa yang termasuk dalam kategori tinggi meningkat dari 37 siswa (52,11%) menjadi 64 siswa (90,14%) pada nilai post-test. Hal ini menunjukkan bahwa kegiatan penyuluhan telah berhasil meningkatkan pemahaman siswa serta memberikan dampak positif bagi siswa dalam menyikapi permasalahan yang ada terkait cara penyimpanan obat yang benar, mengetahui ciri-ciri obat kadaluwarsa dan rusak, serta cara pemusnahannya.

Abstract

ABSTRAK Diabetes melitus (DM) adalah penyakit yang disebabkan gagalnya pankreas memproduksi hormon insulin. Penyakit DM menjadi permasalahan kesehatan yang banyak terjadi di masyarakat Riau. Kegiatan pengabdian ini bertujuan untuk memberikan pemahaman terkait penanganan DM. Metode pelaksanaan kegiatan ini berupa penyuluhan yang didukung dengan adanya pre dan post-test. Kegiatan ini dilaksanakan di kawasan CFD (Car Free Day) Kota Pekanbaru. Alat ukur yang digunakan adalah lembar checklist pre dan post-test serta hasilnya dihitung dengan menggunakan skala Guttmann. Hasil dari kegiatan pengabdian ini adalah sebanyak 30 responden baik laki-laki maupun perempuan mempunyai pemahaman yang sama tentang DM yaitu 82%. Jika dilihat dari segi usia, mayoritas orang dewasa di usia prima memiliki pengetahuan yang lebih tinggi dibandingkan orang yang lebih tua. Mengenai tingkat pendidikan akhir, responden yang berpendidikan tinggi mempunyai pemahaman yang lebih baik (92%). Terkait status pekerjaan, responden yang berstatus bekerja memiliki pengetahuan paling tinggi (84%). Berdasarkan hasil kegiatan terdapat perbedaan yang signifikan antara sebelum dan sesudah kegiatan penyuluhan. Hal ini terlihat dari nilai signifikansi <0,05 (0,000), artinya terdapat perbedaan yang signifikan secara deskriptif antara pre-test dan post-test. Tingkat pengetahuan yang didapatkan adalah rata-rata pengetahuan pre-test (baik) dan post-test (sangat baik). Kesimpulan yang didapat adalah terjadinya peningkatan pengetahuan dari baik menjasi sangat baik.   ABSTRACT Diabetes mellitus (DM) is a disease caused by the failure of the pancreas to produce the hormone insulin. DM is a health problem that occurs in many communities in Riau. This service activity aims to provide an understanding of DM management. The method of implementing this activity is in the form of counseling supported by the pre and post-test. This activity was carried out in the CFD (Car Free Day) area of Pekanbaru City. The measuring instrument used is a pre and post-test checklist sheet and the results are calculated using the Guttmann scale. The results of this service activity are as many as 30 respondents, both men and women, have the same understanding of DM, namely 82%. When viewed in terms of age, the majority of adults in prime age have higher knowledge than older people. Regarding the final level of education, respondents with higher education had a better understanding (92%). Regarding employment status, respondents who were employed had the highest knowledge (84%). Based on the results of the activity, there was a significant difference between before and after the counseling activity. This can be seen from the significance value <0.05 (0.000), meaning that there is a descriptively significant difference between the pre-test and post-test. The level of knowledge obtained is the average knowledge of pre-test (good) and post-test (very good). The conclusion is that there is an increase in knowledge from good to very good.
