Abstract

Cervical cancer ranks among the top causes of cancer-related deaths in women globally. Early detection is vital for improving patient survival rates. The multiclass classification of cervical cell images presents challenges primarily due to the notable variations in cell sizes across different classes. Conventional AI methods for diagnosing cervical cancer often rely on image-resizing techniques that overlook crucial features like relative cell dimensions, which impairs the models' ability to distinguish between classes effectively. This paper presents a novel AI-driven approach that employs constant padding to maintain the natural size differences among cells. Our method utilizes deep learning for both feature extraction and multiclass classification. We assessed the method using the publicly accessible SIPaKMeD dataset. Experimental findings indicate that our approach surpasses traditional image-resizing methods, especially in classes that are more challenging to predict. This strategy highlights AI's potential to improve cervical cancer diagnosis, offering a more precise and dependable tool for early detection. A reliable and precise AI model for diagnosing cervical cancer is crucial for promoting widespread screening and ensuring timely and effective treatment, which can ultimately lower mortality rates. By aiding early and accurate diagnosis, this approach aligns with global health efforts to alleviate the burden of cancer and other diseases, especially in areas with limited access to advanced healthcare services facilities.

Abstract

Abstract

Second-degree burns are tissue injuries that often cause complications and require a long healing time. Previous studies have reported that natural ingredients such as Jatropha multifida L., placenta extract, and Carboxymethyl Chitosan (JM-P-CMC) have the potential to accelerate tissue regeneration, but no combined studies have been conducted on the three. The study aimed to evaluate the effect of the Jatropha multifida L.-Plasenta-CMC combination formulation on the number of fibroblasts as an indicator of tissue regeneration in second-degree burns. This experimental study used a post-test only control group design with 24 male Sprague dawley rats (200–250 g) induced burns on the back using a hot metal plate, then divided into four groups: control (Bioplacenton ® ), JM5%-P-CMC, JM10%-P-CMC, and JM15%-P-CMC. Topical treatment was carried out twice a day for 14 days. Data collection was carried out on days 5, 9, and 14, followed by histological analysis of skin tissue with Hematoxylin Eosin staining to count the number of fibroblasts. The results of the Kruskal-Wallis and Mann-Whitney tests showed a significant increase in the number of fibroblasts in all treatment groups compared to the control, with the highest results in the JM15%-P-CMC group. This study concluded that the combination was effective in enhancing tissue regeneration and has the potential to be developed as an innovative topical therapy based on natural ingredients.

Abstract

Abstract One of the most prevalent diseases in women is breast cancer, which is linked to unchecked cell division via the mTORC signaling pathway. This study sought to determine how Red Bajakah Wood (Spatholobus littoralis) extract affected the in vitro expression of the mTORC protein in MCF-7 breast cancer cells. A true experimental design with post-test only control group was employed, including six treatment groups: negative control, positive control (Tamoxifen), and Red Bajakah Wood extract at IC₅₀ 72-hour concentration. Cells were cultured, treated, and viability assessed using CCK-8 to determine IC₅₀, while mTORC protein levels were measured via ELISA. Results showed that Red Bajakah Wood extract reduced cell viability in a dose- and time-dependent manner, with the lowest IC₅₀ at 72 hours incubation. mTORC levels increased significantly in extract-treated cells compared to negative control, indicating the extract’s effect on the mTORC signaling pathway. Although its cytotoxicity was lower than Tamoxifen, Red Bajakah Wood extract demonstrated potential as a natural-based chemopreventive agent.. Keywords: breast cancer, MCF-7, Red Bajakah Wood extract, mTORC, cytotoxicity, chemoprevention.

Abstract

Introduction: Oxidative stress and inflammation are tightly interconnected processes that drive skin aging, impaired wound healing, and inflammatory skin disorders. Excessive reactive oxygen species (ROS) disrupt skin cell homeostasis, promote cellular senescence, and sustain inflammatory signaling, ultimately compromising skin regeneration. Umbilical cord–derived mesenchymal stem cell (UC-MSC) secretome has recently emerged as a promising cell-free therapeutic approach to modulate these pathological mechanisms. Methods: A narrative literature review was conducted using PubMed, Scopus, ScienceDirect, and Google Scholar to identify studies investigating the role of UC-MSC secretome in regulating oxidative stress and inflammation in skin cells. Search terms included combinations of “UC-MSC secretome,” “extracellular vesicles,” “exosomes,” “oxidative stress,” “inflammation,” “keratinocytes,” “fibroblasts,” and “wound healing.” studies relevant to skin biology were included based on biological relevance and mechanistic insight rather than strict methodological filtering. Results: The UC-MSC secretome, composed of soluble factors and extracellular vesicles enriched with bioactive proteins, cytokines, antioxidant enzymes, and regulatory microRNAs, consistently demonstrates antioxidant and anti-inflammatory effects in keratinocytes, fibroblasts, and melanocytes. Mechanistically, it reduces intracellular and mitochondrial ROS, activates endogenous antioxidant pathways such as Nrf2 and FOXO, preserves mitochondrial function, and NF-κB and MAPK. In vitro and in vivo studies show attenuation of UV-induced photoaging, enhanced wound healing, and protection of melanocytes from oxidative and immune-mediated damage. Conclusion: Current evidence supports UC-MSC secretome as a potent regulator of oxidative stress and inflammation in skin cells. Its cell-free nature offers advantages in safety, scalability, and therapeutic flexibility, highlighting strong translational potential for dermatology and regenerative skin medicine.
