Abstract

Mikroorganisme patogen dapat tersuspensi dalam arus udara dan dapat menempuh jarak yang cukup jauh. Pencegahan terjadinya infeksi melalui udara dapat dilakukan dengan berbagai cara, salah satunya penggunaan masker. Tujuan dari penelitian ini adalah untuk mengetahui efektifitas berbagai variasi masker berdasarkan jumlah pertumbuhan bakteri pada media nutrient agar. Penelitian ini memiliki sifat eksperimental laboratorium dengan desain yang digunakan adalah post test control group design only. Dari ketiga masker uji yang digunakan (surgical masker, masker kain, dan KN95) tidak ditemukan adanya pertumbuhan bakteri pada media nutrient agar setelah diinkubasi 1x24 jam, bakteri hanya tumbuh pada kontrol yaitu hembusan tanpa masker dengan jumlah pertumbuhan 3 ± 0,7 koloni.

Abstract

Escherichia coli infection is caused by contaminated food and drinking water, or direct contact with someone who is sick or with animals carrying the bacteria. Diagnosis of diseases caused by E. coli can be done in several ways, but in general it is done clinically, then strengthened by microscopic examination and culture. Commercial media that is often used for culturing bacteria is Nutrient Agar (NA). NA media is a synthetic medium with a high content of carbohydrates and proteins that support bacterial growth. One ingredient that has a high content of protein and carbohydrates is high-protein wheat flour. The purpose of this study was to determine the potential of high-protein wheat flour as an alternative medium for the growth of E. coli bacteria. This study was a laboratory experimental study using the post test control group design only method using 16 replications and 2 treatments, namely NA medium as a control and high protein wheat flour as an alternative medium. The results showed that the average number of E. coli growth on NA media was 336.5 colonies while on alternative high-protein wheat flour media it was 15.19 colonies. The value of p = 0.000 with a 2-way significance level (α = 0.05) then p <α. So there is a significant difference between the number of E. coli bacteria colonies in NA media and alternative high protein wheat flour media. Alternative medium for high-protein wheat flour takes a very long time to grow E. coli bacteria so it is not maximally used as an alternative medium for the growth of E. coli.

Abstract

Candida is a fungal infection caused by one of the Candida species, the most common being Candida albicans. Diagnosis of candidiasis due to C. albicans can be done in various ways, but the gold standard for examination is by culture. Specimen culture is carried out by planting on mushroom media. Potato Dextrose Agar (PDA) media is the most commonly used medium for culturing C. albicans. The amount of nutrient content in the media affects the level of fertility and incubation time of the microbes grown on the media. In PDA media, the type of sugar (dextrose) used for nutrition is monosaccharide. There are many types of monosaccharides, one of which is fructose. The purpose of this study was to determine the effect of adding fructose to PDA media on the growth of C. albicans. This study was a laboratory experimental study using the post test control group design only method using 16 replications and 2 treatments, namely PDA media without the addition of fructose and PDA media with the addition of fructose. The results showed that the average number of C. albicans growth on PDA media without the addition of fructose was 376.56 colonies while on PDA media with the addition of fructose it was 289.19 colonies. The value of p = 0.000 with a 2-way significance level (α = 0.05) then p <α. So there is a significant difference between the number of C. albicans fungal colonies on PDA media with and without the addition of fructose. The addition of fructose causes the growth of C. albicans to be slower than without the addition of fructose.

Abstract

Infectious diseases due to microbes in Indonesia are mostly caused by fungal infections. One of these microbes is Trichophyton rubrum. There are several ways to diagnose dermatophytosis, but in general it is done clinically, then confirmed by microscopic examination and culture. The commercial media that is often used for culturing mushrooms is Potato Dextrose Agar (PDA). PDA media is a semi-synthetic medium with a high carbohydrate content. One ingredient that contains high carbohydrates is cornstarch. The purpose of this study was to determine the potential of cornstarch as an alternative medium for the growth of the fungus T. rubrum. This research is a laboratory experimental study with the post test control group design only method using 16 replications and 2 treatments, namely PDA media as a control and cornstarch media as an alternative medium. The results showed that the average number of T. rubrum growth on PDA media was 163.94 colonies while on alternative media cornstarch was 35.51 colonies. The value of p = 0.000 with a 2-way significance level (α = 0.05) then p <α. So there is a significant difference between the number of T. rubrum fungus colonies on PDA media and cornstarch alternative media. Cornstarch alternative media can be used as an alternative medium for the growth of T. rubrum but not as optimal as PDA media.
