Abstract

Abstrack In production effort of tiger shrimp (Pennaeus monodon) it had been found several problems that often faced of shrimp farmers in shrimp culture. One of them is the presence of vibriosis disease caused by Vibrio harveyi infection. This study aimed to examine the use of mangrove (S. alba) leaf extract by in vitro to prevent the population growth of bacteria V. harveyi. This research was conducted in the Laboratory of Pathology at Maros Research Institute for Brackish Water Aquaculture. Testing the inhibition of mangrove leaf extract to against bacterial has done in two phases namely qualitative test (inhibition zone) that used concentration were 2.000, 4.000, 6.000, 8.000, 10.000 ppm, control (without extract) and 25 ppm of antibiotic rifampicin as negative control. It the same time that quantitative test (bacterial population) that used concentration were 2.000, 4.000, 6.000, 8.000 ppm, control without extract and DMSO 10%. The results had showed mangrove leaf extract could inhibit the population growth of bacteria V. harveyi. In quality, extract categorized medium at concentration of 8,000-10,000 ppm with inhibition zone from 16.18 to 18.20 mm, and categorized low at 2000-6000 ppm concentration with inhibition zone 9.83 to 12.30 mm. While the quantity of best concentration of mangrove leaf extract in inhibiting the growth of bacteria were 8,000 ppm with total of bacteria was 0 CFU/mL after 8 hours, 6,000 ppm with total of bacteria was 0 CFU/mL after 12 hours, 4.000 ppm with total of bacteria was 4.80 x104 CFU/mL after 24 hours and 2.000 ppm with total of bacteria was 5.17 x107 CFU/mL after 24 hours.                                                                     Abstrak Dalam usaha produksi budidaya udang windu (Pennaeus monodon) ditemukan berbagai permasalahan yang sering menghambat petani dalam melaksanakan kegiatan budidaya di tambak. Salah satu diantaranya adalah adanya serangan penyakit vibriosis yang disebabkan oleh infeksi bakteri Vibrio harveyi. Penelitian ini bertujuan untuk menguji penggunaan ekstrak daun mangrove (S. alba) secara   In Vitro untuk mencegah pertumbuhan bakteri V. harveyi. Penelitian ini dilaksanakan di Laboratorium Patologi Balai Penelitian dan Pengembangan Budidaya Air Payau, Maros. Pengujian daya hambat ekstrak daun mangrove terhadap bakteri dilakukan dengan dua tahapan yakni secara kualitatif (zona hambatan) yang menggunakan konsentrasi 2.000, 4.000, 6.000, 8.000, 10.000 ppm, kontrol (tanpa pemberian ekstrak) dan antibiotik rifampisin 25 ppm serta pengujian secara kuantitatif (kepadatan bakteri) yang menggunakan konsentrasi 2.000, 4.000, 6.000, 8.000 ppm, kontrol dengan tanpa ekstrak dan DMSO 10%. Hasil penelitian menunjukkan ekstrak daun mangrove dapat menghambat pertumbuhan bakteri V. harveyi. Secara kualitas, ekstrak terkategori sedang pada konsentrasi 8.000-10.000 ppm dengan zona hambatan 16,18-18,20 mm,  dan terkategori lemah pada konsentrasi 2.000- 6.000 ppm  dengan zona hambatan 9,83-12,30 mm. Sedangkan secara kuantitas konsentrasi terbaik ekstrak daun mangrove berturut-turut dalam menghambat pertumbuhan bakteri adalah 8.000 ppm dengan total bakteri 0 CFU/mL setelah 8 jam, 6.000 ppm dengan total bakteri 0 CFU/mL setelah 12 jam, 4.000 ppm dengan total bakteri 4,80x104 CFU/mL setelah 24 jam dan 2.000 ppm dengan total bakteri 5,17x107 CFU/mL  setelah 24 jam.

Abstract

Ikan patin (Pangasius hypophthalmus) merupakan salah satu ikan komoditas air tawar di Indonesia yang banyak digemari masyarakat. Budidaya ikan patin adalah alternatif mengantisipasi pola pemijahan ikan patin yang terjadi sekali setahun di alam. Tujuan penelitian untuk mengevaluasi aspek reproduksi induk dan pertumbuhan larva ikan patin siam (P. hypophthalmus) yang diperoleh dari hasil pemijahan secara buatan menggunakan ovaprim. Penelitian dilaksanakan di Balai Perikanan Budidaya Air Tawar (BPBAT) Tatelu, Sulawesi Utara. Tahapan penelitian meliputi persiapan induk, pemijahan induk dan penetasan telur, pemeliharaan dan pengukuran pertumbuhan larva serta kualitas air. Hasil penelitian menunjukkan aspek reproduksi induk berupa fekunditas 297.500 butir, derajat pembuahan 71,4% dan derajat penetasan 8,60%. Laju pertumbuhan spesifik pada larva ikan diamati 4,80% dengan kelangsungan hidup ikan adalah 95,83%. Kualitas air teramati normal sesuai dengan kebutuhan ikan. Pembenihan ikan patin secara buatan menggunakan ovaprim dapat dilakukan sesuai dengan prosedur yang benar.‎Kata Kunci: ikan patin, ovaprim, pembenihan, reproduksiABSTRACTCatfish (Pangasius hypophthalmus) is one of the freshwater commodity fish in Indonesia that is very popular in public. Catfish farming is an alternative to anticipate catfish spawning patterns that occur once a year in nature. The aim of this study to evaluate the reproductive aspec of broodstock and larval growth of catfish (P. hypophthalmus) produced from artificial spawning using ovaprim. The research was conducted in Center of Freshwater Aquaculture (BPBAT) Tatelu, North Sulawesi. The research stages include broodstock preparation, breeding and hatching eggs, maintenance and measurement of larval growth and water quality. The results showed that the reproductive aspec of broodstock such as fecundity of 297,500 eggs, fertilization rate 71.4% and hatching rate 8.60%. The specific growth rate of fish larvae was observed to be 4.80% with fish survival rate 95.83%. The water quality was observed to be normal according to culture fish. Catfish artificially breeding using ovaprim can be conducting according to the correct procedure.Keywords: artificial breeding, catfish, ovaprim, reproductive

Abstract

Penyakit bakterial pada kegiatan budidaya ikan sudah menjadi masalah yang sering dihadapi pembudidaya. Salah satu jenis bakteri yang menyebabkan penyakit pada ikan lele (Clarias sp.) adalah bakteri Aeromonas hydrophila. Tujuan penelitian ini untuk mengevaluasi efek pemberian probiotik B. megaterium PTB 1.4 dan P. pentosaceus E2211 terhadap respons imun dan kelangsungan hidup ikan lele selama uji tantang dengan bakteri A. hyrophila. Probiotik yang digunakan adalah kombinasi 0,5% B. megaterium PTB 1.4 dan 0,5%  P. pentosaceus E2211. Pemberian probiotik dilakukan pada hari ke-0 hingga hari ke-30 melalui pakan. Uji tantang dilakukan pada hari ke-30 hingga hari ke-40. Hasil penelitian menunjukkan bahwa pemberian probiotik dapat berpengaruh terhadap respons imun dan kelangsungan hidup ikan lele dalam menekan infeksi A. hydrophila. Total eritrosit, hemoglobin, hematokrit, total leukosit, aktivitas fagositik dan aktivitas lisozim signifikan (P<0,05) teramati meningkat pada hari ke-35 dan menurun pada hari ke-40. Kelangsungan hidup ikan lele berhasil dipertahankan pada 87,62±4,83%. Total bakteri A. hydrophila meningkat pada hari ke-35 dan menurun pada hari ke-40. Pemberian probiotik B. megaterium PTB 1.4 dan P. pentosaceus E2211 pada dosis 0,5% dapat menekan pertumbuhan bakteri A. hydrophila dan meningkatkan status kesehatan ikan lele.

Abstract

Udang vaname (Litopenaeus vannamei) memiliki keunggulan tahan terhadap penyakit dan produktivitasnya tinggi. Tujuan dari penelitian ini adalah untuk menganalisis secara deskriptif karakteristik kualitas air dan performa pertumbuhan pada budidaya udang vaname secara intensif padat tebar tinggi. Metode penelitian meliputi persiapan tambak, penebaran, transfer udang, menejemen pakan, sampling pertumbuhan, manajemen kualitas air dan penyakit, panen dan pasca panen. Penelitian dilakukan di CV. Dewi Windu selama 105 hari. Hasil penelitian menunjukkan bahwa udang vaname yang dibudidayakan secara intensif memiliki karakteristik kualitas air pH pada kisaran 7-8,3, salinitas 24-37 ppt, suhu 28-32 oC, DO 3-6,2 mg/L, alkalinitas 80-140 mg/L, phosphat 0,6-5 mg/L, nitrit 0-4 mg/L, amoniak 0-0,12 mg/L, dan aminium 0-0,5 mg/L. Performa pertumbuhan yang dihasilkan yakni average body weight (ABW) pada DOC 105 adalah 21,98 gram/ekor, average daily growth (ADG) antara 0,14- 0,35 gram dan kelangsungan hidup 88,41%. Kesimpulan dari hasil penelitian ini adalah bahwa karakteristik kualitas air pada pembesaran udang vaname secara intensif selama satu siklus budidaya menunjukan karakter pola osilasi yang fluktuatif dinamis dari parameter pH, salinitas, suhu, oksigen terlarut, alkalinitas, phosphat, nitrit, amonia, dan amonium. Sedangkan performa pertumbuhan menunjukan hasil yang baik untuk sistem budidaya intensif.Kata kunci: kualitas air, pertumbuhan, sistem intesif, udang vaname.

Abstract

Diseases control in tiger shrimp farming activities needs to be done by applying biosecurity and early detection of IMNV. This study aimed to identify the application of biosecurity and IMNV detection in tiger shrimp hatchery activities at the Hatchery Surya Prima Benur. The method of collecting data on the application of biosecurity at the Surya Prima Benur Hatchery is carried out by observing field activities, interviews and participating in activities directly. IMNV detection in tiger prawn fry was carried out by PCR machine. The results of the study indicated that the application of biosecurity at the research site includes infrastructure biosecurity, personnel biosecurity, biota biosecurity, and environmental biosecurity. Detection of IMNV in 6 samples of fry showed negative IMNV results indicated by the absence of a band on the 139 bp molecule according to the positive control. The results of this study can be used as technical guidelines for the implementation of biosecurity and the IMNV inspection process in tiger shrimp hatchery activities.

Abstract

This study aimed to examine polyculture of seaweed and vaname shrimp in laboratory scale to prevent V. harveyi infection. Study was conducted with a completely randomized design by 3 treatments and 3 repetations. Application of seaweed were 800 grams (P8), 400 grams (P4) and without seaweed (control). Challenge test was carried out for 10 days with bacteria concentration of 106 cfu/ml/ind in 10 shrimp using aquarium containing 100 liters of sea water. The result showed that polyculture of seaweed and shrimp could not maintain the survival rate of shrimp significantly but could suppress the population of V. harveyi in waters. Survival rate respectively from control, P4 and P8 were 60.0%, 73.3%, 76.7%. Population of bacteria in treatment was lower than control on 10th days after seaweed stocking until challenge test. Aplication of seaweed could also accelerate shrimp recovery time from bacterial infections observed in clinical symptoms that return to normal on 9th days after challenge test. Total hemocyte count (THC) were better observed in shrimp cultured seaweed on 10th days after challenge test, and differential hemocytes count (DHC) were better on 5th days after challenge test. Polyculture of seaweed and shrimp has potential to improve shrimp health status.

Abstract

The aim of this study to observe the reproductive performance and larval development of white shrimp broodstock with adequate feed and water quality control programs. The stages of research were preparation of cultivation media, selection of broodstock, spawning, hatching and rearing of larvae. The broodstock that were spawned came from Hawaii with a weight of 35 g/ind for the male broodstock and 40 g/ind for the female broodstock. The broodstock were fed with marine worms and squid. The larvae were given natural diet in the form of Artemia salina, Thallassiosira sp. and Skeletonema costatum and artificial feed. The results showed that the reproductive performance of the broodstock was observed with the number of eggs 102.400.000 eggs from 667 broods, the average fecundity per broodstock were 143.986 eggs, the number of naupli was 75.520.000 individuals, the hatching rate was 60.9%. Larval development starts from the egg stage which undergoes embryogenesis, hatches into naupli 1-6, then becomes zoea 1-3, mysis 1-3 and post-larvae (PL). Length growth from PL1-PL8 was 3.95-8.05 mm with a survival rate of 43.95%. Water quality during rearing is still in the optimum range for the growth of broodstock and shrimp larvae.

Abstract

Teknologi bioflok merupakan sistem pemanfaatan limbah nitrogen anorganik dengan bantuan bakteri probiotik untuk efisiensi pakan dan meminimalkan limbah. Penelitian ini bertujuan untuk laju mengukur pertumbuhan dan konsumsi pakan ikan nila yang dipelihara pada sistem bioflok. Penelitian ini bertujuan untuk mengevaluasi pengaruh kepadatan berbeda terhadap pertumbuhan, laju konversi pakan, kelangsungan hidup, kualitas air dan kadar flok pada kegiatan budidaya ikan nila menggunakan sistem bioflok. Penelitian ini dilaksanakan di Balai Besar Perikanan Budidaya Air Tawar (BBPBAT). Ikan uji yang digunakan adalah benih ikan nila ukuran 20 gram/ekor. Perlakuan yang digunakan adalah kepadatan 100 ekor/m3 (P-100) dan 120 ekor.m3 (P-120). Hasil penelitian menunjukkan bahwa P-120 memiliki pertumbuhan mutlak lebih tinggi yakni 44,14 g dibanding P-100 yakni 23,74 g. Efisiensi pakan terbaik pada P-120 sebesar 99,36% dibandingkan P-100 yakni 61,99%. Feed convertion ratio (FCR) terendah pada P-120 yakni 1,07 dan tertinggi pada P-100 1,34. Nilai kelulushidupan atau survival rate (SR) yang diperoleh 97,8% pada P-100 dan 97,16% pada P-120. Kadar bioflok yang diperoleh pada kedua perlakuan berkisar antara 2-12 mL/L dan kualitas air dalam kisaran normal. Ikan nila yang dipelihara dengan bioflok pada kepadatan 120 ekor/m3 merupakan perlakuan terbaik.

Abstract

Vannamei shrimp and mangrove crabs are export commodities with high economic value. One of the areas that is central to the development of vaname shrimp and mangrove crab cultivation is in Bone Regency, South Sulawesi. The obstacle for vaname shrimp and mangrove crab cultivators is the lack of technical knowledge of environmentally friendly aquaculture. The lack of technology adaptation of cultivators is the reason for the importance of community service activities. This community service activity aims to increase public knowledge about the cultivation of vaname shrimp and mud crab. It is hoped that this activity can improve the welfare of the cultivating community, especially the cultivation of vaname shrimp and mangrove crabs. The methods used include socialization and training, monitoring of production activities and evaluation of activities. Socialization and training on technology and techniques for vaname shrimp cultivation and mud crab fattening. Monitoring is carried out starting from the pre-production, production to post-production stages. Evaluation is carried out to see the results of service activities. The results of the activity include the implementation of the activity stages, namely the formation of a group of vaname shrimp and mud crab cultivators, the implementation of training and outreach to farmers. The location for the cultivation of vaname shrimp and mud crab has been determined in accordance with the requirements for cultivation activities. Implementation of the process of assisting production activities to harvesting and marketing, followed by evaluation. The cultivator group has harvested 400 kg of vaname shrimp with a size of 130 and a mangrove crab of 75 kg with a maintenance period of 100 days. This activity has become the basis for cooperation and the beginning of the formation of synergy between academics, government and the community in Panyiwi Village, Cenrana District, Bone Regency. Community service activities carried out have increased people's income from the results of growing vaname shrimp in traditional ponds and fattening mangrove crabs.

Abstract

One of obstacles in cultivation of lobster Panulirus homarus is availability of lobster seeds in same size. Nursery with a recirculation system is an alternative technology that can be used to make seed in same size. The aim of this study to evaluate the growth performance and water quality of lobster P. homarus reared by using a recirculation system. Research design consisted of 4 treatments and 2 repetation, namely control tank (K) using a flow trough system, treatment tank (SK) equipped with a skimmer, treatment tank (F) equipped with a trickle filter, treatment tank (SKF) equipped with a skimmer and trickle filter. The results showed that the daily growth rate (LPH) and FCR were not significantly different in all treatments (P>0.05). The highest weight was obtained from the control 1.09 g and the highest length in treatment SK was 3.58±0.09 cm. Temperature was relatively stable at 25.6–27.1oC, DO 4.6–6.3 mg/L, salinity 33.4-37.4 ppt, TDS 33.5–36.9 mg/L, pH 7.6–7.9, nitrite (NO2) levels ranged from 0.03 to 4.1 mg/L. The value of nitrate (NO3) during study ranged from 0.9 to 15.5 mg/L. The value of ammonia (NH3) ranged from 0.001 to 0.049 mg/L. Filtration system (skimmer and trickle filter) treatment was able to provide optimal water quality for lobster growth.
