Abstract

Hypertension is a chronic disease that is most often found in society. Hypertension is associated with increased production of superoxide radicals and endothelial dysfunction, which causes oxidative stress characterized by increased levels of MDA. Mango mistletoe contains secondary metabolic compounds such as flavonoids which act as natural antioxidants. The use of antioxidants from manga parasites is an alternative to neutralize the increase in MDA in the body and neutralize free radicals. This study aims to determine the levels of MDA in hypertensive rats (DOCA-Salt) exposed to Mango mistletoe Methanolic Extract (EMBM) for 14 days. This study used the true experimental design method with completely randomized design on 50 male Wistar rats, differentiated into 5 treatments, namely control (-), control (+), treatment 1 dose 50 mg / KgBB, treatment 2 doses of 100 mg / KgBB and treatment 3 doses 200 mg / KgBW. Each treatment had 5 replications. Data analysis used oneway ANOVA and post hoc tests, namely JAMOVI version 1.1.9.0 which was used to determine differences in pulmonary MDA levels between groups and pulmonarymalondialdehyde (MDA) were analyzed by spectrophotometry. The results of this study indicated that pulmonary MDA levels in hypertensive rats (Doca salt) exposed to EMBM showed very significant differences between all groups. This is evidenced by the p value analysis that is <0.05, namely <0.001 so that it can be concluded that EMBM can reduce MDA levels in rats Hypertension. This effect is controlled by EMBM at a dose of 50 mg / KgBB which is the optimum dose in reducing pulmonarymalondialdehyd. Keywords : Hypertension, Mango mistletoe, MDA ABSTRAK Hipertensi merupakan penyakit kronik yang paling kerap ditemukan di masyarakat. Hipertensi berkaitan dengan peningkatan produksi radikal superoksida dan disfungsi endotel, yang menyebabkan stress oksidatif ditandai dengan meningkatnya kadar MDA. Benalu mangga mengandung senyawa metabolic sekunder seperti flavanoid yang berperan sebagai antioksidan alami. Penggunaan antioksidan dari benalu mangga menjadi salah satu alternatif untuk menetralisir terjadinya peningkatan MDA dalam tubuh dan menetralkan radikal bebas. Penelitian ini bertujuan untuk mengetahui kadar MDA pada tikus hipertensi (DOCA-Garam) yang dipapar Ekstrak Metanolik Benalu Mangga (EMBM) selama 14 hari. Penelitian ini menggunakan metode true eksperimental design dengan Rancangan Acak Lengkap pada 50 ekor tikus wistar jantan, dibedakan menjadi 5 perlakuan yaitru control (-), control (+), perlakuan 1 dosis 50 mg/KgBB, perlakuan 2 dosis 100 mg/KgBB dan perlakuan 3 dosis 200 mg/KgBB. Setiap perlakuan terdapat 5 kali ulangan. Analisis data menggunakan uji ANOVA one way dan post hoc yaitu JAMOVI dengan versi 1.1.9.0 yang digunakan untuk mengetahui perbedaan kadar MDA paru antar kelompok dan pulmonarymalondialdehyde (MDA) dianalisis dengan spektrofotometri. Hasil penelitian ini menunjukan bahwa kadar MDA paru pada tikus hipertensi (Doca- Garam) yang dipapar EMBM menunjukan perbedaan yang sangat nyata antara semua perkelompok. hal ini dibuktikan dengan analisis p value yaitu <0,05 yaitu <0,001 sehingga dapat disimpulkan bahwa EMBM dapat menurunkan kadar MDA pada tikus. Hipertensi. Efek ini dikendalikan oleh EMBM dosis 50 mg/KgBB yang merupakan dosis optimum dalam menurunkan kadar MDA Paru. Kata Kunci: Hipertensi, Benalu Mangga, MDA

Abstract

The importance of the presence of bioactive compounds that have the potential as antioxidants from various medicinal plants has become a topic that is often discussed by several researchers today. One of the higher-classification plants that potentially has antioxidant activity is the mango parasite (Dendrophthoe pentandra (L.) Miq), classified as a parasite of the Loranthaceae family. The use of specific endophytic fungi microbes from mango parasites of leaf tissue is expected to produce several bioactive compounds that act as antioxidants needed for large-scale production in a short time without excessive exploitation of nature. This primary research aims to determine the potential antioxidant activity of endophytic fungi extracts from the parasite of mango leaves Integrated Microbiology Laboratory and Halal Center of the Islamic University of Malang. Antioxidant testing using DPPH (2,2-diphenyl-1-picrylhydrazyl) free-radical capture of activity method. The results showed that KEDBM 1 (Aspergillus spp.) isolates had no potential antioxidant activity. While KEDBM 2 (Aspergillus spp.) isolate has the highest antioxidant potential with an IC50 value of 13.71 μg/ml and KEDBM 3 (Colletotrichum spp.) has a high potential as an antioxidant with IC50 of 36.29 μg/ml.Key words: antioxidant, mango balu, DPPH, endophyte

Abstract

Endophytic microbes live in plant tissues and can be extracted to obtain active compounds. One conserved endophytic microbes is endophytic mold, found in plant tissue systems, such as leaves, flowers, branches, and plant roots. The endophytic mold was obtained from the mango parasite (Dendrophthoe pentandra (L.) Miq), a parasitic plant that contains secondary metabolites. This study aimed to determine the antibacterial properties of the endophytic mold isolates extract from mango parasite leaves against Escherichia coli. This research used an experimental method with descriptive quantitative data analysis. The research began with the purification of endophytic mold isolates, production of bioactive compounds, extraction of bioactive compounds, and determination of antibacterial activity by disc diffusion method. The results obtained from this study included four isolates containing antibacterial compounds, including isolates DBM 1, DBM 2, DBM 3, and DBM 4. Extracts of the endophytic mold of mango parasite leaves showed quite good inhibition of the growth of Escherichia coli, with the results of measuring the diameter of the largest inhibition zone of 13.17 mm, which was produced by isolate DBM 1 at a concentration of 75 ppm. Based on the results of antibacterial activity tests on endophytic molds from the leaves of the mango parasite (Dendrophtoe pentandra (L.) Miq), isolates of endophytic fungi can be used as antibacterial agents in the medium renewable category.

Abstract

Rheumatoid arthritis (RA) is an autoimmune disease in which the joint area is inflamed which results in swelling, pain and often causes damage to the inside of the joint. Rheumatoid Arthritis is characterized by decreased Interleukin-6 (IL-6), so that Interleukin-6 (IL-6) inhibitors are needed. This strategy is convincing for controlling RA. Pegagan have to play role in overcoming the problem of joint inflammation. Phytosterols are compounds that are commonly found in pegagan and include antioxidants that are beneficial for health. This study aims to determine the mechanism pathway of pegagan's active compounds in the treatment of RA. This study used an in silico approach by analyzing target proteins and active compounds of pegagan through Lipinski rule of five and molecular docking analysis. This study used 3 phytosterol compounds including campesterol. β-sitosterol. and stigmasterol. The control used in this study was the tartaric acid compound whose receptor molecule was interleukin-6 (IL-6) with PDB ID: 1-ALU. The results of the Lipinski rule analysis found that all the test compounds used met the criteria and could be applied as drugs. Based on the results of molecular docking, stigmasterol has a good binding affinity value and is able to exceed the control compound with the highest binding affinity with score of -6.4 kcal/mol. Visualization results show that there is an interaction formed between the active compound and the target protein so that phytosterl compounds have a role that can be an alternative medicine in the prevention and treatment of RA.

Abstract

Tea Benalu (Scurrula atropurpurea [Bl.] Dans.) and Mango Benalu (Dendrophthoe pentandra L. miq.) containssecondary metabolites with potential antioxidant potential. Antioxidants can inhibit the oxidation process and reduce Total Cholesterol, Triglycerides, Low-Density Lipoprotein (LDL) and increase High-Density Lipoprotein (HDL). The safety of the preparations for the combination of parasites of tea and mango leaves was tested to obtain toxic effects after repeated treatment tests over some time. The purpose of this study was to determine the toxic effects of a combination of methanolic extracts of a combination of tea parasites and mangoes in female rats (Rattus norvegicus) subchronically for 28 days by looking at the results of the clinical biochemical examination on lipid profiles, namely total cholesterol levels, triglycerides, high-density lipoproteins (28%). HDL), and Low-Density Lipoprotein (LDL). The method of this study was experimental with a combination dose of methanol extract of tea parasite leaves and mango parasite leaves at a dose of 250 mg / KgBB, 500 mg / KgBB and 1000 mg / KgBB with 5x replications each treatment. The subjects used white rats (Rattus norvegicus) female Wistar strains aged 6-8 weeks with a minimum body weight of 100 grams. The treatment in this study was in the form of each dose given 5 times a week for 28 days (subchronic toxicity test) orally. Lipid profile levels were analyzed using the one-way ANOVA test. The results showed that the level of control blood lipid profile with a treatment dose of 250 mg / KgBW, 500 mg / KgBW, and 1000 mg / KgBW was not significantly different. The toxicity test of combined extracts of tea leaves and mango parasite leaves of female rat Rattus novergicus on subchronic exposure 28 days with those doses showed no difference tangible between treatments of controls. Tujuan dari penelitian ini untuk mengetahui efek toksik kombinasi ekstrak metanolik kombinasi daun benalu teh dan mangga pada tikus betina (Rattus norvegicus) secara subkronik selama 28 hari dengan melihat hasil pemeriksaan biokimia klinis pada profil lipid, yaitu kadar Cholesterol Total, Trigliserida, High Density Lipoprotein (HDL), dan Low Density Lipoprotein (LDL). Metode penelitian ini adalah eksperimental dengan perlakuan dosis kombinasi ekstrak metanol daun benalu teh dan daun benalu mangga dosis 250 mg/KgBB, 500 mg/KgBB dan 1000 mg/KgBB dengan ulangan 5x tiap perlakuan. Subjek menggunakan tikus putih (Rattus norvegicus) betina strain wistar yang berumur 6-8 minggu dengan berat badan minimal 100 gram. Perlakuan dalam penelitian ini berupa setiap dosis  diberikan 5 kali seminggu selama 28 hari (uji toksisitas subkronik) secara per-oral. Kadar profil lipid dianalisis menggunakan uji One-way ANOVA. Hasil penelitian menunjukkan bahwa kadar profil lipid darah kontrol dengan perlakuan dosis 250 mg/KgBB, 500 mg/KgBB dan 1000 mg/KgBB tidak berbeda nyata. Uji”toksisitas”ekstrak kombinasi daun benalu teh dan daun benalu mangga  tikus betina Rattus novergicus pada paparan subkronik 28 hari dengan”dosis”tersebut menunjukkan hasil tidak”beda”nyata antara perlakuan terhadap”kontrol. ”Hal ini berarti aman dan tidak menimbulkan sifat toksik pada profil lipid tikus wistar betina.Kata Kunci : Profil Lipid, Scurrula atropurpurea, Dendophthoe pentandra, Subkronik, Rattus norvegicus

Abstract

Helmintiasis is a worm disease that often occurs in cows so that it can reduce milk harvesting. One prevention that can be done  with natural ingredients in the form of papaya leaf (Carica papaya). Papaya leaves contain alkaloids, enzyme papain, saponin, flavonoid, and tannin. The purpose of this experiment was to analyze the effect of papaya leaf (Carica papaya) on endoparasites and milk harvesting. This is an experimental research used a pre-test post-test control group design on five dairy cows with one treatment, specifically 70% basal green feed and 30% papaya leaves and one positive control (Albendazole). Method of identification use the qualitative method is, native method (direct slide) and the flotation method (Flotation method). The obtained data were analyzed with a Anova test, it showed that result is (p<0,05). That are able to suppress the growth of endoparasites in cow 1, 2, 3, 4, 5 by 60%, 58%, 50%, 45% , and 34% respectively in the native method and the floating method are 75%, 63%, 56%, 45%, and 34%. Increased harvesting milk for 3 weeks is 2-4 liters.  It was conluded that there was an influence between papaya leaves (Carica papaya) on endoparasites in dairy cows (Bos taurus) as a natural antihelmintic. Salah satu pencegahan alami yang dapat dilakukan yakni dengan memberikan daun pepaya (Carica papaya) sebagai pengobatan alami. Daun pepaya merupakan bahan alami yang mengandung senyawa aktif berupa alkaloid, flavonoid, enzim papain, saponin, dan tanin yang memiliki efek antihelmintik pada sapi perah (Bos taurus). Tujuan dari penelitian ini adalah untuk menganalisa efek daun pepaya (Carica papaya) terhadap endoparasit dan hasil panen susu. Penelitian ini merupakan penelitian eksperimental, menggunakan rancangan penelitian pre test post test control group design pada 5 ekor sapi perah dengan 1 perlakuan yaitu pemberian pakan hijau basal 70% dan daun pepaya 30% dan 1 kontrol positif (Albendazole). Metode pelaksanaan penelitian menggunakan uji feses metode kualitatif yaitu metode natif (Direct slide) dan metode apung (Flotation method). Data yang diperoleh dianalisis dengan uji statistika (Anova) dengan hasil (p<0,05). Hasil penelitian ini menunjukkan bahwa sapi 1, 2, 3, 4, 5 mengalami penurunan endoparasit sebesar 60%, 58%, 50%, 45%, dan 34% secara berturut-turut pada metode natif dan metode apung sebesar 75%, 63%, 56%, 45%, dan 34%. Pemberian daun pepaya pada sapi perah selama 3 minggu dapat meningkatkan panen susu sebesar 2-4 per hari. Sehingga dapat disimpulkan bahwa terdapat pengaruh antara daun pepaya  (Carica papaya) terhadap endoparasit pada sapi perah (Bos taurus) dan hasil panen susu.Kata kunci: Helmintiasis, Daun Pepaya (Carica papaya), Endoparasit, Sapi (Bos taurus),  Hasil panen susu

Abstract

One effort to improve the quality of males is to collect semen to be used for the Artificial Insemination (AI) process. The ejaculate fraction is one of the factors that can affect the quality of fresh spermatozoa in cattle. This study aims to determine the quality of fresh spermatozoa in Simmental cattle and Limousin cattle for various fractions of ejaculate. This study used fresh semen with each cow aged 3-4 years and 5-6 years. This research method used a Completely Randomized Factorial Design with 4 repetitions. Observation of spermatozoa quality was carried out macroscopically and microscopically. The data obtained were analyzed using the F test and Least Significant Different test (BNT). The results showed that semen volume had a significant difference (p<0.05) for different types of cows, ages, and ejaculatory fractions. The color, pH, consistency and concentration of spermatozoa had significant differences (p<0.05) for different ejaculate fractions. There was no significant difference (p>0.05) for motility and abnormalities of spermatozoa for different types of cows, ages and ejaculatory fractions. It can be interpreted that different breeds of cattle and ages can have a significant effect on semen volume, while the ejaculate fraction can have a significant effect on the volume, color, pH, consistency and concentration of spermatozoa. While the different types of cattle, age, and ejaculate fraction did not have a significant effect on the motility and abnormalities of spermatozoa. Keywords : Spermatozoa Quality, Simmental Cattle, Limousin Cattle, Ejaculation Fraction.   ABSTRAK Salah satu upaya untuk meningkatkan kualitas pejantan adalah dengan mengumpulkan semen yang akan digunakan untuk proses Inseminasi Buatan (IB). Fraksi ejakulasi merupakan salah satu faktor yang dapat mempengaruhi kualitas spermatozoa segar pada sapi. Penelitian ini bertujuan untuk mengetahui kualitas spermatozoa segar pada sapi Simmental dan sapi Limousin terhadap berbagai fraksi ejakulasi. Penelitian ini menggunakan semen segar dengan masing-masing sapi berumur 3-4 tahun dan 5-6 tahun. Metode penelitian ini menggunakan Rancangan Acak Lengkap Faktorial dengan 4 kali ulangan. Pengamatan kualitas spermatozoa dilakukan secara makroskopis dan mikroskopis. Data yang diperoleh dianalisis menggunakan uji F dan uji lanjut Beda Nyata Terkecil (BNT). Hasil penelitian menunjukkan bahwa volume semen memiliki perbedaan yang nyata (p<0,05) terhadap jenis sapi, umur, dan fraksi ejakulasi yang berbeda. Warna, pH, konsistensi dan konsentrasi spermatozoa memiliki perbedaan yang nyata (p<0,05) terhadap fraksi ejakulasi yang berbeda. Motilitas dan abnormalitas spermatozoa tidak memiliki perbedaan yang nyata (p>0,05) terhadap jenis sapi, umur, dan fraksi ejakulasi yang berbeda. Dapat disimpulkan bahwa bangsa sapi dan umur berbeda dapat memberikan pengaruh yang nyata pada volume semen saja sedangkan fraksi ejakulasi dapat memberikan pengaruh yang nyata pada volume, warna, pH, konsistensi dan konsentrasi spermatozoa. Sedangkan jenis sapi, umur, dan fraksi ejakulasi yang berbeda tidak memberikan pengaruh yang nyata terhadap motilitas dan abnormalitas spermatozoa. Abstract

Ikan Mas (Cyprinus carpio L.) merupakan salah satu komoditas sektor perikanan air tawar yang terus berkembang pesat. Benih ikan mas yang unggul dalam kualitas dan kuantitas tidak lepas dari peranan kegiatan manajemen pemberian pakan dan manajemen kualitas air. Penelitian ini bertujuan untuk mengetahui korelasi antara kualitas air dan laju pertumbuhan maupun sintasan benih ikan mas pada media dengan pemberian dosis pakan yang berbeda. Penelitian dilakukan dalam laboratorium menggunakan Rancangan Acak Lengkap (RAL), dengan 3 perlakuan dan 1 kontrol. Setiap perlakuan memiliki ulangan sebanyak 3.  Hasil penelitian menunjukkan adanya hubungan antara parameter kualitas air dengan laju pertumbuhan maupun sintasan benih ikan mas. Hasil menunjukkan bahwa hubungan suhu dengan pertumbuhan bobot maupun panjang ikan maupun sintasan menunjukkan arah yang positif yaitu dengan nilai yang kuat dan sangat kuat. Sedangkan hubungan pH dengan pertumbuhan bobot dan panjang ikan maupun sintasan juga menunjukkan arah yang positif yaitu dengan nilai sedang dan kuat. Dan untuk hubungan DO dengan pertumbuhan bobot dan panjang ikan juga menunjukkan arah yang positif yaitu dengan nilai kuat dan sangat kuat.

Abstract

Red pomegranate leaves (Punica granatum L.) is one of the plants that has many benefits and potential as herbal medicine. Red pomegranate leaves have bioactivities such as alkaloids, flavonoids, phenols, saponins, tannins and terpenoids so that they can be used as an alternative antioxidant. This study aims to determine the comparison of the amount of total phenol content in red pomegranate leaves extract using maceration and digestion extraction methods. Total phenol content was determined using the Folin Ciocalteau method with a spectrophotometer UV-Vis. The research method carried out was descriptive experimental quantitative and data analysis was carried out using one way ANOVA. The results of the determination of total phenol content in gallic acid obtained a regression equation y = 0.012x + 0.545 with R2 = 0.9981. The results showed that the total phenol content in the ethanol extract of red pomegranate leaves was greater with the maceration method of 91.83 (µg GAE/g sample) compared to the digestion method of 83.27 (µg GAE/g sample). It can be concluded that the two methods used on red pomegranate leaf samples have no significant difference in total phenol content.

Abstract

Indonesians have been utilising plants as traditional raw materials, especially herbal plants, since the time of the struggle until the current independence. Mistletoe mango which belongs to the Loranthaceae family, leaves of mistletoe mango is considered a medicinal plant. Phytochemical analysis shows that these plants contain a number of secondary metabolites, such as flavonoids, saponins, tannins, alkaloids, glycosides, and inulin. Antioxidants are compounds that can prevent oxidation reactions by binding free radicals and highly reactive molecules. The DPPH method is a simple, easy, rapid and sensitive antioxidant activity test method that uses only a small sample. The aim was to determine the comparison of the antioxidant activity of mistletoe mango leaves extract (Dendrophthoe pentandra (L) Miq) with ethanol and methanol solvents, as well as analyzing the 50% inhibitin of DPPH. The results showed that the antioxidant activity of mistletoe mango leaves extract using ethanol solvent was 15,286 ppm, while using methanol solvent was 93,456 ppm. It was concluded that antioxidant activity showed a very strong category and there was a significant difference.
