Abstract

Acne is a facial skin problem that is generally experienced by 75-80% of adolescents as a result of excessive production of oil glands and sweat, inflammatory mediators. The acnecausing bacteria, Propionibacterium acnes, involves KAS III and JNK 1 proteins, so it is expected to be an effective anti-inflammatory and antibacterial strategy. Cucumber leaves are known to have biological activity as a candidate for acne medication. This study aims to detect secondary metabolites in cucumber leaves through histochemical and in silico analysis. Histochemical analysis was carried out by preparing fresh leaves through lower leaf incisions by dropping secondary metabolite detection reagents and then observing the color changes microscopically. The in silico test aims to determine the physicochemical properties, pharmacokinetic properties and interactions of the active compounds with KAS III and JNK1 as acne drug targets through molecular docking. Histochemical analysis showed that cucumber leaves positively contained secondary metabolites, namely terpenoids, flavonoids, alkaloids, tannins, and phenols. Meanwhile, it was known that isovitexin, cucurbitacin B & C had fairly good physicochemical properties, but only isovitexin and cucurbitacin C had the best pharmacokinetic properties. Based on the results of molecular docking, there are the same amino acid bonds between isovitexin and the control at the JNK1 receptor, namely the amino acids Ile:32, Val:40 and Leu:168 and the binding affinity value is low so it is predicted to be the most effective in inhibiting JNK1. Therefore, cucumber leaves open up a new potential as a candidate for natural acne preparations. Keywords: acne, cucumber, secondary metabolites, histochemical, in silico

Abstract

Tea mistletoe and mango mistletoe are plants known to possess strong antioxidant activity. The antioxidants in these plants play a role in combating free radicals, inhibiting cell oxidation, and reducing the risk of cellular damage. The active compounds present in tea and mango mistletoe have the potential to reduce malondialdehyde (MDA) levels caused by free radical exposure. This study aimed to evaluate the effect of tea and mango mistletoe (BTBM) administration on MDA levels in male Wistar rats induced with deoxycorticosterone acetate (DOCA)-salt. This study used 25 male Wistar rats, divided into five groups: normal, control (DOCA-salt), and three treatment groups receiving BTBM at doses of 50, 100, and 200 mg/kgBW. The study lasted 28 days, beginning with BTBM administration for 14 days, followed by DOCA-salt induction alongside BTBM administration from day 15 to day 28. MDA levels were measured using the Thiobarbituric Acid Reactive Substances (TBARS) method with a UV-Vis spectrophotometer at a wavelength of 531 nm. Data were statistically analyzed using one-way ANOVA, followed by a Post Hoc Tukey test to determine differences between groups. The results showed that BTBM administration at a dose of 50 mg/kgBW significantly (p<0.05) reduced MDA levels compared to the control group. Thus, it can be concluded that the combination of tea and mango mistletoe has potential as an antioxidant agent in reducing MDA levels, with 50 mg/kgBW as the optimal dose in the DOCA-salt-induced rat model.

Abstract

Foeniculum vulgare and Alyxia reinwardtii are two herbal plants frequently co-utilized by Javanese people in Indonesia to treat various diseases, as evidenced by ancient manuscripts about traditional medicine (Jamu). However, the potential of the combination of kaempferol and coumarin, the major bioactive compounds derived from each plant, to inhibit BCL-2 and BCL-XL anti-apoptotic proteins in cancer has not yet been investigated. Therefore, this study aimed to identify the inhibitory activity of kaempferol, coumarin, and their combinations on BCL-2 and BCL-XL through in silico studies. The physicochemical properties of both compounds were predicted using the SwissADME web server. Meanwhile, the docking activity prediction of these compounds on BCL-2 and BCL-XL was performed using the molecular docking method with HEX 8.0.0 CUDA. Docking visualization was performed using BIOVIA Discovery Studio 2020 Client. The results of this study indicated that kaempferol could bind the binding groove of both BCL-2 (-246.40 kcal/mol) and BCL-XL (-245.76 kcal/mol), whereas coumarin only interacted with the binding groove of BCL-XL (-160.61 kcal/mol). The combination of the two compounds exhibited a stronger interaction with BCL-2 (-248.50 kcal/mol) and BCL-XL (-260.43 kcal/mol) compared to each compound individually. Therefore, the combination of these compounds is predicted to exhibit greater anticancer potential than either kaempferol or coumarin alone. Nevertheless, further extensive studies are required to validate the findings of this study.

Abstract

SARS-CoV-2 is a new strain of coronavirus (CoV) that was identified in Wuhan in 2019. This virus is known to have the ability to reduce human immunity. Kelor (Moringa oleifera) is a potential natural resource in Indonesia, which is very abundant and contains several metabolic compounds such as phenolics, flavonoids, saponins, cytokines, and caffeoylquinic acid, which was reported to show antioxidants, antibacterial and antiviral. This study aims to predict the biological activity, physicochemical properties, toxicity, and affinity-interactions of the active compounds of M. oleifera leave. The active compounds of M. oleifera were obtained from the KNApSAcK and PubChem. Analysis of the bioactivity of the compounds using the Way2Drug Pass Online. Analysis of drug-likeness and toxicity using the Lipinski web server and pkCSM. Docking is done using Autodock vina software to analyze the interaction of the compounds with Mpro. The results indicate that the compound astragalin is the compound with the highest affinity value, namely -8.7 (kcal/mol), compared to lopinavir as a control compound with an affinity value -6.6 (kcal/mol). The types of bonds in astragalin compounds are hydrogen bonds with amino acids Glutamine 127 and Arginine 298. From these results, it is predicted that astragalin compounds have the highest potential as alternative drugs to increase body immunity against the COVID-19.
