Abstract

DNA markers and Multiplex-PCR have emerged as methods for species detection in processed meat products. The primary objective of this study is to design multiplex primer sequences for pork, rat, beef, and chicken, generating distinguishable amplicons through agarose gel electrophoresis for halal detection in processed meat products. Primer design involved utilizing mitochondrial genomic data and the NCBI-Primer BLAST site to obtain specific pork and beef primer sequences. In silico simulations, including single and multiplex-PCR, were conducted using Primer Pooler. In vitro validation encompassed Single-PCR and Multiplex-PCR annealing temperature optimization, using samples of chicken, beef, pork, and rat as well as processed meat products like meatballs, sausages, and nuggets. In vitro validation demonstrated that the halal marker gene's multiplex primer efficiently amplified the target sequence, specifically at the optimal annealing temperature of 58°C. Amplicons from beef (1,217 bp), pork (860 bp), rat (622 bp), and chicken (272 bp) primers could be distinguished on a 1.5% agarose gel. The study's results can aid in cost-effective and rapid halal testing and authentication of processed meat products, offering advantages over PCR with a single primer.

Abstract

This study aims to analyze the effect of the Project-Based Learning (PjBL) model, adapted to the VARK learning styles, on student well-being in biology learning, particularly in the context of digestive system material. The research used a quantitative quasi-experimental design with control and experimental groups. Results indicated that the average Student Well-Being score for the control group was 48.3%, while for the experimental group, it reached 51.7%. These findings highlight that implementing the VARK-based PjBL model can improve student well-being and contribute to better learning outcomes by accommodating diverse learning styles and reducing learning-related anxiety. Furthermore, the model promotes collaborative learning and meaningful engagement, leading to a more supportive and enjoyable educational experience. The study recommends the adoption of innovative teaching methods that prioritize student well-being and engagement to create a more effective and enjoyable learning environment.

Abstract

The use of plastic packaging is difficult to decompose by microbes, this raises the idea of making edible films in food packaging. Cassava peel starch can be the main component material in making this edible film, because its content can be used as an edible film making material and can reduce cassava peel waste. The aim is to understand and recognize the effectiveness of making edible film from cassava peel starch because the utilization of cassava peel as agroindustrial waste of cassava processing can be useful to reduce the amount of cassava peel waste and the use of plastic. The research method used is the literature study method. Cassava peel is the most effective material for making edible film.  Researchers should conduct further research related to the use of natural materials that can make edible films more durable than before.

Abstract

The aim of this research was to analyze the influence of practical-based learning model on the concept of regulation system of the science process skills and concept comprehension of student. Research method was a quasi-experimental with the matching pretest-posttest design. The research population consisted of all students in class XI in one of senior high school  in West Sumatra with a total population of 150 students. Sampling technique was a cluster random sampling. Sample consisted of 26 students of class experiment through practical-based learning and 26 students of class control through verification  practical learning. The instrument consist of a science process skills test and concept comprehension test. The data was analyzed using the Mann-Whitney test, Normalized gain. The results showed that there was a significant difference in science process skills and concept comprehension between the experimental and the control class. Herewith, it has been concluded that the practical-based learning model on the concept system of regulation improvement the science process skills and concept comprehension.

Abstract

The prevalence of prostate cancer cases in men is expected to continue increase. In 2040, it is estimated that there will be 2.293.818 new cases and a 1.05% increase in the death rate due to prostate cancer. Eurycoma longifolia Jack roots extract (ELE) has potential as an alternative treatment. This study aims to analyze ELE potential as an anti-prostate cancer agent through in silico assay and in vitro assays on the prostate cancer cell line (PC-3). ELE compounds were docked against Casp-3, Casp-8, HAX-1, p27, and PTEN. In vitro assays on PC-3 cells were used, namely cell viability (WST-8), ROS levels; cell cycle; and cell apoptosis (flow cytometry), PC-3 cell senescence (μ-Galactosidase staining), Casp-3; Casp-8; HAX-1; p27; and PTEN gene expression (qRT-PCR). All proteins target were successfully docked with ELE compounds and presented binding interactions. ELE is known to reduce viability, intracellular ROS levels, live cells, necrosis, and reduce HAX-1 gene expression, and inhibit the cell cycle G0/G1 phase. ELE can also increase inhibition, senescence, late and early apoptosis, and Casp-3, Casp-8, p27, and PTEN gene expression. ELE 100 μg/mL is the most effective concentration. ELE has potential as an anti-prostate cancer agent through apoptosis, cell cycle, and antioxidant pathways

Abstract

Tujuan kegiatan pengabdian ini adalah  sosialisasi tentang daur ulang limbah organik menjadi ekoenzim  untuk menjaring data tentang kepedulian masyarakat terhadap limbah organik dan kebermanfaatan ekoenzim di kalangan masyarakat. Metode yang dilakukan adalah sosialisasi tatap muka, paparan secara online dan diskusi grup komunitas di media sosial.  Jumlah mitra yang terlibat 21 orang wanita dari kelompok PKK (Pemberdayaan Kesejahteraan Keluarga) di Desa Giri Mekar, Ujung Berung, Bandung. Pada kegiatan sosialisasi tersebut para peserta melakukan praktek membuat ekoenzim. Sedangkan penyampaian paparan secara online dan media sosial dilakukan untuk mencapai target responden dari generasi muda dan masyarakat umum dengan jumlah peserta lebih banyak (524 responden).  Responden terdiri dari 82,3% wanita dan 17,7% pria. Angket penjaringan data menunjukkan bahwa masyarakat mengetahui tentang limbah organik (92,4%), ekoenzim (52,9%); kebermanfaatan ekoenzim untuk pupuk (64,1%); dan untuk lain-lain (44,8%). Hasil ini menunjukkan bahwa masih ada masyarakat yang belum melakukan daur ulang sampah organic menjadi ekoenzim. Walaupun sosialisasi ekoenzim terus dilakukan oleh para relawan melalui beberapa metoda. Promoting Ecoenzymes Based on Organic Waste to Increase Awareness in Protecting the Environment The purpose of this service activity is to socialize about recycling organic waste into eco enzymes to collect data about public awareness of organic waste and the benefits of eco enzymes among the community. The methods used are face-to-face socialization, online exposure, and community group discussions on social media. The number of partners involved was 21 women from the PKK (Family Welfare Empowerment) group in Giri Mekar Village, Ujung Berung, Bandung. In the socialization activity, the participants practiced making eco enzymes. Its use online and social media is carried out to reach the target respondents from the younger generation and the general public with a larger number of participants (524 respondents). Respondents consisted of 82.3% women and 17.7% men. The survey data shows that the public knows about organic waste (92.4%), eco enzymes (52.9%); the usefulness of eco enzymes for fertilizers (64.1%); and for others (44.8%). These results indicate that there are still people who have not recycled organic waste into eco enzymes. Although the socialization of eco enzymes continues to be carried out by volunteers in several ways.

Abstract

Background: Yogurt is a dairy product produced by fermenting milk using starter cultures such as Lactobacillus bulgaricus and Streptococcus thermophilus bacteria. The use of these bacteria is due to their role in producing lactic acid. With the involvement of these bacteria, the liquid milk coagulates and develops a distinctive acidic taste. Methods: The research method employed in this article is qualitative research using a literature review approach. Finding: Based on the study results, it was found that cow's milk undergoes an increase in protein and carbohydrate levels after being transformed into yogurt. Goat's milk experiences an increase in carbohydrate and calcium levels post-yogurt production. Meanwhile, oat milk shows an increase in ash concentration and protein levels. Conclusion: The comparative results indicate that each yogurt from different sources offers distinct nutritional advantages. Therefore, it is important to consider the body's nutritional needs before consumption.

Abstract

Wound healing often requires specialized interventions to accelerate recovery due to prolonged inflammation and limited regenerative factors. Umbilical Cord Mesenchymal Stem Cells secretome (UCMSCs) comprises various cytokines and growth factors that can promote wound healing. This study aims to analyze the potential of a secretome-based hydrogel as a wound-healing agent using BJ fibroblast cells induced by lipopolysaccharide (LPS) and scratch injury as an inflammation model. The secretome hydrogel was formulated using Carbopol, Hydroxypropyl Methylcellulose (HPMC), Hydroxyethyl Cellulose (HEC), and secretome. Cytotoxicity was conducted using the WST-8 assay, while cell migration was evaluated through a scratch assay. Tumor Necrosis Factor-???? (TNF-????), Nuclear Factor kappa-B (NF-????B), and Interleukin-8 (IL-8) gene expression were analyzed via qRT-PCR. Additionally, malondialdehyde (MDA) levels were measured for oxidative stress assessment, whereas Connective Tissue Growth Factor (CTGF) and Transforming Growth Factor-????1 (TGF-????1) levels were quantified using ELISA and colorimetric assays. The secretome hydrogel exhibited no cytotoxic effects on BJ fibroblast cells and significantly enhanced cell migration. Moreover, it reduced the TNF-????, IL-8, and NF-????B expression, indicating anti-inflammatory activity. The hydrogel also decreased MDA levels while increasing TGF-β1 and CTGF expression, suggesting antioxidant properties and enhanced tissue regeneration in the inflammatory model. The secretome-based hydrogel presents a promising therapeutic approach for promoting chronic wound healing by modulating inflammation, reducing oxidative stress, enhancing tissue regeneration, and stimulating fibroblast migration.

Abstract

In cells, many phenomena occur such as metabolism to protein synthesis, these become abstract and cause students to have difficulty understanding them. A good learning experience is needed so that students can understand the concept correctly. One learning strategy that provides a learning experience to discover and understand concepts is the Think Talk Write learning strategy. This study uses a pre-experimental design method. The subjects in this study were students of class XI science at Muhammadiyah 5 Rancaekek high school. Mastery of concepts measured in this study were C1 (memorization), C2 (understanding), C3 (application), and C4 (Analysis). Based on the results of data processing, the pretest value was 47.22 while the posttest value was 65.28. The results of the analysis using the paired sample TTest obtained from the pretest and posttest values of 0.00, this value indicates that sign <α = 0.00 <0.05.
