Abstract

Wabah Covid-19 mulai pada tahun 2020 lalu tidak hanya menyerang kesehatan masyarakat, namun juga menyebabkan krisis dari segi ekonomi. Khususnya pada lansia yang memiliki kondisi fisiologis yang lebih rentan terhadap kejadian Covid-19, maupun untuk melakukan aktivitas bernilai ekonomis. Pengembangan usaha berbasis produk pangan fungsional menjadi salah satu solusi untuk mendapat manfaat kesehatan dan ekonomis sekaligus. Pengabdian masyarakat dilakukan dengan tujuan analisis prospek kelayakan usaha bagi komunitas lansia sebagai dasar pengembangan lebih lanjut. Sebagai metode kegiatan ditempuh dalam dua tahap, yaitu pelatihan dan pendampingan dalam pembuatan fermented garlic pada komunitas lansia, serta tahap kedua yaitu analisis bisnis dengan model canvas yang terdiri dari sembilan kategori komponen bisnis. Hasilnya, tahap pertama kegiatan yang berupa pelatihan dan pendampingan pembuatan fermented garlic pada kelompok lansia di kelurahan Kalisari, kecamatan Mulyosari, Surabaya mendapat timbal balik yang baik dimana komunitas lansia berhasil membuat fermented garlic yang sesuai. Hasil analisis model usaha dengan pendekatan metode canvas, produk fermented garlic sebagai produk pangan fungsional yang memiliki keunggulan pada manfaat kesehatan, serta memiliki peluang pengembangan usaha dan keuntungan bernilai ekonomis dengan mengaplikasikan sembilan komponen yang terdapat dalam bisnis model canvas.

Abstract

ABSTRAK Latar belakang: Intensitas kegiatan industri penepungan ikan lele menghasilkan berbagai produk samping dalam jumlah besar, yaitu tulang/duri, minyak, dan kulit. Hasil-hasil samping tersebut jika tidak diolah akan menjadi limbah organik yang mengganggu lingkungan. Oleh karena itu, pada kegiatan pengabdian masyarakat ITB 2022 yang dilakukan bersama PT Rumah Inovasi Natura bertujuan untuk memanfaatkan salah satu limbah tersebut, yaitu minyak ikan. Minyak ikan lele sebagaimana minyak ikan lainnya, mempunyai aroma yang sangat tidak sedap sehingga tidak disukai, padahal kandungan senyawa aktifnya sangat bagus untuk pertumbuhan dan perkembangan kecerdasan. Tujuan: Tujuan kegiatan pengabdian masyarakat ini adalah melakukan penjernihan minyak ikan lele menggunakan bentonit agar dihasilkan minyak yang lebih jernih dan tidak berbau. Selanjutnya, minyak tersebut diolah menjadi permen yuppy sehingga lebih disukai oleh anak-anak. Metode: Kegiatan pengabdian tersebut dikemas dalam berbagai bentuk kegiatan: penyuluhan dan pelatihan kepada masyarakat di kabupaten Tulung Agung, meliputi masyarakat dari kelompok PKK, IKM (industri kecil menengah), murid SMKN 1, serta murid PAUD. Khusus untuk murid PAUD, mereka diberikan permen yuppy minyak ikan dan nasihat pentingnya makan minyak ikan untuk kepintaran. Hasil: Hasil kegiatan pengabdian masyarakat mendapatkan respon yang sangat baik dari berbagai kalangan masyarakat yang diikutsertakan pada program ini. Kesimpulan: Kegiatan pengabdian ini menghasilkan produk pangan yang berpotensi digunakan sebagai sarana suplementasi minyak ikan lele yang mengandung berbagai bioaktif yang baik untuk membantu meningkatkan perkembangan kecerdasan anak. Kata kunci: Minyak Ikan Lele, Permen, Bentonit, Tulung Agung, Kecerdasan Mental ABSTRACT Background: The intensity of the activity of catfish flour industry produces various by-products in large quantity, namely bones/thorns, oil, and skin. These by-products, if not treated, will become organic waste that disturbs the environment. Therefore, the ITB 2022 community service activity carried out with PT Rumah Inovasi Natura aims to utilize one of these wastes, namely fish oil. Catfish oil, like other fish oils, has a very unpleasant aroma that is not liked, even though the content of active compounds is very good for the growth and development of intelligence. Objective: The purpose of this community service is to purify catfish oil using bentonite so that the oil produced is clearer and odorless. Furthermore, the oil is processed into sweet candy so that it is preferred by children. Methods: The service activities are packaged in various forms of activities: counseling and training for the community in Tulung Agung district, including people from the PKK group, IKM (small and medium industries), SMKN 1 students, and PAUD students. Especially for PAUD students, they were given fish oil yuppy candy and advice on the importance of fish oil for intelligence. Results: The results of community service activities received a very good response from various groups of people who were included in this program. Conclusion: This service activity produces food products that can be used as a means of supplementing catfish oil which contains various good bioactives to help improve the development of children's intelligence. Abstract

Natural ingredients can have extraordinary potential as alternative medicines due to their accessibility and cost-effectiveness. Application of these ingredients should consider solubility and permeability, which determine the success of pharmaceutical characteristics formulation and biological activity indication. In this context, physical manipulation, specifically particle size reduction, is an effective strategy to address these issues. Previous research has explored active compounds in the stilbenoid group, found in the outer skin, hard shell, and endosperm of melinjo (Gnetum gnemon L.) seeds, functioning as antioxidant. Based on the potential as antioxidant, stilbenoid compounds, including resveratrol, contained in melinjo seed hardshell have shown significant pharmacological effects. Therefore, this research aimed to investigate the potential of melinjo seed hardshell extract as a natural antioxidant alternative by modifying the particle size through a grinding process to obtain nanoparticles. The analysis was carried out using ball milling to enhance the solubility of melinjo seed hardshell extract by increasing the saturated solubility and surface area of the particles. The results showed that the total phenol content and the antioxidant power increased significantly (p < 0.05) after ball milling. Melinjo seed hardshell nanoextract is reported herein as a promising source of natural antioxidant from local Indonesian plants.

Abstract

Silymarin is a natural compound from Silybum marianum which has potential to treat various liver diseases, but it has low bioavailability. This study aimed to develop silymarin-carbon dot nanoemulsion, analyze its parameters, and its interaction with various cell lines. Nanoemulsion oil phase was prepared from castor oil: cremophore RH40 (surfactant): PEG400 (co-surfactant) with a ratio 1:8:1. Mild stirring method was applied for nanoemulsion preparation followed by characterization including, visual appearance, droplet size, polydispersity index, zeta potential, entrapment efficiency, and physicochemical stability under normal storage condition for 2 weeks. Toxicity determined by IC50 using 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium] method using NIH/3T3, MCF-7, and MDA-MB cells. Nanoemulsion penetration ability was compared to silymarin solution on MCF-7 cell using confocal microscope after 3 and 6-hours incubation. The results of droplet measurement, zeta potential, polydispersity index, and entrapment efficiency was 27,95 ± 4,31 nm, 0,296 ± 0,09, -11,81 mV, and 91,92 ± 0,04%, respectively. According to stability data, the nanoemulsion showed no significant changes in the entrapment efficiency as well as the physical characteristic. The IC50 value of silymarin nanoemulsion and solution was 30,67µg/ml and 45,40µg/ml for MCF-7, and was 37.50 µg/ml and 39.14 µg/ml for MDA-MB. The nanoemulsion containing silymarin is potential to penetrate MCF-7 cell line at concentration of 12.5 µg/ml both after three and six-hours incubation. The developed nanoemulsion is a promising system to improve the therapeutic value of sylimarin.

Abstract

Mulut merupakan habitat bagi sekitar 700 spesies bakteri yang dapat memicu gangguan jika kebersihan mulut tidak terjaga. Salah satu upaya menjaga kebersihan mulut adalah dengan menggunakan mouthwash atau obat kumur. Saat ini masih banyak beredar obat kumur di pasaran yang mengandung alkohol, padahal penggunaan alkohol dalam obat kumur dapat menimbulkan beberapa efek samping setelah pemakaian. Di sisi lain, terdapat bahan alam yang memiliki aktivitas antibakteri terhadap bakteri mulut seperti Salvadora persica dan Syzygium aromaticum. Minyak cengkeh merupakan minyak atsiri yang kurang stabil, sehingga pada penelitian ini dikembangkan formula nanoemulsi minyak cengkeh dengan penambahan ekstrak siwak. Formula akhir nanoemulsi diuji aktivitas antibakterinya terhadap bakteri dari saliva mencit galur BALB/c melalui penentuan konsentrasi hambat minimum (KHM). Pada formula akhir, diperoleh nanoemulsi dengan ukuran 22,4 nm dengan indeks polidispersitas 0,385, zeta potensial -2,1 mV,dan pH 7,15. Hasil uji aktivitas antibakteri menunjukkan bahwa nanoemulsi yang ditambahkan dan tidak ditambahkan ekstrak siwak memiliki KHM terhadap bakteri saliva mencit sebesar 6,25 mg/ml. Hal ini disebabkan karena KHM dari minyak cengkeh adalah 6,25 mg/mL, sedangkan dari ekstrak siwak adalah 12,5 mg/ml sehingga jumlah ekstrak dalam formula tidak dapat menurunkan KHM nanoemulsi. Hasil uji stabilitas menunjukkan bahwa nanoemulsi stabil terhadap 2 siklus beku-cair dan masih dapat diterima pada penyimpanan selama 4 minggu dalam suhu 4°C. Oleh karena itu, dapat dikatakan bahwa formula yang dikembangkan memiliki aktivitas antibakteri terhadap bakteri saliva mencit BALB/c dan memiliki stabilitas yang cukup baik selama 4 minggu dengan suhu penyimpanan 4°C.

Abstract

Zebrafish serve as a pivotal model for bioimaging and toxicity assessments; however, the toxicity of banana peel-derived carbon dots in zebrafish has not been previously reported. The aim of this study was to assess the toxicity of carbon dots derived from banana peel in zebrafish, focusing on two types prepared through hydrothermal and pyrolysis methods. Banana peels were synthesized using hydrothermal and pyrolysis techniques and then compared for characteristics, bioimaging ability, and toxicity in zebrafish as an animal model. Pyrolysis-derived banana peel and hydrothermal-derived banana peel showed blue emission under ultraviolet light, indicating excitation-dependent behavior. To test their potential application for bioimaging, a soaking method was used using zebrafish that showed fluorescence intensity in the eyes, abdomen, and tail of zebrafish. Toxicity comparison showed that pyrolysis-derived banana peel had lower toxicity with 50% lethal concentrations (LC50) of 1707.3 ppm than hydrothermal-derived banana peel (LC50 993 ppm) in zebrafish. Both types of carbon dots showed significant differences (p<0.05) in hatching rates at 96 and 120 hours of exposure. Of the two methods for carbon dot synthesis from banana peel, the pyrolysis method had a higher toxicity threshold than the hydrothermal method, as indicated by the LC50 value and the number of zebrafish embryos that died, hatched delayed, and experienced malformation during their development.

Abstract

Current anti-breast cancer drugs have limited efficacy and often cause severe side effects, highlighting the need for bioactive agents that could overcome these limitations. Curcumin, a phenolic compound from Curcuma domestica, has antineoplastic activity but has low solubility in physiological media, while moringa oil is a key component of the oil-phase nanocarrier and also possesses anticancer properties. The aim of this study was to develop a moringa oil-based nanocarrier system containing curcumin and to analyze its anticancer effects on MDA-MB-231 cell lines, focusing on the underlying mechanisms involving B-cell lymphoma 2 (Bcl-2) and Bcl-2-associated X (Bax) proteins. Additionally, the study investigated the side effects of the nanocarrier system following acute administration in animals. The anticancer effects were evaluated in vitro using MDA-MB-231 cell lines, while the acute toxicity assessment was conducted in healthy female Wistar rats. The nanocarrier system was formulated using moringa oil, Cremophor RH40, and PEG 400. Its cytotoxicity against MDA-MB-231 cells was assessed using the 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay. DNA fragmentation, apoptosis, and the expression of Bax and Bcl-2 proteins were analyzed via terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assays, flow cytometry, and western blotting. Acute toxicity was further evaluated in female Wistar rats. The results demonstrated that the moringa oil-based nanocarrier system containing curcumin inhibited cell proliferation and induced apoptosis in MDA-MB-231 cells. Curcumin suppressed tumorigenesis by modulating Bcl-2 and Bax protein expression. Our data indicated that the combination of curcumin and moringa oil in a nanocarrier system had greater anticancer potential than either component alone. Moreover, administration of the nanocarrier system did not result in any clinically significant changes in body weight, behavior, or organ weight indicative of toxicological effects. No treatment-related histopathological abnormalities were observed at terminal necropsy. In conclusion, this novel combination of curcumin and moringa in nanocarrier system has better anticancer potential; nevertheless, further studies are needed to confirm this in cancer animal models.

Abstract

Introduction: The use of essential oils in aromatherapy is widespread. However, few studies have explored the effects of smoke from the evaporation of commonly used essential oils. While essential oils are promoted for various benefits, prolonged exposure to inhaled particles from essential oil smoke may pose potential health risks. This study aimed to examine the effects of essential oil vapors on mice. Methods: This was an experimental study investigating the effects of different treatments on lung immunohistopathology, endothelial nitric oxide synthase (eNOS) expression, serum nitric oxide (NO) levels, and serum surfactant protein-D (SP-D) as an inflammation marker in mice. A total of 40 adult male Mus musculus mice (25–30 g) were randomly divided into four groups. Inflammation models were established by exposing the mice to a gas mixture containing vegetable glycerin, propylene glycol, and vitamin E acetate solution. Following inflammation induction, the mice received seven-day interventions with 0.9% NaCl solution, Eucalyptus globulus essential oil (EgEO), and citronella essential oil (CtEO), alongside an untreated inflammatory group. Results: The CtEO group intervention showed significant increases in eNOS expression (P=0.001) but no significant increase in NO compared to the other groups. The correlation analysis of eNOS expression in lung cells, serum NO, and SP-D levels was not significant, P>0.05 (p=0.160; p=0.115; p=0.234). Abstract

Awareness of the importance of cultural preservation should be a fundamental concern for every Indonesian citizen, particularly in the face of the rapid cultural influences brought about by globalization and advancements in internet technology. This is especially relevant for the people of Riau Province, which is rich in historical heritage and cultural diversity. The development of SITARI (Sistem Informasi Cagar Budaya Riau) represents a strategic initiative aimed at preserving the cultural assets of Riau Province through the facilitation of cultural heritage reporting, heritage data collection, and 360-degree virtual tours.During the development process, a Focus Group Discussion (FGD) was conducted involving representatives from relevant government agencies across the 12 regencies and municipalities in Riau Province. The objective was to identify existing challenges and gather input regarding system development needs. In parallel, a public survey was carried out to capture community perspectives on the difficulties faced in reporting suspected cultural heritage objects. The system was developed using the waterfall model approach. Evaluation results indicate that the SITARI application operates effectively and performs optimally without errors. It is deemed relevant in supporting cultural literacy, heritage preservation, and the promotion of tourism in Riau. The user interface and layout received positive feedback, indicating potential for further development. Notably, the 360-degree Virtual Tour feature was highly appreciated. The application also adheres to OWASP security standards, with minor low-risk vulnerabilities identified for future improvement.
