Abstract

One of the efforts to improve the quality and the conservation of local fruits like durian requires characteristic certainty of durian fruits according to the needs of plant breeders and tastes of consumers. Some durian germplasm at Hortimart Agro Center in Central Java have been successfully characterized. There are 43 accessions with 18 characters covering 64 character states of fruit. The dendrogram showed that those 43 accessions split into 2 groups. Durian Kartomarmo separated from the other 42 accessions. Durian Kartosumarmo has a unique characteristic on its fruit, which has an apex with irregular shape (mammiform). Some durians which have good taste, such as durian Bismo, Cokro, Janoko, and Petruk were grouped together with durian Monthong. The coeffient similarity in 43 accessions of the durian fruit is between 0.44-0.86. The diversity of accessions based on the fruit morphology showed that these 43 accessions were different and not a duplicate of different accessions. It shows that each of accession should be categorized as a different cultivar since there are many other characteristics that have not been observed.

Abstract

issue cultures that have passed callus phase can induce somaclonal variation, of which the intensity was influenced by the addition of selective agents to culture media. Somaclonal variations of peanut plant obtained from in-vitro cultured and in-vitro selected somatic embryos using PEG was not yet understood. The objectives of this research were 1) to identify the qualitative variants of peanut plant var. Kelinci obtained from in-vitro cultured and in-vitro selected somatic embryos using PEG, 2) to estimate the factors that control the qualitative variants. The non-selected and the selected variant somatic embryos of peanut were germinated and cultured. From fertile R0 lines, sufficient number of R1 and R2 progenies were grown for evaluation. The results showed that phenotypic variations of qualitative characters can be observed among R0, R1 and R2 generations of somaclonal lines. Variant phenotypes of qualitative characters can be observed, these include wide branching, excessive branching, leaf variegation, leaflet number abnormality, leaf pointed tip, ‘rosette’ leaf, complete sterility and partial sterility. The data indicated that wide branching, excessive branching, leaflet number abnormality, partial sterility and complete sterility were genetically controlled, while variant phenotypes of ‘rosette‘ leaf, leaf variegation, and leaf pointed tip were epigenetically controlled. Key words: somaclonal variant, qualitative characters, in vitro selection, in vitro culture

Abstract

Feronia limonia (L.) Swingle better known as wood apple is an identity plant of Rembang Regency, Central Java Province, Indonesia. However, the population is very limited. In order to identify the conservation efforts, this ethnobotany study was aimed to determine traditional knowledge on the existence, economic value, utility, breeding technique and conservation of wood apple. The survey was conducted on a total of 102 local people from 10 sub-districts which were determined by proportionally stratified random sampling. Data were collected by questionnaires and individual interviews and then were analyzed descriptively. The results showed that all of respondents knew the figure of wood apple, but only 79.50% understood its status as an identity plant. Most of respondents (96%) stated that the fruits have low economic value, 80.39% respondents have utilized ripe fruit as ingredients of beverages and stems as firewood. This utilization was only equal to 29.72% compared to the potential usability based on research results. It was concluded that the traditional knowledge about the existence, economic value, and breeding technique were good and on the contrary, the knowledge about the plants usability was bad. The results will form a basic knowledge for in-depth studies to provide a government policy of Rembang Regency in order to develop an efficient strategy of management and conservation of wood apple as plant identity.

Abstract

Carica pubescens Lenne & K. Koch, an endemic species in Dieng mountains, must be conserved. The in vitro conservation has been developed, but sub-culture period needs to be extended. This study aimed to obtain a more efficient in vitro conservation protocol of C. pubescens. The research was carried out experimentally by using a completely randomized factorial design with three factors, namely decreasing in storage medium concentration (75% and 50% of MS medium), temperature (4oC and 8C), and irradiation duration (8 hours/day and 16 hours/day). Shoots were kept in the storage medium for 6, 9 and 12 months, then their viability were tested by growing them in the regeneration medium. Data were analyzed by Analyses of Variance and Least Significant Difference Test. The results showed that medium concentration of 50% of MS, the temperature of 8 C, and 16 hours/day of irradiation were able to suppress the C. pubescens growth in vitro storage for six months and could maintain its viability in the regeneration medium. Based on these results the medium concentration of 50% of MS, the temperature of 8 C and 16 hours/day irradiation can be used for in vitro conservation of C. pubescens without sub-culture for six months. Abstract

Carica pubescens Lenne & K. Koch is a rare species that need to be conserved. The research aim was to develop a slow growth method of in vitro conservation medium through determining some effects of nutrition decreasing availability in the conservation medium on growth and survival of explants. Establishing epicotyls reached from in vitro seed germination was grown on diluted basic medium of 75% MS (Murashige and Skoog), 50% MS, 25% MS, while osmoregulator compound of mannitol and sorbitol was added to the full MS medium in several concentrations. The treatments were arranged in a completely randomized design with three replications. The epicotyls were grown at storage medium for 12 and 16 weeks, then their survival were evaluated at regeneration medium and rooting medium. The diluted basic medium and osmoregulator addition were evaluated for its influence in retarding the culture growth in terms of improved survival over the period of 16 weeks. Data analyzed by one way analysis of variance and Duncans multiple range test. The results showed that the decreasing of nutrition concentration suppressed the growth of the epicotyls until 16 weeks after conservation. Epicotyls taken from 16 weeks after conservation could grow on the regeneration medium. The best survival was shown by the 75% MS, 50% MS and supplementing of 20 g/l mannitol treatments. Based on these results, 50% MS medium is recommended for storage C. pubescens for 16 weeks with no sub-culture. Eksplan berupa epikotil kecambah in vitro.
