Abstract

The widespread use of antibiotics in the livestock industry serves both as a preventive measure against diseases and as a feed additive. However, excessive antibiotic use has led to antibiotic resistance, where bacterial exposure to antibiotics renders them resistant to one or more antibiotic agents. This study investigates the presence of antibiotic resistance in Escherichia coli isolated from chicken meat sold in traditional markets in Yogyakarta. An experimental method was employed, using ECB and EMBA media for bacterial isolation. Gram staining was performed to characterize the morphology of E. coli, followed by molecular testing using the uspA gene. Antibiotic susceptibility testing was conducted using the Kirby-Bauer disk diffusion method. A total of 10 chicken meat samples were collected from five traditional markets in Yogyakarta. The antibiotics tested included ampicillin, ciprofloxacin, and chloramphenicol. The results indicated resistance in all 10 isolates. Sample codes G1 and G2 were resistant to all three antibiotics. Samples P1, P2, and L1 exhibited resistance to ampicillin and chloramphenicol, with intermediate resistance to ciprofloxacin. Sample L2 showed resistance to ampicillin, intermediate resistance to ciprofloxacin, and sensitivity to chloramphenicol. Sample B1 exhibited resistance to all antibiotics tested. Sample B2 was resistant to ampicillin and ciprofloxacin, with intermediate resistance to chloramphenicol. Samples K1 and K2 were resistant to ampicillin and chloramphenicol but sensitive to ciprofloxacin. These findings highlight the growing concern of antibiotic resistance in bacterial strains isolated from chicken meat, emphasizing the need for stricter regulations on antibiotic use in the poultry industry.

Abstract

Parasit adalah organisme yang hidup pada tubuh organisme lain dan umumnya menimbulkan dampak negatif pada organisme yang ditempatinya. Salah satu parasit yang kerap menyerang ikan lele dari golongan protozoa adalah Trichodina sp. yang menyebabkan gatal. Pengendalian penyakit Trichodinasis ini dapat dilakukan dengan menggunakan obat kimia, namun cara ini dapat mencemari lingkungan dan produk makanan. Oleh karena itu, penelitian ini dilakukan untuk penanggulangan parasit menggunakan bahan alami diantaranya dengan menggunakan infusa daun jambu biji. Daun jambu biji mengandung tanin, alkaloid, fenol dan saponin sebagai antiparasit. Penelitian ini bertujuan untuk mengetahui pengaruh infusa daun jambu biji terhadap penurunan prevalensi ikan lele yang terserang parasit Trichodina sp. dan intensitas parasit Trichodina sp. Variabel bebas dari penelitian ini adalah konsentrasi infusa daun jambu biji (Psidium guajava L.) yang digunakan, yaitu 2,5 mL/L; 5 mL/L; 10 mL/L dan kontrol (tanpa infusa daun jambu biji). Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 4 perlakuan 5 kali ulangan sehingga diperoleh 20 unit percobaan. Tahapan penelitian meliputi infundasi daun jambu biji, penginfeksian parasit ke ikan, perendaman, dan pengamatan. Data yang diperoleh dianalisis menggunakan Analisis Varians (ANOVA) dan dilanjutkan uji Duncan jika terdapat beda nyata. Data ini dikomputerisasi menggunakan program SPSS. Berdasarkan hasil penelitian, pemberian infusa daun jambu biji memiliki pengaruh nyata terhadap prevalensi ikan lele yang terserang parasit Trichodina sp. dan intensitas parasit Trichodina sp., konsentrasi infusa daun jambu biji yang berbeda paling nyata adalah 10 mL/L.

Abstract

Karakteristik kombucha terbagi menjadi karakteristik kimiawi dan biologi. Kombucha merupakan minuman fermentasi yang kaya akan kandungan antioksidan. Mentimun dipercaya bermanfaat sebagai sumber antioksidan yang dapat mencegah radikal bebas. Penelitian tentang aktivitas antioksidan kombucha dengan berbagai substrat sudah banyak dilakukan, tetapi penelitian tentang aktivitas antioksidan kombucha mesokarp mentimun (Cucumis sativus L.) belum pernah dilakukan. Oleh karena itu, penelitian ini dilakukan untuk mengetahui karakteristik serta aktivitas antioksidan kombucha mesokarp mentimun pada hari ke-6 dan 12. Fermentasi dilakukan selama 6 dan 12 hari, kemudian masing-masing sampel diuji karakteristiknya, seperti pH, berat kering scoby (Symbiotic Culture of Bacteria and Yeast), kadar gula pereduksi, serta total asam. Aktivitas antioksidan kombucha mesokarp mentimun diuji melalui metode DPPH, kemudian dihitung nilai IC50 nya. Hasil menunjukkan pH dan kadar gula pereduksi mengalami penurunan, sedangkan berat kering scoby dan total asam mengalami peningkatan. Aktivitas antioksidan kombucha mesokarp mentimun menunjukkan nilai IC50 hari ke-6 sebesar 3,5 ppm dan hari ke-12 sebesar 4,4 ppm. Kombucha mesokarp mentimun memiliki antioksidan sangat kuat yang dapat dimanfaatkan sebagai pengobatan alternatif.

Abstract

Fragaria x ananassa or strawberry has significant economic value worldwide. However, its cultivation faces various challenges. One example was fungal diseases, such as anthracnose, caused by Colletotrichum acutatum. Disease management generally relies heavily on synthetic fungicides, raising concerns about environmental sustainability and human health risks. Therefore, more environmentally friendly alternatives are needed, such as the use of biological agents. This study aims to assess the potential of endophytic fungi from strawberry plants as natural biocontrol agents against Colletotrichum acutatum. The study began with the isolation of endophytic fungi from strawberry tissues and their characterization morphologically. The antagonistic activity of endophytic fungi against colletotrichum acutatum was evaluated using three methods: a dual culture assay to assess direct inhibition, a cell-free filtrate assay to analyze the effect of fungal metabolites, and a volatile organic compound (VOC) assay to inhibit the growth of pathogenic pathogens. In this study, 20 fungal isolates were successfully isolated. A total of 15 showed antagonism against Colletotrichum acutatum. Isolate A1 showed potent inhibition in the dual culture test by effectively antagonizing Colletotrichum acutatum. Isolate D1 produced extracellular compounds that reduced the growth of Colletotrichum acutatum by 52.28% ± 3.21% in the cell-free filtrate assay. At the same time, isolate D3 proved most effective in the VOC assay, suppressing the growth of pathogenic colonies by 71.35% ± 3.37%. Thus, endophytic fungi, especially isolates A1, D1, and D3, have the potential to serve as an environmentally friendly alternative to combat anthracnose disease on strawberries.

Abstract

This study aims to isolate and determine the antagonistic activity of lime's endophytic mushrooms on Candida albicans. Antagonism tests were performed in vitro with dual culture methods. The study obtained 14 isolates that were successfully isolated from the stem, roots, leaves, and peels of lime fruit. A total of 8 isolates could inhibit Candida albicans through antibiosis mechanisms and healthy competition. The isolates with the highest inhibition percentage were KD2 and MA1 (42.85%).

Abstract

Aeromonas hydrophila bacteria can cause Motile Aeromonas Septicemia (MAS), a disease that impacts freshwater fish. The turmeric rhizome contains numerous bioactive compounds that act as antibacterials and might be utilized to inhibit A. hydrophila. This research aimed to identify the phytochemicals in turmeric (Curcuma domestica) infusion, evaluate its antibacterial activity against A. hydrophila, and define the toxicity effect of turmeric rhizome infusion. The agar dilution method was used to measure antibacterial activity, whereas the Brine Shrimp Lethality Test (BSLT) was used to assess toxicity. The research confirmed the presence of phytochemicals such as tannins, alkaloids, flavonoids, phenols, and saponins in a turmeric rhizome infusion. According to the findings, the infusion from turmeric rhizome effectively inhibited the growth of A. hydrophila at concentrations between 750-1000 ppm. In toxicity tests, the LC50 value of the turmeric rhizome infusion against shrimp larvae was 381.18 ppm. Hence, the turmeric rhizome infusion has the potential to be further developed for the prevention and treatment of fish infected with A. hydrophila in freshwater fish farming.

Abstract

Aeromonas hydrophila is an infectious agent that triggers Motile Aeromonas Septicemia (MAS) in freshwater fish, including catfish, leading to significant economic losses in aquaculture. This study aims to evaluate the antibacterial activity of Mango Bacang (Mangifera foetida L.) leaf infusion against A. hydrophila and its effect on catfish mortality. The research methods included the preparation of the infusion, phytochemical screening, antibacterial activity testing, and assessing the infusion's impact on catfish mortality. Phytochemical screening revealed that Mango Bacang leaf infusion contains alkaloids, flavonoids, phenols, tannins, and saponins, contributing to its antibacterial properties. The well-diffusion technique used to examine antibacterial effects demonstrated that the infusion at concentrations of 5–100% produced significant inhibition zones against A. hydrophila, with strong activity observed at a 5% concentration (14.38 ± 1.67 mm). However, toxicity testing on catfish indicated increased mortality with higher infusion concentrations, reaching 50% at 5% concentration. These findings suggest that Mango Bacang leaf infusion has potential as a natural antibacterial agent to control A. hydrophila, but its use requires precise dosage management to ensure safety for catfish. Further studies are necessary to optimize effective and environmentally friendly concentrations and explore applications on a broader scale.

Abstract

Peternak lebah kelulut di Terengganu menghadapi permasalahan dalam memanfaatkan media digital secara profesional, di mana mereka selama ini hanya mengandalkan media sosial seperti TikTok tanpa memiliki platform yang terstruktur dan kredibel untuk promosi produk. Tujuan pelatihan ini adalah untuk meningkatkan kapasitas digital para peternak dengan membekali mereka keterampilan dasar dalam membuat dan mengelola website menggunakan Google Sites sebagai media pemasaran digital yang lebih efektif. Pelatihan dilaksanakan dalam tiga tahapan, yaitu persiapan materi dan identifikasi kebutuhan peserta, pelaksanaan pelatihan secara praktikal melalui demonstrasi dan pendampingan langsung, serta evaluasi melalui pre-test dan post-test. Hasil menunjukkan peningkatan signifikan dalam pemahaman dan keterampilan peserta, di mana seluruh peserta yang sebelumnya belum memahami cara membuat website kini mampu membuat, mengelola, dan mengintegrasikan website mereka secara mandiri dengan media sosial dan marketplace. Efektivitas pelatihan ini diukur melalui peningkatan skor post-test dibandingkan pre-test serta keberhasilan peserta dalam menghasilkan website sebagai produk akhir. Dengan demikian, pelatihan berbasis praktik langsung terbukti efektif dalam meningkatkan literasi digital peternak dan memperluas jangkauan serta kredibilitas pemasaran produk mereka.

Abstract

Getih-getihan (Rivina humilis) merupakan tanaman liar yang sering ditemukan tumbuh di sekitar pekarangan rumah atau lahan kosong. Daun dari tanaman getih-getihan diketahui mengandung berbagai senyawa aktif, salah satunya adalah flavonoid yang memiliki sifat antiinflamasi sehingga sangat berpotensi dalam mempercepat proses penyembuhan luka. Penelitian ini bertujuan untuk mengetahui efektivitas berbagai konsentrasi nanogel ekstrak daun getih-getihan dalam membantu proses penyembuhan luka robek pada hewan uji tikus (Rattus norvegicus). Metode dalam penelitian ini  yang digunakan untuk menguji efektivitas konsentrasi nanogel ekstrak daun getih-getihan yaitu dengan menggunakan analisis eksperimental. Analisis data dilakukan dengan pengujian ANOVA untuk melihat perbedaan yang signifikan, dan kemudian dilanjutkan dengan uji lanjutan dengan analisis Duncan-Multiple Range Test (DMRT) untuk melihat kelompok perlakuan yang paling efektif. Hasil pengujian efektivitas nanogel ekstrak daun getih-getihan dengan konsentrasi 2 %, 4%, dan 6% b/v pada tikus (Rattus norvegicus) menunjukkan nanogel yang paling efektif dalam penyembuhan luka adalah pada konsentrasi 6%. Karena pada hari ke 6 menunjikan bahwa kondisi bekas luka sayatan sudah tertutup sempurna. Dengan demikian, dapat disimpulkan bahwa nanogel ekstrak daun getih-getihan dengan konsentrasi 6% merupakan formulasi paling efektif dalam mempercepat penyembuhan luka robek. Hasil penelitian ini menunjukkan potensi besar tanaman liar untuk dapat dikembangkan menjadi produk obat yang berbasis herbal sehingga aman dan ramah lingkungan.
