Abstract

Kombucha adalah minuman fermentasi yang terbuat dari teh manis dan difermentasi oleh koloni bakteri dan ragi, dikenal sebagai SCOBY (Symbiotic Culture of Bacteria and Yeast). Minuman ini dikenal memiliki berbagai manfaat kesehatan, seperti meningkatkan sistem pencernaan dan kekebalan tubuh. Dusun Gamping Lor, Sleman, Yogyakarta, merupakan pemukiman padat penduduk yang sebagian besar anggotanya adalah ibu rumah tangga. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk memberdayakan ibu-ibu rumah tangga di Dusun Gamping Lor melalui pelatihan pembuatan kombucha. Pelatihan ini dirancang dengan kombinasi teori dan praktik, meliputi pengenalan kombucha, proses fermentasi, dan teknik pemasaran. Hasil pelatihan menunjukkan bahwa seluruh peserta (100%) merasa pelatihan ini bermanfaat, memberikan wawasan baru dalam pengolahan makanan, dan sebagian besar peserta (80%) tertarik untuk memperdalam pengetahuan mereka tentang kombucha. Selain itu, 75% peserta menunjukkan minat untuk menjual produk kombucha. Pelatihan ini berhasil meningkatkan pengetahuan dan keterampilan peserta, serta memotivasi mereka untuk mengembangkan potensi ekonomi keluarga melalui produksi kombucha.

Abstract

Pemberdayaan masyarakat merupakan strategi penting dalam meningkatkan kesejahteraan dan kemandirian komunitas. Desa Ambarketawang, Kecamatan Gamping, Kabupaten Sleman, Yogyakarta, memiliki potensi besar dalam produksi kelapa yang dapat dimanfaatkan untuk pembuatan Nata de Coco. Program ini bertujuan memberikan pelatihan kepada anggota PKK Desa Ambarketawang dalam proses pembuatan Nata de Coco sebagai alternatif penghasilan tambahan dan optimalisasi pemanfaatan limbah air kelapa. Metode pelaksanaan meliputi sosialisasi, pelatihan teori dan praktik, serta pendampingan dan monitoring. Hasil program menunjukkan peningkatan keterampilan dan pengetahuan anggota PKK, pemanfaatan sumber daya lokal, serta peningkatan kesadaran akan kesehatan. Hasil kuesioner menunjukkan antusiasme peserta yang tinggi dalam mengembangkan produk berbasis Nata de Coco. Rekomendasi mencakup peningkatan pelatihan lanjutan, kolaborasi dengan pihak eksternal, dan pengembangan riset serta inovasi untuk diversifikasi produk. Program ini membuktikan bahwa PKK dapat berperan penting dalam meningkatkan kesejahteraan dan kemandirian ekonomi masyarakat desa.

Abstract

 AbstrakObesitas disebabkan adanya kelebihan kalori dalam tubuh. Penurunan obesitas dapat dilakukan dengan cara menghambat penyerapan lemak sebagai sumber utama kelebihan kalori. Salah satu cara yang dapat dilakukan untuk menghambat penyerapan lemak adalah dengan menghambat aktivitas lipase pankreas. Penelitian ini dilakukan untuk menentukan aktivitas inhibitor lipase ekstrak metanol daun mangga arum manis (Mangifera indica L. cv Arum manis) dan kweni (Mangifera odorata Griff.). Tahapan penelitian diawali dengan ekstraksi menggunakan metode maserasi lalu diikuti dengan uji kadar total fenol, kadar flavonoid, dan uji aktivitas inhibitor lipase ekstrak daun mangga arum manis serta kweni. Hasil uji kadar total fenol menunjukkan total fenol pada ekstrak daun mangga arum manis dan kweni berturut-turut yaitu 246,94 mg GAE/g dan 176,11 mg GAE/g. Kadar flavonoid daun mangga arum manis, yaitu 129,95 mg.QE/g, sedangkan daun mangga kweni yaitu 26,50 mg.QE/g. Masing-masing ekstrak diuji kemampuannya dalam menghambat lipase pankreas secara in vitro. Nilai IC50 ekstrak daun mangga arum manis dan kweni, yaitu 61,55 µg/mL dan 79,98 µg/mL. Nilai tersebut lebih tinggi dibandingkan kontrol positif, yaitu orlistat (18,01 µg/mL). Dengan demikian, ekstrak daun mangga arum manis dan kweni dapat menghambat aktivitas lipase pankreas dan berpotensi untuk dikembangkan sebagai antiobesitas.AbstractObesity is caused by excess calories in the body. Reducing obesity can be done by inhibiting the absorption of fat as the main source of excess calories. One way that can be done to inhibit fat absorption is by inhibiting pancreatic lipase activity. This research was conducted to determine the lipase inhibitor activity of methanol extract of arumanis (Mangifera indica L. cv Arumanis) and kweni (Mangifera odorata Griff.) mango leaves. The research stages began with extraction using the maceration method, followed by testing the total phenol content, flavonoid content, and testing the lipase inhibitor activity of arumanis and kweni mango leaf extracts. The results of the total phenol content test showed that the total phenol in Arumanis and Kweni mango leaf extracts was 246.94 mg GAE/g and 176.11 mg GAE/g, respectively. The flavonoid content of Arumanis mango leaves is 129.95 mg.QE/g, while that of Kweni mango leaves is 26.50 mg.QE/g. Each extract was tested for its ability to inhibit pancreatic lipase in vitro. The IC50 values of Arumanis and Kweni mango leaf extracts are 61.55 µg/mL and 79.98 µg/mL. This value is higher than the positive control, namely orlistat (18.01 µg/mL). Thus, Arumanis and Kweni mango leaf extracts can inhibit pancreatic lipase activity and have the potential to be developed as an anti-obesity agent.

Abstract

Sidomulyo Village, situated in Bantul Regency, Indonesia, is confronted with economic challenges, particularly among its predominantly female population who face limited educational and economic opportunities. Recognizing the village's abundant natural resources, this community service program facilitated by Kelompok Wanita Tani (KWT) Amanah aimed to empower local women through ecoprint batik making. The program focused on equipping participants with skills in ecoprint techniques using locally sourced plant materials. Through stages of socialization, training, mentoring, and evaluation, the participants learned to create vibrant fabric patterns without chemical dyes, promoting environmental sustainability and preserving local biodiversity. Positive feedback highlighted the program's effectiveness in enhancing participants' skills and knowledge, with a strong interest shown in further developing ecoprint skills and exploring entrepreneurial opportunities, including online marketing. This study underscores the transformative impact of community-based initiatives in promoting women's economic independence and sustainable development through traditional craft practices.

Abstract

Sidomulyo Village in Bantul Regency, Yogyakarta, holds untapped potential in its rich biodiversity, particularly for economic empowerment among women, who comprise 53% of the population. The Women Skills Academy (WSA) were established with the aim of facilitating women's empowerment in Sidomulyo Village. In a previous program, training on ecoprint creation was conducted but several challenges were identified. These included limited diversity in ecoprint materials and designs, which restricted the ability to highlight the unique local identity of Sidomulyo Village. Furthermore, participation was predominantly limited to members of the Kelompok Wanita Tani (KWT), emphasizing the need to engage a wider spectrum of Women Skills Academy (WSA) members. To address these issues, the current program focuses on enhancing ecoprint techniques while incorporating e-commerce training. This integrated approach aims to empower participants with the skills needed to achieve economic independence and better represent the cultural richness of the village. The program included socialization, preparation, training, mentoring, and evaluation stages. The participants, guided by the community service team, significantly improved their knowledge and skills in both ecoprint production and online marketing, as shown by pre- and post-training evaluations. Participants successfully created ecoprint products using natural resources and learned to market these products via e-commerce platforms. Despite these achievements, challenges remain in diversifying materials, enhancing design uniqueness, and fully utilizing digital marketing. This program underscores the importance of leveraging local resources and providing continuous support to develop innovative, eco-friendly products that contribute to sustainable economic growth in rural communities.

Abstract

Kombucha, a traditional fermented beverage made from tea, sugar, and a kombucha culture, is known for its unique flavor, aroma, and health benefits. The fermentation process, typically lasting between 8 and 12 days, influences the beverage's physical and chemical properties. While traditionally brewed from tea, alternative substrates like rosella flowers (Hibiscus sabdariffa L.) are also used. Rosella flowers, rich in vitamins and bioactive compounds, have demonstrated potential in combating obesity and exhibiting antioxidant properties. This study aimed to evaluate the chemical and biological characteristics of rosella flower kombucha, particularly its antioxidant and lipase inhibitor activities. The fermentation process led to significant biochemical changes, including a decrease in pH and reducing sugar content, an increase in microbial biomass, and a reduction in ascorbic acid content. The results revealed that rosella flower kombucha had a higher total phenol content and better DPPH radical scavenging activity compared to rosella flower infusion, indicating enhanced antioxidant potential. Additionally, rosella kombucha demonstrated more effective lipase inhibition, suggesting its potential benefits for weight management and metabolic health. These findings emphasize rosella kombucha's potential as a healthful functional beverage, recommending further research in these areas.

Abstract

Strawberries (Fragaria x ananassa Duch.) are a valuable horticultural crop in Indonesia, but their productivity is threatened by anthracnose disease caused by Colletotrichum acutatum Simmonds. Traditional control methods often involved synthetic fungicides, which posed risks of pesticide residues and environmental harm. This study investigated Actinomycetes isolated from various strawberry plant organs and their rhizosphere as potential biological control agents. The isolation processes included the collection of Actinomycetes from strawberry plant organs and the rhizosphere, along with the regeneration of C. acutatum. The isolates were identified through observations of their morphological characteristics. Screening of antagonistic activity was performed using dual-culture assays. Over 10 Gram-positive Actinomycetes strains were successfully isolated and characterized. Two isolates exhibited the highest antagonistic activity against C. acutatum, with inhibition percentages of 67.17% and 55.80%, respectively. Data analysis revealed that these isolates demonstrated significant biocontrol potential through mechanisms such as competition and antibiosis. The results suggested that Actinomycetes could serve as effective, eco-friendly alternatives to chemical pesticides in strawberry cultivation. Future research should focus on optimizing these strains for broader agricultural applications.

Abstract

Madu Kelulut merupakan produk lebah tanpa sengat yang memiliki nilai ekonomi tinggi, namun pengelolaan penjualannya masih menjadi tantangan bagi peternak. Pengelolaan penjualan hanya terbatas pada pencatatan manual pada buku catatan. Pemahaman terhadap analisis penjualan belum memadai, sehingga strategi untuk peningkatan penjualan masih belum mampu dilakukan. Pelatihan analisis penjualan ini bertujuan untuk membekali komunitas peternak lebah madu Kelulut di Terengganu, Malaysia, dengan keterampilan dasar dalam pencatatan dan analisis penjualan menggunakan perangkat lunak sederhana. Kegiatan ini dilaksanakan melalui metode ceramah, diskusi interaktif, dan praktik langsung. Hasil evaluasi menunjukkan peningkatan signifikan dalam pemahaman peserta, di mana sebelum pelatihan, seluruh peserta (100%) belum mengetahui konsep analisis penjualan, namun setelah pelatihan, semua peserta telah memahami dan menguasai materi yang diberikan. Selain itu, mayoritas peserta (86%) menilai metode yang diajarkan mudah diterapkan, dan seluruh peserta (100%) menganggap pelatihan ini sangat bermanfaat bagi usaha mereka. Tingginya antusiasme dan respons positif terhadap pelatihan menunjukkan efektivitas metode yang digunakan. Dengan meningkatnya keterampilan dalam analisis penjualan, peternak dapat mengelola usaha mereka secara lebih efisien, meningkatkan strategi pemasaran, serta mendukung pencapaian SDG 12 terkait konsumsi dan produksi yang berkelanjutan.

Abstract

Resistensi antibiotik merupakan isu kesehatan penting yang berkaitan dengan penyebaran bakteri resisten dari peternakan ayam ke lingkungan. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi Escherichia coli dari perternakan ayam serta menentukan profil kepekaan antibiotiknya. Sampel diambil dari peternakan ayam di Bantul, Yogyakarta. Penelitian diawali dengan isolasi bakteri E. coli, diikuti pewarnaan Gram serta identifikasi molekuler dengan gen uspA. Uji sensitivitas antibiotik dilakukan menggunakan metode Kirby-Bauer terhadap antibiotik ampisilin, tetrasiklin dan kloramfenikol. Hasil menunjukkan bahwa 4 dari 8 isolat (A2, A4, B2, B3) merupakan positif bakteri E. coli, ditandai dengan koloni hijau metalik pada EMBA dan pita DNA sepanjang 884 bp pada hasil elektroforesis gen uspA. Uji resistensi menunjukkan seluruh isolat resisten terhadap ampisilin (100%), sedangkan terhadap tetrasiklin dan kloramfenikol menunjukkan hasil yang lebih rendah (50%). Temuan ini mengindikasikan adanya potensi penyebaran bakteri resisten dari peternakan ayam sehingga diperlukan pengawasan penggunaan antibiotik dan pemantauan resistensi secara berkala.

Abstract

Red spinach is widely cultivated and consumed by the public because it has high antioxidants, vitamins (A, C, E), protein, carbohydrates, fats, minerals, iron, magnesium, manganese, potassium and calcium. P giving POC can increasered spinach biomass. Nutrients in POC have been decomposed so that they are easily absorbed by plants. This study aims to analyze the giving of POC Lemna sp. on growth and biochemical content of red spinach and determine the concentration of POC Lemna sp. effective for red spinach (Amaranthus tricolor L.). The study used a completely randomized design with 3 replications and 3 treatments. The independent variable in this study was the concentration of liquid organic fertilizer Lemna sp. (A=0%, B=5%, and C=10%). Data were analyzed by ANOVA test and DMRT test at 5% level. The results showed that the administration of POC Lemna sp. gave significantly different results on the A (0%) treatment of plant height, stem diameter, and flavonoid parameters. Giving POC Lemna sp. can increase the growth and flavonoid content of red spinach. The concentration of liquid organic fertilizer Lemna sp. 5% effective for the growth of red spinach (Amaranthus tricolor L.).
