Abstract

The banana commodity is one of the prioritized commodities in Gunung Kidul Regency. The Young Farmers' Studio and Youth Organization “Solid Berkarya” in Serut Village are trying to develop banana plants by managing critical land into productive land using smart-farming irrigation systems. The concept of smart farming includes using technology such as sensors, connected devices (IoT), data analytics, artificial intelligence, and agricultural management software to create an efficient farming system. The irrigation system needs to apply optimization of timing and intensity tailored to the needs of the plants. The geographical terraced land conditions require unique methods to maximize agricultural yields. In banana cultivation, geographical conditions, climate, soil conditions, and water availability are crucial, so the community's knowledge and skills in irrigation on critical land must be improved. Therefore, banana irrigation training is conducted in this community service, especially related to water availability optimization. In this training, the participant's level of knowledge is measured by conducting pre-tests and post-tests related to irrigation optimization materials. From the test results, it can be seen that there is an increase in community knowledge from 6.44 before training to 9.0 after training. Thus, there is an increase in knowledge by 53.4%. Abstract

As the COVID-19 pandemic comes to an end, education has returned to normal with in-person learning. The good practices in online learning conducted during the pandemic can be maintained to support practical activities, for example, using a virtual laboratory. The limitations of laboratory facilities and the skills of educators and laboratory staff in developing practical materials have made Biology practicals minimal and monotonous. Based on discussions with partners, there is a main problem that needs to be addressed through this program, which is the limited laboratory facilities in terms of tools and materials, resulting in minimal implementation of Biology practical activities in schools. The objective of this community service is to enhance the knowledge, skills, and empowerment of Biology teachers at Muhammadiyah high schools in Yogyakarta City in conducting practicals using virtual labs. This community service method is carried out by training on Biology practical learning online using virtual labs. This program is conducted in several sessions, each lasting 200 minutes, from June to October. Each session consists of a pre-test, material delivery, Q&A discussions, and a post-test. It can be concluded that the community service activities ran smoothly and provided benefits in the form of increased knowledge and skills of participants in Biology practical learning using virtual labs.

Abstract

Actinomycetes form associations with plants through colonizing plant tissues (endophytes) or by residing in the soil around some plants' roots (rhizosphere). Actinomycetes are known to produce antibacterial compounds. This study aimed to investigate the antibacterial activity of actinomycetes associated with the clove plant (Syzygium aromaticum) against Staphylococcus aureus and Escherichia coli. Actinomycetes were isolated from clove plants and the rhizosphere, and their antimicrobial activity against Staphylococcus aureus and Escherichia coli was evaluated using the agar plug method, where the presence of transparent zones around 10-day-old actinomycete growth indicated inhibition of bacterial growth. Four isolates showed inhibitory effects against Staphylococcus aureus, while only three isolates, B.4, T.3, and T.4, demonstrated inhibitory activity against Escherichia coli, as indicated by the presence of inhibition zones. Isolate T.3 exhibited the highest inhibition zone of 8.5 mm against S. aureus, whereas B.4 displayed the highest inhibition zone of 7.7 mm against E. coli. In conclusion, the actinomycetes found in clove plants (Syzygium aromaticum) demonstrate antibacterial properties against Staphylococcus aureus and Escherichia coli, indicating their potential as antibacterial agents.

Abstract

The fermentation process of kombucha has become a focus of halal product research due to its ethanol content. Processing kombucha into powder form can be an alternative to address the ethanol content in liquid kombucha, as it has undergone a drying process, making it a potential halal-certified functional drink. The objective of this study was to compare the quality of liquid and powdered kombucha. The research was conducted from March to September 2024 at the Biotechnology Laboratory, Muhammadiyah University of Bandung. The method used was the Complete Randomized Design (CRD) method. The parameters analyzed included ethanol content, total BAA (Bacterial Acetic Acid), pH, organic acid content, and sugar content. In the production of powdered kombucha, optimization of maltodextrin and gum arabic usage was also conducted to achieve the best solubility and taste. The best powdered kombucha was obtained with a maltodextrin-to-gum arabic ratio of 3:2. In the research, liquid kombucha contained ethanol at an average of 0.09%. In comparison, no ethanol was detected in the powdered kombucha (0%). Total BAA in powder decreased after the drying process (p<0.05). The pH value of kombucha in liquid form (4.4) also differed significantly from powdered (3.2) (p<0.05). However, the levels of organic acids and sugars in liquid and powdered kombucha did not differ significantly (p>0.05). The sugar content in liquid and powdered kombucha was lower than that in kombucha in general.

Abstract

Obesity is a condition where there is excessive fat in the body. Weight loss can be accomplished by inhibiting fat absorption through the inhibition of pancreatic lipase activity, a key enzyme in fat metabolism. Natural products from plants contain various phytochemical compounds that can act as pancreatic lipase inhibitors. This research was conducted to determine the lipase inhibitor activity of the methanol extract of Mangifera foetida leaves. Mangifera foetida leaves were extracted and then the total phenolic and flavonoid levels were determined. The extract was tested for its ability to inhibit pancreatic lipase in vitro. The inhibitory activities of the extract and Orlistat were measured using p-nitrophenyl palmitate as a substrate at concentrations of 31.25, 62.5, 125, 250, and 500 µg/mL. The results showed that the extracts of young leaves (YL) and mature leaves (ML) contained phenol and flavonoid compounds. The IC50 values of young leaf extract (YL) and mature leaves (ML) were 45.22 and 35.50 µg/ml, respectively. In conclusion, the Mangifera foetida leaf extract can be promoted as a good source of anti-obesity compounds

Abstract

Endophytic fungi can produce bioactive compounds that are useful as antimicrobials. This study evaluates the antimicrobial potential of culture filtrate extracts derived from endophytic fungi isolated from the medicinal plant Tinospora crispa. Isolation was carried out from the roots, leaves, and stems of T. crispa, which were then identified based on the ITS gene. The culture filtrate was extracted using ethyl acetate and assessed for antimicrobial activity using the disc diffusion method against Escherichia coli, Staphylococcus aureus, and Candida albicans. A total of 3 endophytic fungal isolates were isolated and identified as Acrocalymma vagum, Diaporthe tulliensis, and Colletotrichum truncatum. The results showed that all culture filtrate extracts of the fungal endophyte isolates exhibited varying antimicrobial activity, with the highest antibacterial activity demonstrated by C. truncatum isolates against Escherichia coli and Staphylococcus aureus. The most significant anticandida activity was by D. tulliensis isolates. Endophytic fungi of medicinal plant T. crispa can be developed as a source of antimicrobial agents, especially to overcome the increasing antibiotics resistance.
