Abstract

Bakteri endofit adalah bakteri yang hidup didalam tanaman sehat tanpa menyebabkan perubahan morfologi ataupun menimbulkan penyakit. Bakteri endofit membantu meningkatkan kesehatan dan produktivitas tanaman dengan cara menekan bakteri patogen dan meningkatkan pertumbuhan tanaman. Penelitian ini bertujuan untuk mengisolasi dan mengkarakterisasi kemampuan bakteri endofit yang terdapat pada buah dan batang tanaman tomat cherry (Solanum lycopersicum var. cerasiforme) dalam menghasilkan hormon asam indol asetat (AIA). Hasil proses isolasi bakteri diperoleh 8 isolat bakteri endofit, yaitu 3 isolat dari buah tomat matang (isolat M1, M2, M3), 3 isolat dari buah tomat mentah (isolat Mt1, Mt2 dan Mt3) dan 2 dari batang tanaman tomat cherry (isolat Bt1 dan Bt2). Beberapa bakteri endofit yang berhasil diisolasi memiliki bentuk coccus, bentuk (form) koloni sirkuler, permukaan (elevation) koloni convex, tepi (margin) koloni entire dan berwarna putih. Semua isolat mampu memproduksi hormon AIA kecuali isolat M1 dan Mt3. Bakteri ini berpotensi untuk meningkatkan produktivitas tanaman tomat cherry melalui hormon AIA yang dihasilkan.

Abstract

Lidah buaya terkenal akan beragam khasiat yang dimilikinya. Tanaman ini sangat cocok tumbuh pada iklim tropis Indonesia. KWT (Kelompok Wanita Tani) An-Naba’ sebagai salah satu organisasi kemasyarakatan di Desa Ambarketawang, Sleman berfokus pada peningkatan ekonomi masyarakat setempat melalui budidaya tanaman lokal. Salah satu tanaman yang saat ini digalakkan untuk dibudidayakan adalah lidah buaya. Lidah buaya dipilih karena sangat mudah untuk ditanam dan memiliki beragam manfaat. Saat ini kendala yang masih dihadapi KWT An-Naba’ Desa Ambarketawang adalah sulitnya memasarkan hasil tanaman mereka tersebut. Pelepah lidah buaya hanya dihargai dengan harga yang murah. Salah satu cara untuk membantu meningkatkan daya jual pelepah lidah buaya yaitu dengan mengolah lidah buaya tersebut menjadi berbagai macam olahan yang memiliki nilai ekonomis tinggi. Lidah buaya dapat diolah menjadi berbagai macam olahan minuman dan makanan yang memiliki nilai gizi tinggi. Tujuan dari program pengabdian masyarakat ini yaitu meningkatkan pengetahuan anggota KWT An-Naba’ tentang manfaat, kandungan gizi serta potensi lidah buaya serta meningkatkan keterampilan anggota KWT An-Naba’ dalam mengolah lidah buaya menjadi berbagai produk makanan. Produk makanan tersebut diantaranya yaitu teh Aloe vera, nata de Aloe vera, selai Aloe vera dan manisan Aloe vera. Program pengabdian masyarakat ini telah dilaksanakan pada bulan Mei-Agustus 2017 di Balai Desa Ambarketawang, Kecamatan Gamping, Kabupaten Sleman, D.I.Yogyakarta. Program pengabdian dilakukan dengan menggunakan metode penyuluhan dan pelatihan. Kegiatan diawali dengan penyuluhan tentang manfaat dan khasiat tanaman lidah buaya kemudian dilanjutkan dengan pelatihan tentang cara pembuatan olahan lidah buaya. Peserta sangat antusias dalam mengikuti pelatihan. Peningkatan keterampilan masyarakat melalui pelatihan pengolahan produk lidah buaya ini diharapkan mampu mendorong kemajuan perekonomian masyarakat dan tumbuhnya industri rumah tangga di Desa Ambarketawang.

Abstract

WOMEN FARMERS EMPOWERMENT OF AMBARKETAWANG VILLAGE IN HOUSEHOLD WASTE TREATMENT. People in Ambaraketawang village have begun to have awareness about the dangers of burning rubbish and littering. However, awareness of sorting and processing household waste is still not done. Garbage is only collected and finally transferred to the TPA (Tempat Pembuangan Akhir). The purpose of this activity is to provide counseling, training and assistance in processing household waste. This activity is an effort to reduce and manage household waste into something that has a use value, especially for members of the Kelompok Wanita Tani (KWT) in processing household waste. The activity was divided into several sessions including counseling on waste sorting, training on making organic liquid fertilizer, training on making organic liquid fertilizer from local microorganisms, training on composting using the Takakura method, counseling on product quality standards, and counseling on product packaging. The results obtained from this activity are increasing knowledge and skills of participants in the processing of household waste through community empowerment.

Abstract

In the era of industrial revolution 4.0, many human activities depend on the gadget and the internet connection. We can observe the daily activities of human life right now. When they wake up in the morning, they usually start by using their handphones or gadget. This condition motivates educators to build a digital module in order to use the positive point of view of the Industry Revolution 4.0 era. In this chance, the researchers develop two topics that are very important for human life in Industry Revolution 4.0, whicht are, human literacy and technology literacy. Both of the topics were derived into two-courses. The course of development of students (Perkembangan Peserta Didik) will be representing the human literacy topics and the course of introduction to Biotechnology (pengantar bioteknologi) will be representing the technology literacy topic. The research is research and development. Furthermore, the products of this research are flipbooks that were uploaded on the Learning Management System managed by Universitas Ahmad Dahlan.

Abstract

Fungi that grow in plant tissue without harming the host are called endophytic fungi. This fungus has the potential as a biological agent (biocontrol) in controlling plant diseases. One of the fungi that can cause disease in banana plants is Fusarium oxysporum, which can cause Fusarium wilt disease. Endophytic fungi have antagonistic properties in the form of nutritional competition and antibiosis. These antagonistic properties can be used as biological agents. The purpose of this study was to isolate and test the antagonistic activity of the endophytic fungus of banana plant (Musa paradisiaca L.) against the pathogenic fungus Fusarium oxysporum. This research is an experimental study, which begins with the isolation of endophytic fungi and then continues with the antagonism activity test. The results of the isolation obtained 4 isolates of endophytic fungi, namely: 1) isolate D1 (Phytium sp.); 2) isolate B1 (Aspergillus sp.); 3) isolate A1 (Trichoderma sp.); and 4) isolate A2 (Penicillium sp.). Based on the antagonism activity test, isolate A1 showed the best results in inhibiting the fungus Fusarium oxysporum through the mechanism of space and nutrient competition, while isolates that showed antibiotic activity were isolate A2. The results of the inhibition test using the dual culture method showed that, 2 isolates (B1 and A2) could inhibit the growth of the fungus Fusarium oxysporum. The highest percentage of inhibition was owned by isolate A2. Based on its ability to inhibit Fusarium oxyaporum, endophytic fungal isolates have the potential to be further developed as Fusarium oxysporum control agents.

Abstract

Banyak penyakit yang penyebarannya diperantarai oleh lalat (Musca domestica). Lalat  merupakan salah satu vektor mekanis dalam penyebaran penyakit. Saat ini, bakteri resisten antibiotik menjadi masalah serius bagi kesehatan global. Penyebaran bakteri ini salah satunya dapat diperantarai oleh lalat. Hal ini dapat menyebabkan menurunnya kualitas hidup masyarakat sekitar. Tujuan dari penelitian ini adalah untuk menganalisis keberadaan bakteri resisten antibiotik yang berasosiasi dengan lalat dewasa (M. domestica) yang dikumpulkan dari tempat pembuangan sampah di sekitar kampus Universitas Ahmad Dahlan, Banguntapan, Yogyakarta. Tahapan penelitian yang dilakukan yaitu isolasi bakteri yang berasosiasi dengan lalat dewasa kemudian menguji kepekaan bakteri tersebut terhadap antibiotik kanamycin, chloramphenicol, amoxycillin, tetracycline, ciprofloxacin, gentamicin, dan erythromycin. Dari 12 lalat yang ditangkap, bakteri yang diperoleh sebanyak 20 isolat. Persentase resistensi masing-masing isolat terhadap antiobiotik yang diujikan berdasarkan pengukuran zona hambat yang diperoleh yaitu kanamycin 0%, chloramphenicol 25%, amoxycillin 25%, tetracycline 50%, ciprofloxacin 0%, gentamicin 0%, dan erythromycin 75%. Hasil analisis antibiotik menunjukkan bahwa semua isolat bakteri yang diperoleh telah resisten terhadap satu ataupun dua antibiotik yang diujikan.Kata Kunci:Musca domestica; Resistensi antibiotik; Bakteri

Abstract

Warga Kecamatan Gamping Kabupaten Sleman telah memiliki kesadaran mengenai pengelolaan sampah oleh karena itu, sungai yang mengalir di wilayah tersebut cukup bersih. Sungai ini kemudian dimanfaatkan oleh Pokdarwis Mina Guyangan 05 yang ada di Gamping Lor untuk merintis Wisata Sungai dengan menebar ikan Nila Merah (Oreochromis niloticus). Permasalahan utama yang dihadapi oleh mitra saat ini adalah adalah kurangnya pengetahuan dan keterampilan mitra dalam diversifikasi jenis ikan yang dipelihara, pengendalian parasit ikan, serta kurangnya penataan lingkungan sekitar sungai untuk rintisan wisata sungai. Kegiatan pengabdian ini dilaksanakan dengan metode penyuluhan tentang diversifikasi jenis ikan dan pengendalian parasit ikan, serta dalam bentuk penyuluhan dan pelatihan budidaya dan pemanfaatan tanaman untuk ditanaman di sekitar sungai. Kegiatan yang dilakukan terbukti dapat meningkatkan pengetahuan mitra berdasarkan hasil tes. Kegiatan memang belum memberikan dampak ekonomi secara langsung tetapi telah memberikan dampak sosial berupa meningkatnya kesadaran menjaga kebersihan air dan memanfaatkan tanaman sekitar sungai untuk membuat pakan dan pengendali parasit. Kata kunci: diversifikasi ikan, gamping lor, pokdarwis mina guyangan 05, parasit ikan, wisata sungai ABSTRACT Residents of Gamping Subdistrict, Sleman Regency already have awareness about waste management, therefore, the rivers that flow in the area are quite clean. This river was then used by Pokdarwis Mina Guyangan 05 in Gamping Lor to pioneer River Tourism by stocking Red Tilapia (Oreochromis niloticus). The main problems faced by partners today are the lack of knowledge and skills of partners in diversifying the types of fish that are kept, controlling fish parasites, and the lack of environmental management around the river for pioneering river tourism. This activity is carried out with counseling methods on fish species diversification and fish parasite control, as well as in the form of counseling and training on cultivation and use of plants for planting around rivers. The activities carried out are proven to increase partner knowledge based on test results. The activity has not yet had a direct economic impact, but has had a social impact in the form of increasing awareness of maintaining water cleanliness and utilizing plants around the river to make feed and control parasites. Abstract

Disposable diaper waste takes hundreds of years to decompose and cause environmental pollution if disposed of directly into landfills or rivers. Therefore, innovation in the processing of disposable diaper waste and increasing public awareness for the processing of disposable diaper waste at the household level are needed. The purpose of this community service is to provide counseling and training to members of the Aisyiyah Branch (PCA) Gamping, Yogyakarta in managing disposable diaper waste into planting media. This activity was attended by 15 people from the board and members of PCA Gamping as well as local community leaders. The result of this community service activity is that there is an increase in the knowledge and skills of participants in processing disposable diaper waste into planting media. Through this activity, it is hoped that it will be useful to help reduce environmental pollution due to disposable diaper waste.

Abstract

One of the main issues still faced by Indonesia is organic waste. This problem can be overcome by processing organic waste into eco-enzymes. Eco-enzymes contain alcohol and acetic acid, so they can be used as disinfectants and floor cleaners, dishwashing liquids, and detergents. This study aims to determine sensory characteristics such as aroma and color preferred by subjects for eco-enzyme production with the addition of yeast (Saccharomyces cerevisiae) and fruit peel variations. All eco-enzymes were made from organic waste in the form of fruit peel mixed with sugar and water in a ratio of 3:1:10. To determine the effect of yeast addition, eco-enzymes were made with orange peel with 2 variations, namely: 1) with the addition of yeast; and 2) without the addition of yeast. In the analysis of the effect of fruit peel variations, eco-enzymes were made with 6 variations, namely: 1) orange peel; 2) orange peel and pineapple peel; 3) orange peel and banana peel; 4) orange peel and papaya peel; 5) orange peel and apple peel; and 6) a mixture of all fruit peels. After one month of fermentation, the obtained eco-enzyme products were analyzed for their sensory properties. According to the observations, the aroma characteristic of the obtained eco-enzymes is that all eco-enzyme products, on average, have a normal fermentation sour aroma, with the exception of eco-enzyme products with added yeast, which have a sharper alcohol aroma. Observations on the sensory properties of color showed that eco-enzymes have a characteristic color ranging from dark brown to light brown. Overall, most of the panelists preferred the aroma and color of eco-enzymes produced without the addition of yeast. Meanwhile, for different fruit peel variations, the panelists preferred the mixture of fruit peels compared to other variations.

Abstract

Tuberculosis still becomesa major health problem in the world. This infectious disease is caused by Mycobacterium tuberculosis (Mtb). Novel anti-tubercular drug is urgently needed to counter multidrug resistant cases and Mtb's spread. The cytoplasmic domain of PhoR histidine kinase, a part of the two-component system PhoR-PhoP in Mtb, is one of the potential candidates for anti-tubercular drug target.Three dimensional protein of drug target is needed to screen potential drug candidate using rational drug design approaches. Previous studies have successfully characterized and isolated putative cytoplasmic domain of PhoR (CytoPhoR) from Mtb strain H37Rv. This study aimed to clone, overexpress and purify of CytoPhoR protein. CytoPhoR was fused with thioredoxin protein in expression vector pET32b and overexpressed in Escherichia coli (E.coli)BL21 (DE3) as soluble fraction by induction  1 mM IPTG. Purification of his-tagged CytoPhoR was carried out using IMAC Ni-NTA Agarose his-tag affinity column. SDS-PAGE analysis showed that another protein was co-purified (~35 kDa) along with the CytoPhoR protein. Subsequent protein purification using DEAE-ion exchange column generate a strong single band of 37 kDa on SDS–PAGE which is indicated as CytoPhoR protein. The purified CytoPhoR protein was successfully obtained and can be used for further analysis on determining three dimensional structure of CytoPhoR protein.
