Abstract

Abstract

Heavy metals have been used in various areas around the world especially in the industrial sector. Heavy metals contamination is very dangerous for ecosystem because of its toxicity for some organisms. Cadmium (Cd) is a dangerous metal pollutant that can cause remarkable diverse of toxic effects, in particular for humans and animals. The use of bacteria as bioremediation agents has been widely studied because more efficient, less cost, and environmentally friendly strategy. This present study aimed to isolate and identify Cd-resistant bacteria from the industrial disposal site. Wastewater samples were collected from disposal site of agar flour industry in Lawang Malang, East Java. The collected wastewater effluent was analyzed for physicochemical properties. Isolation of Cd-resistant bacteria was carried out using serial dilution. Bacterial isolates were observed and tested for their effects on the content of Cd. The content of Cd was tested daily using Atomic Absorption Spectroscopy (AAS) for seven consecutive days. Data was analyzed using one-way ANOVA (p < 0.05) and Tukey test. Characterization of potential bacterium was performed using bacterial identification kit. Four bacteria isolates have been successfully isolated from the wastewater sample. There was a statistically significant difference between groups as determined by one-way ANOVA (F = 1229.62, p = 0.00). A Tukey post hoc test revealed that all conditions are significantly different from each other. The content of Cd in wastewater sample was statistically significantly lower after taking the A isolate (3.39 mg/L, p = 0.00), B Isolate (1.47 mg/L, p = 0.00), C Isolate (1.15 mg/L, p = 0.00), and D isolate (1.95 mg/L, p = 0.00) compared to the control treatment (5.11 mg/L, p = 0.00). Two of the most potential isolates identified as Pseudomonas flourescens (C isolate) and Enterobacter agglomerans (B isolate).

Abstract

Cellulolytic bacteria are bacteria that have the ability to hydrolyze cellulose complexes into smaller oligosaccharides and eventually become glucose. Glucose is used as a carbon and energy source for bacterial growth. This study was conducted to isolate the cellulose degrading bacteria from banana peel compost that produce cellulose enzymes based on the clear zone that visible around the colony. The cellulolytic activity was determined by the ability of bacteria to hydrolyze the Carboxymethyl Cellulose (CMC) substrate. Determination of cellulolytic activity is known based on cellulolytic index calculation, the diameter total minus the diameter of the colony and divided by the diameter of the colony. The result of five bacterial isolates was found but only one bacterium had the potential to be a cellulose degradation. Based on the Microbact Gram-Negative Identification System, the bacterium is Burkholderia cepacia. These bacteria have an important role in nature as decomposers of various complex compounds, such as cellulose, hemicellulose, lignin, and pectin.

Abstract

A survey and exploratory study were conducted to analyze Lake Tempe as a resource for contextual learning (CTL). The first phase of the study included exploring the lake’s potential through interviews with the people who live in the lake’s coastal region. Quadrat transects were done to identify aquatic plants around the lake. A feasibility analysis was conducted to see the lake’s potential as a learning resource. Field data were collected and compared to the environmental biology (EB) curriculum, responses, and learning outcome. The data were analyzed descriptively and tested using a paired-sample t-test. The results showed differences in the three stations’ physicochemical parameters and decreased quality of the water. It was found that anthropogenic activity had a significant contribution to the lake’s pollution. Nine species of six aquatic plant families were found in the lake. The survey revealed that the lake was feasible as a learning resource with criteria easy to access, safety, time efficiency, costs, and the material's suitability for EB courses. The t-test showed a difference in the students’ scores between before and after exploration practices. These findings suggest that Lake Tempe can serve as a resource for CTL in EB courses.
