Abstract

The concentration of three heavy metals Pb, Cd and Hg ware measured in Bone River, Gorontalo sediment. The heavy metal pollution on sediment in Bone River have not been explored longitudinally. The samples were taken in July 2021 at 8 stations.  The trend of metals ware observed in sediment as PbCdHg. The level of studied metals Cd and Hg did not exceed the safe limit by ANZECC, OSQG LEL and CCME TEL meanwhile Pb was met the limit by by ANZECC. However, the investigated showed that fluctuations due to differences in the characteristics of each location. Contamination factor (CF) and geoaccumulation index (Igeo) demonstrated that the sediment samples were low contamination and lightly polluted. Contamination by heavy metals in the Bone River implies that the conditions are much frightening for the biota and residents around the river.

Abstract

Abstract Humpback grouper (Cromileptes altivelis) is a commodity to be cultivated because it has a high price, but the grouper highly susceptible to Vibriosis (Vibrio harveyi). The research objective was to see the function of immune cells of interleukin-4 (IL-4) are formed due to the induction of immunogenic proteins of Vibrio harveyi. Research procedures include Vibrio harveyi immunogenic protein isolation, SDS-PAGE electrophoresis, elektroelusi and dialysis, clinical exam of Vibrio harveyi immunogenic proteins, and immunohistochemical to look at immune cells (cells IL-4) with using a secondary antibody labeling of anti IL-4 biotin conjugate. The results showed that Vibrio harveyi protein found have immunogenic properties is 51.16 kDa. This protein is an adhesin protein. Protein 51.16 kDa was purified by electroelution and the protein will be used as a vaccine administered by intraperitoneal injection in grouper (Cromileptes altivelis) at a dose of 31.6 ug protein / 150 g of fish weight. In organ tissues of the brain, eyes and kidneys of induced fish by immunohistochemical examination, It was detected Immune cells (cells IL-4) using a secondary antibody labeling of anti IL-4 biotin conjugate. Tues formed IL-4 can be seen on the network grouper indicated by a change in tissue color with an inverted microscope examination of the golden brown color is formed on the IL-4 immune cells. It show the cross-reaction between 51.16 kDa immunogenic protein induced in fish with anti IL-4 biotin conjugate. The conclusions is the protein of Vibrio harveyi with molecular weight of 51.16 kDa is an immunogenic protein which able to induce formation of IL-4 in grouper.

Abstract

Abstract The aim of these study was to know plankton (phytoplankton and zooplankton spesies and density at Sengguruh dam, Karangkates, Lahor, Wlingi Raya and Wonorejo, and to estimated trophic status these dams use phytoplankton and zooplankton density approachment. These study used descriptive methode with survey technic. Data wich is taken from each dam is taken from four different station that suggested the representated dam condition. The result showed according to the range phytoplankton density: at Karangkates between 144-236 ind/ml is clustered as oligotrophic, the range phytoplankton density at sengguruh between 217-313 ind/ml is clustered as oligotrophic. At Lahor dam between 5.35612.704 ind/ml is clustered as mesotrophic, at Wlingi Raya dam between 74-1480 ind/m' is clustered as mesotrophic to eutrophic, while at Wonorejo dam between 48-288 ind/ml is clustered as oligotrophic waters. According to the range zooplankton density: at Karangkates dam between 28.153-132.056 ind/lt is clustered as mesotrophic, Lahor dam between 4.996-14.275 ind/lt is clustered as mesotrophic, Wlingi Raya dam between 623-3.114 ind/lt is clustered as oligotrophic to mesotrophic, and Wonorejo dam between 2-6 ind/lt is clustered as mesotrophic. Abstract

HighlightCatfish aquaculture wastewater contains organic matter that exceeds the quality standard of 68.75 mg / L and 72.48 mg / L.Tutut Jawa snail (F. javanica) can reduce organic matter in waste by up to 92%.Susuh Kura snail (S. testudinaria) can reduce organic matter in waste by up to 87%.Catfish aquaculture wastewater treatment with snails only takes 8 hours. Abstract The residual or wastewater from the cultivation process that is discharged directly into common waters can reduce the quality of the waters because it contains a lot of organic material from organism feces and feed residue, so it needs management first. The purpose of this study was to determine the ability of Javanese snails and freshwater gastropods to reduce total organic matter in catfish culture waste. The research method used was the experiment with 2 factor Factorial completely randomized design, namely the measurement time (4, 8, 12, and 16 hours) and the abundance of the Tutut Java snail and freshwater gastropods (100%, 75%, 50%, 25%, and control) with 5 repetitions. Tutut Java snails were taken from the pond at UPR Sumbermina Dau and brought to the laboratory. The results obtained in the treatment with the addition of  Tutut Java snail, the total organic matter content decreased at 75% snail density by 90% (from 68.75 mg / L to 5.31 mg / L) at the 12th hour. The treatment with the addition of Freshwater Gastropods at a density of 25%, the total organic matter content decreased by 87% (from 72.48 to 9.35 mg / L) at the 8th hour. Water quality during the study was in a condition that could tolerate the Tutut Java snails and freshwater gastropods. The conclusion is that the addition of Tutut Java snail and / or freshwater gastropods can reduce the total organic matter content to the water quality standard.

Abstract

Caulerpa lentillifera is known to have an antibacterial benefit based on various research findings. The study aims to analyze the potential of the Caulerpa lentillifera water extract in the bacterial barrier power isolated from the lele infected wound disease. The methods carried out included extracting Caulerpa lentillifera water extract. The study was conducted using an experimental method with a Complete Random Scheme (RAL) consisting of control, 50 mg/l and 100 mg/ l on each extract. The results showed that the water extract of Caulerpa lentillifera showed significant increases in the barrier strength at doses of 50 mg / l and 100mg/l with diameters of 7.8 mm and 8.4 mm respectively in bacteria isolated from the disease-affected fish wounds. On the other hand, on the result of the bacterial barrier that is isolated from the living media, the lele obtains a low barrier. This is due to the bioactive activity of the antibacterial water extract Cauleroa lentillifera specific to bacteria. It can be concluded that Caulerpa lentillifera is also capable of inhibiting the growth of the bacteria causing the lily's wound

Abstract

Caulerpa lentilifera merupakan makroalga atau sering disebut dengan anggur laut yang mudah ditemui di Indonesia. Berbagai aktivitas manfaat dari Anggur laut telah ditemukan untuk kegiatan akuakultur salah satunya yakni dalam peningkatan sistem imun ikan. Kajian penggunaan caulerpa sebagai imunostimulan yang efektif perlu dilakukan penelitian untuk menentukan dosis yang tepat untuk meningkatkan imune ikan. Uji toksisitas akut merupakan uji awal yang diperlukan untuk mengetahui dosis lethal ekstrak caulerpa. Hasil uji toksisitas akut dijadikan sebagai dasar penentuan dosis pada uji sub kronis yang selanjutnya akan diketahui dosis efektif dalam peningkatan imunitas ikan. Tujuan penelitian ini adalah untuk menganalisis toksisitas akut ekstrak pelarut yang berbeda yakni pelarut metanol dan pelarut water extract dan menguji pada kadar glukosa darah ikan sebagai indikator tingkat stress ikan. Metode yang digunakan dalam ekstraksi dengan menerapkan metode maserasi dengan pelarut metanol dan pelarut water extract. Uji toksisitas yang dilakukan yaitu metode penentuan LC50-96jamdengan analisis probit.  Metode ini dilakukan diawali dengan uji pendahuluan untuk menentukan ambang lethal atas dan ambang lethal bawah. Selanjutnya dilakukan uji sesungguhnya untuk menentukan nilai LC50-96jam . Pada proses uji toksisitas dilakukan pengamatan kadar glukosa darah ikan komet menggunakan kit blood glucose. Pada penelitian juga dilakukan pengukuran kualitas air seperti suhu, pH dan oksigen terlarut (DO). Hasil penelitian menunjukkan toksisitas akut tertinggi pada ekstrak Caulerpa lentilifera dengan pelarut Metanol sebesar 0,761 ppm. Selanjutnya kadar gula darah tertinggi didapatkan pada ekstrak Caulerpa lentilifera dengan pelarut Metanol sebesar 102 mg/dL pada konsentrasi 0,24 ppm. Parameter kualitas air menunjukkan bahwa pengukuran suhu, pH, oksigen terlarut (DO) didapatkan nilai masih dalam ambang batas untuk mendukung hidup ikan.   Caulerpa lentilifera is a sea grape like-macroalga that grows well in Indonesia. Many beneficial activities of sea grapes have been found for aquaculture, such as to increase fish immune system. However, Caulerpa lentilifera potency as an effective immunostimulant need to evaluate further to determine the proper dose to increase fish immunity. Acute toxicity test is the initial test needed to determine the lethal dose of Caulerpa lentilifera extract. The results then used as the basis to define the dose in the sub-chronic test and further to decide the effective dose in increasing fish immunity. The purpose of this study was to analyze the acute toxicity of different solvent extracts (methanol and water extract) and to evaluate the blood glucose levels of fish studied as an indicator of fish stress levels. The extraction method used was the maceration method . Toxicity test  was carried out to determine LC50-96hours with probit analysis. The toxicity preliminary test was performed to define the upper and the lower lethal threshold. The further toxicity test was carried out to determine the value of LC50-96hours. In the toxicity test process, the blood glucose levels of comet fish were observed using a blood glucose kit. In this study water quality parameters such as temperature, pH, and dissolved oxygen (DO) were also  asesses. The results showed that the highest acute toxicity was found at Caulerpa lentilifera extracted with methanol at 0.761 ppm. Furthermore, the highest blood sugar levels  at 102 mg/dL were found in Caulerpa lentilifera extracted with methanol of at a concentration 0.24 ppm. Water quality parameters indicate that the temperature, pH, and dissolved oxygen (DO) values ​​are still within the threshold to support fish life.

Abstract

Masalah yang sering muncul dalam kultivasi vanamei adalah serangan penyakit. Salah satu penyakit semangka yang sering menyebabkan kegagalan tanaman untuk kematian massal adalah Vibriosis. Vibriosis adalah infeksi pada kerang yang disebabkan oleh bakteri Vibrio (Vibrio alginolyticus, Vibrio harveyi, dan Vibrio parahaemolyticus). Caulerpa ini diketahui memiliki aktivitas biologis, seperti anti-hipertensi, antioksidan, anti-kanker, dan aktivitas anti-mikroba. Tujuan dari penelitian ini adalah untuk mengetahui potensi antibakteri masing-masing ekstrak dari Caulerpa lentillifera. Metode penelitian adalah eksperimental, desain yang digunakan adalah RAL dengan Antibakteri Aktivitas Tes, yang mencakup Uji ekstrak air, etanol dan methanol terhadap bakteri Vibrio alginolyticus, Vibrio harveyi, Vibrio parahaemolyticus. Bakteri dalam penelitian ini adalah bakteri Vibriosis. (Vibrio alginolyticus, Vibrio harveyi, Vibrio parahaemolyticus). Tes ini dilakukan untuk menentukan apakah pelarut telah mempengaruhi pembentukan diameter zona penghalang. Pada piring (media) untuk divaksinasi dengan bakteri uji. Sebuah kertas disk steril 8 mm ditempatkan di atas medium untuk kemudian disegel dengan ekstrak kasar 10 μl dengan konsentrasi ekstrak 25; 50; 100; 200 mg / disk. Petri cangkir dikemas dalam kemasan plastik dan disimpan di inkubator pada suhu 370 C selama 1-3 hari dan diulang 3 kali. Hasil penelitian menunjukkan bahwa diameter zona hambat rata-rata tertinggi ditemukan dalam perlakuan dengan ekstrak Air, Etanol dan Metanol dari Caulerpa pada dosis 100 mg/L sedangkan hasil zona hambat terendah ditemukan pada perlakuan ekstrak Air, Etanol dan Metanol Caulerpa 12,5 mg / L.The frequent issue encountered in the cultivation of vanamei is the occurrence of disease outbreaks.   Vibriosis is one of the diseases that often leads to mass death in watermelon plants.   Vibriosis is an infection in shellfish caused by the Vibrio bacteria.   The three species are Vibrio alginolyticus, Vibrio harveyi, and Vibrio parahaemolyticus.   Caulerpa is known to possess biological activities, such as antihypertensive, antioxidant, anticancer, and antimicrobial activities.   The research method employed was experimental, utilising a Randomised Complete Block Design (RCBD) with Antibacterial Activity Test. This test encompassed the examination of water, ethanol, and methanol extracts against Vibrio alginolyticus, Vibrio harveyi, and Vibrio parahaemolyticus bacteria.   The bacteria used in this study are Vibriosis bacteria.   The three species are Vibrio alginolyticus, Vibrio harveyi, and Vibrio parahaemolyticus.   This es tis conducted to determine whether the solvent has influenced the formation of the barrier zone diameter.   On the plate (medium) for inoculation with test bacteria.   A sterile 8 mm disc paper is placed on the medium and then sealed with a crude extract of 10 μl with extract concentrations of 25, 50, 100, and 200 mg/disk.   The petri dishes are packaged in plastic packaging and stored in an incubator at a temperature of 370°C for 1-3 days, and this process is repeated 3 times.   The research findings indicate that the highest average inhibitory zone diameter was found in the treatment with extracts of Water, Ethanol, and Methanol from Caulerpa at a dosage of 100 mg/L, while the lowest inhibitory zone diameter was found in the treatment with extracts of Water, Ethanol, and Methanol from Caulerpa at a dosage of 12.5 mg/L.

Abstract

The MMD 1000 Village Strategic Community Service Grant Program aims to address the issues that the village community is currently facing, such as the low level of community participation in PKK activities, the need to improve the nutrition of low-income families, and the underutilization of narrow yards. So that through the MMD 1000 Village Strategic Community Service Grant Programme activities, this will offer solutions to these problems, including outreach and demonstration plots along with the assistance of aquaponic cultivation systems. The methods that will be applied to this activity include the following: 1) providing understanding to community leaders and target audiences, especially activity partners; 2). Counselling and training to increase knowledge about the implementation of an appropriate aquaponic cultivation system, 3). Direct practice or application of the aquaponic cultivation system, including assembling the design of aquaponics tools, setting tools and tanks, water preparation and monitoring water quality, stocking fish seeds, planting vegetable seeds, and raising fish and vegetables together 4). Evaluation to monitor success in implementing activities, 5). Evaluate the impact of activities directly on increasing fish and vegetable consumption and the impact on the family economy through fish and vegetable production. This activity increases the community's ability to manage narrow land by implementing an aquaponic system, which has an impact on improving family nutritional welfare and increasing economic income and is expected to be implemented by the wider community in Dawuhan Village and other villages in Malang Regency.
