Abstract

This study aimed to evaluate the effect of spinach (Amaranthus hybridus) extract containing phytoecdysteroids on molting duration, growth performance, feed efficiency, and carbonic anhydrase (CA) enzyme activity in mud crabs (Scylla sp.) reared in a Crab Apartment Box (CAB) system. A Completely Randomized Design (CRD) was applied with four levels of spinach extract doses (0, 300, 700, and 1000 ng/g body weight) and three replications. The results showed that the administration of spinach extract significantly shortened molting duration, with the highest dose (1000 ng/g) reducing the average molting period to 11.83 days compared to 39.17 days in the control group. Although absolute weight gain decreased at higher doses, specific growth rate (SGR) and feed efficiency improved, and feed conversion ratio (FCR) declined. While weight gain percentage slightly decreased at higher doses, overall metabolic efficiency improved. Enzymatic analysis revealed that CA activity peaked at 300 ng/g, suggesting optimal ionic regulation during post-molt recovery at this dose. However, higher doses caused a decline in CA levels, potentially due to regulatory feedback or physiological stress. These findings demonstrate the potential of spinach extract as a natural molting accelerator and physiological modulator in soft-shell crab production.

Abstract

Ranu Grati is a volcanic lake situated in Grati District, Pasuruan Regency, East Java, Indonesia. The rapid expansion of floating net cage aquaculture in Ranu Grati has raised concerns about water quality deterioration and its impact on fish health. This study aimed to evaluate the hematological profile of tilapia (Oreochromis niloticus) as a bioindicator for assessing water quality in floating net cage cultivation systems. The purpose of this research was to examine the hematological profile of tilapia fish alongside a series of water quality parameter measures as an environmental factor influencing fish life. Sampling was conducted with three repetitions every three weeks from September to November 2024 at five distinct locations, which were deemed representative of each group of floating net cages distributed around the Ranu Grati region. The Canonical Correspondence Analysis (CCA) approach was utilized to ascertain the relationship between hematological profile of tilapia and water quality. The data analysis indicates that the water quality at Stations 2 and 3 in the research area was somewhat polluted, but Stations 1 and 5 were heavily polluted. The elevated pollution levels at Stations 1 and 5 are attributed to the significant quantity of floating net cages in the vicinity relative to other stations. The investigation indicated that low dissolved oxygen levels and elevated ammonia concentrations impacted the hematological parameters of Tilapia fish. All water quality characteristics influenced the status of leukocytes, erythrocytes, and hemoglobin at moderate quantities. Moreover, micronuclei and hematocrit levels were significantly affected by elevated concentrations of ammonia, temperature, BOD, and pH, whereas DO, TDS, and TSS influenced lower values.

Abstract

This study investigates the effects of dietary supplementation of spinach (Amaranthus sp.) extract, rich in phytoecdysteroids, on the molting process and physiological parameters of mud crabs (Scylla sp.) cultivated in crab apartment boxes integrated with a recirculating aquaculture system. This experiment was designed using a completely randomized design (CRD) with four different doses of spinach extract as treatments (0, 300, 700, and 1000 ng g-1 of crab) and three replications. Some parameters measured were molting duration, hormone levels, including ecdysteroid and molt inhibiting hormone (MIH), reactive oxygen species (ROS), and water quality. The results showed that higher doses of spinach extract significantly shortened the molting duration (p < 0.05), with the fastest molting observed at 1000 ng g-1 (11.83 ± 1.26 days). Increased spinach extract doses also significantly affected MIH and ROS levels. Specifically, MIH levels increased from 12.05 ng mL-1 in the control to 20.88 ng mL-1 in treatment D, while ROS levels rose from 116.80 to 147.33 µmol mL-1. Overall, the administration of phytoecdysteroids from spinach presents a promising, eco-friendly approach to enhance soft-shell crab production efficiency, although careful dose optimization is necessary to balance production acceleration with animal welfare.

Abstract

The aquaculture sector plays an important role in supporting food security and improving the community's economy. Aquaculture, especially in rural areas, has become one of the main pillars in providing protein sources for the community and also driving the local economy. This community service activity aims to improve the skills and independence of fish farmers in the Fish Farmers Group (PAKWALI) of Bakalan Village, Pasuruan Regency through the innovation of alternative floating paste feed based on local potential. The main problems faced by partners are the high cost of commercial feed and the lack of knowledge about making independent feed. The methods used include counseling, training, and assistance in making alternative feed. The main innovation is the development of floating paste feed that uses local raw materials such as tofu dregs and bran. The results of the activity showed an increase in the skills of partners in producing independent feed, with production cost efficiency reaching 40.3% compared to commercial feed. This alternative feed also increases the efficiency of nutrient absorption in fish and reduces feed waste. In conclusion, this program has succeeded in increasing the independence of fish farmers, reducing production costs, and encouraging more environmentally friendly and sustainable cultivation practices.

Abstract

Lempuk fish is a native Indonesian fish that is endemic in the Ranu Grati tectonic lake, Pasuruan Regency, East Java. This research aims to provide information in efforts to domesticate lempuk fish, especially on reproductive aspects related to the microscopic characteristics of gonads and reproductive type of Lempuk fish (Gobiopterus sp). This research uses the primary sexual characteristics method with gonadal histology. The variables observed included the male to female sex ratio, the histological structure of male and female gonads, and the reproductive type of lempuk fish. All data was analyzed descriptively, qualitatively and quantitatively by comparing with related literature. The samples taken in this research were 39 Lempuk Fish (Gobiopterus sp). The samples were then preserved in 10% formalin. The number of samples for histological analysis was 20 individuals, while for morphometric and meristic analysis there were 8 individuals. The results of the research show that morphologically the lempuk fish has a transparent body shape and there is a central line on the body with black or brown pigment and has a total length of around 2-3 cm. Lempuk fish have a type of reproduction that develops asynchronously (asynchronous) because the development of oocytes in the ovaries of female fish shows a variety of developmental stages, namely the initial growth stage (nuclear and perinuclear chromatin), cortical alveolar, vitellogenesis, and the mature stage. The sex ratio is predominantly female with a ratio of 0.64: 0.36. The histological structure of male and female gonads has histological differences that reflect different reproductive functions, where the ovaries play a role in egg cell production, while the testes are responsible for sperm formation. An imbalance in the sex ratio indicates the importance of carrying out domestication steps in order to prevent extinction or continuous population decline.

Abstract

The increasing presence of microplastic contamination in shrimp ponds poses a serious threat to the health of whiteleg shrimp (Litopenaeus vannamei). This study aimed to analyze the relationship between microplastic abundance in gills and intestines with the level of histopathological damage in whiteleg shrimp cultivated in ponds located in Probolinggo, East Java, Indonesia. A quantitative descriptive method was used with Spearman correlation analysis. Shrimp samples were collected from three pond locations and histologically examined to assess tissue damage scores. The results showed a very strong and significant correlation between microplastics abundance and gill damage (r = 0.815; p-value = 0.007), intestinal degeneration (r = 0.885; p-value = 0.002), necrosis (r = 0.804; p-value = 0.009), and inflammation (r = 0.688; p-value = 0.041). These findings indicated that microplastics significantly contribute to structural damage in the respiratory and digestive organs of shrimp. This study highlighted the importance of effectively managing microplastic pollution in sustainable aquaculture systems.

Abstract

Intensive ponds often experience a decrease in water quality and cause ponds to bloom due to the fertility of the pond bottom due to the buildup of organic material originating from leftover feed, feces and the remains of dead shrimp. The aim of this research is to determine the relationship between water quality and phytoplankton abundance in intensive pond types with different densities in Bayeman Village, Probolinggo Regency. The research was carried out from January to March 2023. Samples were taken using the purposive sampling method in 6 different pond plots. Phytoplankton analysis was carried out at the Laboratory of the Sand Source Freshwater Fisheries Technical Implementation Unit and the Hydrobiology Laboratory of the Fish Resources Section of the Faculty of Fisheries and Marine Sciences. Measurement of in situ water quality parameters, namely temperature, pH, brightness, DO, TDS, light intensity, salinity and ex situ nitrate, phosphate and ammonia using spectrophotometric methods. The correlation test results of physico-chemical parameters related to phytoplankton abundance have a strong and heterogeneous relationship only with the orthophosphate parameter

Abstract

Brantas River is habitat to a fish species known as the bader fish (Barbonymus altus). Research aims to evaluate the hematological profile of the bader fish as a means to assess the quality of the Brantas River in the Blitar region. Determining the waste flow from community activities that influences blood quality of bader fish is another goal of this research. To achieve this, various water quality indices were measured in the river and hematological parameters measured in the fish. Data collection was conducted once every week for three weeks during 17 January to 17 March 2022. The measurements aimed to observe any changes in the fish's hematological profile over this period. Two data analysis techniques were employed for the research: Pollution Index (IP) and Canonical Correlation Analysis (CCA). IP measurements reveal that all three stations share a common assessment, classifying them as having light pollution. When analyzing correlation measurements through CCA, it was found that erythrocytes exhibited a robust positive correlation with a pH value of 0.549, while conversely, DO (Dissolved Oxygen) showed a strong negative correlation with leukocytes with value -0.717. In summary, the water conditions in the Brantas region remain conducive for the habitat of bader fish.

Abstract

The Brantas River has many functions for both humans and the aquatic organisms in it. The type of fish that is often found in the Brantas River in Surabaya is the red bader fish (Barbonymus altus). Hematology is a sensitive parameter and can be used as a marker in assessing water conditions. The aim of this research is to analyze the hematological profile of red bader fish (Barbonymus altus) to assess the condition of the Brantas River, Surabaya. The research was carried out at 3 locations on the Brantas River in Surabaya with 3 sampling times with an interval of 1 month starting in January – March 2022. The parameters observed were water quality (temperature, TSS, TDS, pH, DO, BOD, ammonia and heavy metals Hg) and hematological parameters (erythrocytes, hemoglobin, hematocrit, leukocytes and micronuclei). Analysis of the relationship between water quality parameters and hematology using the Correspondence Correlation Analysis (CCA) method. The results show that there are water quality parameters that do not meet quality standards and there are abnormal hematological parameters. CCA analysis shows that there is a correlation between water quality and hematological parameters with moderate concentrations (erythrocytes, hemoglobin and hematocrit) and strong concentrations (leukocytes and micronuclei).
