Abstract

Penanganan benih jabon merah oleh pengelola persemaian permanen Anduna, Konawe Selatan, Sulawesi Tenggara masih terbatas. Oleh karena itu, perlu upaya peningkatan pengetahuan para pengelola melalui bimbingan teknis. Tujuan pelaksanaan bimbingan teknis adalah untuk meningkatkan pengetahuan dan kapasitas pengelola persemaian permanen tentang ekstraksi dan perkecambahan benih jabon merah. Bimtek ini telah dilaksanakan pada tanggal 11 Mei 2024 bertempat di Persemaian permanen Anduna, Konawe Selatan dengan jumlah peserta 30 orang. Metode yang digunakan dalam program ini adalah metode Technology Transfer. Hasil pengabdian menunjukkan bahwa terjadi peningkatan pengetahuan dan kapasitas pengelola persemaian terkait pengenalan jabon merah, teknik ekstraksi dan perkecambahan benih jabon merah. Selain itu, tersedia benih jabon merah untuk kebutuhan produksi bibit jabon merah di persemaian permanen BPDAS Sampara.han produksi bibit jabon merah di persemaian permanen BPDAS Sampara.Abstarct. Handling of red jabon seedlings by managers of the Anduna permanent nursery, South Konawe, Southeast Sulawesi is still limited. Therefore, efforts are needed to increase managers' knowledge through technical guidance. The aim of implementing technical guidance is to increase the knowledge and capacity of permanent nursery managers regarding extraction and germination of red jabon seeds. This technical guidance was carried out on 11 May 2024 at the Anduna permanent nursery, South Konawe with a total of 30 participants. The method used in this program is the Technology Transfer method. The results of the service show an increase in the knowledge and capacity of nursery managers regarding the introduction of red jabon, extraction techniques and germination of red jabon seeds. Apart from that, red jabon seeds are also available for the production of red jabon seedlings at the BPDAS Sampara permanent nursery.

Abstract

Native arbuscular mycorrhizal fungi (AMF) have been shown to effectively enhance the growth and nutrient uptake of Vitex cofassus grown on post-asphalt mining soils. However, their potential needs to be further evaluated across various mining substrates and different tree species. This study aimed to assess the effects of native AMF inoculation on the early growth performance of Albizia saponaria seedlings cultivated on post-asphalt and post-nickel mining soils under greenhouse conditions. A completely randomized design was employed with seven treatments: uninoculated control, Racocetra crispi, Glomus intraradices, Glomus sp., Glomus sp.-LW10, Glomus sp.-SW10, and Mycofer IPB/AMF mixture. Parameters observed after three months of growth included plant height, AMF root colonization percentage, total dry weight, mycorrhizal inoculation effect (MIE), and the uptake of phosphorus (P), nitrogen (N), calcium (Ca), and nickel (Ni). The results indicated that native AMF inoculation significantly improved early growth and total dry weight of  A. saponaria seedlings. The highest MIE values were observed in the  G. intraradices treatment, reaching 92.73% and 82,9%. Native AMF inoculation also enhanced the uptake of P, N, Ca, and Ni in plant tissues. These findings suggest that local AMF isolates can be effectively utilized to improve seedling quality and accelerate the success of revegetation programs on degraded post-mining lands in Indonesia.

Abstract

This study aimed to investigate the effect of gold mining on soil properties. Soil samples were taken from the post-gold mining land, the property of PT Panca Logam Nusantara and PT Alam Buana Indonesia, and a nearby natural forest in Bombana, Southeast Sulawesi Province. The next step focused on specifying soil pH, total nitrogen (TN) and carbon (TC) concentration, C/N ratio, available phosphorus (P) concentration, cation exchange capacity (CEC), and exchangeable K, Na, Mg, Ca, Fe, Mn, Cd and Pb concentration, texture and spore amount, AMF resource and AMF colonization. The result shows that the pH in post-gold mining soil was higher than that in natural forest soil. Meanwhile, TN, TC, available P, and CEC of post-gold mining soil got lower compared with these of natural forest soil. The texture in the post-mining soil was clay loam, while that in natural forest soil was clay. Total of 10 AMF species belonging to five genera and three families were found in a post-gold mining area. Soil pH, CEC, soil texture, Mn, and total Fe had a negative relation with AMF colonization and spore count, while organic C, total N, C/N ratio, P2O5 and silt had a positive relation. Sand was proven to have a strong and positive correlation with the amount of AMF species. Adding organic matter and fertilization as well as applying mycorrhizal biofertilizers, were urgently required to support the effort in restoring post-gold mining soil.

Abstract

Kayu putih merupakan salah satu tanaman dari famili Myrtaceae yang dapat menghasilkan minyak atsiri. Jenis ini dapat dikembangkan sebagai tanaman rehabilitasi lahan yang dapat diintegrasikan dengan produksi minyak kayu putih. Untuk mendukung keberhasilan penanaman maka perlu didukung oleh input pemupukan. Tujuan dari penelitian ini adalah mengetahui respon pertumbuhan tanaman kayu putih terhadap pemberian campuran pupuk kandang sapi dan arang sekam padi. Penelitian dilaksanakan selama 6 bulan (Juli-Desember tahun 2022) bertempat di Kebun Raya Universitas Halu Oleo, Kecamatan Kambu, Kota Kendari, Provinsi Sulawesi Tenggara. Rancangan penelitian mengunakan Rancangan Acak Kelompok (RAK) yang terdiri atas empat taraf yaitu kontrol, 1 kg, 2 kg, dan 3 kg campuran pupuk kandang dan arang sekam dengan masing-masing perlakuan diulang 3 kali . Hasil penelitian menunjukan bahwa pemberian pupuk kandang sapi dan arang sekam meningkatkan pertumbuhan tanaman kayu putih, dimana takaran pupuk kandang sapi 3 kg efektif meningkatkan pertambahan jumlah daun dan diameter batang pada tanaman kayu putih umur 6 bulan di kebun raya UHO.

Abstract

Arbuscular mycorrhizal fungi (AMF) are one type of mycorrhizal association with plant roots. AMF have a fungal structure in the form of arbuscules, vesicles, and hyphae, which are found in roots and spores in the soil. The research aimed to determine the diversity of arbuscular mycorrhizal fungi on the rhizosphere of Xanthostemon petiolatus in the Botanical Gardens of Universitas Halu Oleo (UHO), Kendari. Studies that have been conducted show that Nona wood has a symbiotic relationship with AMF. The soil and plant root sampling method was carried out at the Botanical Gardens UHO Kendari, randomly (non-proportionally) based on the distribution of plants. Isolated plant roots and identification of AMF were carried out at the Plastic House of the Indonesian Mycorrhiza Association (AMI) Kendari Branch, Southeast Sulawesi Province, which took place during May-June 2024. The results of the research showed that X. petiolatus roots were colonized by AMF, as evidenced by the discovery of internal hyphae and external hyphae. A total of 6 species of AMF from the genus Glomus (5 species) and the genus Acaulospora (1 species) were found. The dominant type is Glomus. Diversity is included in the low category (H ' = 0.235). This type of AMF has the potential to be developed as a biofertilizer.

Abstract

The propagation of Racocetra crispa using tree-like legume host plants has not yet been widely undertaken. This study aims to investigate the sporulation of R. crispa in single-spore cultures using legume tree host plants, namely sengon (Paraserianthes falcataria), albizia (Albizia saponaria), johar (Cassia siamea), cajanus (Cajanus cajan), and lamtoro (Leucaena leucocephala). The study was conducted at the Greenhouse of the Indonesian Mycorrhiza Association (AMI) Southeast Sulawesi Branch and the Forestry Department Laboratory of Halu Oleo University from November 2025 to February 2026. The study employed a completely randomised design (CRD) with 5 treatments and 3 replicates. The variables observed included culture success, AMF colonisation of the roots, and the number of spores formed at 3 months, 5 months, and after drying 10 g of medium. The results of the study showed that the A. saponaria host plant treatment yielded the best results, with a culture success rate of 86.67%, the highest AMF colonisation of 35.83%, and the highest number of spores at 5 months and after drying 10g of medium. Conversely, L. leucocephala showed the lowest results, with a culture success rate and spore count of 0%. The best host plant for the propagation of Racocetra crispa is A.saponaria.

Abstract

Waterlogged condition is one of the environmental stresses unfavorable for plants and mycorrhizal fungi. Arbuscular Mycorrhizal (AM) species that are tolerant tend to be more adaptive to waterlogged conditions. Besides increasing plant tolerance, the documented effects of AM fungi (AMF) inoculation on wetland plant performance remain inconsistent, and AMF application may also cause plant growth depression under waterlogging. This study aimed to examine the morpho-anatomical adaptation, survival, growth, biomass, and nutrient uptake of Nauclea orientalis (L.) seedlings from different habitats inoculated with Arbuscular Mycorrhizal Fungi under waterlogged conditions for 90 days. Experimental procedures included seed germination, preparation of AMF inoculum, inoculation, and waterlogging treatment. The results showed that N. orientalis seedlings formed lenticels (100%), adventitious roots (41%), and aerenchyma under waterlogged conditions. Seedlings originating from dryland habitats, both mycorrhizal and non-mycorrhizal, had lower height growth rate compared to those from other habitat types. Similar patterns were observed in relative growth rate for shoot (RGRs), root (RGRr), total (RGRt), and root-to-shoot ratio. Treatments of swamp habitat seedlings without waterlogging and with waterlogging, as well as savannah seedlings under waterlogged conditions, showed higher total plant dry weight and RGRt than other treatments. In general, total nitrogen concentration in roots and shoots was highest in the interaction between swamp habitat seedlings and non-mycorrhizal treatment under waterlogging. However, seedlings from dryland habitats required AMF under waterlogging to improve biomass and nitrogen accumulation in roots. Independently, waterlogging increased average height gain (18%), stem diameter (46%), leaf area (40%), leaf length (17%), and leaf width (21%). Nonetheless, waterlogged treatment decreased the number of leaves (9%) and AMF colonization (71%). The results indicate that seedlings from temporary swamp habitats demonstrated better growth performance and higher biomass under waterlogged conditions, regardless of AMF inoculation.

Arbuscular mycorrhizal fungi (AMF) are fungi from division Glomeremycota within the kingdom Fungi, which have a broad ecological range and symbiosis with a variety of vegetations, including vegetations in gold post-mining areas or gold tailings. The study of identifying types of AMF in gold tailings in Southeast Sulawesi is still limited. The purpose of this study was to identify the types of AMF in the rhizosphere of adaptive plants in gold tailings land in Bombana District, Southeast Sulawesi. Soil samples were collected from adaptive plants in Perseroan Terbatas (PT) Panca Logam Makmur, Bombana District, Southeast Sulawesi Province, Indonesia. AM Fungi spores were isolated and identified based on their morphological characteristics (shape, size, color, attached hyphae, and spore ornamentation). Five types of AMF were identified, namely Glomus aggregatum, Sclerocystis sinuosa, Acaulospora scrobiculata, A. tuberculata, Scutellospora pellucida, and two genera that had not been identified up to species, Glomus sp. and Scutellospora sp. In addition to spores, the symbiosis of AM Fungi with adaptive plants was also demonstrated by the structure of AM Fungi in plant roots with a rate of 74% colonization. S. pellucida and S. sinuosa were recorded as new species in Indonesia and Sulawesi, respectively. While A. tuberculata is a type of AMF that enriches AMF diversity in Southeast Sulawesi.

Arbuscular mycorrhizal fungi to enhance the growth of tropical endangered species Pterocarpus indicus and Pericopsis mooniana in post gold mine field in Southeast Sulawesi, Indonesia. Biodiversitas 22: 3844-3853. Gold mining activities contribute to the national economy, but have a serious impact on forest and environmental degradation and pose a threat to tree species in the tropics. Reforestation of post-gold mining with tropical legume tree species is threatened with extinction. Arbuscular mycorrhizal fungi are effective in conservation of endangered plants and restoration of degraded land. The objective of the study was to evaluate the effect of native AM Fungi inoculation on the growth of Pterocarpus indicus and Pericopsis mooniana in nurseries and post-gold mining fields. P. indicus and P. mooniana seedlings were inoculated with Glomus claroideum Schenk & Smith, Glomus coronatum Giovann., and mixed AMF (G. claroideum, G. coronatum). Uninoculated seedlings were used as control treatment, and they were maintained for 4 months under greenhouse conditions. After 4 months in greenhouse, seedlings were transferred to post-gold mine and planted for 4 months. The percentage of AMF colonization, plant growth, nutrient content and uptake of N, P, K, Fe, Mn were measured after 4 months both in the greenhouse and the field. The percentage of AMF colonization under greenhouse conditions in P. indicus and P. mooniana ranged 5.67-75.3% and 2.2-41.2%. All AMF colonization tended to have higher shoot height, leaf numbers and nodules, plant dry weight and N, P, K content under greenhouse conditions. Shoot height, stem diameter, leaf dry weight, N, P, K, Mn and Fe under field conditions had higher inoculated seedlings than control four months after planting. AMF could be used to conserve endangered tree species in post-gold mining reforestation in the tropics.

Effect of pre-sowing treatment on the germination and early growth of Kalappia celebica Kosterm.: an endemic and vulnerable tree species of Sulawesi, Indonesia. Biodiversitas 23: 4297-4302. Kalapi (Kalappia celebica) is an endemic tree species of Sulawesi and is threatened with extinction, so it requires conservation efforts to preserve the tree species. The research aims to determine the effects of seed pre-sowing methods on the germination of K. celebica. Matured seeds of kalapi were collected from several mother trees at the Lalona Village, Kolaka Regency, Southeast Sulawesi, Indonesia. The seeds were treated with six pre-germinations treatments (control, nicking, nicking and soaking in cool water (20°C) for 12 h, nicking and soaking in cool water for 24 h, soaked in hot water (80°C) for 24 h till the water-cooled and soaked in cool water for 48 h. Three replication and each replication using 25 seed were used in this research, so the total seed were 450 seeds. The randomized completely design was used to test the effect of pre-germination treatments on seed germination parameters at the greenhouse. The germination parameters calculated were the first day of germination, last day of germination, germination period, germination capacity, mean germination time (MGT), and mean daily germination (MDG). The results showed that pre-germination treatment affected all seed germination parameters, which significantly increased the germination capacity. The germination capacity with the nicking was 94,67%, nicking and soaking in cool water (20°C) for 12 h (94,67%) followed by nicking and soaking in cool water (20° C) for 24 h (90,67%). In all treatments, germination started 5 to 10 days after sowing and was completed from 4 to 16 days later. The seed treatment by nicking and soaking in cool water for 12 d exhibited the lowest MGT (5.56 days), the highest MDG (0.95 germinate day-1), the highest seedling collar diameter (1.64 mm), and the highest leaf number (1.95). Therefore, the study recommends nicking followed by soaking in cool water for 12 h as an ideal pre-germination treatment to promote germination and seedling growth of kalapi tree species for supported conservation activity.
