Abstract

Wheat is an important agricultural commodity in Indonesia, yet the country remains heavily reliant on imports to meet domestic demand. The diverse agroclimatic conditions in the highlands and lowlands pose significant challenges for national wheat production. This study aimed to evaluate the performance of wheat lines in two distinct altitude environments: Cipanas (1120 m above sea level) and Cisarua (600 m above sea level).  An augmented randomized complete block design (RCBD) assessed 50 F6 wheat lines derived from the HP1744/”Selayar” cross. The results revealed significant variability in agronomic traits attributable to altitude differences. However, genotype-by-environment (GxE) interactions were found to be non-significant for all evaluated characters. Broad-sense heritability was classified as medium for the grain-filling period in Cipanas and seed weight of the main panicle in Cisarua, while all other traits exhibited low heritability. Based on the stress sensitivity index (SSI), 14 lines (28%) were identified as tolerant to higher temperatures in the medium-altitude environment, particularly in terms of seed weight of the main panicle. These findings are of utmost importance as they underscore the need to consider genetic and environmental factors in developing wheat cultivars suitable for Indonesia’s diverse agro-climatic conditions.

Abstract

Somatic embryogenesis, a crucial plant regeneration method, has become indispensable for crop improvement, particularly for species reliant on somatic cell manipulation techniques. Optimization of this process necessitates an understanding of the developmental stages involved. This study investigates the histological aspects of somatic embryogenesis in Musa acuminata ssp. malaccensis derived from immature zygotic embryos. Through detailed histological analysis, we aimed to elucidate the morphological changes and cellular organization occurring during the various stages of somatic embryogenesis, from induction, culture proliferation, and somatic embryo development to plantlet conversion. The initial stages of embryogenesis, characterized by nodules, were primarily composed of meristematic cells with high cell division activity. These cells contained tetrad-like structures that could develop into distinct two- and four-celled proembryoids or proembryogenic aggregates. Our histo-anatomical analysis revealed that embryogenic cultures proliferated through multiple pathways simultaneously: somatic embryo budding, proembryo formation, and pro-embryonic mass formation from both internal and peripheral cells. At the stage of somatic embryo development, embryos with a well-defined protoderm layer, containing cells with prominent nuclei and dense cytoplasm, potentially regenerate into plantlets. Furthermore, histological examination revealed the presence of procambium within mature somatic embryos, which subsequently developed into the vascular system of the complete plantlet

Abstract

Durio testudinarius, known as Durian Kura-Kura, is a native Bornean durian species in the Malvaceae family with fruit at the base of the main stem. Recent advances in genomic-based next-generation sequencing are being used to conserve germplasm and enhance plant breeding. This method rapidly and cost-effectively sequences plant genomes, allowing for simple sequence repeat markers to be constructed in silico. This study aimed to design and evaluate novel, simple sequence repeat markers utilizing next-generation sequencing microsatellite data of D. testudinarius. A total of 20 sequences containing di-, tri-, tetra-, and penta-nucleotide motif repeats were selected and designed to generate primers using Primer3 online software. The PCR results of the twenty primers tested on D. testudinarius and 19 genomic DNA samples of species from the Malvales order showed successful amplification. These newly developed simple sequence repeat markers are crucial for genetic population analyses, determining population structure, and enhancing plant breeding programs and conservation strategies for D. testudinarius and its relative's germplasm resources.

Abstract

Wild banana Musa acuminata ssp. malaccensis, an ancestor of cultivated bananas, possesses valuable genetic diversity, including resistance genes to fusarium wilt, and demonstrates high environmental adaptability. These traits are important for pre-breeding programs, whether by conventional breeding, which is a lengthy process, or by taking advantage of somatic cell manipulation techniques such as somatic hybridization, which requires an efficient plant regeneration system like somatic embryogenesis. We have established an efficient and comprehensive protocol for somatic embryogenesis of this wild Musa using immature zygotic embryo explants covering culture induction and proliferation, somatic embryo development, and subsequent plant conversion. Embryogenic culture was induced on a complex-modified MS medium supplemented with 1 mg.L-1 2,4-D auxin or 5 mg.L-1 picloram. The embryogenic cultures proliferated in the form of granular or nodular structures, which was best obtained by reducing the picloram concentration to 1 mg.L-1 and combining it with the same concentration of 2,4-D at a half-strength macro-nutrient of basal medium. Embryo development from embryogenic cultures and regeneration into shoots. Proembryos as granular structures spontaneously matured into early-stage somatic embryos upon withdrawal of the strong auxin inducer. Increasing the sucrose and gelling agent concentrations in the growth medium improved somatic embryo formation from embryogenic cultures. The frequency of shoot formation from developed somatic embryos was increased by incorporating 0.5 mg.L-1 BA and 0.5 mg.L-1 GA3 into the regeneration medium.

Abstract

The selection of the early generation is a viable strategy for the rapid development of new plant varieties. In mungbeans (Vigna radiata L. Wilczek), the selection process is carried out to obtain high seed yield, early maturity, and sufficiently large seed size. To enhance this process, the extension of the combined-eigenanalysis selection index method (CESIM) with transgressive segregation variables can be used to fix multiple-trait transgressive segregates. Therefore, this research aimed to (1) develop an eigenvalue-based selection index method in information from relatives analysis capable of fixing multiple-trait transgressive segregates in the early generation, (2) predict expected and realized multiple-trait selection responses, and (3) verify the presence of multiple-trait transgressive segregates in the early generation of mungbeans. The material used consisted of the F3 generation population and its selection outcomes in F4, originating from the crossbreeding of mungbean varieties Gelatik × Lasafu Lere Butsiw. The empirical breeding values (EBV) between and within families were obtained using the BLUP values of the F3 generation from the mixed linear model with a nested design and log-normal distribution. The EBV values between families and dummy variables of transgressive segregate families were used in the selection process with CESIM. Furthermore, analysis of variance in EBV values within families in the F4 generation was applied to verify the presence of multiple-trait transgressive segregates. The results showed that the selection from the best CESIM equation yielded index scores with a determination coefficient R2 = 97.76% and an expected selection response of 136.62. The verification process confirmed the presence of 9 families as multiple-trait transgressive segregate families in the early generation.

Abstract

Kacang bambara merupakan salah satu jenis legum yang dibudidayakan di Indonesia dan telah dikembangkan untuk mendapatkan genotipe unggul melalui seleksi galur murni. Tujuan penelitian ini adalah mengevaluasi keragaan karakter kuantitatif dan kualitatif galur-galur kacang bambara asal lanras Sumedang serta untuk memperoleh korelasi antar karakter pada galur-galur tersebut. Penelitian ini dilaksanakan di Kebun Percobaan Cikarawang pada Januari–Juli 2022. Rancangan penelitian ini adalah RKLT dua ulangan dengan 14 galur asal lanras Sumedang, satu galur asal lanras Sukabumi dan satu populasi Sumedang yang belum diseleksi. Satu galur asal Sukabumi telah diseleksi galur murni dan digunakan sebagai pembanding. Hasil penelitian ini menunjukkan bahwa tinggi tanaman, umur berbunga, bobot 100 biji berbeda nyata antar galur yang diamati. Bobot 100 biji memiliki nilai duga heritabilitas arti luas kategori tinggi. Koefisien keragaman genetik seluruh karakter berada pada kategori rendah–sedang. Karakter-karakter pada komponen hasil saling berkorelasi positif. Umur berbunga berkorelasi negatif terhadap karakter tinggi tanaman, jumlah daun dan lebar kanopi pengukuran pertama, bobot polong total, bobot polong bernas, dan bobot 100 biji. Tipe kanopi, bentuk daun, bentuk polong, warna polong, tekstur polong serta warna dan corak biji menunjukkan keragaman antara galur uji dengan galur pembanding. Abstract

Indonesia is rich in biodiversity, including rice (Oryza sativa L.), one of the most important and widely cultivated crops. However, the utilization of local Indonesian rice germplasm in breeding programs remains limited due to the lack of reliable characterization data. Therefore, it’s essential to establish a reliable method to ensure variety protection by breeders. In addition, understanding the relationship between morphological traits and genetic diversity is crucial for a targeted breeding study aimed at evaluating genetic diversity among 12 local rice genotypes from Indonesia through agronomic traits and molecular analysis using start codon targeted (SCoT) markers. Statistical analyses, including cluster and diversity indices, were applied to interpret the data. Characterization was conducted based on the descriptors from The International Rice Research Institute (IRRI) and Bioversity International, while molecular analysis employed 23 selected SCoT primers. The effectiveness of the markers was assessed through polymorphism percentage, PIC values, and cluster analysis. A polymorphism level of 69% was observed, with PIC values ranging from 0.3610 to 0.4646, suggesting that the SCoT markers used provided adequate informativeness for diversity analysis. The study revealed high genetic variability, with heritable traits and informative SCoT markers, demonstrating their potential for pre-breeding and diversity analysis.

Abstract

Java’s tea plants have efficacy to treat diabetes. Sinensetin is a chemical compound in Java’s tea that acts as diabetes medicine. The experiment objective was to study diversity of agronomy characters and sinensetin content of ex situ collections of Java’s tea from West, Central, and East Java regions. Java’s tea was harvested when population has flowering about 70% and tested sinensetin content on leaf part. The experiment was conducted with a completely randomized block design with 18 accessions and one control (purple flower accession). The result of study showed that there were significant differences in the accumulation of height plant increase while 8 weeks after planting, secondary branches and secondary branches internode number; leaf length, width and area index; average stems and leaf dry weight per 4.41 m2 and sinensetin levels among some accessions. Banyumas 1, Sumbersari and Kraksaan accessions have agronomy and production characters except for sinensetin levels were better than control (Leaf sinensetin levels based on leaf dry weight increased from low to medium but decreased in high category) Based on comparison between accession and leaf dry weight category, purple flower accession has higher sinensetin levels (0.043%) than another accession, but only reaches 43% of Indonesian Herbal Pharmacopoeia minimum standard (There were no significant differences in sinensetin accession production tested either by comparison of accession or accession categories). Abstract

Perakitan suatu varietas tanaman memerlukan informasi genetik mengenai sifat yang diinginkan. Salah satu cara untuk memperoleh informasi genetik yaitu melalui pendugaan komponen genetik dan nilai heritabilitas. Percobaan ini yang bertujuan memperoleh informasi komponen ragam dan nilai heritabilitasnya pada 2 populasi tanaman cabai rawit. Masing-masing populasi P1, P2, dan F1 ditanam dengan jumlah 40 tanaman, BCP1 dan BCP 2 sebanyak 100 tanaman dan F2 sebanyak 300 tanaman. Hasil pada populasi IPB C285 x IPB C290 menunjukkan nilai heritabilitas arti luas kategori tinggi untuk semua karakter kecuali pada karakter diameter buah. Jumlah buah per tanaman dan tebal daging buah memiliki nilai heritabilitas arti sempit kategori tinggi. Hasil pada populasi IPB C321 x IPB C290 menunjukkan heritabilitas arti luas kategori tinggi untuk semua karakter kecuali karakter bobot per buah. Karakter panjang buah termasuk dalam kategori tinggi untuk heritabilitas arti sempit. Rasio ragam aditif lebih besar daripada ragam non aditif pada populasi IPB C285 x IPB C290 untuk karakter jumlah buah per tanaman, bobot per buah, panjang buah, diameter buah, tebal daging buah dan umur panen sedangkan pada populasi IPB C321 x IPB C290 yaitu karakter jumlah buah per tanaman, panjang buah, tebal daging buah dan diameter buah. Metode seleksi yang bisa digunakan pada kedua populasi ini yaitu metode pedigree sehingga kegiatan seleksi bisa dilakukan pada generasi awal. Populasi IPB C285 x IPB C290 metode seleksi pedigree dilakukan pada karakter jumlah buah per tanaman dan bobot per buah sedangkan pada populasi IPB C321 x C290 metode seleksi pedigree dilakukan pada karakter panjang buah. Abstract

Indonesia merupakan negara produsen serta konsumen buah melon (Cucumis melo L.) yang tinggi, tetapi impor benih melon dari luar negeri masih cukup besar. Hal tersebut membuka peluang pemulia tanaman untuk merakit varietas hibrida unggul. Varietas melon hibrida yang diharapkan adalah yang memiliki kualitas buah yang baik dan produksi yang tinggi. Penelitian ini bertujuan mempelajari karakteristik beberapa genotipe melon dan memilih genotipe potensial untuk materi perakitan varietas hibrida baru. Genotipe yang digunakan ialah enam genotipe dari koleksi Pusat Kajian Hortikultura Tropika (PKHT) IPB (IPB G1, IPB G30, IPB G41, IPB 240, IPB 283, Glamour-S1), tiga galur inbrida PKHT IPB (IPB M13, IPB M23, IPB M21), dan satu varietas pembanding (Alisha F1). Genotipe IPB G1 dan IPB 240 memiliki keunggulan penampakan buah menarik dengan keunikan berupa corak pada kulit buah genotipe IPB G1 dan jala yang tebal dengan intensitas tidak terlalu rapat pada genotipe IPB 240. Genotipe IPB G41 memiliki kandungan padatan terlarut total yang tinggi dan rasanya sangat manis. Genotipe Glamour-S1 memiliki keunggulan penampakan buah menarik serta buah yang besar. Karakter bobot buah berkorelasi positif dengan diameter buah dan panjang buah. Kandungan PTT berkorelasi positif dengan umur panen. Berdasarkan analisis korelasi, genotipe potensial adalah yang memiliki ukuran buah yang besar dan umur panen yang cukup. Genotipe yang memiliki potensi sebagai materi pemuliaan adalah IPB G1, IPB 240, IPB G41, dan Glamour-S1. Kata kunci: genotipe potensial, karakteristik melon, pemuliaan melon
