Abstract

ABSTRACTSelection of transgressive segregants is considered as one effective way of obtaining peanut genotypes with high yield on early segregation. The objective of the study was to estimate genetic parameters and to identify transgressive segregants of peanut family in F3 generation. The experiment was conducted at Leuwikopo, Dramaga, Bogor on August-Desember 2014. The genetic materials evaluated were 218 family F3 generation from 5 biparental population (Jerapah/GWS79A1, GWS79A1/Zebra, GWS79A1/Jerapah, Zebra/GWS79A1 and Zebra/GWS18) and 4 commercial varieties as checks (Gajah, Jerapah, Sima, and Zebra). The experiment was conducted in an augmented randomized complete block design with 4 replications for the checks. The results showed that quantitative characters had continuous distribution and they were controlled by many genes with additive, dominance, complementary epistasis, or additive epistasis gene action. Total number of pod and number of pod had high heritability and moderate genetic coefficient of variation. Selection with 10% intensity showed that 22 families had large total number of pods while six families had larger total number of pods than both parents. Family U2-39 estimated was identified as a putative transgressive segregant with high total number of pod and had smaller variance within family than Sima variety. Abstract

Oil palm is a perennial oil crop that contribute the most important source of vegetable oil in the world. Oil palm breeding cycle take a long period, therefore molecular marker-assisted selection (MAS) is required to shorten the selection time. This MAS require the associated marker to desired trait, particularly for yield and the component that as a complex trait and depend on genetic background. The aim of this study was to obtain SSR marker that associated with average of bunch weight (ABW). Plant material was used in this study is Tenera population derived from Deli Dura x AVROS Pisifera. Seven SSR markers from public database were associated to ABW trait. Molecular data analysed for SSR markers profile which consisted of allelic diversity, heterozygosity level, and polymorphism information content (PIC). Association between SSR markers to ABW traits was performed with single marker analysis using one way analysis of variance. The results showed that SSR markers were able to amplify DNA with two to three number of alleles with the average of 2.3 alleles per locus. ABW trait for this population was significantly associated with mEgCIR3428 marker. This marker may be used in the selection of Deli Dura and AVROS Pisifera palm on the next selection cycle.Keywords: Average of bunch weight, AVROS Pisifera, Deli Dura, single marker analysis

Abstract

ABSTRACTThe fruit of ornamental chili functioned as ornamental as well as for consumption, and therefore it requires diversity of traits for selection process. Information on inheritance pattern of the traits are needed for effective selection. The ideotype of ornamental chili are shortened internode that form a bouquet of flowers, erect fruit orientation and contain anthocyanins for attractive appearance. The research was aimed to study qualitative characters inheritance associated with shortened internode, fruit orientation and anthocyanins content. This study used 6 population including female parent (P1) which has anthocyanin in flower parts, the male parent (P2) bird pepper which has the character of a shortened internode, F1, F1R, BCP1, BCP2, and F2. Data were analyzed with Chi-square test to determine the Mendelian ratio in the F2 population. The results showed that there were characters that was controlled by one gene or two genes. Shortened internode and erect fruit orientation were controlled by a single recessive gene with a ratio of 1:3. On the other hand the color of anthocyanin in the anther stem was controlled by one dominant gene with a ratio of 3:1. The characters controlled by two genes that were dominant and recessive epistasis was the color of anthocyanins in the anther with the ratio 13:3.Keywords: Mendelian ratio, antosianin, shortened internode

Abstract

Penelitian tentang respon cabai rawit terhadap salinitas perlu dilakukan untuk pengembangan varietas yang toleran salinitas. Penelitian ini bertujuan untuk mengetahui respon morfologi dan fisiologi empat genotipe cabai rawit pada kondisi cekaman salinitas. Penelitian dilakukan di kebun percobaan Tajur 2 IPB Bogor pada bulan Oktober 2019-Maret 2020. Rancangan kelompok lengkap teracak dua faktor dengan lima kelompok sebagai ulangan digunakan dalam percobaan ini. Faktor pertama yaitu genotipe cabai rawit (Sigantung, CR10, CSR1, dan CRK1) dan faktor kedua adalah taraf salinitas (0-1, 2-4, 5-7, dan 8-10 mS cm-1). Hasil yang diperoleh menunjukkan bahwa salinitas secara signifikan berpengaruh terhadap peubah morfologi dan fisiologi tanaman cabai rawit. Salinitas 8-10 mS cm-1 pada peubah morfologi menyebabkan penurunan hasil tertinggi pada jumlah bunga (74.65%), jumlah daun (71.23%), bobot buah (58.48%), dan panjang akar (49.81%). Selain itu, pada konsentrasi 8-10 mS cm-1 juga menyebabkan penurunan hasil tertinggi pada peubah fisiologi. Penurunan hasil tertinggi terjadi pada konduktansi stomata (29.37%), laju transpirasi (26.47%), konsentrasi CO2 interseluler (21.83%),dan indeks klorofil (20.19%). Berdasarkan nilai rata-rata indeks sensitivitas cekaman (ISC), genotipe Sigantung (1.2) dan CSR1 (1.0) termasuk katagori peka salinitas (ISC >1.0). Akan tetapi, CR10 dan CRK1 memiliki nilai rata-rata ISC sebesar 0.8, sehingga termasuk ke dalam katagori moderat toleran salinitas (0.5<ISC<0.1). Abstract

Upaya untuk menghasilkan varietas gandum tropika dengan potensi hasil baik dan adaptif di agroekosistem Indonesia terus dilakukan. Tujuan penelitian ini adalah memperoleh informasi tentang keragaan dan mengukur nilai parameter genetik genotipe F2:3 gandum, Materi genetik yang digunakan adalah galur-galur segregan transgresif putatif generasi F2:3 hasil seleksi pada generasi F2 dari famili G1/Se, G2/Se, G3/Se, dan Ja/Se. Penelitian dilaksanakan pada bulan Juni hingga November 2018 di Kebun Percobaan Balai Penelitian Tanaman Hias, Cipanas, Jawa Barat (1,100 m dpl). Penelitian disusun dalam rancangan kelompok lengkap teracak dengan perbesaran. Lima varietas tetua digunakan sebagai pembanding. Pengamatan dilakukan pada karakter agronomi dan komponen hasil. Keempat famili F2:3 gandum menunjukkan keragaan yang melebihi tetua terbaiknya pada karakter panjang malai, jumlah biji per tanaman, dan jumlah anakan produktif. Nilai duga heritabilitas arti luas dan nilai koefisien keragaman genetik yang tinggi terdapat pada karakter jumlah anakan produktif, jumlah biji malai utama, bobot biji malai utama, bobot 100 butir, jumlah biji per tanaman dan bobot biji per tanaman. Hasil verifikasi menunjukkan tidak terdapat galur-galur yang terkonfirmasi sebagai segregan transgresif. Seleksi berdasarkan karakter bobot biji per tanaman menghasilkan 30 galur F2:3 dari empat famili dengan diferensial seleksi mencapai 59%.
