Abstract

Cancer is a type of chronic disease which is the main cause of death in the world with 19.3 million new cases in 2020. Current cancer treatment includes surgery, chemotherapy, radiotherapy, hormone therapy, antiangiogenesis inhibitors, stem cell therapy, and others. Long-term cancer treatment can cause other health problems, so alternative anti-cancer therapy with the fewest side effects is needed. One natural ingredient that is known to prevent and act as anti-cancer therapy is Zingiber officinale. Zingiber officinale contains gingerol which functions as an anti-oxidant, anti-inflammatory, anti-bacterial, anti-cancer, anti-tumor and anti-mutagenic. The anticancer effects of gingerol are known to be effective in cancers of the liver, stomach, mouth, prostate, breast and ovaries. This literature aims to delve deeper into the role of gingerol as an anti-cancer compound. The strategy employed in the article search involved using electronic databases such as Google Scholar and PubMed. Keywords utilized included "cancer," "red ginger," "gingerol," "Zingiber officinale," and "anticancer." The articles selected were those published within the last 10 years. The study results indicate that gingerol is beneficial in oral, breast, lung, colorectal, cervical, and prostate cancers. The mechanism of action of gingerol involves pathways such as the PI3K/AKT, JAK/STAT, apoptosis, and ROS proliferation pathways. Gingerol is known to be effective as an anti-cancer agent and has the potential to become one of the alternative anti-cancer treatments.

Abstract

Breast cancer is the leading cause of death in women. Although hormonal therapy has produced positive results in HR+ breast cancer, resistance to this therapy remains a challenge in the treatment of breast cancer. Various mechanisms have been proposed as an explanation of resistance in breast cancer, one of which is the activation of the PI3K/AKT/mTOR pathway. The PI3K/AKT/mTOR pathway is a transduction pathway that regulates many important aspects in both normal cells and cancer cells, including cell proliferation, apoptosis, cell morphology and migration, protein synthesis, and integrase metabolism. Preclinical dan clinical data show inhibition of this pathway can improve survival, especially in resistant hormonal therapy models. The use of mTOR inhibitors (everolismus) has been approved by the FDA as a combination therapy with exemestane for postmenopausal women with HR+/HER2-breast cancer.

Abstract

Breast cancer has the highest incidence in women, covering 2.3 million new cases per year and a mortality rate of 23%. Therapeutic strategies applied in breast cancer treatment include surgery, chemotherapy, endocrine therapy, and radiation. All of these therapies have relatively high side effects, so alternative therapies are needed that are more effective with minimal side effects. One of the herbs known to be effective in treating breast cancer is Curcumae Rhizoma from Curcumae longa extract. The essential components of this drug are Curcumol and Curcumin. Literature searches were obtained from various online portals such as Pubmed, Google Scholar and Science Direct using the keywords "breast cancer", "Curcumae", "Curcuma Rhizoma" and "herbal medicine". Curcuma Rhizomae can inhibit the formation and spread of cancer cells by utilizing anti-angiogenic effects, disrupting the cell proliferation cycle, and inducing apoptosis. In Indonesia, Curcumae or turmeric is commonly used as a spice ingredient. Its anti-cancer potential can be used as a breast cancer therapy in Indonesia.

Abstract

Dysmenorrhea is a common occurrence in most women. The pain arising from dysmenorrhea ranges from mild to moderate and can disrupt daily activities and performance. Many women practice self-medication to alleviate this pain. This self-medication practice is influenced by several factors, one of which is the level of knowledge. This study aims to explore the relationship between the level of knowledge among female students and self-medication behavior in dysmenorrhea. This study used a cross-sectional design with a random sampling of female students from SMK Negeri 2 Palembang. The total sample involved 41 students. Data were collected and analyzed using chi-square and Mann-Whitney U tests. The majority of students experienced dysmenorrhea (87.8%) with an average Visual Analog Scale (VAS) score of 4.6. The duration of pain varied, with most experiencing pain for 2-3 days after menstruation. Almost all students (97.6%) had good or sufficient knowledge about dysmenorrhea. However, only a few students practiced self-medication (11.1%), with paracetamol being the most commonly used medication. The analysis showed a significant relationship between knowledge level and self-medication behavior (p=0.002, OR: 12.681). The level of knowledge among female students is significantly associated with self-medication behavior in dysmenorrhea. Better health education on dysmenorrhea management and the use of safe medication needs to be strengthened to reduce the risk of side effects and improve the well-being of students.

Abstract

Pulmonary tuberculosis (TB) remains the second leading cause of death from a single infectious agent. The number of people with pulmonary TB in the world still increased, and TB treatment success percentage in Indonesia has not reached the target until now. Genetic polymorphisms are defined as DNA variations seen in ≥1% of the population. The study of genetic polymorphisms can help identify disease risk, predict drug efficacy and safety, and determine the right chemotherapeutic agent with a dose that suits each patient. Various genetic polymorphisms associated with pulmonary TB susceptibility have been found, including polymorphisms of the NRAMP1, HLA-DRB1, vitamin D receptor, TNF-α, and IFN-γ genes. In addition, several genetic polymorphisms associated with TB treatment were also found, such as SLCO1B1, NAT2, SLC2A9, and ABCB1 genes. Detection of genetic polymorphisms in each individual can help predicts TB risk and improve TB treatment success and reduce side effects that may occur due to TB treatment.

Abstract

Background: Gout arthritis is caused by the deposition of monosodium urate crystals in the joints, which can cause significant pain, swelling, and recurring inflammation, particularly in the big toe. Inflammatory mediators are critical to the beginning and duration of the inflammatory response in gout arthritis. Gout is mediated by TNF-α, ILs, and the NLRP3 inflammasome, which promotes macrophages, monocytes, and neutrophils, leading to inflammatory reactions. TNF-α acts as a potent inflammatory mediator by stimulating the expression of other inflammatory cytokines, adhesion molecules, and matrix metalloproteinases (MMPs). TNF-α interacts with particular receptors and activates intracellular signaling pathways, leading to leukocyte recruitment, NLRP3 inflammasome activation, and the release of other inflammatory mediators. Objective: This article is a review article in which the researchers examine the current state of understanding regarding the role of TNF-α as a major inflammatory mediator in gout arthritis, deepen the understanding of TNF-α in gout inflammation, and explore its potential as a therapeutic target. Method: It involvedsystematically searching relevant databases, selecting studies based on inclusion criteria, and synthesizing findings to identify trends, gaps, and the current state of knowledge on the topic. Result: The results of this review article indicated that TNF-α plays a crucial role in joint inflammation by triggering the release of pro-inflammatory cytokines and stimulating neutrophil infiltration into the joints. Conclusion: Thus, Inhibiting TNF-α can reduce inflammation and improve symptoms in people with gout arthritis.
