Abstract

The high cost and limited availability of fishmeal in aquaculture feed formulations have prompted the search for sustainable and affordable alternative protein sources. This study aimed to evaluate the effects of substituting fishmeal with silk worm (Tubifex sp.) meal on the growth performance, feed utilization, and survival rate of Nile tilapia (Oreochromis niloticus) fry. A completely randomized design (CRD) was used with four dietary treatments: P1 (control, 0% Tubifex), P2 (15%), P3 (25%), and P4 (35%), each with three replications. Fish were fed twice daily for 30 days, and growth parameters were measured weekly. Results showed that the inclusion of Tubifex meal significantly improved the final weight, specific growth rate (SGR), average daily gain (ADG), and feed efficiency (FE), particularly in P3 and P4. The highest SGR (2.28%/day), ADG (0.016 g/day), and FE (61.93%) were recorded in the P4 group. Survival rate also improved, reaching 82.22% in P4 compared to 51.11% in the control. Water quality parameters remained within optimal ranges, indicating no adverse environmental impact from Tubifex inclusion. The findings suggest that Tubifex meal is a viable protein source that can replace fishmeal by up to 35% in tilapia fry diets. Its application offers nutritional, economic, and ecological benefits, making it a promising component in sustainable aquafeed development.

Abstract

Microcapsules are small round particles with uniform walls or layers around them that generally have a diameter from 0.2 to 5000 µm. Microencapsulation is a process that converts solid or liquid materials into capsule form which aims to protect and maintain active ingredients from environmental influences such as humidity, high temperature, light exposure, or oxidation processes. Microcapsule formation can be done using the crosslink method. Microcapsule size is affected by the stirring speed of the encapsulator machine. During microencapsulation of fish meal with eggs and gelatin using the crosslink method, the stirring speed is 3000 rpm, 3500 rpm, 4000 rpm, 4500 rpm, and 5000 rpm. In the microencapsulation process of fish meal with eggs and gelatin, the smallest average diameter at a speed of 5000 rpm, which is 18.220 ± 3.3 µm and has a yield of 26% while the largest diameter at a speed of 3000 rpm, which is 33.53 ± 4.8 µm and has a yield of 28.4%. Based on the results of the Tukey SPSS test, the speeds of 3000 rpm (33.53 ± 4.8 µm), 3500 rpm (30.25 ± 4.4 µm), 4000 rpm (26.30 ± 3.5 µm), 4500 rpm (22.50 ± 4.1 µm), and 5000 rpm (18.220 ± 3.3 µm) were very significantly different (P <0.05) in each treatment.

Abstract

The growing demand for catfish in the market is prompting farmers to consistently enhance their production capacity. Several obstacles are encountered during production growth, such as pathogen attacks, suboptimal growth, and low survival rates. This study aimed to determine the effectiveness of Minaraya probiotics with different doses in feed on the growth and immune response of catfish. The method used in this study was an experimental method with a completely randomized design (CRD) consisting of four treatments: control (without probiotics), and treatments P1, P2, and P3 with the addition of Minaraya probiotics at 2, 5, and 10 mL per 2.5 kg of feed, respectively, each with three replications. The parameters observed included survival rate, SGR, DGR, FCR, PER, AF, RB, and leukocyte differentiation. The results showed that the survival rate in all treatments was relatively high, ranging from 95% to 98.89%. The results also showed that the addition of Minaraya probiotics significantly (p<0.05) increased growth parameters (SGR and DGR) and feed efficiency (FCR and PER), with the best dose being in treatment 3 (P3) at a dose of 10 mL/2.5 kg of feed. On the other hand, the non-specific immune response also increased in the treatment given Minaraya probiotics, as indicated by increased phagocytic activity, respiratory burst, and proportion of lymphocytes, neutrophils, and monocytes, as well as an increase in the total number of erythrocytes after Aeromonas hydrophila infection. The application of Minaraya probiotics in a sustainable cultivation system can improve catfish productivity and health.

Abstract

Sekuen gen GH A. bicolor memiliki similaritas nukleotida sebesar 98,49-99,14% jika dibandingkan dengan gen GH ikan sidat A. anguilla, A. australis, dan A. japonica. The growth hormone (GH) gene needs to be studied to support the domestication of the Anguilla bicolor eel in Indonesia. This study aimed to isolate and identify the GH coding gene and analyze its expression in samples of the eel A. bicolor. Primers for amplification of the GH gene of A. bicolor eels were designed based on GH gene sequences from several other eel species available in GenBank. Pituitary and brain cDNA samples of eel were used for GH gene amplification. The amplification results were sequenced and the results were analyzed using BLAST analysis, multiple sequence alignment, hormone signature, and phylogenetic analysis. The GH gene expression analysis was carried out using the real-time PCR technique with the delta-delta CT method on 14 eel samples (23.1 ± 19.6 g; 24.5 ± 4.2 cm). The 486 bp DNA fragment was successfully amplified and sequenced. The GH gene sequence of A. bicolor has a nucleotide similarity of 98.49-99.14% when compared with the GH gene of other eel species A. anguilla, A. australis, and A. japonica. This partial sequence was found in silico to code 155 amino acids (aa) GH protein. The amino acid sequence of the A. bicolor GH protein is very similar to that of other eel species with only three amino acid differences and forms one branch on the phylogenetic tree. The expression level of the GH gene in eel samples had high variations. The results of this study could be a basis for further studies, especially regarding the role of the GH gene in the growth of the eel A. bicolor.

Abstract

Photoperiod affects fish reproduction as it regulates activities of the endocrine glands, which produce the hormones needed for geonadal growth and development, gametogenesis, and reproductive cycles. This study aimed to determine the effects of photoperiod on the hard-lipped barb's reproductive performance by exposing the fish to three photoperiod treatments (light hour: L, darks hour: D), namely 14L:10D (control), 8L:16D (short photoperiod) and 18L:6D (long photoperiod), with four aquaria, each containing 9 fish, serving as replicates. The fish were kept under these photoperiods for 8 weeks. Liver activity, the observable variable in the study, was evaluated by measuring vitellogenin gene expression. Normalized data were then subjected to ANOVA, followed by Tukey's range test. The hard-lipped barb's vitellogenin cDNA was found have a 1136 bp sequence and the vitellogenin precursors encoded cDNA comprising 378 amino acids. The vitellogenin gene in each experimental group saw a significant increase on average when exposed to longer photoperiods (P<0.05), and the highest levels of vitellogenin gene expression occurred under long photoperiods (LP, 18 h light:6 h dark). These results indicate that longer photoperiods stimulate and improve the hard-lipped barb's reproductive performance.
