Abstract

Latar belakang: Kanker paru merupakan salah satu penyebab utama kematian akibat kanker di dunia. Pengetahuan dini mengenai kesehatan saluran pernapasan dan faktor risikonya, terutama di kalangan remaja, sangat penting untuk pencegahan. Tujuan: Kegiatan Pengabdian kepada Masyarakat (Pengmas) ini bertujuan untuk meningkatkan pemahaman siswa-siswi SMAN 1 Gondang, Kabupaten Nganjuk, mengenai topik tersebut. Metode yang digunakan adalah penyuluhan kesehatan secara offline dengan media presentasi (PPT), dilengkapi dengan pre-test dan post-test untuk mengukur efektivitas intervensi. Sebanyak 316 siswa berpartisipasi. Hasil menunjukkan peningkatan pemahaman yang signifikan, ditandai dengan kenaikan nilai rata-rata pre-test sebesar 57.29% menjadi 77.69% pada post-test, dengan peningkatan keseluruhan sebesar 20.40%. Peningkatan tertinggi terjadi pada soal tentang bahaya rokok elektrik atau vape (40.02%). Kesimpulan: Kegiatan ini membuktikan bahwa edukasi kesehatan langsung efektif dalam meningkatkan kesadaran remaja akan kesehatan paru dan bahaya faktor risikonya, sehingga dapat menjadi modal untuk perilaku hidup sehat di masa depan.

Abstract

Objective to compare the inflammatory response between platelet rich plasma 20% and autologous serum 20% on cornea after sodium hydroxide exposure. Methods a true experimental study with two groups post-test design. A total of 18 rabbits were used in this study and allocated into two groups. After collecting intravenous blood samples from both group, platelet rich plasma 20% and autologous serum 20%, respectively, were obtained by centrifugation and dilution. Alkali burns were inflicted on the central cornea of each rabbit's right eye by applying a round filter paper, 7.0 mm in diameter, soaked in 1N NaOH for 60 s. Clinical outcome of the inflammation were observed everyday for seven days. On the seventh day, corneal tissue was collected for histopathological examination to evaluate amount of PMN neutrophils that infiltrate the central cornea. All data were statistically analyzed for difference between the study groups. Resuls statistical analysis shows statistically difference in clinical inflammatory feature of conjunctival hyperemia at day 3 until day 7, in which the group that received PRP 20% got lower conjunctival hyperemia grading compared to the group that received AS 20%. Other clinical features, corneal opacity and epithelial defect, shows no statistically difference between two groups. Histopathological examination shows lower amount of PMN neutrophil infiltration to the central cornea on PRP group, compared to AS group. Conclusio Platelet-rich plasma 20% eyedrop can be used as one of adjuvant therapies and has better control of inflammatory response towards alkaline injury of the cornea during acute phase.

Abstract

Latar Belakang. Pengetahuan komprehensif tentang anatomi, fisiologi, dan kondisi patologis yang memengaruhi sistem reproduksi wanita sangat penting untuk meningkatkan kesadaran akan kesehatan reproduksi. Siswa SMA, sebagai bagian dari generasi muda, diharapkan memiliki pemahaman yang memadai di bidang ini. Tujuan kegiatan ini adalah untuk meningkatkan pemahaman tentang kesehatan dan keganasan reproduksi wanita pada siswa di SMAN 1 Gondang-Nganjuk. Metode. Program pengabdian kepada masyarakat dilaksanakan dengan metode presentasi dan diskusi. Materi yang disampaikan meliputi anatomi, fisiologi, faktor risiko penyakit reproduksi, keganasan saluran reproduksi, serta pentingnya deteksi dini dan pemeriksaan kesehatan rutin. Media pembelajaran terdiri dari presentasi LCD, foto, video, dan leaflet. Peningkatan pengetahuan dinilai menggunakan kuesioner pretes dan postes. Hasil.  Kegiatan dilaksanakan pada 16 Juli 2025 yang diikuti oleh lebih dari 300 orang. Sebanyak 326 siswa mengikuti pretes, sementara 331 siswa mengikuti postes. Proporsi jawaban benar meningkat dari 59,23% pada pretes menjadi 67,88% pada pos tes. Kesimpulan. Terdapat peningkatan pemahaman tentang kesehatan dan keganasan reproduksi wanita pada siswa SMAN 1 Gondang Nganjuk sebesar 8,65%. Kegiatan ini direkomendasikan untuk dilaksanakan secara rutin di sekolah setahun sekali pada siswa baru sebagai upaya untuk mencegah penyakit menular seksual dan kanker.

Abstract

Latar belakang: Kanker serviks merupakan masalah kesehatan global dengan angka morbiditas dan mortalitas yang tinggi, termasuk di Indonesia. Deteksi dini melalui pemeriksaan Pap smear masih menjadi metode yang sering dipakai, namun akurasi hasilnya sangat bergantung pada teknik pengambilan sampel yang adekuat. Tujuan: Pengabdian masyarakat ini bertujuan untuk Meningkatkan pengetahuan dan keterampilan tenaga kesehatan di Puskesmas Ngariboyo dalam teknik pengambilan sediaan Pap smear yang memadai, dan Meningkatkan kesadaran dan pengetahuan masyarakat tentang deteksi dini kanker serviks. Metode: Metode kegiatan terdiri dari dua bagian utama. Pertama, pelatihan bagi tenaga kesehatan yang meliputi webinar dan praktikum offline untuk mengukur adekuasi sediaan berdasarkan Sistem Bethesda. Kedua, penyuluhan dan skrining offline bagi 95 warga dan 101 peserta pemeriksaan Pap smear. Hasil Hasil pelatihan menunjukkan peningkatan signifikan pengetahuan tenaga kesehatan (nilai 80-100 meningkat dari 58,82% menjadi 82,35%). Penyuluhan juga berhasil meningkatkan pengetahuan masyarakat (nilai 80-100 meningkat dari 80% menjadi 94,74%). Hasil skrining menunjukkan 100% sediaan adekuat dan 100% hasil Negative for Intraepithelial Lesion or Malignancy (NILM). Kesimpulan: Disimpulkan bahwa program ini efektif dalam meningkatkan kapasitas tenaga kesehatan dan kesadaran masyarakat, sekaligus menghasilkan data dasar skrining kanker serviks yang berkualitas bagi wilayah Ngariboyo.

Abstract

Human papillomavirus (HPV) infection is one of the most common sexually transmitted infections. HPV high risk (HPVHR) were HPV16,18 related with invasive penile carcinomas, and HPV low risk (HPVLR) were HPV6,11 related to anogenital warts. Male infection is usually asymptomatic that it would be explain increasing the incidence of HPV associated cancers. Identification HPV genotype is very important for predicting the development of the diseases, to be benign or malignant cancer. The objective of this study was to identify the genotype of HPV that infect men with anogential warts. This research used 12 biopsy specimens from men patient with anogenital warts at Outpatient clinic of Department Dermatology and Venereology, Dr. Soetomo General Hospital period 2016-2017. The specimens were diagnozed by pathologist and HPV gentoyping was done to detect 40 HPV genotype including HPVHR and HPVLR. The result showed that 58% (7/12) were positive for HPVLR and 42% (5/12) were positive for HPV LR/HR. The genotype HPV that infected men patient with anogenital warts is HPVLR (HPV6,11) and HPVHR (HPV18,51,52,82) with single infection of HPVLR or mutiple infection HPVLR/LR or HPVLR/HR. The infection of HPVHR would be develops to be malignant transformation. It suggested that HPV genotype needs to be checked the for the anogenital warts cases for predicting the development of the diseases.

Abstract

Approximately 20-30% of all cervical cancer cases are adenocarcinoma and adenosquamous carcinoma. Around 70% of all of these types of cancer are related to infection of Human Papillomavirus (HPV). This study evaluated the distribution of HPV genotype in cervical adenocarcinoma and adenosquamous carcinoma. A cross-sectional study was conducted at the Department of Anatomic Pathology, Dr. Soetomo General Academic Hospital, Surabaya, Indonesia, from January to December 2015. The sample were 22 formalin-fixed paraffin-embedded (FFPE) of cervical adenocarcinoma tissues and adenosquamous carcinoma tissues. FFPE was used for DNA extraction and followed with HPV genotyping to detect 40 genotypes of HPV, including low risk (LR) and high risk (HR) HPV. The histopathological types of adenocarcinomas were adenocarcinoma NOS and mucinous adenocarcinoma, while the adenosquamous carcinoma types were adenosquamous carcinoma and adenosquamous carcinoma glassy. All of the specimens were infected by HPV. In cervical adenocarcinoma, the infection was by HPV 6, 11, 16, 18, 31, 45, 68B, and 72, and in adenosquamous carcinoma by HPV 6, 16, 18, 45, and 59. HPV 18 was predominant, which was found in 13/22 (59.1%) in adenocarcinoma and 19/22 (86.4%) in adenosquamous carcinoma. Single infection and multiple infections in adenocarcinoma were 13/22 (59.1%) and 9/22 (40.9%), while in adenosquamous carcinoma were 21/22 (95.5%) and 1/22 (4.5%) respectively. The most common HR HPVs found in this study were HPV 18, HPV 45, HPV 16 and LR HPV are HPV 11, HPV 6.

Abstract

Ethanol extract of leaves and petiole of pegagan (Centella asiatica) is potential as antimicrobial and antifertility. This study was to investigate whether ethanol extracts of leaf and petiole of Centella asiatica has potential as an antimicrobial and can decrease sperm quality and spermatogenic cells of mice. This was a randomized controlled study using Centella asiatica plant from which we made ethanol extract. The extract of the leaves and petiole of Centella asiatica was subjected to antimicrobial test using the bacteria S. aureus, S. epidermidis, and A. aerogenes. Then the observation was done after a 24-hour-old bacterial cultures by measuring the diameter of the clear zone in mm. Thereafter, the extract was given to randomly grouped mice that were divided into two groups receiving the leaf extract in one group and the petiole exract in another for 35 days. On day 36 the mice were sacrificed for spermatozoa quality and testicular histology examination. Statistical analyses used in this study were ANOVA and LSD tests. There was inhibition zone difference between leaf and petiole (p=0.000). Inhibition zone between bacteria S. epidermidis, S. aureus and A. aerogenes showed no difference (p=0.198). Inter-dose inhibition zone showed difference (p=0.000). Centella asiatica leaf extract showed inhibition zone better than that of Centella asiatica petiole extract. In spermatozoa quality test, there was effect of Centella asiatica leaf extract on sperm motility (p<0.05), viability (p<0.05), morphology (p<0.05) and concentration (p<0.05). No effect of pegagan petiole extract was found on sperm motility (p<0.05), viability (p<0.05), abnormal morphology (p<0.05) and concentration (p<0.05). Highest degradation of spermatozoa quality was obtained from Centella asiatica petiole extract at a concentration of 150 mg/kg bw. Spermatogenic cell test results showed no effect of Centella asiatica leaf extract in decreasing spermatogonia (p=0.000), spermatocytes (p=0.000), and spermatids (p=0.000) counts. No effect of pegagan petiole extract in decreasing the number of spermatogonia, spermatocytes and spermatids. The highest reduction spermatogenic cells of seminiferous tubules was obtained by Centella asiatica petiole extract in a concentration of 150 mg/kg bw. In conclusion: Centella asiatica leaf and petiole extract has antimicrobial potency as contra-ceptives

Abstract

Colorectal carcinoma has high morbidity and mortality rate worldwide, in which over 90% are adenocarcinoma. Colorectal carcinoma is one of the most cancer that metastasize to the lymph node. Angiogenesis have an important role in tumor growth and metastasis. Hypoxia is the trigger factor for angiogenesis. Hypoxia induced factor-1 (HIF-1) is one critical protein directly reacting to hypoxia. HIF-1alpha, a HIF-1 subunit, is an important regulator of angiogenesis. This study analyzed HIF-1a expression in colorectal adenocarcinoma with and without regional lymph node metastasis. This study was to prove that HIF-1a has a role in regional lymph node metastasis in colorectal adenocarcinoma. An analytical observational study was conducted on thirty formalin fixed paraffin-embedded colorectal adenocarcinoma tissues from Anatomic Pathology Laboratory of Dr. Soetomo Hospital. Detection of HIF-1alpha expressions were performed with immunohistochemistry method, using HIF-1alpha antibody. It were statistically analyzed using Mann-Whitney and Kruskal- Wallis methods. There were no significant differences in the expression of HIF-1alpha in colorectal adenocarcinoma with or without lymph node (LN) metastasis (p>0.05). As a conclusion, this study showed that HIF-1alpha has no role in LN metastasis in colorectal adenocarcinoma.

Abstract

Highlights: 1. In this study, new primers designed using the semi-nested polymerase chain reaction (PCR) method were utilized to identify MAGE-A11 and MAGE-A12 expressions in specimens collected from core biopsy, forcep biopsy, and bronchoalveolar lavage. 2. The histopathological analysis revealed positive expressions of MAGE-A11 and MAGE-A12 in specimens diagnosed with non-small cell lung cancer (NSCLC) as well as in specimens with no malignant cells. 3. This study provides evidence indicating that the detection of messenger ribonucleic acid (mRNA) of MAGE-A11 and MAGE-A12 by nested reverse transcription PCR can improve the accuracy of lung cancer diagnosis. Abstract The melanoma antigen gene (MAGE) belongs to the group of cancer-testis antigens that are exclusively expressed in germ cells but may be re-expressed in cancer cells. The highly expressed MAGE-A subfamily in lung cancer may potentially be a diagnostic and prognostic biomarker. This study aimed to identify MAGE-A11 and MAGE-A12 expressions in lung tumors obtained from core biopsy, forceps biopsy, and bronchoalveolar lavage specimens. A cross-sectional observational study was conducted on 90 patients clinically diagnosed with lung tumors. These patients received core biopsy, forceps biopsy, and bronchoalveolar lavage interventions after ethical approval was obtained. The complementary deoxyribonucleic acid (cDNA) quality was assessed by the polymerase chain reaction (PCR) of glyceraldehyde-3-phosphate dehydrogenase (GAPDH). The assessment was performed to ascertain if all specimens exhibited positive PCR amplification of the GAPDH gene. MAGE-A11 and MAGE-A12 were identified through a semi-nested reverse transcription PCR. The positive results were detected by measuring the PCR products, with MAGE-A11 and MAGE-A12 at base pairs (bp) of 858 and 496 in the first and second rounds, respectively. The expressions of MAGE-A11 and MAGE-A12 were observed in 3 (3.33%) and 40 (44.44%) out of 90 specimens, respectively. The prevalence rate of non-small cell lung cancer (NSCLC) was 31.11% (28/90). Among these cases, 3.57% (1/28) showed the expression of MAGE-A11, while 32.14% (9/28) exhibited the expression of MAGE-A12. Sixty-two (68.89%) out of 90 patients were diagnosed with no tumor cell malignancy. Out of 62 cases, 2 (3.23%) exhibited the expression of MAGE-A11, while 31 (50%) demonstrated the expression of MAGE-A12. MAGE-A11 and MAGE-A12 were detected in NSCLC and certain specimens with a pathological diagnosis that indicated the absence of malignant cells. In conclusion, MAGE A11 and MAGE A12 have potential markers to improve the pathological diagnosis of lung cancer. Further investigation is necessary to explore the expression of MAGE-A in correlation with lung cancer progression.

Abstract

Background: Lead acetate exposure through oral route has a significant impact on lung tissue as lead can induces the formation of reactive oxygen species (ROS), resulting in decreased glutathione peroxidase (GPx) and triggering inflammatory responses. The induction of exogenous antioxidants may prevent this mechanism. This study aimed to evaluate the protective effect of Tamarillo ethanol extract on GPx expression and inflammatory reactions in the lung tissue of mice induced with lead acetate.Methods: This true experimental laboratory study utilized stored formalin-fixed-paraffin-embedded (FFPE) lung tissue  from 30 male mice (Mus musculus). The samples were divided into five intervention groups, each consisting of six mice. Immunohistochemistry and histopathology assessments were performed at 100x and 400x magnification to evaluate the GPx expression using the H-score and to assess inflammatory reactions based on five scoring parameters, which were summed to obtain the total lung injury score.Results: Lead exposure significantly increased inflammatory reactions, particularly in the hemorrhage parameters (p=0.041). A significant increase in GPx expression was observed following lead acetate administration (p=0.027). Administration of Tamarillo ethanol extract at a dose of 400 mg/kgBW increased GPx expression compared with the positive control group (p=0.027).Conclusion: Tamarillo ethanol extract, especially at a dose of 400 mg/kgBW, significantly increases GPx expression in lead acetate-induced lung tissue of mice. This finding highlights the potential role of Tamarillo extract as an antioxidant source to reduce lead-induced lung injury, supporting broader efforts to promote healthy lifestyle practices that protect against environmental toxin exposure.
