Abstract

Highlights:1. In this study, new primers designed using the semi-nested polymerase chain reaction (PCR) method were utilized to identify MAGE-A11 and MAGE-A12 expressions in specimens collected from core biopsy, forcep biopsy, and bronchoalveolar lavage. 2. The histopathological analysis revealed positive expressions of MAGE-A11 and MAGE-A12 in specimens diagnosed with non-small cell lung cancer (NSCLC) as well as in specimens with no malignant cells. 3. This study provides evidence indicating that the detection of messenger ribonucleic acid (mRNA) of MAGE-A11 and MAGE-A12 by nested reverse transcription PCR can improve the accuracy of lung cancer diagnosis.   AbstractThe melanoma antigen gene (MAGE) belongs to the group of cancer-testis antigens that are exclusively expressed in germ cells but may be re-expressed in cancer cells. The highly expressed MAGE-A subfamily in lung cancer may potentially be a diagnostic and prognostic biomarker. This study aimed to identify MAGE-A11 and MAGE-A12 expressions in lung tumors obtained from core biopsy, forceps biopsy, and bronchoalveolar lavage specimens. A cross-sectional observational study was conducted on 90 patients clinically diagnosed with lung tumors. These patients received core biopsy, forceps biopsy, and bronchoalveolar lavage interventions after ethical approval was obtained. The complementary deoxyribonucleic acid (cDNA) quality was assessed by the polymerase chain reaction (PCR) of glyceraldehyde-3-phosphate dehydrogenase (GAPDH). The assessment was performed to ascertain if all specimens exhibited positive PCR amplification of the GAPDH gene. MAGE-A11 and MAGE-A12 were identified through a semi-nested reverse transcription PCR. The positive results were detected by measuring the PCR products, with MAGE-A11 and MAGE-A12 at base pairs (bp) of 858 and 496 in the first and second rounds, respectively. The expressions of MAGE-A11 and MAGE-A12 were observed in 3 (3.33%) and 40 (44.44%) out of 90 specimens, respectively. The prevalence rate of non-small cell lung cancer (NSCLC) was 31.11% (28/90). Among these cases, 3.57% (1/28) showed the expression of MAGE-A11, while 32.14% (9/28) exhibited the expression of MAGE-A12. Sixty-two (68.89%) out of 90 patients were diagnosed with no tumor cell malignancy. Out of 62 cases, 2 (3.23%) exhibited the expression of MAGE-A11, while 31 (50%) demonstrated the expression of MAGE-A12. MAGE-A11 and MAGE-A12 were detected in NSCLC and certain specimens with a pathological diagnosis that indicated the absence of malignant cells. In conclusion, MAGE A11 and MAGE A12 have potential markers to improve the pathological diagnosis of lung cancer. Further investigation is necessary to explore the expression of MAGE-A in correlation with lung cancer progression.

Abstract

Highlights The histopathological types of OSSN were significantly associated with age and occupation. The study's findings enhance the understanding of OSSN in Indonesia and emphasize the importance of UV exposure, occupation, and age in its development.   Abstract Background:  Ocular Surface Squamous Neoplasia (OSSN) is a spectrum of tumors affecting the conjunctiva and cornea, in which a more invasive type of OSSN has a higher recurrence rate. As a tropical country, Indonesia is constantly exposed to intense ultraviolet (UV) radiation, the main risk factor for OSSN. Despite this, there are very few studies regarding OSSN in Indonesia. Objective:  This study aimed to analyze the association between sociodemographic characteristics such as age, sex, and types of occupation (outdoor and indoor) and histopathological types of OSSN to fill the gap in research and contribute to early diagnostic strategies. Material and Method: Sociodemographic and histopathological data were extracted from the medical records of patients who were histopathologically diagnosed with OSSN at Dr. Soetomo General Hospital from 2017 to 2021. Data from 88 samples that met the inclusion and exclusion criteria were classified and analyzed with descriptive statistics and a two-sided Fisher's exact test using SPSS version 27.0 (IBM Corp., Armonk, N.Y., USA). Result: The exact test results of the two-sided fisher revealed a statistically significant relationship between age (p = 0.0004) and occupation type (p = 0.049) with histopathological types of OSSN. However, no significant association was found between sex and histopathological types of OSSN (p = 0.130). Conclusion: Age and occupation were found to have a significant association with the histopathological types of OSSN, reinforcing the need for further exploration of its strength and nature to be considered by future researchers.

Abstract

Introduction: Habit of consuming excess calories and sedentary lifestyle can cause oxidative stress which is known to be involved in the pathophysiological process of various diseases including diseases of the gastrointestinal mucosa. Increased Reactive Oxygen Species (ROS) due to a high-calorie diet can damage lipids, proteins, and DNA oxidation of the mucosa and lead to inflammation, apoptosis, damage of mucosal and its barrier. Moderate-intensity physical exercise is known to have health benefits, improve quality of life, reduce the risk of death, and prevent the occurrence of diseases related to oxidative stress. The aim of this study was to analyze the effect of high-calorie diet and moderate-intensity physical exercise on gastric histopathological features of mice. Methods: This research was a true experimental study with posttest only control group design. Female Balb/C strain were randomly assigned into three groups, the control group/K1 (n=12), high-calorie diet group/K2 (n=12), and high-calorie diet and moderate-intensity physical exercise group/K3 (n=9). High-calorie diet in the form of dextrose (D40) was given ad libitum using sonde in a dose of 3-5% of calories or 0.0325 mL/gBW. The moderate-intensity physical exercise was carried out by swimming three times a week for 15 minutes using 6% gBW loading. After 4 weeks of treatment, the mice were sacrificed and their stomach were sampled and prepared for microscopic analysis. Results: The histopathological examination of the gastric mucosa showed the average number of inflammatory cells was 11.60 for K1, 11.25 for K2, and 14.5222 for K3. The Shapiro-Wilk test revealed that the data on inflammatory cells infiltrations were not normally distributed (p<0.05), so a non-parametric difference test was conducted using the Kruskal-Wallis test. Based on the Kruskal-Wallis test results, it was obtained that p>0.05, which means no significant difference was found between the three study groups. Conclusion: There was no effect of a high-calorie diet and moderate-intensity physical exercise on the gastric histopathological features of mice (Mus musculus) in this study

Abstract

Excessive release of interleukin-6 (IL-6) during the progression of coronavirus disease 2019 (COVID-19) induces cytokine storms, resulting in multi-organ damages including liver injury, similar in nature with mechanism of viral hepatitis. Systemic IL-6 has been associated with the incidence of liver injury among COVID-19 patients; however, studies on IL-6 expression in the liver tissue are completely lacking. The aim of this study was to measure the IL-6 expression in the liver tissues and to determine its correlation with the degree of liver injury in fatal COVID-19 patients. Through this first cross-sectional study, IL-6 expression was measured through immunohistochemical staining and the degree of liver injury was identified based on level of serum alanine aminotransferase (ALT). The Spearman correlation test was used to identify the correlation between IL-6 expression and the degree of liver injury. A total of 47 deceased COVID-19 patients were included and IL-6 expression was observed in all post-mortem liver specimens, ranging from mild to strong expression. Liver injury at various degrees (mild to severe) was found in more than half (59.5%) of the cases. The Spearman correlation analysis suggested a statistically insignificant correlation between liver IL-6 expression and the degree of liver injury (r=0.152; p=0.309). In conclusion, even IL-6 expression was observed in all post-mortem liver specimens, there was an insignificant correlation between IL-6 expression in the liver tissue with the degree of liver injury among fatal COVID-19 patients, suggesting that IL-6 was not the only main factor contributing to liver damage in COVID-19 patients.

Abstract

Breast cancer is the most common cancer found in the world and this cancer is the second cause of death due to cancer after lung cancer in Indonesia. This high mortality rate can be prevented by carrying out early detection. However, this is still not done well, especially in communities living in remote areas with low to medium economic and educational levels. The aim is to convey knowledge about breast cancer and training in the self-examination of the breast. It was held on Tuesday, 12 September 2023 at the Ngluyu Community Health Center, Nganjuk, from 07.00 to 14.00 WIB and was attended by 111 people. Data on breast cancer risk factors was collected through questionnaires. There were 96 participants who filled out the questionnaire, aged between 22-63 years. Risk factors data showed that 3.13% did not have children, 15.63% breastfed children less than 6 months, 5.21% were exposed to X-rays more than once a year; 1.04% had breast tumors, and 4.17% had family members suffering from cancer. In addition, 89.58% were over 25 years old and 42.71% had doing self-examination of breast. There was an increase in public understanding at the Ngluyu Community Health Center from 77.70% to 84.12%, namely an increase of 6.42%. This can increase public understanding about breast cancer and procedures to self-examination of breast and contribute in early detection of breast cancer as well as reducing the incidence of breast cancer. The results of this counseling can be used as recommendations by health centers in the Nganjuk district area.
