Abstract

NFT hydroponic is a plant cultivation technology that allows plant roots to grow on a shallow nutrient layer on the narrow field. The goal of this study was to evaluate the best NPK 16-16-16 fertilizer concentration and planting media, as well as the combination of the best NPK 16-16-16 fertilizer concentration and planting media, on the growth and yield of pakchoy (Brassica rapa L). This study used a split plot in factorial randomized block design. The first factor is the concentration of AB mix fertilizer as a control at 1400 ppm (N1) and the concentration of NPK fertilizer, namely: 758 ppm (N2), 1000 ppm (N3). The second factor element is the type of planting medium, which consists of 6 media: rockwool (M1), cotton (M2), sponge (M3), fern (M4), charcoal husk (M5), and cocopeat (M6), with 2 repetitions and 18 treatment combinations. Plant height (cm), number of leaves (pieces), leaf width (cm), leaf length (cm), wet weight of sample plants (grams), and dried weight of plants (grams) were the observation variables. In comparison to an NPK nutrient concentration of 1000 ppm, providing an NPK fertilizer concentration of 758 ppm resulted in better growth and yield in the parameters of number of leaves, wet weight, and dry weight of pak choy plants. Using husk charcoal as a planting medium resulted in superior growth and results in the plant height, number of leaves, leaf width, leaf length, and wet weight of pakchoy plants.

Abstract

Dendrobium Gradita 31 has superior characteristics that are in demand by the market in Indonesia, so seeds with sameness quality, the same characteristics as the parent and in large quantities are needed. Tissue culture is one solution to support the availability of seeds in large quantities and have a high level of sameness quality. The process of regulating the type of ZPT and concentration in the culture media does not always produce the same regeneration pathway. This research aims to determine the regeneration pathway and to obtain the best type and concentration of cytokinin to support its regeneration pathway. This study used a completely randomized design with 3 replications. The first factor is the type of cytokinin, BA and Tdz. The second factor is the concentration of Benzyladenine and Thidiazuron, 1, 2 and 3 mg/l.. The data was analyzed using analysis of variance, and mean value was tested using tukey test 5%. The results of this study indicate that the regeneration pathway in the formation of adventitious shoots was indirect embryogenesis. The use of Tdz 3 mg/l was able to produce the highest percentage of somatic embryo formation and the highest number of somatic embryos. The fastest shoot formation time, shoot formation percentage and the highest number of shoots per clump were found BA 2 mg/l.

Abstract

The use of liquid organic fertilizer with added AB Mix nutrients is an alternative solution to reduce the use of AB Mix nutrients. The aims of the research were to study interaction between the application of liquid organic fertilizer and the hydroponic system on the growth and yield of pak choy and the effect of concentration of liquid organic fertilizer and hydroponic systems on the growth and yield of pak choy. This experiment used Split Plot Design, which consist 2 factors, main plot was concentration of liquid organic fertilizer. Sub plot was hydroponic systems. Data obtained were analyzed with F test, and were continued with LSD at 5 percent when significant. Results showed that there was no interaction between the concentration of liquid organic fertilizer and hydroponic system for all observed parameters. The concentration of 25% liquid organic fertilizer + 75% AB Mix and concentration of 45% liquid organic fertilizer + 55% AB Mix gave better results on the parameters of height, number of leaves and fresh weight of pak choy plants grown in a hydroponic system when compared with a concentration of 65% liquid organic fertilizer + 35 % AB Mix. The NFT hydroponic system is better in the parameters of height and number of leaves of pak choy plants when compared to the DFT hydroponic system.

Abstract

Acclimatization is a critical stage in plant micropropagation. Many plantlets resulting from tissue culture cannotsurvive when acclimatized. External environmental conditions will affect the ability of acclimatized plants to survive. Apart from climatic conditions (temperature and sun intensity), the type of media and its composition of the planting mediacan also influence the ability of plantlets to survive and grow. This research aims to: 1) see effect of media plantingcomposition on acclimatization of fig micro shoot-tip cutting, and 2) find the best composition of acclimatization mediatreament for the success and growth of acclimatized fig from micro shoot-tip cutting. The research was carried out using aRandomized Block Design with 7 treatments, and each treatment was repeated 4 times. Each experimental unit was plantedwith 6 cuttings of fig plants resulting from in vitro multiplication. The data were subjected to analysis of variance, anddifferences between acts were tested with Tukey Test at the 5% level. The results showed: 1) The media plantingcomposition were success to influence acclimatization of fig micro shoot-tip cutting, 2) The best composition ofacclimatization media treament for the success and growth of fig micro shoot-tip cutting is top soil+cocopeat, and 3) Thehighest percentage of survival rate on fig micro shoot-tip cutting acclimatization (75%) obtained on top soil+cocopeatmedia followed by top soil media (45%).

Abstract

Abstract

The objective of plant hybridization is to generate alternative superior phenotypes with the aim of decreasing the dependence on wild orchid exploitation in nature. Although interspecies crosses in Dendrobium are very likely to occur and there is a low chance of experiencing bud depression, interspecies crosses have the potential to encounter several problems, including failure of embryo development resulted to the seeds unable to germinate. Media formulation is the most essential component in the success of in vitro culture of orchid seeds. Therefore, this study aimed to obtain new Dendrobium hybrids and the best media formulation for germination and seedling growth. A total of three experiments were conducted including hybridization, the impacts of basic media namely (1) MS, (2) VW, and (3) foliar fertilizer both with and without the addition of tryptone, on seedling germination. The results showed that there was no compatibility barriers observed in all crosses. Foliar fertilizer + tryptone media was recommended for orchid seedlings because it produced better growth and roots up to 8 months after planting (MAP). For best results on Dendrobium hybrid seedlings, MS + tryptone media up to 6 MAP was used followed by deflasking into VW + tryptone to stimulate optimal root growth before hardening.
