Abstract

Background: Diabetes mellitus is a metabolic disease, characterized by hyperglycemia due to disturbance in both insulin secretion and function. One of theurapeutic approaches is to reduce blood glucose levels by inhbiting α-/β-glucosidase and α-amylase involved in carbohydrate digestion. Thus, inhibition of these enzymes play important role in the treatment of diabetes mellitus. Roselle (Hibiscus sabdariffa L.) has been known to have several medicinal properties and potency as an antidiabetics agents. This reseacrh aimed to observe antidiabetic properties of roselle ethanol extract (REE) towards α-glucosidase, β-glucosidase and α-amylase.Materials and Methods: REE was done with maceration technique using diluent of 70% ethanol. Antidiabetic properties were measured by inhibitory activity of α-amylase, α-glucosidase and β-glucosidase.Results: REE was able to inhibit α-/β-glucosidase and α-amylase in the highest concentration with inhibition percentage of 72.68, 47.34 and 73.08% respectively, and were comparable with Acarbose of 81.49, 50.97, 73.08%. The median inhibitory concentration (IC50) of α-/β-glucosidase and α-amylase of REE were 15.81, 41.77, 18.09 μg/mL respectively, and Acarbose were 9.45, 22.57, 3.64 μg/mL respectively. Conclusions: REE inhibits α-/β-glucosidase and α-amylase.Keywords: Roselle, Acarbose, α-glucosidase, β-glucosidase, α-amylase, antidiabetic

Abstract

cause of death and disability globally. One of the causes is associated with cell signaling disorders, neuronal apoptosis, inflammation, and the deposition of aggregate proteins. The N-methyl-D-aspartate (NMDA) receptor is a glutamate receptor that, when activated, causes synaptic dysfunction and leads to neuronal death. Phytoestrogen compounds are able to replace the role of estrogen in maintaining body homeostasis, including in the CNS.  Objectif: This study aims to determine the role of phytoestrogen compounds in inhibiting NMDA activation (1PBQ), which causes neurodegenerative diseases. Methods: The method used is molecular docking with the AutoDockTools 1.5.6 program. The prediction of pharmacokinetic and pharmacodynamic properties used SwissADME, while the toxicity used pkCSM and ProTox II. Results:  The results of docking using the 1PBQ protein showed that the compounds α-amyrin, β-amyrin and eudesmin had the best binding potential compared to 17β-estradiol which was the positive control. Pharmacokinetic and pharmacodynamic tests showed that the three compounds had good permeability and strong lipophilicity, so they could penetrate cell membranes, and were not toxic, except for eudesmin, which was included in class IV in the toxicity test using ProTox II. Conclusion :   α-amyrin and β-amyrin compounds have the potential to treat neurodegenerative diseases against NMDA receptors (1PBQ).

Abstract

The Clitoria ternatea flower, known as bunga telang in Indonesia, is commonly mixed with food and beverages to provide a natural blue colour. Aside from its popular culinary use, it is a traditional medicine in Indonesia for diseases in the eyes, urinary tract and skin, as well as functioning as an anti-toxin. Furthermore, recent advances in science and technology have revealed that the C. ternatea flower contains a high level of polyphenol compounds that possess anticancer activity, including saponins, tannins, steroids, triterpenoids, kaempferol, and quercetin. This review aims to identify and analyse recent articles regarding the phytochemical activities of C. ternatea flower extract as an anticancer agent. The literature on main databases from 2011 to 2021 was searched systematically using the keywords “Anticancer activity of Clitorea ternatea” and “Phytochemical activities of Clitorea ternatea flower extract against cancer cells”. The various extracts of C. ternatea flower display a moderate cytotoxic, IC50 = 21 µg/mL - 200 µg/mL, for many cancer cell lines, such as MCF-7, MDA-MB-231, CaoV-3, HEp-G2 in aquadest extract and the DLA cell line in petroleum ether extract. The bioactive compounds responsible for the anticancer effect include ternatins, delphinidin, kaempferol, quercetin, sitosterol, and tocopherols. In addition, there have been no reports of any toxic effect on normal cells (Hs27) and oral consumption in mice. According to many studies, the extract is active on multi-molecular targets, with the most conclusive effect on polymerase enzymes, whose inhibition can be an important therapeutic strategy to treat hyperproliferation in cell cancer. Therefore, the findings suggest a potential application of C. ternatea for cancer treatment.

Abstract

Leukemia, characterized by abnormal leukocyte proliferation, ranks ninth in Indonesia as the most common cancer. While treatments such as chemotherapy and radiation effectively target cancer cells, they also risk damaging healthy blood cells. This has spurred interest in exploring low-toxicity herbal compounds as potential therapies, with mangosteen peel emerging as a widely researched option. Nanotechnology, which has the potential to enhance the bioavailability of herbal compounds, is also a focus of extensive research. This study objective was to assess the impact of Mangosteen Peel Nanocrystal (MPN) on HL-60 leukemia cells by analyzing various parameters, including cytotoxicity, reactive oxygen species (ROS) levels, senescence, and gene expression changes. MPN was prepared with high-speed milling and characterized using particle size analyzers, microscopy, and stability assessments. HL-60 cells were cultured and subjected to MPN treatment. Cytotoxicity was evaluated using WST-8 assays, ROS levels were assessed using flow cytometry, and senescence analyses using Senescence-Associated b-Galactosidase Staining. AKT and FLT-1 gene expression were determined via qRT-PCR. MPN has been successfully characterized as a nanoparticle based on size, stability, and morphology. MPN has an impact on leukemia cells by increasing cytotoxicity, decreasing ROS levels, inducing senescence, and modulating AKT and FLT-1 gene expressions. The findings suggest potential implications for MPN in targeting leukemia cells. The study sheds light on the promising effects of MPN in leukemia cell models, indicating its potential applications in targeting cancer cells, inducing senescence, decreasing ROS levels, and modulating gene expressions related to cell survival and proliferation.

Abstract

External aging factors such as UV exposure, pollution, and lifestyle choices contribute to skin aging, resulting in deep wrinkles, fine lines, and rough skin, which can lower self-confidence. Plant extracts have been widely studied for their antiaging potential, while Salmon DNA has shown promise in stimulating collagen production. This study explores the formulation of a serum combining Centella asiatica, Curcuma longa, Aloe vera, Rosa centifolia, and Salmon DNA for its antiaging effects on injured human fibroblast cells. The serum was formulated using extracts from C. asiatica, C. longa, A. vera, R. centifolia, and Salmon DNA. Antioxidant activity was evaluated with the DPPH method, cytotoxicity using the WST-8 assay, and gene expression through qRT-PCR for COL1A1, TGF-β1, HYAL-1, and FGF-2. The serum exhibited weak antioxidant activity (IC50 = 373.33 µg/ml) and reduced cell viability at high concentrations. Gene expression analysis revealed increased expression of COL1A1, TGF-β1, and FGF-2, along with reduced HYAL-1 expression in injured BJ cells. The formulated serum shows potential as an antiaging agent, promoting collagen production and reducing hyaluronidase activity.

Abstract

Jatijejer Village, located between the slopes of Mount Penanggungan and Mount Welirang, has significant potential for cultivating aromatic plants, particularly lemongrass (Cymbopogon citratus) and citronella (Cymbopogon nardus). These plants are sources of essential oils with various benefits, including uses in aromatherapy, antibacterial, and anti-inflammatory applications, and are widely utilized in the cosmetics and healthcare industries. Despite this potential, the community of Jatijejer Village previously only utilized lemongrass as raw material, yielding suboptimal economic value and welfare improvement. This community service program aims to enhance the welfare of Jatijejer Village through the production and diversification of essential oil-based products. Activities included training in steam distillation techniques, quality measurement of essential oils, and the development of derivative products such as inhalers and diffusers. Evaluation results showed a significant improvement in participants' knowledge and skills. The average pre-test score of 58.4 percent increased to 97.2 percent in the post-test. Additionally, the essential oils produced were successfully developed into value-added products, such as inhalers and diffusers, which have begun to be marketed locally. Solid waste from the distillation process was also utilized as organic fertilizer, reinforcing the program's sustainability. This training effectively improved the skills, income, and economic independence of the Jatijejer Village community.

Abstract

ABSTRACT Ischemic stroke is the sudden development of focal neurological deficit due to inadequate blood supply to an area of the brain, which can be thrombotic or embolic. CAD (Coronary Artery Disease) is pathological process characterized by the formation of atherosclerotic plaque followed by rupture, ulceration, or erosion. CAD can take the form of CCS (Chronic Coronary Syndrome). The 2018 AHA/ASA Guidline Antiplatelet Therapy used for ischemic stroke is Aspirin at a dose of 160mg-300mg 1x1. In the 2019 ESC Guidline, CCS patient therapy uses DAPT (Dual antiplatelet). In a 2021 AHA meta-analysis compared with aspirin alone, short-term DAPT within 24 hours after mild-moderate ischemic stroke reduced the risk of recurrent stroke at the expense of a higher risk of major bleeding. Here we present a case of a 76 year old female patient with ischemic stroke and CCS who used DAPT. Complaints of sluggishness, weakness in the left half of the body, acute dysarthria onset on the first day, when waking up. Hemiplegia S 1 week ago onset of waking up and being treated at Mataram Regional Hospital. The patient received the antiplatelet Aspirin 160mg 1x1 a day then switched DAPT (Aspirin 80mg 1x1 and Clopidogrel 75mg 1x1) due to a history of CCS.
