Abstract

Background: Inflammation is a protective biological response to tissue injury or infection; however, excessive inflammation may lead to tissue damage and chronic disease. Matoa fruit peel (Pometia pinnata J.R.Forst. & G.Forst.) contains bioactive compounds, including Flavonoids, saponins, tannins, and phenolics, which are known to have anti-inflammatory properties, are present in the plant. Objective: This study sought to assess the anti-inflammatory efficacy of the ethanol extract derived from the matoa fruit peel through the utilization of a carrageenan-induced paw edema model in male white rats. Methods: In an experiment, we used male white rats (Rattus norvegicus) and split them into five groups. We had a negative control, a positive control, and three treatment groups. The treatment groups got ethanol extract at different doses—100, 200, and 300 mg/kg of body weight. To get the inflammation going, we gave them a 1% carrageenan injection under their skin. Paw edema volume was measured periodically using a plethysmometer to determine the anti-inflammatory effect. Results: The initial volume of edema of the whole group was homogeneous (0.06 ± 0.00–0.01 mL). Ethanol extract of matoa fruit peel at doses of 100, 200, and 300 mg/kgBB showed anti-inflammatory activity by gradually reducing edema until the 6th hour. The dose of 100 mg/kgBB provided the highest edema inhibition effect (42.06 ± 8.36%), followed by the 300 mg/kgBB dose (41.27 ± 13.75%) and 200 mg/kgBB (31.75 ± 2.75%), while positive control showed the largest inhibition (61.11 ± 9.62%). Statistical analysis showed that all treatment groups differed significantly from negative controls (p < 0.05), but still had lower effectiveness than positive controls.Conclusion:Ethanol extract of matoa fruit peel demonstrates promising anti-inflammatory activity in carrageenan-induced male white rats and may serve as a potential natural source for anti-inflammatory drug development. Further studies are needed to identify the active compounds, clarify the molecular mechanisms, and evaluate the extract's safety.

Abstract

Penelitian ini bertujuan untuk menguji efektivitas ekstrak etanol daun awar-awar (Ficus septica) sebagai antianemia pada mencit putih jantan (Mus musculus) yang diinduksi menggunakan natrium nitrit (NaNO₂). Penelitian menggunakan metode pretest-posttest control group dengan rancangan eksperimental melibatkan lima kelompok perlakuan yaitu kontrol positif (tablet tambah darah), kontrol negatif (Na-CMC 0,5%), serta tiga kelompok ekstrak etanol daun awar-awar dosis 10, 20, dan 40 mg/kgBB. Mencit diinduksi anemia selama 14 hari dengan NaNO₂ dan pengukuran kadar hemoglobin dilakukan menggunakan Hb meter. Hasil skrining fitokimia menunjukkan ekstrak mengandung flavonoid sebagai komponen utama yang diduga berperan dalam aktivitas antianemia. Hasil analisis paired t-test menunjukkan terdapat penurunan signifikan kadar hemoglobin setelah induksi pada semua kelompok (P < 0,05). Namun, analisis dan uji independen sampel t test menunjukkan tidak terdapat perbedaan yang signifikan antara kelompok perlakuan ekstrak etanol daun awar-awar dengan kelompok kontrol, sehingga ekstrak etanol daun awar-awar pada dosis yang diberikan belum terbukti efektif meningkatkan kadar hemoglobin pada model anemia mencit. Penelitian ini merekomendasikan agar penelitian lanjutan mempertimbangkan pengendalian asupan nutrisi dan eksplorasi dosis maupun durasi pemberian yang lebih variatif untuk mendapatkan hasil yang lebih optimal.

Abstract

Background : Hyperglycemia is a metabolic condition characterized by an increase in blood glucose levels above normal limits and is the main characteristic of diabetes mellitus, a chronic disease with a prevalence that continues to increase in various countries. The akway plant (Drimys cf piperita.) is endemic to the Sorong Mountains, Southwest Papua . Its leaves are known to contain various bioactive compounds such as flavonoids, tannins, saponins, and alkaloids. Purpose: This study aims to test the ability of akway leaf extract to lower blood sugar levels in vivo in white mice that were previously raised glucose levels through glucose solution. Method : This study design is an experimental laboratory with five treatment groups, namely negative control (Na-CMC 0.5 mg), positive control (glibenclamide 3 mg), and three groups of akway leaf extracts, each at doses of 100, 200, and 400 mg/kg BW. Hyperglycemia was induced for 7 days using a glucose solution. Blood glucose measurements were performed at 30, 60, 90, 120, and 150 minutes using a glucometer. Data analysis used One-Way ANOVA followed by a post -LSD test. Results: Akway leaf extract at a dose of 200 mg/kg BW provided the highest reduction in blood glucose, namely 54.24%, which was statistically significant (<0.05) and even better than glibenclamide (51.15%). Meanwhile, a dose of 400 mg/kBW actually produced lower effectiveness (45.85%). Implications: Akway leaf extract has been proven to have antihyperglycemic activity with an optimal dose of 200 mg/kgBW.

Abstract

Background: Red fruit (Pandanus conoideus L.) is an endemic plant of Papua that has been traditionally used to treat various diseases. Its biological activities are associated with the presence of secondary metabolites, particularly flavonoids and alkaloids. Objective: This study aimed to identify the secondary metabolites and evaluate the effect of different extraction methods on the total flavonoid and alkaloid contents of ethanolic extracts of red fruit (Pandanus conoideus L.) stem core. Methods: Extraction was performed using three different methods, namely maceration, Soxhlet extraction, and reflux extraction with ethanol as the solvent. Secondary metabolites were identified qualitatively, while total flavonoid and alkaloid contents were determined spectrophotometrically. Results: Phytochemical screening revealed the presence of alkaloids, flavonoids, terpenoids, steroids, and tannins in the ethanolic extract of red fruit stem core. The total flavonoid contents obtained by maceration, Soxhlet extraction, and reflux extraction were 218.966 mgQE/g (21.89%), 104.023 mgQE/g (10.40%), and 247.701 mgQE/g (24.77%), respectively. The total alkaloid contents obtained by maceration, Soxhlet extraction, and reflux extraction were 145.770 mgAE/g (14.58%), 65.767 mgAE/g (6.58%), and 76.433 mgAE/g (7.64%), respectively. Conclusion: The extraction method affected the total flavonoid and alkaloid contents obtained from the sample. Reflux extraction produced the highest total flavonoid content (247.701 mgQE/g), whereas maceration produced the highest total alkaloid content (145.770 mgAE/g). Therefore, reflux was the most effective method for flavonoid extraction, while maceration was the most effective method for alkaloid extraction from the stem core of red fruit (Pandanus conoideus L.).

Abstract

Akar laka (Dalbergia parviflora Roxb.) is a traditional medicinal plant used by communities in North Maluku to treat various types of wounds and skin conditions; however, scientific research on the antifungal activity of this plant’s stem is still limited. This study aims to identify the content of secondary metabolites and evaluate the antifungal activity of akar laka stem extract against the growth of Trichophyton rubrum in vitro. This is a laboratory experimental study conducted using the maceration method with 95% ethanol as a solvent to obtain a concentrated extract, followed by phytochemical screening to identify the classes of secondary metabolites. Antifungal activity testing was performed using the disk diffusion method with extract concentrations of 5%, 10%, and 15%, with ketoconazole used as the positive control and aqua pro injection as the negative control. The results showed that from 200 grams of crude drug, 43 grams of concentrated extract were obtained with a yield of 21.5%. Phytochemical screening results indicated the presence of alkaloids, saponins, and tannins, while flavonoids were not detected. In the antifungal activity test, the extract did not form an inhibition zone at any of the tested concentrations against T. rubrum, whereas the positive control exhibited a very strong inhibition zone of 26.7 mm. Based on the research results, it can be concluded that the extract of the akar laka stem (D. parviflora Roxb.) contains several secondary metabolites but did not exhibit antifungal activity against T. rubrum at the tested concentrations.

Abstract

Background: Papua has high biodiversity with significant potential as a source of traditional medicinal plants, including Nelambo suon (Rubiaceae), which has been empirically used by local communities. However, scientific information regarding the active compounds in its stem bark remains limited. Objective: This study aimed to identify the secondary metabolite groups present in the ethanol extract of Nelambo suon stem bark through phytochemical screening. Methods: The research was conducted experimentally using the maceration method with 80% ethanol as the solvent. The stem bark simplicia was extracted and qualitatively screened for alkaloids, flavonoids, tannins, and saponins using specific reagents. Results: The results showed that the ethanol extract of Nelambo suon stem bark contained alkaloids, flavonoids, tannins, and saponins, indicated by characteristic color changes, precipitate formation, and foam production. Conclusion: These findings suggest that Nelambo suon stem bark is a potential source of bioactive compounds and may be further developed as a herbal medicinal material based on local wisdom.
