Abstract

Escherichia coli (E. coli) is a normal flora and a facultative anaerobic bacterium that plays an essential role in the humoral nutritional metabolism of broiler intestines. This study aims to determine the effect of salix (Salix tetrasperma roxb) extract 50 and 100 mg/litre in drinking water of E. coli colonies in the broiler intestines experiencing heat stress. The sample is 12 intestines of broiler strain MB-90 aged 28 days. The method used a completely randomized design. The study by user three treatments: P0 was not given salix extract in drinking water; P1 and P2 were given salix extract 50 and 100 mg/litre in drinking water. The data were analyzed using One-way ANOVA and were continued with the Duncan test. We can conclude that the salix extract given to broiler chickens with heat stress could affect many E. coli colonies. This result can be seen from the doses given, indicating that the number of E. coli colonies in chicken intestines that experience heat stress depends on the dose of salix extract. The higher dose is given, the fewer colonies in the chicken intestine

Abstract

This study aims to investigate the effect of celery leaf infusa on decreasing  level of blood glucose in rat (Rattus norvegicus). Twenty five rats were divided into 5 groups namely first treatment group (P1) as negative control. Second treatment group (P2) as positive control, rats were induced with alloxan. Third treatment group (P3) rats were induced with alloxan and given 5% celery leaf infusa. Fourth treatment group (P4) rats were induced with alloxan and given 10% celery leaf infusa. Fifth treatment group (P5) rats were induced alloxan and given 15% celery leaf infusa for 14 days. The level of blood glucose of the rat was determined before treatment, after given aloksan and after given celery leaf infusa. Data were analyzed using Analysis of Varians (ANOVA). Results showed that administration of celery leaf infusa  for 14 days, show significant effect to decreasing level of glucose in rat (P0.01). It can be concluded that the administration of celery leaf infusa  concentrated 5%, 10% and 15% for 14 days show significant effect to decreased level of glucose in rats.

Abstract

This research aimed to determine the total amount of Staphylococcus aureus (S. aureus) contamination in beef sold in traditional market in Banda Aceh. Total of 16 beef samples were used in this research, 2 samples from Beurawe market, 10 samples from Peunayong market, and 4 samples from Seutui market. Samples were cultured on manitol salt agar. The number of S. aureus colonies was calculated using colony counter, and the result was compared to the limit of microbial contamination based on Indonesian National Standard (SNI). The result of this research showed that the highest level of S. aureus contamination was derived from Beurawe market. It is concluded that all beef sold by seller from traditional market in Banda Aceh were contaminated by S. aureus.Key words: Staphylococcus aureus, meat, traditional market, number of contamination

Abstract

This study was aimed to determine the level of Density Figure of Aedes spp mosquito larvae with parameter House Index (HI) Container Index (CI), and Breteau Index (BI) at Jeulingke Village, Syiah Kuala Subdistrict, Banda Aceh. This research was conducted by using Single Larva Method. Single Larvae Method is a method used to take a single larva by using a plastic scoop on each container that is found to have a larva and visually observed by using a flashlight. The sample used consisted of 120 houses taken randomly from 6 hamlets. The larval data were obtained from various types of containers tabulated into tabular form, then the larvae index was calculated to determine the density of the larvae based on the Density figure criteria. The results showed that from 120 houses found 702 containers in Jeulingke Village with 20 positive larval houses and 20 containers that positively contained larvae such as 19 bath containers and used tires by 1 container so that the index of larvae obtained with House Index is 17% , Container Index of 3%, Breteau Index 20, so as to obtain Density Figure value in the range of numbers 2-4. The results show that the Density Figure in Jeulingke Village is in the medium category.

Abstract

This study was aims to determine the fat content of depik fish (Rasbora tawarensis) collected from Laut Tawar Lake Central Aceh. The samples used were fresh fish from traditional catching around Laut Tawar lake with the weight of 12.45 and 12.74 gram. The samples were analyzed by Soxhlet method with two repetitions. The result of this study showed that the fat content of depik fish was 6,50%. It can be concluded that depik fish from Laut Tawar lake Central Aceh had average of fat content of 6,50%.Key words: Rasbora tawarensis, Laut Tawar, Aceh Tengah

Abstract

Penelitian ini bertujuan mengetahui pengaruh ekstrak etanol sarang semut (Myrmecodia sp.) terhadap gambaran histopatologi ginjal mencit  (Mus musculus) jantan yang hiperurisemia. Penelitian ini menggunakan 25 ekor mencit jantan dengan umur 10 minggu yang secara klinis dinyatakan sehat. Secara acak seluruh mencit dibagi menjadi 5 kelompok perlakuan, masing-masing kelompok perlakuan terdiri atas 5 ekor mencit. Kelompok K0 adalah kelompok kontrol negatif yang diberikan pakan standar, K1 adalah kelompok kontrol positif yang diberikan pakanstandar dan diinduksi pakan purin tinggi, K2 adalah kelompok yang diberikan pakan standar, diinduksi pakan purin tinggi dan diterapi allopurinol, K3 dan K4 adalah kelompok yang diberikan pakan standar, diinduksi pakan purin tinggi dan diterapi dengan ekstrak etanol sarang semut masing-masing dengan dosis 100  dan 200  mg. Setelah perlakuan selesai pada hari ke-22,  organ ginjal diambil untuk dibuat preparat histologi dan diwarnai dengan metode haematoksilin eosin. Data yang diperoleh dianalisis dengan analisis varian (Anova). Rata-rata tubulus proksimal yang mengalami penyempitan pada kelompok K0; K1; K2: K3; dan K4 masing-masing adalah 32,95±1,37; 67,74±1,81; 75,53±2,68; 24,11±2,61; dan 13,50±2,90. Pemberian ekstrak etanol sarang semut berpengaruh sangat nyata (P0,01) terhadap gambaran histopatologi ginjal mencit jantan dalam memperbaiki penyempitan lumen tubulus proksimal mencit jantan yang diinduksi pakan purin tinggi.  Dapat disimpulkan bahwa pemberian ekstrak etanol sarang semut dengan dosis 200 mg memiliki efektivitas lebih baik dibandingkan dengan dosis 100 mg dalam memperbaiki penyempitan lumen tubulus proksimal.

Abstract

This study aimed to detect the Second Internal Transcribed Spacer (ITS-2) encoding gene of Sarcoptes scabiei from rabbits. ITS-2 is a highly conserved gene and always used to identify and differentiate host species infected by S. scabiei. Isolates of S. scabieiwere collected from samples of rabbit skin scrapings suspected of being infected with S. scabiei. The rabbits were acquired from Surabaya, Sidoarjo, Pasuruan, Mojokerto and Nganjuk. Detection on the presence of S. scabiei was carried out microscopically then further confirmed by PCR method and visualized using electrophoresis. PCR identification results showed DNA bands at 304 base pairs. The results showed that samples were infected by S. scabiei, determined by the presence of ITS-2 encoding gene. The ITS-2 encoding gene was chosen as the main focus in this study because it was easily amplified that made analysis and identification easier. The high cases of scabies in Indonesia can be a warning to prevent infection of S. scabiei, especially in pets that have the potential as a reservoir in spreading diseases.

Abstract

           The purpose of this research was to identified the compounds in ethanol extract of soursop flower and determined the antibacterial activity of the soursop flower extract on Salmonella enteritidis. The phytochemistry screening was done to identify the secondary metabolite compounds of this extract. The concentrations used of ethanol extract of soursop flower were 10%, 20%, 30%, 40%, 50%, with ampicillin 10 μg/disc as the positive control and 10% Dimethyl Sulfoxide (DMSO) as the negative control. The antibacterial activity test of soursop flower ethanol extract against Salmonella enteritidis was held in vitro using the paper disc diffusion method by measuring the diameter of the inhibition zone. The phytochemistry screening showed that the ethanol extract of soursop flower had secondary metabolites such as alkaloids, phenolic, and flavonoid. The result of the antibacterial activity test showed that there was no inhibition zone (bright zone) at various concentrations. So, it can be concluded that soursop flower ethanol extract (Annona muricata L.) contained the secondary metabolites such as alkaloids, phenolic, and flavonoid, and also this extract had no antibacterial activity against Salmonella enteritidis

Abstract

This study was aimed to find out the morphology and carbohydrates content of lingual and sub-lingual glands of duck (Anas plathyrinchos). Sample used in this study was lingual and sublingual glands of a one year old female duck. Anatomy morphology and histology of lingual and sublingual glands were observed after processed into histological preparation and stained with hematoxylin-eosin (HE) then stained further with alcian blue (AB) pH 2.5 and periodic acid Schiff (PAS) to detect carbohydrate contents. The results showed that the lingual gland of duck cannot be observed due to its location below the epithelial layer of the tongue. While the sublingual gland of duck has an elongated shape and not granulated like typically glands, located at ventral tongue (dorsoventral lingual) between os mandibulae and covered with musculus digastricus venter oral. Staining with AB pH 2.5 and PAS showed that cytoplasm of secretory cell and secrete of lingual and sublingual glands contain acidic and neutral carbohydrates which indicated a positive reaction with intensity ranging from weak (+) to strong (+++). This study concluded that the lingual and sublingual gland found in female duck is a complex tubular gland with mucous secretory cells type. ____________________________________________________________________________________________________________________

Abstract

Penelitian ini bertujuan mengetahui kadar proksimat lemak pada ikan nila yang diberi suplemen tepung daun jaloh yang dikombinasi kromium dan dipapar stres panas. Sebanyak 30 ekor ikan nila dengan panjang 1,3-1,5 cm, dibagi menjadi enam perlakuan dengan lima kali ulangan yang terdiri atas pakan komersil pada suhu 29±1° C (P1S1), pakan komersil yang dikombinasi tepung daun jaloh pada suhu 29±1° C(P2S1), pakan komersil yang dikombinasi tepung daun jaloh dan kromium suhu pada 29±1° C (P3S1), pakan komersil pada suhu 35±1°C (P1S2), pakan komersil yang dikombinasi tepung daun jaloh pada suhu 35±1° C (P2S2), dan pakan komersil yang dikombinasi tepung daun jaloh dan kromium pada suhu 35±1° C (P3S2). Setelah 15 hari, dihitung kadar proksimat lemak menggunakan metode Soxhlet dan Folsch. Hasil penelitian menunjukkan bahwa rataan kadar proksimat lemak menurun. Berdasarkan uji statistik rancangan acak lengkap faktorial menunjukkan adanya pengaruh yang sangat nyata (P0,01) oleh suhu dan pakan serta interaksi suhu dengan pakan juga berpengaruh sangat nyata (P0,01) terhadap kadar proksimat lemak. Dari hasil penelitian disimpulkan bahwa pakan komersil yang dikombinasikan daun jaloh dengan kromium dapat menurunkan kadar proksimat lemak ikan nila yang terpapar stres panas.
