Abstract

The Muslim community has difficulties in determining the halal status of animal-derived items and their derivatives due to their extensive prevalence. Islamic customers are increasingly becoming more selective and are demanding halal certification for culinary goods. Developing a way to identify porcine DNA in goods is of utmost importance. The Cyt b gene is being used to evaluate swine DNA samples by using primer candidates. The process of generating primers utilizing bioinformatics tools, such as NCBI, MegaXI, Primer3Plus, SnapGene Viewer, and Net Primer websites, is utilized to assess and analyze the effectiveness of laboratory research. The study concluded that the primers effectively amplified pig DNA, but they were unsuccessful in amplifying chicken or beef DNA. After conducting in silico experiments, a total of 7 possible primers were generated. The most advantageous pair of primers was identified, which includes the forward primer 5'-AACATCCGAAATCACACCC-3' and the reverse primer 5'-AGAATGATATTTGTCCTCAGGG-3'. The efficacy of these primers was evaluated in a controlled laboratory environment. Results from the laboratory experiments demonstrate that these particular primers have the ability to amplify the Cyt b gene from the Sus scrofa species at a temperature of 58°C, producing a DNA fragment that is 415 base pairs long. DNA sequencing is essential to verify that the amplified DNA band matches to the Sus scrofa Cyt b gene. Keywords:          PCR, Porcine, Primer, Cyt b, Halal

Abstract

Alpha-lipoic acid as a co-antioxidant can increase the effectiveness of green tea extract as antiaging in the NLC system by increasing stability. This research aims to prove that the preparation of NLC green tea extract with alpha-lipoic acid do not irritate the skin. The method used is making an NLC system using High Shear Homogenization with F1 (without alpha-lipoic acid), F2 (1% alpha-lipoic acid) and F3 (1.5% alpha-lipoic acid) which were tested for irritation in vivo using the back skin of mice. Then observations were made to count the number of inflammatory cells and scoring was carried out. The results obtained were that all formulas had a scoring value of 1 (average number of inflammatory cells 8-15) and were categorized as diffuse inflammatory cells with low density because they were below 50 and the sig value. > 0.05 which means the preparation group is the same as normal. In conclusion, all formulas, both F1, F2 and F3, do not cause irritation. Penelitian ini bertujuan membuktikan sediaan NLC ekstrak teh hijau dengan asam alfa-lipoat tidak iritasi pada kulit. Metode yang digunakan yaitu pembuatan sistem NLC menggunakan High Shear Homogenization dengan F1 (tanpa asam alfa-lipoat) , F2 (1% asam alfa -ipoat) dan F3 ( 1,5% asam alfa-lipoat ) yang diuji iritasi secara in vivo dengan menggunakan kulit punggung mencit. Kemudian dilakukan pengamatan menghitung jumlah sel radang dan dilakukan skoring. Hasil yang didapatkan yaitu pada semua formula memiliki nilai skoring 1 (rata-rata jumlah sel radang 8-15) dan masuk kategori sel radang menyebar dengan kepadatan rendah karena dibawah 50 serta nilai sig. > 0,05 yang berarti kelompok sediaan sama dengan normal. Abstract

Skin wrinkles are caused by the cleavage of collagen fibers by the major collagenase enzyme matrix metalloproteinase-1 (MMP-1). Peptide-based matrix metalloproteinase-1 inhibitors present a potential new therapeutic strategy to prevent MMP-1-mediated collagenolysis, which is induced by UV-induced MMP-1 overexpression. This in silico study investigated the MMP-1 inhibitory capacity of carefully selected and well-designed hexapeptides. A molecular docking method was employed to screen 14 hexapeptides. The target ligands were observed to interact non-covalently. The LYYYGP peptide sequence was found to form three hydrogen bonds (H-bonds) with amino acid (AA) residues in the catalytic domain of MMP-1. Additionally, hydrophobic interactions between these peptides and the protein were observed at certain sites. The maximum binding affinity was observed for the LYYYGP sequence (binding free energy, ?G = -9.00 kcal/mol). These sequences exhibited binding energies comparable to those of the LSYYGP sequence (-8.48 kcal/mol) and YSGYYP sequence (-8.27 kcal/mol). The YSGYGY sequence demonstrated a lower binding affinity for the target receptor, with ?G = -5.59 kcal/mol. This study indicates that specific peptide sequence components are crucial for forming hydrogen bonds that link proteins to peptides. Further investigation, encompassing chemical synthesis and laboratory experimentation, is requisite to substantiate the efficacy of the two hexapeptides.

Abstract

Jarak Merah (Jatropha gossypifolia) is widely grown in Indonesia. Its leaves are used to treat wounds, ulcers, itching, fever, stomach pain and swelling. To be developed into traditional medicine, research was conducted with phytochemical tests, antioxidant tests, and toxicity tests. Ethanol-water was used to dissolve non-polar and polar compounds in red castor leaves. The highest yield was obtained from ethanol-water extract (0:1) which was 31.2%. Qualitative phytochemical testing detected the presence of tannins, flavonoids, phenols, alkaloids, saponins, steroids, while terpenoid compounds were not detected. The highest phenolic compound content was obtained from ethanol-water extract (2:1) 175.714 mg GAE/g. The highest flavonoid content was obtained from ethanol-water extract (2:1) 11.982 mg QE/g. The highest tannin content was obtained from ethanol-water extract (0:1) 1.339 mg TAE/g. Strong antioxidant activity was shown by ethanol-water extract (1:0) with IC50 17.97 ppm. Ethanol-water extract (1:2) showed toxicity in the highly toxic category with LC50 22.45 ppm. Daunnya digunakan untuk mengobati luka, bisul, gatal-gatal, demam, sakit perut dan bengkak. Untuk dapat dikembangkan menjadi obat tradisional, dilakukan penelitian dengan uji fitokimia, uji antioksidan dengan metode penangkapan radikal DPPH, dan uji toksisitas dengan metode BSLT. Digunakan etanol-air untuk melarutkan senyawa non polar dan polar dalam daun jarak merah. Rendemen tertinggi diperoleh dari ekstrak etanol-air (0:1) yaitu 31,2%. Pengujian fitokimia secara kualitatif mendeteksi adanya senyawa tanin, flavonoid, fenol, alkaloid, saponin, steroid, sedangkan senyawa terpenoid tidak terdeteksi. Kandungan senyawa fenolik tertinggi diperoleh dari ekstrak etanol-air (2:1) sebanyak 175,714 mg GAE/g. Kandungan flavonoid tertinggi diperoleh dari ekstrak etanol-air (2:1) sebanyak 11,982 mg QE/g. Kandungan tanin tertinggi diperoleh dari ekstrak etanol-air (0:1) sebanyak 1,339 mg TAE/g. Aktivitas antioksidan yang kuat ditunjukkan oleh ekstrak etanol-air (1:0) dengan IC50 17,97 ppm. Abstract

Children in the school-age group are in the process of transitioning from childhood to adolescence. The period of childhood is crucial for establishing a strong basis for the development of high-quality individuals, and oral health plays a significant role in determining the quality of human resources. Dental caries is a frequently encountered issue in teeth and oral health. Childhood dental caries can result in discomfort, impairing the ability to chew (mastication), impacting food consumption, and contributing to inadequate nutritional health. The objective of this study is to establish the correlation between the def-t dental caries index and the nutritional status of children aged 6-9 years in SDN 003 Sungai Kunjang, Samarinda City. This study is a cross-sectional research conducted on a group of 163 students. The data underwent processing and analysis through the utilization of the Chi-Square test. The findings indicated that the prevalence of dental caries among children aged 6-9 years in SDN 003 Sungai Kunjang was relatively low (43.5%). Additionally, the most prevalent category of nutritional status was found to be normal, accounting for 49.6% of the sample. The correlation between dental caries and nutritional status was found to be statistically insignificant, with a p-value of 0.802 (p > 0.05). The study reveals no significant correlation between the def-t dental caries index and nutritional status among children aged 6-9 years in SDN 003 Sungai Kunjang, Samarinda City. Keywords:          Dental caries, nutritional status, young individuals   Abstrak Anak sekolah merupakan usia peralihan dari anak-anak menjadi remaja. Anak usia sekolah merupakan masa untuk meletakkan landasan kokoh bagi terwujudnya manusia yang berkualitas dan kesehatan rongga mulut merupakan faktor penting yang menentukan kualitas sumber daya manusia. Permasalahan yang umum terjadi pada gigi dan mulut adalah karies gigi. Karies gigi yang terjadi pada anak akan menyebabkan munculnya rasa sakit sehingga menyebabkan terganggunya fungsi pengunyahan (mastikasi) yang mempengaruhi asupan makan dan membuat status gizi menjadi buruk. Tujuan penelitian ini untuk mengetahui hubungan indeks def-t karies gigi dengan status gizi pada anak usia 6-9 tahun di SD Negeri 003 Sungai Kunjang Kota Samarinda. Penelitian ini merupakan penelitian dengan pemilihan desain cross sectional dengan banyak sampel 163 siswa. Data diolah dan dianalisis dengan uji Chi-Square. Hasil penelitian menunjukkan distribusi karies terbanyak pada anak usia 6-9 tahun di SDN 003 Sungai Kunjang adalah rendah (43,5%) dan distribusi kategori status gizi terbanyak adalah normal (49,6%). Korelasi karies gigi dengan status gizi menunjukkan p = 0,802 (p > 0,05). Disimpulkan bahwa tidak terdapat korelasi yang bermakna antara indeks def-t karies gigi dengan status gizi pada anak usia 6-9 tahun di SD Negeri 003 Sungai Kunjang Kota Samarinda. Abstract

Borax is a chemical used in making detergent and insect repellent. However, there is still a lot of use of borax as an additional ingredient in food with the aim of preservative and improving texture, if borax enters the human body it can cause toxic effects. This research aims to identify and measure borax levels in meatballs and wet noodles sold at the Traditional Market. Identification of borax is carried out using qualitative methods, namely organoleptic tests, numeric paper, and flame, while quantitative determination of borax levels can be done using the visible spectrophotometric method. The analytical method validation parameters carried out are linearity, LOD, LOQ, accuracy, and precision. In this study, the maximum wavelength obtained was 546.8 nm and incubation was 35 minutes. It was identified that 8 samples of wet noodles sold at the Traditional Market contained borax at levels of 9,458.04 - 26,279.46 mg/kg. These results show that the use of borax in food is still occurring and violates the regulations set by BPOM regulations. Namun, masih banyak penyalahgunaan boraks sebagai bahan tambahan pada makanan bertujuan sebagai pengawet dan memperbaiki tekstur. Jika boraks masuk ke dalam tubuh manusia dapat menimbulkan efek toksik. Penelitian ini bertujuan untuk mengidentifikasi dan mengukur kadar boraks pada bakso dan mie basah yang dijual di Pasar Tradisional. Identifikasi boraks dilakukan dengan metode kualitatif yaitu uji organoleptik, kertas tumerik, dan nyala api, sedangkan penentuan kadar boraks secara kuantitatif dapat dilakukan dengan menggunakan metode spektrofotometri visibel. Parameter validasi metode analisis yang dilakukan adalah linieritas, LOD, LOQ, akurasi, dan presisi. Pada penelitian ini, panjang gelombang maksimum yang diperoleh adalah 546,8 nm dan inkubasi 35 menit. Teridentifikasi 8 sampel mie basah yang dijual di Pasar Tradisional mengandung boraks dengan kadar yaitu 9.458,04-26.279,46 mg/kg. Hasil ini menunjukkan masih terjadi penggunaan boraks pada makanan dan melanggar peraturan yang ditetapkan oleh peraturan BPOM. Abstract

Diabetes mellitus is frequently described as a chronic metabolic condition characterized by hyperglycemia. Diabetes symptoms induce sleep disturbances, thereby compromising the participant’s quality of life. The primary aim of this study was to establish a correlation between sleep quality and quality of life among type 2 diabetic patients at Pasundan Community Health Centers, Samarinda. The study design that applied in this study was an observational analytics with a cross-sectional approach. Data was obtained through respondent identity forms, PSQI questionnaires, and DQOL-BCI questionnaires. This study involved 58 participants with type 2 diabetes mellitus who were chosen through accidental sampling method and fulfilled the researcher's specific criteria for inclusion. The outcome showed 91.4% had poor sleep quality, while 56.9% had good quality of life. Furthermore, the Pearson test revealed a significant correlation between sleep quality and quality of life (p-value = 0,022, r = -0,301). Sebanyak 537 juta penderita diabetes tersebar secara global. Manifestasi klinis yang beragam pada diabetes mellitus berdampak negatif terhadap kualitas tidur dapat memengaruhi kualitas hidup penderita. Penelitian ini bertujuan untuk mengidentifikasi korelasi kualitas tidur terhadap kualitas hidup pada penderita diabetes mellitus tipe 2 di Puskesmas Pasundan Samarinda. Pendekatan penelitian yang diterapkan adalah analitik observasional dengan desain potong lintang. Data dikumpulkan dari formulir identitas responden, formulir PSQI (The Pittsburgh Sleep Quality Index), dan formulir DQOL-BCI (The Diabetes Quality of Life Brief Clinical Inventory). Sampel dari penelitian ini melibatkan 58 penderita diabetes mellitus tipe 2 yang didapatkan menggunakan teknik accidental sampling menurut kriteria inklusi peneliti. Hasil penelitian menunjukkan sebanyak 91,4% memiliki kualitas tidur buruk dan 56,9% memiliki kualitas hidup baik. Peneliti memakai uji Pearson dan diperoleh nilai p sebesar 0,022 dan r sebesar -0,301. Kesimpulan didapatkan adanya korelasi bermakna antara kualitas tidur dengan kualitas hidup penderita diabetes mellitus tipe 2. Abstract

Diabetes mellitus (DM) is a metabolic disease that can cause damage to several organs. IDF projects an increase in DM by 2045 to 46%. The study aimed to evaluate the treatment of patients with type 2 DM and the potential for drug interactions. The research design was observational and retrospective data collection on prescriptions and medical records of type 2 DM patients at Banguntapan 1 Health Center, January- February 2023, a total of 125. The results of the study were that most type 2 DM patients were female gender (62%), age 46-65 years (70%), and hypertension comorbidities (78%). The most widely used antidibetic was the metformin-glimepiride combination (83%). The conclusions of the study were dose accuracy (100%), accuracy of the time interval of administration (98.4%), and potential drug interactions (66%), with the most interactions being metformin and amlodipine (71%). IDF memproyeksikan peningkatan DM pada tahun 2045 menjadi 46%.  Tujuan penelitian ini untuk mengevaluasi pengobatan pasien DM tipe 2 serta potensi interaksi obat. Desain penelitian adalah observasional dan pengambilan data secara retrospektif pada resep serta rekam medik pasien DM tipe 2 di Puskesmas Banguntapan 1, bulan Januari-Februari 2023, sejumlah 125. Hasil penelitian yaitu pasien DM tipe 2 terbanyak adalah jenis kelamin perempuan (62%), usia 46-65 tahun (70%) dan penyakit penyerta hipertensi (78%). Antidibetik yang paling banyak digunakan adalah kombinasi metformin-glimepirid (83%). Kesimpulan penelitian adalah ketepatan dosis (100%), ketepatan interval waktu pemberian (98,4%), serta potensi interaksi obat (66%), dengan interaksi paling banyak adalah metformin dan amlodipin (71%).
