Abstract

The Indonesian government officially announced a positive case of a patient exposed to COVID-19 in 2020. The purpose of the study was to determine the characteristics of the community in Kiringan Hamlet, calculate the willingness to pay (WTP) value for temulawak and determine the factors that influence the WTP value during COVID-19. The method used descriptive-analytical non-experimental, which involved distributing questionnaires to consumers and herbal medicine producers in Kiringan Village, Bantul, Yogyakarta. The number that met the inclusion criteria was 276 respondents. The primary data obtained were then analyzed descriptively, followed by a WTP analysis using the contingent valuation modeling (CVM) approach and a factor analysis using regression analysis. The results showed that the age distribution obtained from 69 respondents was 36-45 years old (25%), the majority of female gender with 156 respondents (57%), education level was high school with 137 respondents (50%), 172 people (62%) with income level below 1,000,000.00 IDR, the average WTP value of temulawak was 10,956.52 IDR. In contrast, the total WTP value of temulawak reached 3,024,000.00 IDR. Occupation factors (p=0.004), number of family members (p=0.040), and duration of consumption (p=0.005) also affected the WTP of temulawak.

Abstract

Soil-Transmitted Helminth infection (STH) such as ascariasis, has become one of the most persistent parasitic diseases in most developing countries, including Indonesia. Galangal, turmeric, and ginger plants are known to provide various of pharmacologically active substances which commonly used as antibacterial, aphrodisiac, and antiparasitic. This research was aimed to study the anthelmintic activity of ethanol extract of ginger, turmeric, galangal in single extract as well as their combination against Ascaris suum Goeze as a model. The anthelmintic activity was analyzed by determining the profile of time of paralysis (TOP), time of death (TOD), and the IC50­ of the inhibition activity from single extracts and extract combinations as well as the combination index (CI). The results showed that galangal, turmeric, and ginger ethanol extract have anthelmintic activity with IC50 7.66, 22, and 13,14 mg/mL, respectively. Meanwhile, the IC50 values for the extract combination of galangal with turmeric (KL), galangal with ginger (JL), and turmeric with ginger (JK) were 10.22; 3.92; and 34.93 mg/mL, respectively. The synergistic combination properties were shown by combination of galangal with turmeric extracts and galangal with ginger extracts with the combination index (CI) 0,67 and 0,38, respectively.

Abstract

Coronavirus Disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), is a contagious illness transmitted from person to person. The main protease (Mpro) of SARS-CoV-2 is a promising target for therapeutic intervention. Noni (Morinda citrifolia L.), a plant commonly used in traditional medicine in Indonesia for treating COVID-19, has been investigated for its potential anti-SARS-CoV-2 compounds. However, the specific bioactive compounds targeting SARS-CoV-2 remain unidentified. This study aimed to identify and predict the activity of compounds in Noni against Mpro through pharmacophore modeling, molecular docking, drug-likeness, and pharmacokinetic analysis using LigandScout®, Discovery Studio Visualizer®, AutoDock®, SwissADME, Pre-ADMET, and Toxtree®. Among the 40 compounds analyzed, quercetin emerged as the most promising candidate, with a pharmacophore fit score of 41.05, a binding energy (ΔG) of -7.79 kcal/mol, and interactions with amino acid residues GLY143A and MET165A, similar to the native ligand. These findings suggest that quercetin from M. citrifolia has the potential to be developed as a novel anti-SARS-CoV-2 therapeutic agent.

Abstract

Produk antistunting menjadi inovasi dalam mengatasi permasalahan malnutrisi, namun keterbatasan pemodelan hewan uji menjadi salah satu hambatan dalam pengembangan produk tersebut. Penelitian ini bertujuan menciptakan hewan model malnutrisi melalui formulasi pakan rendah protein. Formulasi pakan rendah protein dari tepung maizena dan tapioka diberikan kepada hewan uji yang dibagi ke dalam 3 kelompok, yaitu kelompok normal, kelompok maizena dan kelompok tapioka. Induksi pakan dilakukan 15 hari dimana kelompok normal diberikan pakan CP551, sedangkan dua kelompok uji masing-masing diberikan pakan formulasi. Perubahan bobot badan tikus dianalisis selama 15 hari perlakuan. Parameter malnutrisi, yaitu kadar hemoglobin, albumin dan protein total pada darah hari ke-0 (sebelum perlakuan) dan hari ke-15 (setelah perlakuan) diukur dan dibandingkan. Hasil pengamatan bobot badan menunjukkan kedua pakan formulasi rendah protein dapat menciptakan kondisi malnutrisi setelah dibandingkan terhadap kelompok normal (p<0,05). Persentase penurunan kadar hemoglobin, albumin dan protein total terbesar ditunjukkan oleh kelompok maizena dengan nilai penurunan masing-masing sebesar 5,68%; 12,70% dan 7,64% dibandingkan kelompok tapioka dengan penurunan masing-masing sebesar 0,53%; 2,30% dan 4,74%. Namun, tidak terdapat perbedaan signifikan perubahan bobot badan antara dua kelompok uji (p>0,05). Formulasi pakan rendah protein dari tepung maizena dan tapioka dapat menciptakan kondisi malnutrisi pada hewan uji dengan formulasi terbaik diberikan oleh pakan maizena.

Abstract

Antibacterial resistance in pathogenic bacteria and their contamination in water are contributing to the spreading of infectious diseases. As a result, isolation and antimicrobial susceptibility testing for drinking water samples are becoming increasingly important in addressing the human health hazards associated with ingesting contaminated water. Therefore, this study was aimed to identify enterobacteria strains in drinking water refill obtained from 20 refill water stations in Jatinangor, West Java, Indonesia and investigated the potential antibacterial activity of rubber plant (Ficus elastica Roxb. ex Hornem) leaf extracts on identified strains of the isolated enterobacteria. Bacterial isolation was accomplished using membrane filtration method and the bacterial identification was performed by observing the cell and colonies morphologies, biochemical testing approach and concluded using the computer program Global Infectious Diseases and Epidemiology Network (GIDEON). The antimicrobial susceptibility test was carried out using the Kirby-Bauer diffusion technique. From 20 water samples, we isolated 30 single colonies which composed of four strains of identified Enterobacteriaceae, as follows: Citrobacter freundii (nine samples), Salmonella typhi (six samples), Serratia marescens (six samples) and Escherichia coli (nine samples) with percentage: 30%, 20%, 20% and 30%, respectively. The extract can inhibit all Enterobacteria isolates, however, among those strains, E. coli was the most susceptible Enterobacteria to be inhibited by F. elastica leaf extract. The current study suggests that this plant potentially to be used as a novel antibacterial agent against Enterobacteria strains as the major gastrointestinal tract pathogenic microbes.

Abstract

Quercetin is a flavonoid compound with the highest antioxidant activity and has the potential to treat various diseases. However, quercetin has poor solubility in water which causes low bioavailability of the drug in the body. One of the modifications that can be made to improve the solubility of a drug is solid modification, for example cocrystals. Cocrystal is a crystalline system between drug and coformer which interacts through hydrogen bonding. Glycolic acid is a compound that can function as a coformer because it has functional groups that have the potential to interact through hydrogen bonds with quercetin molecules. Quercetin-glycolic acid cocrystals were prepared in two molar ratios, namely 1:1 and 1:2, and through two methods of preparation, namely solvent evaporation and slurry. The results of the crystallinity analysis and thermal analysis of the sample indicated that a new solid phase was formed in the sample that was specific and different from the two constituent components. Samples with a molar ratio of 1:2 and the slurry preparation method are known to be samples with the most optimal new crystalline phase formation.

Abstract

The low level of Artemisinin in Artemisia annua L. is associated with its biosynthetic pathway. Genetic engineering offers a potential solution to increase its metabolites. The research objectives were to transform the double bond reductase (dbr2), one of the critical genes in the biosynthesis of Artemisinin, and p19, an anti-silencing gene, into A. annua L. by the mediation of Agrobacterium tumefaciens and assess their impact on metabolites production. The dbr2 and p19 genes were verified by analyzing PCR products. Plasmids pCAMBIA1303-DBR2-P19 were transformed into A. tumefaciens and subsequently introduced into the leaves from tissue culture of A. annua L, using vacuum infiltration and assessed by GUS assay. UPLC-ESI-MS/MS measured artemisinic acid (AA), dihydroartemisinic acid (DHAA), and Artemisinin contents. The transformation in A. tumefaciens was successful using a freeze-thaw method. The tissue culture of A. annua L. has been infected by A. tumefaciens using vacuum infiltration. Based on the GUS histochemical assay, the gene has been successfully inserted in the leaves with an efficiency of 48.2%. The result from UPLC-ESI-MS/MS showed that the level of Artemisinin in the transformation sample with and without pCAMBIA-dbr2-p19 was detected but not quantifiable while in wild-type leaves were quantified at 0.008% in fresh weight. The AA and DHAA were not detectable but only in the wild type. The transformation was successful, but the quantification of DHAA and AA was unsuccessful because of the low quantity in the samples.

Abstract

Labu kuning merupakan tanaman dengan sumber antioksidan tinggi dan antiinflamasi. Kandungan senyawa zat aktif dalam labu kuning yang memiliki aktivitas beragam menjadi hal yang menarik untuk dikembangkan sebagai kandidat kosmetik antiaging. Labu kuning dilakukan formulasi dalam bentuk masker gel peel off untuk meningkatkan efektivitasnya dengan formulasi sediaan yang aplikasinya mudah, mudah dibersihkan dan menarik. Tujuan penelitian dilakukan untuk membuat formulasi dan melakukan evaluasi sediaan masker gel peel off enkapsulasi ekstrak labu kuning. Enkapsulasi ekstrak labu kuning dibuat dalam bentuk sediaan masker gel peel offl dan dilakukan pengamatan parameter fisikakimia. Uji stabilitas sediaan dilakukan dengan pengamatan parameter fisikakimia dievaluasi selama 1 bulan, uji sentrifugasi dan cycling test untuk melihat stabilitas sediaan. Pengujian iritasi dilakukan menggunakan hewan uji kelinci dengan melihat respon udem dan iritasi. Masker gel peel off enkapsulasi ekstrak labu kuning stabil pada uji sentrifugasi dan penyimpanan selama 1 bulan stabil ditunjukkan tidak ada perubahan pada parameter organoleptis, homogenitas, daya sebar, waktu kering, viskositas, pH. Pengujian cycling test selama 6 siklus tidak menunjukkan perubahan organoleptis, homogenitas, daya sebar, daya lekat, waktu kering, viskositas dan pH. Uji iritasi menunjukkan bahwa masker gel peel off labu kuning tidak memiliki efek iritasi pada kulit punggung kelinci dengan pengamatan selama 72 jam dilanjutkan pengamatan 15 hari. Formulasi masker gel peel off enkapsulasi ekstrak labu kuning stabil dan tidak mengiritasi.

Abstract

Udang putih (Litopenaeus vannamei) merupakan salah satu spesies Crustacea yang cangkangnya mengandung senyawa karotenoid. Senyawa ini merupakan antioksidan alami yang berpotensi tinggi. Penelitian ini bertujuan untuk mengetahui karotenoid total pada ekstrak cangkang L. vannamei dan mengevaluasi aktivitas antioksidannya secara in vitro. Ekstrak diperoleh dengan cara maserasi menggunakan n-heksan. Tingkat total karotenoid dalam ekstrak ditentukan setara dengan β-karoten. Spektrofotometer UV-Visible mengukur ekstrak dan larutan standar referensi pada panjang gelombang maksimum β-karoten (462 nm). Dua metode berbeda digunakan dalam menentukan aktivitas antioksidan ekstrak; metode peredaman radikal 1,1-diphenyl-2-picryl-hydrazyl (DPPH) dan metode ferric reducing antioxidant power (FRAP). Hasil penelitian menunjukkan bahwa karotenoid total dalam ekstrak cangkang L. vannamei adalah 5,87 mg/g, setara dengan β-karoten. Konsentrasi hambat setengah (IC50) ekstrak dengan metode DPPH adalah 27,59 µg/mL menunjukkan potensi sedang sebagai antioksidan dibandingkan dengan kuersetin standar sebesar 1,28 µg/mL. Kapasitas antioksidan ekstrak dengan metode FRAP dinyatakan dalam mg setara kuersetin (QE) per g ekstrak (mgQE/g), dan hasilnya menunjukkan potensi penghambatan sebesar 1,65 mgQE/g ekstrak. Dengan demikian, dapat disimpulkan bahwa ekstrak cangkang udang vannamei sangat potensial sebagai sumber karotenoid alami dengan efek antioksidan yang memadai.

Abstract

Sonneratia alba or rambai leaves are a plant from East Kalimantan that the community has used for generations as a cold powder. Antioxidants are needed to ward off free radicals, while bacteria are one of the causes of acne. Propolis also shows potential as an antioxidant and antibacterial agent. This research evaluated whether the combination of Sonneratia alba leaves extract and Heterotrigona itama propolis can increase antioxidant and antibacterial activity and whether this combination has synergistic potential. The S. alba leaves and H. itama propolis were macerated using ethanol as the solvent. The antioxidant activity test using the DPPH method with three comparisons of 1:1, 1:2, and 2:1 produced IC50 values of 12.669 µg/ml, 32.756 µg/ml, and 19.694 µg/ml, respectively. The IC50 value for ascorbic acid is 12.293 µg/ml. The results show that the combination of S. alba leaves and H. itama propolis ethanol extract has very strong antioxidant activity. The antibacterial activity test against Staphylococcus aureus bacteria with three concentrations of 60%, 80%, and 100% showed inhibition zones of 10.5 mm (medium category), 16 mm, and 17.5 mm (strong category), respectively.
