Abstract

Lead (Pb) is a heavy metal that can cause poisoning and accumulate in the human body. Determination of lead levels by using AAS the condition is that the sample must be a solution, then it is necessary to destruct first. There are two types of destruction that can be done, namely wet destruction using acid reagents to decompose samples and dry destruction by heating or destruction using very high temperatures. HNO3 oxidizing agent and HNO3 mixture with HClO4 are the main oxidizing agents that can provide the best destructive results compared to other oxidizing agents. This study aims to determine the best oxidizing and destructive substances present in the hair using Atomic Absorption Spectrophotometry (AAS). The research was conducted at surabaya regional health laboratory in April - May 2021. This research is comparative research. The sample used is the hair of gelam gas station operator officer, Candi. There are two ways of preparing samples that can be done in pb analysis on hair, namely proses wet and dry destruction. The result of destruction is determined by the level of lead using Atomic Absorption Spectrophotometry (AAS). The results of the analysis pno dry destructionHNO3 as a whole has rentang lead levels 0.1281-0.1958 mg / L. Dry destruction HNO3 : HClO4 secara overall has rentang lead levels 0.1457-0.2159 mg /L. HNO3 closed wet construction has lead content range 0.1431-0.2034 mg/L and wet destructive closed HNO3 : HClO4 has lead content range 0.1080-0.5371 mg/L. Based on these results, it can be known that all hair samples tested in this study contain lead levels that are still within normal limits or within the tolerance limit of < 12 μg/g.

Abstract

Background: Sepsis can affect all age, but is most common in young infants and neonates. Young infants and neonates are more susceptible to infection because their immune systems are immature, unable to localize infections, and lack the IgM needed to protect the body against bacterial infections. When experiencing neonatal sepsis, there is often an increase in leukocytes in response to infection and inflammation, where inflammation can also cause a decrease in albumin in the blood. Objective: This study aims to analyze the relationship between albumin levels and leukocyte counts in the incidence of neonatal sepsis. Methods: The research method used analytic correlation with a cross sectional approach. A sample of 31 patients with neonatal sepsis who were treated at Waluyo Jati hospital, Probolinggo Regency in December 2022 - April 2023 were sampled using saturation sampling method. Results: The results of the study on neonatal sepsis based on laboratory examinations mostly occurred in conditions of leukocytosis (45.2%) and hypoalbuminemia (77.4%), while the results of the study based on Pearson correlation test analysis obtained a significant value (p) of 0.737 (α=0.05). Conclusion: there is no relationship between albumin levels and leukocyte counts in neonatal sepsis.

Abstract

Proses penuaan dapat memengaruhi kesehatan fisik lansia dan meningkatkan risiko berbagai gangguan kesehatan, termasuk hipertensi dan asam urat. Kegiatan ini berlangsung pada tanggal 14 Juni 2023 bertempat di Balai RW 2 Kelurahan Kertajaya, Surabaya dan dihadiri 59 lansia yang berdomisili di RW 1 dan RW 2 Kelurahan Kertajaya Kecamatan Gubeng Surabaya. Jenis kegiatan yang dilaksanakan adalah pengukuran status gizi, asam urat dan senam hipertensi. Hasil pemeriksaan mengungkapkan 57,6% lansia memiliki kadar asam urat yang tinggi dan mayoritas memiliki kelebihan berat badan, sehingga lansia perlu mengendalikan tekanan darah dengan pengobatan yang diresepkan oleh dokter, perubahan gaya hidup seperti diet rendah garam, olahraga teratur, dan menghindari stres.

Abstract

Gas stations are a source of lead exposure that comes from vehicle gas emissions and petrol fuel vapors. Gas station officers, both operators and nonoperators, have a risk level of lead exposure that can enter the body and then accumulate and excreted through urine by 75-80%. Lead exposure can endanger the health of gas station officers. The purpose of this study was to analyze differences in lead levels in the urine of gas station operators and nonoperators in Kademangan District, Probolinggo City. This research is an observational study with a cross sectional method which was conducted at the Surabaya Health Laboratory Center in October 2020 - June 2021. The sample used was 15 operators and 15 nonoperators who were taken by purposive sampling. The results showed the average value of lead levels in the urine for operators 0.0007213 ppm, indoor nonoperators 0.0006386 ppm and outdoor nonoperators 0.0008263 ppm. The conclusion of this study was that there was no significant difference in lead levels in the urine of gas station operators and nonoperators.

Abstract

Infeksi protozoa usus masih menjadi salah satu permasalahan di dunia, terutama di negara-negara berkembang dan beriklim tropis. Angka insidensi kasus protozoa usus di Indonesia mencapai 10-18%. Infeksi protozoa usus dapat menyerang anak-anak dan orang dewasa (Deza et al., 2018). Di Negara berkembang tingkat pendidikan masih rendah dan iklim tropis merupakan faktor risiko infeksi protozoa usus. Hasil penelitian Charisma dkk. (2020), menyebutkan bahwa pada pemeriksaan sampel feses menunjukkan sebanyak 20,8% positif adanya kista protozoa usus. Didukung oleh penelitian Simatupang et al., (2013) yang melaporkan sebanyak 40% kasus infeksi didominasi oleh protozoa usus daripada cacing usus (STH) pada anak usia SD di Kedung Cowek, Surabaya. Rendahnya pengetahuan masyarakat terkait penyakit parasit masih menjadi salah satu masalah. Dengan demikian, pemberian edukasi kebersihan personal pada mereka menjadi salah satu solusi untuk menurunkan angka infeksi parasit usus (Fransisca et al., 2015). Dengan demikian, peneliti akanmengidentifikasi keberadaan protozoa usus pada sampel feses yang bertujuan untuk mengetahui prevalensi protozoa usus dan gambaran kebersihan personal pada anak usia sekolah dasar di desa Ngingas Barat, kecamatan Krian kabupaten Sidoarjo. Dari hasil pretes dan postes terdapat terdapat peningkatan para siswa dalam menjawab soal dari 2.16 menjadi 3.66. Soal yang paling sulit dijawab oleh siswa adalah nomor 4 yaitu tentang protozoa. Setelah mendapat materi tentang protozoa usus, para siswa mampu menjawab soal nomor 4 pada postes. Peningkatan pengetahuan tentang protozoa usus sebagai penyebab diare diharapkan dapat mengubah sikap dan tindakan para siswa menjadi lebih baik lagi untuk mencegah terjadinya penyakit tersebut. Satu minggu kemudian setelah diadakan kegiatan penyuluhan para siswa dilakukan pemeriksaan Feses lengkap untuk diperiksa adanya infeksi protozoa. Hasil pemeriksaan laboratorium dinyatakan semua siswa negatif dari infeksi protozoa atau 0%.

Abstract

Pemeriksaan Hapusan darah tepi (HDT) merupakan pemeriksaan untuk melihat morfologi sel-sel darah secara mikroskopis. Salah satu metode pemeriksaan hapusan darah tepi yang sering digunakan adalah pewarnaan giemsa. Pengenceran larutan giemsa secara teoritis menggunakan larutan penyangga buffer phosphat. Pengenceran giemsa sering menggunakan larutan buffer phosphat karena buffer phosphat mudah didapatkan, namun harganya cukup mahal. Kandungan phosphat yang melebihi batas 2 mg/L juga dapat berpengaruh terhadap keseimbangan ekosistem perairan sehingga dapat mencemari lingkungan bila terus digunakan. Penelitian ini bertujuan untuk menganalisis perbedaan morfologi sel-sel darah pada pemeriksaan HDT dengan pewarnaan giemsa menggunakan larutan buffer dan aquabidest dalam upaya penentuan larutan alternatif yang bisa digunakan sebagai larutan pengencer giemsa yang lebih murah dan mudah didapatkan. Penelitian ini merupakan penelitian eksperimental dengan studi cross sectional yang membandingkan kualitas morfologi sel darah sediaan HDT dengan pewarna giemsa yang diencerkan dengan buffer phosphate dan aquabidest. Sampel diambil dari 1 orang koresponden penelitian, untuk kemudian dibuat sediaan HDT masing-masing sebanyak 17 buah (pengencer buffer phosphat), dan 22 buah (pengencer aquabidest), dan diamati dengan mikroskop perbesaran lensa objektif 100x. Kualitas sediaan HDT giemsa dengan pengencer buffer phosphat menunjukkan hasil “baik” sebanyak 13 buah (76,47%), dan hasil “kurang baik” sebanyak 4 buah (23,53%), sedangkan kualitas sediaan HDT giemsa dengan pengencer aquabidest menunjukkan hasil “baik” sebanyak 18 buah (81,82%), dan hasil “kurang baik” sebanyak 4 buah (18,18%). Berdasarkan uji Mann-Whitney U, didapatkan P-Value 0,222. Kualitas sediaan HDT pewarnaan giemsa dengan pengencer buffer phosphat tidak menunjukkan perbedaan yang signifikan dengan yang menggunakan pengencer aquabidest (p>0,05).

Abstract

Subclinical inflammation and almost every indication of systemic inflammation, which is defined by elevated levels of inflammatory markers in the blood, are seen in patients with diabetes mellitus (DM). Understanding the relationship between inflammatory markers and glycemic control is essential for creating comprehensive management strategies to avoid diabetes-related complications. The aims of this study were to determine the correlation between erythrocyte sedimentation rate (ESR) and high-sensitivity C-reactive protein (hs-CRP) and glycated hemoglobin (HbA1c) in DM patients. This was an analytical observational study with a cross-sectional design that was conducted on 35 DM patients from the DM prolanis group at the Bangkalan Regency First-Level Health Facility. HbA1c and hs-CRP levels were measured using the fluorescence immunoassay (FIA) method, while ESR was assessed using the Westergren method. Statistical analysis was performed using the Spearman correlation test. Regarding the biomarkers, all respondents (100%) had HbA1c levels >8.0%, 43% had elevated hs-CRP levels associated with cardiovascular risk, and 80% had abnormal ESR values. A significant relationship was found between ESR and HbA1c (p = 0.019; r = 0.394), but no significant relationship was observed between hs-CRP and HbA1c (p = 0.351; r = 0.163). The relationship between inflammation and glycemic control in DM can be understood through the correlation of ESR, hs-CRP, and HbA1c. These findings offer practical value, particularly in monitoring disease progression and managing diabetes-related complications. Future research should investigate the effects of interventions aimed at reducing inflammation, e.g lifestyle modifications or anti-inflammatory medications, on ESR, hs-CRP, and HbA1c levels.

Abstract

As people age, the physical state of the human body will gradually deteriorate due to a decrease in the quantity of cells present, leading to a gradual reduction in functionality referred to as the aging process. In elderly humans (aged more than 60 years) excessive cholesterol levels and high blood pressure are one of the causes of diseases such as heart disease which has been recognized by WHO as the number one cause of death in the world. The buildup of fatty deposits on the linings of blood vessels has the potential to lead to obstructions within the blood vessels. The objective of this initiative is to enhance understanding regarding the hazards associated with cholesterol. This outreach effort took place in July 2023, conducted at Posyandu Abiyoso, Surabaya Health Polytechnic. The activity consists of several processes 1). conducting a survey of the location of the Abiyoso posyandu of the Surabaya Health Polytechnic, 2). Opening registration and taking participants' personal data, 3) conducting participant health interviews, 4). Checking cholesterol levels and blood pressure of participants. From the community service activities carried out, it was found that 42 of the 65 (64.6%) participants who took part in the examination at the integrated service post had high cholesterol levels, and 60 of the 65 (92.3%) participants had high blood pressure or hypertension. By conducting health checks on cholesterol levels and blood pressure, there is an expectation that the society will come to realize and comprehend. the dangers of hypertension and hypercholesterolemia so that the community can prevent the occurrence of sustainable diseases.

Abstract

Background: Peripheral blood smear examination (HDT) is a test to observe the morphology of blood cells microscopically. A peripheral blood smear is a simple method, and its examination is widely available in laboratories. The advantage of peripheral blood smear examination is its ability to assess various components of peripheral blood cells, such as cell morphology (erythrocytes, leukocytes, and platelets). One of the commonly used methods for peripheral blood smear examination is Giemsa staining. The process of Giemsa staining fixation aims to preserve the cell structure and its components before the staining is performed. The precise fixation time is key to obtaining optimal staining results, which allows for accurate observation of cell morphology. The aim of this study is to analyze the morphology of leukocyte profiles in peripheral blood smears stained with Giemsa based on variations in fixation time to determine the optimal time. Methods: This research was conducted in August-September 2024. The sample in this study was whole blood taken from the academic community of the Department of Medical Laboratory Technology and examined with a peripheral blood smear using Giemsa staining, and analyzed using descriptive statistical tests with the SPSS for Windows 22 software. Results: The research results showed that at a fixation time of 3 minutes, 60% were good and 40% were less good; at 5 minutes, 100% were good; at 10 minutes, 100% were good; at 15 minutes, 40% were good and 60% were less good; and at 20 minutes, 20% were good and 80% were less good. Conclusion: The best time variation in Giemsa staining using phosphate buffer is fixation for 5 minutes and 10 minutes, where all preparations (5 preparations) are 100% in the good category.
