Abstract

In ruminants, interferon-tau (IFN–τ)  is well known as an important pregnancy factor. Interferon-tau as antiluteolytic cytokine is secreted from the tropoblast during the time of implantation. About 10 – 15% of pregnancy losses in cows were possibly due to the lack of IFN–τ to maintain corpus luteum. Interferon-tau induces some early pregnancy factors which are expressed around 15 days post insemination. This has led that IFN–τ to be a strong candidate for the development of ruminant early pregnancy tests. The aim of this paper is to review the existing literatures on the roles of IFN–τ in biology reproduction as well as researches that have been carried out on reproductive biotechnology especially in regards to the development of early pregnancy test for ruminant. It can be concluded that administration of IFN–τ is able to improve reproductive performance of female ruminants experimentally. Interferon-tau and its derivates can be used to develop a rapid test for early pregnancy diagnostic.   Key words: Interferon-tau, early pregnancy test, ruminant

Abstract

Nasopharyngeal Carcinoma, an aggressive cancer in the head and neck region is mostly diagnosed at an advanced stage and thus has a poor prognosis. Cytokeratin Fragment 21-1 (CYFRA 21-1) is one of the oncomarkers discovered in saliva. For developing a diagnostic kit, CYFRA 21-1 antibodies and immunogens are needed. Natural CYFRA 21-1 immunogen is difficult to obtain, so it must be made in-silico using bioinformatics. We aim to predict CYFRA 21-1 immunogenic epitope to produce antibodies polyclonal against CYFRA 21-1, which can be used to develop NPC diagnostics. The immunogenic epitopes were predicted and chosen based on antigenicity, surface accessibility, and hydrophilicity, then characteristic was analyzed and evaluated. The epitope candidates were compared with other saliva protein biomarkers to find if there were cross-reactions. CYFRA 21-1 consists of 57 amino acids, where two immunogenic epitopes (C3 and D2) were chosen. The Ramachandran Plot of both epitopes shows that 100% of the amino acids were in the favoured area. Epitopes C3 and D2 have no cross-reaction with other protein biomarkers. The predicted immunogenic epitopes have the potential as antigen to produce antibodies for developing saliva-based immunodiagnostics to early diagnose NPC patients.

Abstract

Penyuluhan manajemen ternak sapi diberikan kepada kelompok ternak Iye Gati diDesa Sukadana Kecamatan Pujut Lombok Tengah untuk meningkatkan keterampilan dalampemeliharaan, perkandangan, kesehatan, dan reproduksi sapi potong. Keberhasilanpeternakan bergantung pada pembibitan, pakan, dan manajemen pemeliharaan, denganfaktor non-genetik berkontribusi besar terhadap efisiensi reproduksi. Pengabdian inibertujuan untuk mengevaluasi efektivitas penyuluhan manajemen pemeliharaan ternak sapiyang dilakukan di Kelompok Ternak Iye Gati di Desa Sukadana Kecamatan PujutKabupaten Lombok Tengah. Kurangnya pengetahuan peternak dalam pemeliharaanmenjadi tantangan utama. Penyuluhan dan pelatihan berperan penting dalam meningkatkanwawasan peternak serta mendukung ketahanan pangan dan pertanian berkelanjutan (SDGspoin 2). Kegiatan pengabdian masyarakat ini dilakukan melalui survei, wawancara,observasi, pemaparan materi, dan diskusi untuk meningkatkan produktivitas peternakan diDesa Sukadana. Hasil penelitian menunjukkan bahwa penyuluhan yang diberikan berhasilmeningkatkan pengetahuan dan keterampilan peternak dalam aspek manajemenpemeliharaan. Peternak yang mengikuti penyuluhan secara aktif menunjukkan peningkatansignifikan dalam praktik beternak yang lebih efisien dan berkelanjutan. Selain itu, hasilevaluasi juga mengungkapkan bahwa penyuluhan membantu mengatasi beberapa masalahyang dihadapi peternak, seperti kekurangan pakan dan penyakit ternak. Kesimpulannya,penyuluhan manajemen pemeliharaan ternak sapi di Kelompok Ternak Desa Sukadana telahmemberikan dampak positif terhadap peningkatan produktivitas dan kesejahteraanpeternak. Rekomendasi untuk penyuluhan selanjutnya adalah peningkatan frekuensi danvariasi materi penyuluhan, serta pengembangan program pelatihan yang lebih komprehensifuntuk menjawab kebutuhan spesifik peternak di wilayah tersebut.

Abstract

The utilization of local feedstuffs is a cost-effective strategy to reduce production expenses in animal husbandry. Enhancing the nutritional quality of these feedstuffs through fermentation presents a viable approach. This study investigates the efficacy of cellulolytic bacteria in degrading fiber in three local feed materials: duckweed (Lemna minor), water hyacinth (Eichornia crassipes), and lamtoro (Leucaena leucocephala). We employed a completely randomized block design with feed types as the block and levels of starter culture as treatment. Pediococcus pentosaceus, identified for its cellulase enzyme production with a clear zone of 4 cm on starch media, was utilized as the starter culture. The experimental procedure involved spraying Pediococcus pentosaceus onto duckweed, water hyacinth, and lamtoro, followed by incubation. The results indicated that Pediococcus pentosaceus effectively produced cellulolytic enzymes and significantly reduced crude fiber in duckweed by 18.18%, in water hyacinth by 9.17%, and in lamtoro by 4.23% (p < 0.05). Additionally, the crude protein content increased in all samples: by 4.69% in duckweed, 2.71% in water hyacinth, and 2.13% in lamtoro (p < 0.05). Among the treatments, the optimal outcome was achieved with 3 ml of Pediococcus pentosaceus. This study demonstrates the potential of Pediococcus pentosaceus for improving the nutritional profile of local feedstuffs through fermentation.

Abstract

Foot and Mouth Disease (FMD) is a viral disease that affects livestock and can cause significant economic losses.  Vaccination has been recognized as the primary strategy for FMD prevention, but vaccine development, especially conventional vaccine production, has time, cost, and effectiveness limitations. Using software-based immunoinformatics methods has cost and time efficiency for simulation development and calculations in vaccine development research.  This study aimed to design FMD vaccine candidates using an epitope-based in-silico approach, focusing on identifying potential epitopes of the pathogen that causes FMD. The in silico approach was used to analyze FMD virus genome sequences from UniProtKB (https://www.ebi.ac.uk/Tools/msa/clustalo) and the National Library of Medicine application (https://www.ncbi.nlm.nih.gov).  Furthermore, the prediction of FMD virus epitopes was analyzed using the Immuno-Epitope database, and the prediction of peptide antigenicity levels using VaxiJen 2.0 software. The analysis identified eight potential epitope candidates, such as CSIQKR, TEFGFHPNA, EIRPMEKVRA, YEGVEL, SFARRGT, APGLPWALQGKRRGALIDFESGTV, MASLEDKGKPF, and TLPTSFNYGAI with antigen binding affinities of 2. 9424nM, 1.65nM, 1.4702nM, 1.2966nM, 1.2643nM, 1.1967nM, 1.1921nM and 1.0143nm respectively. These epitopes are expected to form the basis for developing more selective and safe peptide vaccines. By focusing on epitopes, the resulting vaccine can improve the effectiveness and safety of vaccination and accelerate the development of new vaccines for FMD prevention.
