Abstract

Sorgum merupakan salah satu jenis tanaman pangan yang berpotensi sebagai pangan fungsional dan memenuhi diversifikasi pangan nasional. Pengembangan sorgum dalam meningkatkan hasil dan kualitas tekstur nasi yang dihasilkan masih perlu dilakukan untuk meningkatkan palatabilitas. Penelitian ini bertujuan untuk memperoleh informasi mengenai keragaman genetik karakter agronomi dan kadar amilosa pada populasi F3 hasil persilangan Pulut 3 × Kawali. Penelitian menggunakan metode Single Seed Descent (SSD). Hasil penelitian menunjukkan keragaman karakter kualitatif maupun kuantitatif berdasarkan analisis Nilai Tengah, Khi-Kuadrat, Skewness and Kurtosis, Korelasi, dan Heritabilitas arti luas. Kandungan amilosa yang bertipe waxy mengikuti nisbah Mendel berdasarkan pengujian pewarnaan iodin pada endosperma biji. Perbedaan karakter yang ditemukan pada F3 belum berbeda nyata dibandingkan tanaman tetua. Aksi gen yang ditemukan yaitu dominan, epistasis, dan aditif dengan jumlah gen yang banyak atau sedikit. Beberapa karakter yang ditemukan memiliki keragaman yang tinggi dan keeratan hubungan yang rendah. Pengelompokan genotipe tanaman berdasarkan karakternya dapat digunakan dalam melakukan seleksi pada generasi lanjutan. Kata kunci: keragaman, klaster, pewarnaan iodin, Single Seed Descent

Abstract

This research is a preliminary yield test of the F7 lines crossing from Numbu x Samurai 2. The aims are to obtain selection criteria, a relationship between vegetative characters and yield and biomass characters, and to obtain vegetative characters that have a direct influence on character Yield and biomass and have high production potential. This study used 20 F7 lines and 4 check lines grown using a Randomized Complete Block Design. The data were analyzed for variance and to estimate variance component analysis, broad sense heritability, genetic diversity coefficient, correlation analysis, cross-trace analysis, and selection index. The results showed that to obtain lines with high biomass, attention should be put on the characters of plant height, panicle diameter, and weight while selecting lines with high yields needs to pay attention tocharacters of the number of leaves at harvest and panicle weight. These characters show wide genetic variation, high broad sense heritability values, and have a high correlation and direct influence on biomass and yield. Therefore, they were potent selection criteria for sorghum plant breeding programs. The F7 lines selected for high biomass and yield based on the selection index were NS021, NS126, NS107, NS123, NS112, NS020, NS015, NS119, NS121 and NS011. Penelitian ini merupakan bagian dari uji daya hasil pendahuluan galur-galur F7 hasil persilangan Numbu x Samurai 2. Tujuannya adalah memperoleh kriteria seleksi hasil dan biomassa, memperoleh hubungan antar karakter vegetatif dengan karakter hasil dan biomassa, memperoleh karakter vegetatif yang berpengaruh langsung terhadap karakter hasil dan biomassa dan memperoleh galur sorgum yang memiliki potensi produksi tinggi. Penelitian ini menggunakan 20 galur F7 dan 4 galur pembanding yang ditanam menggunakan Rancangan Kelompok Lengkap Teracak. Data dianalisis ragam, analisis komponen ragam, heritabilitas arti luas, koefisien keragaman genetik, analisis korelasi, analisis sidik lintas dan Indeks seleksi. Hasil analisis menunjukkan bahwa untuk mendapatkan galur dengan biomassa tinggi perlu memperhatikan karakter tinggi tanaman, diameter dan bobot malai. Sedangkan seleksi galur dengan hasil tinggi perlu memperhatikan karakter bobot malai. Karakter-karakter tersebut menunjukkan ragam genetik yang luas, nilai heritabilitas dalam arti luas yang tinggi, dan mempunyai korelasi dan pengaruh langsung besar terhadap biomassa dan hasil, sehingga merupakan karakter seleksi yang sangat baik untuk program pemuliaan tanaman sorgum. Galur F7 yang terseleksi dengan biomassa dan hasil tinggi berdasarkan indeks seleksi adalah NS021, NS126, NS107, NS123, NS112, NS020, NS015, NS119, NS121 dan NS011.

Abstract

Taoge atau kecambah merupakan salah satu jenis sayuran yang banyak dikonsumsi masyarakat Indonesia. Taoge salah satunya dihasilkan dari perkecambahan biji kacang hijau (Vigna radiata L.). Taoge mengandung beberapa zat yang berperan sebagai antioksidan di antaranya fitosterol, fenol, flavonoid, vitamin E, niasin, vitamin C, dan mineral. Taoge memiliki potensi produksi yang cukup menjanjikan mengingat banyaknya manfaat yang terkandung dalam sayuran tersebut. Penelitian ini bertujuan mendapatkan informasi karakter biji dan taoge, pendugaan potensi taoge serta mendapatkan galur potensial untuk dikembangkan menjadi taoge pada galur-galur kacang hijau IPB. Penelitian dilaksanakan pada bulan Februari sampai Maret 2023 di Laboratorium Pemuliaan Tanaman, Departemen Agronomi dan Hortikultura. Materi genetik kacang hijau terdiri atas 17 galur kacang hijau IPB dan dua varietas nasional (Vima 1 dan Vimil 1) sebagai pembanding. Pengujian kecambah dilaksanakan berdasarkan rancangan kelompok lengkap teracak (RKLT) yaitu 19 genotipe dengan tiga ulangan. Penelitian menggunakan 30 butir kacang hijau per satuan percobaan. Pengamatan dilakukan terhadap karakter kuantitatif dan kualitatif taoge dan biji galur kacang hijau IPB. Hasil penelitian menunjukkan pengaruh nyata genotipe terhadap karakter panjang taoge, bobot satuan taoge, bobot total taoge dan bobot 100 butir. Galur kacang hijau IPB yang memiliki potensi unggul produksi taoge adalah F7-VR10/V1-49, F7-Lom2/129-37, F7-VR10/V1-27, F7-Lom2/129-34, F7-VR10/V1-13, F7-Lom2/129-49, dan F7-VR10/V1-29. Abstract

  Sorghum is one of high value commodities for food and energy security due to its wide adaptation. The objectives of this research were to study some genetic parameters of sorghum agronomic traits grown under acid soil conditions and to select aluminum-tolerant sorghum mutant lines.  The selection was conducted in an augmented design with 61 lines at the field research station of B2TP-BPPT Lampung. Genetic materials used were mutant lines developed throught gamma irradiation.  The results showed that there was significant differences among the mutant lines for  some agronomic characters observed.  The heritability estimates for agronomic characters were classified as intermediate to high.  Selection based on seed weight and biomass production resulted three best sorghum mutan lines i.e ZH30-29-07, ZH30-30-07, and ZH30-35-07.  These lines showed the best yield and biomass production under acid soil condition.   Key words:  sorghum, acid soil, heritability, selection

Abstract

This research was aimed to develop selection criteria based on correlation and path coefficient analysis.  The experiment was carried out at CETROFS (Center for Tropical Fruit Studies) field station of Bogor Agricultural University, Tajur.  The result showed that plant height, pseudostem diameter, number of leaves, weeks to harvest,  finger number per plant, and mean of  finger weight were positively correlated with bunch weight. Pseudostem diameter,  number of leaves, finger number per plant have strong direct effect on bunch weight,  with path coefficient 0.321, 0.264, 0.297, and 0.722, subsequently.  However, the plant height showed strong indirect effect on bunch weight  through  pseudostem diameter. These characters  are useful as a selection criteria for developing new variety of banana  with higher bunch weight   Key words: path coefficient, selection criteria, banana

Abstract

High-yielding variety is one of the targets in soybean breeding program. High-yielding variety could be developed through simultaneous selection. Simultaneous selection  using selection index was more efficient  than selection based on an individual trait only.  Objective of  the research was to select agronomic characters for construction selection index  on 11 soybean populations F6 generation based on correlation analysis, path analysis, and heritability.  The correlation analysis  showed that  number of branch,  number of total nod, number of filled pod, number of unfilled pod, number of total pod, and percentage of filled pod have positively and significantly correlated with  yield per plant.  Based on  path analysis, characters number of branch, number of unfilled pod, and number of filled pod have positive direct on yield per plant.   The result also showed that  number  of filled pod has maximum positive direct effect on yield per plant.  Generally, number of total nod, number of filled pod, number of total pod, and percentage of filled pod have higher heritability  than other characters.   Key words: Soybean, correlation, path analysis, heritability, selection index

Abstract

 A study was conducted to reevaluate 15 upland lines previously selected in a field experiment and nutrient culture experiment or tolerance to aluminum toxicity and potassium efficiency in the plastic house of Center for Crop Improvement Studies, Tajur, Bogor. The experiment was conducted in pot culture using Red Yellow Podzolic soil from Jasinga, Bogor. Two level of AI stress (no AI stress and AI stress with pH 4.8, Al saturation 80%, and exchangeable Al 16.6 mei/IOOg) and two levels of potassium fertilizer (6 kg K2Olha and 60 K2Olha) were applied. The study showed that the upland rice lines performed poorer under AI-tress condition compared to under non stress condition in plant heigl at harvest, total number of  tiller, number of productive tiller, length of spikelet and grain weight/pot. The performance of the upland rice lines was also significantly different under different level of potassium fertilizer in total number off tiller and nun1ber of productive tiller. Lines that showed significant difference in its performance under AI-stress and non stress condition were consiidered as tolerant lines. The result showed that Lalantik Bamban, Sigundil,CT65 I 0-24-3- I, Hawara Bunar, Ketombol, and Grogol were classified as AI-tolerant and K-efficient lines. Key word: Upland rice, Aluminium, Potasium

Abstract

Anther culture provides rapid route in obtaining pure lines in a single generation through producing green haploid plants that may be spontaneously doubled. This technique has been used for crop improvement especially in rice. The objective of this research was to determine regeneration ability of eight F1s derived from crossess between upland and new plant types of rice and from their four parents through anther culture. Completely randomized design with 25 replications was used in this research. Treatments consisted of four parent lines/varieties i.e. P1 (Fatmawati and BP360E-MR-79-2), P2 (Fulan Telo Gawa and Fulan Telo Mihat) and eight F1s obtained from reciprocal crosses of P1 and P2. Callus induction medium was based on N6 medium + 2.0 mg L-1 NAA + 0.5 mg L-1  kinetin + 10-3  M Putrescine, while regeneration medium was based on MS + 0.5 mg L-1  NAA + 2.0 mg L-1  kinetin + 10-3  M Putrescine. The result indicated that F1 derived from Fatmawati x Fulan Telo Gawa (5.00% green plants per total anther) and their reciprocal (3.80% green plants per total anther) crosses were the most responsive genotypes in rice anther culture (had high anther culture ability). The F1 genotypes were more effective to produce green and doubled haploid plants in rice anther culture than their parents. From this research, 161 double haploid plants (29.81%) from total acclimated green plantlets were obtained.  Keywords: anther culture, upland rice, new plant type of rice

The study was conducted in the greenhouse of the University Farm, Bogor Agricultural University from August to October 2009. The experiment was carried out as a Factorial experiment in a Completely Randomized Design with three replications. The fi rst factor was sorghum genotypes consisted of Numbu (tolerant) and B-75 (sensitive), and the second factor was combination of lime and phosphorous fertilization consisted of: no lime-no P (R1), no lime-low P (R2), no lime-suffi cient P (R3), lime-no P (R4), lime-low P (R5) and lime-suffi cient P (R6). The results showed that all variables were signifi cantly affected by the interaction of media and genotype, except for length of shoot and total P uptake. Under low P and Al toxicity, sorghum variety Numbu showed a higher nutrient effi ciency than B-75 with ability of forming greater dry mass. The sensitive genotypes showed a higher nutrient uptake but a lower P nutrient use efficiency under Al toxicity and P defi ciency. Total P uptake and P nutrient use effi ciency were highly correlated with root growth and dry material accumulation. Abstract

ABSTRAKPerbaikan hasil kacang hijau dapat dilakukan melalui seleksi langsung maupun seleksi tak langsung berdasarkankriteria seleksi dari satu atau beberapa sifat kuantitatif sebagai indikator seleksi. Penelitian ini bertujuan untuk memilihindikator seleksi bagi hasil biji dan menentukan kriteria seleksi bagi indikator seleksi tersebut pada populasi generasi awalkacang hijau berumur genjah. Penelitian menggunakan rancangan acak tak lengkap berblok bersekat-tersarang 1-tahap.Analisis lintas dan regresi aditif serta genotipik masing-masing digunakan untuk memilih dan menentukan indikator seleksi dan kriteria seleksi. Hasil analisis pengaruh langsung memperlihatkan jumlah biji bernas dan umur berbunga merupakanindikator seleksi bagi hasil biji dengan kriteria seleksi 200 biji pada kisaran umur berbunga 40-46 hari, untuk memperolehhasil > 12 g tanaman. Sebaliknya, hasil analisis pengaruh tak langsung memperlihatkan tinggi tanaman, jumlah cabangdan lama hari panen dapat pula dimasukkan sebagai indikator seleksi. Kriteria seleksi bagi masing-masing indikator seleksiini adalah dua cabang dengan lama hari panen 16-20 hari dan tinggi tanaman sekitar 85 cm untuk menghasilkan 200 biji.Bila dipilih tanaman yang lebih pendek, kriteria seleksi tinggi tanaman 75-80 cm dapat digunakan, namun lama hari panenmenjadi 22-26 hari.-1Kata kunci: analisis regresi, analisis lintas, BLUP, seleksi generasi awal
