Abstract

Penelitian ini dilakukan untuk mengetahui efek antihiperglikemia infusa kangkung darat (Ipomoea reptans P.j pada mencit Swiss jantan yang diinduksi streptozotocin (STZ) dengan parameter pengukuran kadar glukosa darah puasa (KGDP) dan berat badan. Sebanyak 30 ekor mencit Swiss janlan berat 20 - 30 g dibagi menjadi 6 kelompok (N=5). Kelompok I (kontrol normal) yang tidak diberi perlakuan. Kelompok II (kontrol negatif) diberi STZ dosis 40mg/kgbb secara intra peritoneal selama 5 hari berturut-turut. Keiompok III (kontrol positif) diberi metformin 3,9 mg/20gBB. Kelompok IV;V:VI diberi infusa kangkung darat dosis 44,64mg/20gBB; 89,28mg/20gBB; 178,56mg/20gBB. Kangkung darat diberikan secara p.o selama 7 hari berturut-turut yaitu pada hari ke-13 sampai hari ke-19 seEelah pemberian STZ. Pengukuran KGDP pada hari ke-0 (sebelum pemberian STZ), hari ke-13, hari ke-20 dengan cara diarnbi darah secukupnya melalui ekor mencit Swiss jantan yang sudah dipuasakan sebelumnya dan diukur menggunakan glukotest . Data dianalisis dengan analysis of covarian (ANCOVA) dan analysis of varian (ANOVA) (p=0:05). Hasil penelitian menunjukkan bahwa berat badan mencit diabetes rata-rata mengalami kenaikan dibandingkan kelompok kontrol normal sebesar 0,2g/hari dan mencit Swiss jantan mengalami penurunan KGDP setelah diberikan infusa kangkung darat (Ipomea reptans P.) dosis 44,64mg/20gBB; 89,28rng/20gBB; 178,56mg/20gBB sebesar 21,27%; 34,44%: 31,18%.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh pra-perlakuan prednison terhadap profil bioavailabilitas teofilin. Hewan coba yang dipergunakan dalam penelitian ini adalah tikus Wistar jantan yang dibagi dalam 2 kelompok yaitu kelompok kontrol dan perlakuan dimana setiap kelompok terdiri atas 5 ekor tikus. Kelompok kontrol diberikan teofilin dosis 45 mg/kg BB tikus sedangkan kelompok perlakuan diberikan prednison dosis 3,6mg/kg BB tikus sekali sehari selama 7 hari dan pada hari ke-8 diberikan prednisolon. Cuplikan darah diambil sebanyak 0,25 ml dari vena lateral ekor. Penetapan kadar fenitoin dilakukan dengan metode HPLC. Berdasarkan data kadar obat dalam darah lawan waktu dilakukan perhitungan parameter bioavalabilitas teofilin yang meliputi Cpmax, tmax and AUC0-∞,. Hasil penelitian menunjukkan pemberian pra perlakuan prednison menurunkan secara signifikan nilai tmax dari teofilin sampai dengan 51,19% (p<0,05).

Abstract

Honey and curcumin is widely consumed by children as food supplements. Honey is useful as an antioxidant and antibacterial, while curcumin  is useful as an antioxidant, antiinflammatory,  antibacterial,  antiviral. Both  health  supplements  are  not  infrequently used in conjunction with drugs, especially paracetamol. This study aims to determine the effect of curcumin and honey on the pharmacokinetic parameters of paracetamol in male rats.  In  this  study,  test  animals  were  divided  into  3  groups:  control,  treatment  I  and treatment  II.  In  the  control  group  mice  were  given  paracetamol  150mg  /  KgBW, treatment  group  I  rats  were  given  curcumin  18  mg  /  KgBW,  1  hour  and  then  given paracetamol 150mg  / KgBW whereas  the  treatment group  II  rats given a dose of honey 7.65 mL / KgBW along with giving paracetamol 150mg KgBB . The results showed that administration of curcumin on the pharmacokinetics of paracetamol in mice can decrease the  primary  parameters  of  the  elimination  phase  of  CLT  for  68.25%,  increasing  the secondary parameter t 1 / 2 at 193.72%, thus increasing the derivative parameters AUC0-inf for 88.36%, and MRT of 155.17% (p

Abstract

Angiogenesis is a neovascularization process which its function is to supply nutrient and oxygen for cell survival. Pasak bumi is one of the Indonesian original plants having anti cancer activity. Extract of methanol, buthanol, kloroform, and water akar pasak bumi was proved to have cytotoxic effect on cancer cell culture. This research aim to know activity of antiangiogenic extract of metanol akar pasak bumi at chorioallantoic membrane of chicken embryo. Antiangiogenic activity test conducted by divide 8-9 days old egg embrio into 8 groups. Group I as paper disc control, group II as bFGF control, group III as bFGF control + DMSO 0.8% solvent, group IV, V, VI, VII, and VIII as  groups of treatment. Each groups of treatment were gave by bFGF 1ng/µL extract of metanol akar pasak bumi respectively with concentration 20, 40, 60, 80 and 100 µg/mL concentration. After 3 days incubation, then the chorioallantoic membrane macroscopic and microcospically analized. The antiangiogenic activity extract of metanol akar pasak bumi at chorioallantoic membrane bFGF induce at used concentration, that is start from 40 µg/mL concentration. The increases of the concentration also increase the prohibition activity to angiogenesis.

Abstract

Pasak bumi is one of the Indonesian original plants having anti cancer activity. One compound of akar pasak bumi, i.e eurycomanone was proven to have cytotoxic effect on cancer cell culture by apoptosis induction on HeLa cells. Therefore, this study was aimed to isolate and identify euycomanone from akar pasak bumi and to look at the anti angiogenic activity of the compounds on corio alantois membrane (CAM) of chicken embryo induced by bFGF.The akar pasak bumi was ground and was macerated with methanol. The macerate was fractioned by mean of VLC. Six fractions was obtained. The second fraction exhibited two spots on a TLC system, one spot was cochromatographed with eurycomanone as the reference standart and another spot exhibited blue fluorecence at 254 nm. This second fraction, then, was chromatographed on a preparative scale TLC to separate isolate A. Anti angiogenic test was performed for Isolate A at concentration series of 5, 10, 15, 20, 40 μg/mL respectively. Egg embrio (8-9 days) were divided into 8 groups. The first group was implanted paper disc only, the second group were given bFGF on Tris HCl buffer and the third group were given bFGF (on Tris HCl buffer) + 0,8 % DMSO. The fourth to eighth group were induced by 10 ng bFGF and the test preparation at the respective concentration. After incubation at 39 °C for 3 days, each egg was opened, the content of each was aspired and discarded, the CAM sticked to its shell was examined macroscopically and microscopically for the formation of new blood vessels.Based on spectral data of UV, IR, 1H NMR, 13C NMR and MS of isolate A and comparison with the spectral data of the standart, the isolate A was eurycomanone. Eurycomanone inhibited angiogenesis at concentration of 10 μg/mL.Key words: antiangiogenic, angiogenesis, E.longifolia, eurycomanone,CAM.

Abstract

ABSTRAK Kangkung darat (Ipomea reptans Poir) adalah salah satu tanaman di Indonesia yang secara empiris telah dimanfaatkan oleh masyarakat sebagai obat untuk terapi Diabetes Melitus (DM).  Riset sebelumnya telah dilakukan untuk memperoleh data ilmiah dari pemanfaatan kangkung darat sebagai terapi DM, dan pengembangannya dalam bentuk sediaan Self Nano Emulsifying Drug Delivery System (SNEDDS). Penelitian ini bertujuan untuk mengetahui sitotoksisitas sediaan SNEDDS ekstrak daun kangkung terstandar dengan menggunakan metode MTT (Methyl-Thiazolyl-Tetrazolium). Pengujian MTT dilakukan dengan cara well plate diplotkan terlebih dahulu untuk sampel uji 70 sumuran (well), kontrol sel 6 sumuran, dan kontrol media 6 sumuran.  Terakhir tiap sumuran ditambahkan stopper SDS sebanyak 100µl. Plate dibungkus alumunium foil dan diinkubasi semalaman pada suhu ruang. Hari berikutnya dilakukan pembacaan absorbansi sampel, kontrol media, dan kontrol sel menggunakan ELISA reader. Uji sitotoksik SNEEDS kangkung pada sel vero diperoleh hasil bahwa SNEEDS kangkung tidak menyebabkan kematian pada sel uji. Pada pengujian sel dilakukan pemaparan sampel excipient dari SNEEDS kangkung sebagai pembanding, dengan hasil menyebabkan kematian <50% sel uji pada dua kadar tertinggi dari sampel excipient. Namun pada hasil uji SNEEDS kangkung diperoleh data bahwa toksisitas excipient pada kadar tersebut tidak mempengaruhi timbulnya toksisitas pada sampel SNEEDS kangkung. Sehingga berdasarkan uji in vitro, SNEEDS kangkung tidak toksik terhadap sel vero. Kata kunci: Ipomoea reptans, sitotoksisitas, SNEDDS  ABSTRACT Kangkong (Ipomea reptans Poir) is a type of Indonesian plant empirically utilized by many people to treat diabetes mellitus (DM). A study has been conducted to obtain scientific data from the use of kangkong for DM treatment as well as to develop kangkong in SNEDDS preparation. This current study aimed to examine the cytotoxicity of kangkong leaf standardized extract in SNEDDS preparation through the MTT (Methyl-Thiazolyl-Tetrazolium) method. MTT assay was performed by initially plotting well plates for test sample (70 wells), control cell (6 wells), and control medium (6 wells). As much as 100µl SDS stopper was added into each well, and then plates were wrapped in aluminum foil and incubated all night at ambient temperature. On the following day, the absorbance of test sample, control medium, and control cell was identified using ELISA reader. The cytotoxicity test of kangkong SNEDDS on Vero cell lines showed that kangkong SNEDDS did not cause cell death. The cell was tested through exposure of excipient sample from kangkong SNEDDS as a comparison, resulting in <50% cell death by the two highest concentrations of excipient sample. However, the test result of kangkong SNEDDS indicated that excipient toxicity at such concentrations did not affect kangkong SNEDDS. Abstract

Telah dilakukan penelitian untuk mengetahui nilai parameter spesifik dan nonspesifik standardisasi ekstrak kangkung darat hasil budidaya di wilayah Sardonoharjo, Sleman serta potensinya sebagai antioksidan. Sampel yang digunakan dipanen pada saat usia kangkung darat ±25-30 hari setelah penanaman. Ekstraksi dilakukan dengan metode maserasi menggunakan pelarut etanol 96%. Parameter spesifik terdiri dari uji organoleptik, pola kromatografi, kadar senyawa marker dan parameter nonspesifik terdiri dari uji bobot jenis,uji kadar air, uji kadar abu total, uji kadar abu tidak larut asam, uji cemaran logam, uji cemaran mikroba, uji cemaran kapang dan khamir, perkiraan angka koliform, dan uji sisa pelarut etanol. Hasil standardisasi ekstrak menunjukkan nilai pengukuran berturut-turut untuk parameter kadar air 16,45±0,05%, bobot jenis ekstrak 3,26±3,37x10-3 g/mL, kadar abu total 4,52± 0,77%. Tidak terdapat sisa pelarut etanol di dalam ekstrak, serta angka cemaran mikroba, angka kapang dan khamir, angka koliform, serta cemaran logam timbal (Pb) dan kadmium (Cd), di dalam ekstrak di bawah standar batas maksimal yang ditetapkan BPOM. Hasil pengukuran dengan KLT densitometri menunjukkan nilai kadar β-karoten di dalam ekstrak sebesar 5,7% (b/b). Hasil Uji aktivitas antioksidan dengan menggunakan 2,2-diphenyl-1- picrylhydrazyl (DPPH) menunjukkan nilai IC50 ekstrak daun kangkung sebesar 178,3 μg/ mL.

Abstract

Bupivacaine is used as a local anesthetic with Epidural Lumbal Anasthesia (ELA) for Caesarean Section (CS) in Indonesia. During pregnancy, there are changes in pharmacokinetic parameter values, as a result of changes in the physiology of the body. This study was designed to determine the pharmacokinetic parameters of isobaric bupivacaine 0.5% with non-compartment model and to monitor its effectiveness. The study was conducted on 7 normotensive pregnant patients who treated with 0.5% bupivacaine doses 75 mg at Integrated Central Surgery Building in Dr. Sardjito Hospital. Blood samples (3 mL) from each patients were drawn at the following intervals: 0, 15, 20, 25, 30, 45, 60, 75 and 90 minutes, then the plasma bupivacaine consentration were measured using HPLC and pharmacokinetic parameters were calculated with non-compartment models, i.e. AUC(0-~), AUMC(0-~) and the MRT. Monitoring the effectiveness was conducted by Pinprick test. Relationship of AUC(0-~) with maximum sensory block was analyzed by Pearson correlation. The results of pharmacokinetic parameters of isobaric bupivacaine 0.5% for 7 normotensive pregnant patients with non-compartment model were AUC(0-~) 101,25 ± 15,07 μg.mL-1.min; AUMC(0-~) 6.431,92 ± 2.212,96 μg.mL-1.min2 and MRT 63,74 ± 20,52 minutes. Bupivacaine 0,5% doses 75 mg as an anesthetic for CS with ELA in 7 normotensive pregnant patients was effective for pain blockade, with maximum sensory block was T6. Abstract

Preeclamsia patient experienced deterioration of glomerolus filtration and plasma protein concentration. Bupivakain is bound strongly with the plasma protein that is 95%. This research was aimed to know the pharmacokinetics profiles of isobaric bupivacaine with the noncompartemen model in preeclampsia patients who underwent Sectio Caesarea (SC) with lumbar epidural technique in RSUP Dr. Sardjito and to know correlation between protein levels and pharmacokinetics profiles of isobaric bupivacaine in preeclampsia patients. Firstly, albumin concentration of patients was checked in the laboratory and then isobaric bupivacaine 0.5% in 15ml (75mg) was given by lumbar epidural technique. The samples of blood were taken through veins vessels at 0; 15; 20; 25; 30; 45; 60 and 90 minutes. The concentration of 0.5% isobaric bupivacaine in those samples of blood was determined with High Performance Liquid Chromatography (HPLC) methods. Furthermore, pinprick test was taken to measure block sensoric effect in T10-T6 areas. Pharmacokinetics parameters of bupivacaine (AUC, AUMC, MRT) were calculated based on data of bupivakain concentration in the blood versus time. The results of this research showed that the AUC0-∞ (total AUC) was 116±37,6 (μg/ml) minutes; AUMC0-∞ (total AUMC) was 9187,5 (μg/ml) minutes2; and MRT0-∞ was 102,5 minutes. Average block sensoric effect was at T8. There was no linear correlation between the protein levels and pharmacokinetics profiles of bupivacaine (p>0,05). The conclusion was that there was no linear correlation between the protein levels and pharmacokinetics profiles of bupivacaine that was given by lumbar epidural in preeclampsia patientswho underwent section caesarea.Keywords : bupivakaine, pharmacokinetic profile, lumbar epidural, section caesarea.

Abstract

Telah dilakukan penelitian mengenai peningkatan derajat kesehatan melalui promosi kesehatan Pola Hidup Bersih Sehat (PHBS) di Dusun Sawahan Desa Pendowoharjo, Kecamatan Sewon, Kabupaten Bantul dengan metode penyuluhan dan pemantauan serta pengamatan tanpa sepengetahuan subjek uji. Penelitian ini bertujuan untuk mengetahui pengaruh dari pelaksanaan promosi kesehatan Pola Hidup Bersih Sehat (PHBS) terhadap kesehatan masyarakat Dukuh Sawahan. Hasil penelitian menunjukkan bahwa frekuensi penyuluhan dan pemantauan Pola Hidup Bersih dan Sehat (PHBS) terhadap warga dusun Sawahan menunjukkan korelasi yakni meningkatnya indikator Pola Hidup Bersih dan Sehat (PHBS) yang tercapai. Tercapainya indikator Pola Hidup Bersih dan Sehat (PHBS) menunjukkan peningkatan kesadaran warga akan pentingnya lingkungan sehat dan perilaku sehat sehingga meningkatkan derajat kesehatan warga Dusun Sawahan.
