Abstract

Profil sidik jari kromatografi lapis tipis (KLT) dapat dimanfaatkan untuk kendali mutu bahan baku maupun produk tumbuhan obat. Penelitian ini bertujuan mengembangkan metode analisis sidik jari rimpang temu mangga (Curcuma mangga) untuk kendali mutunya. Rimpang kering temu mangga diekstraksi menggunakan metanol dan dianalisis sidik jari KLT. Kloroform:etil asetat (8.5:1.5) merupakan fase gerak optimum yang memisahkan 11 pita dari rimpang temu mangga dengan keterpisahan yang baik dan memiliki pita khas biru tua (Rf 0.36). Demetoksikurkumin terdeteksi pada rimpang temu mangga dengan warna jingga kecoklatan pada UV 366 nm (Rf 0.48) setelah diderivatisasi dengan pereaksi asam sulfat 10%. Pengujian spesifitas terhadap rimpang lain dalam satu genus, rimpang temu mangga menunjukan pola yang berbeda. Validasi metode analisis sidik jari KLT memenuhi kriteria keberterimaan, sehingga metode ini dapat digunakan untuk kendali mutu rimpang temu mangga.

Abstract

This research aimed to determine the active fraction and profile of active compounds in Manonjaya sSnakefruit skin extract that can be used as antidiabetic and the antidiabetic activity using zebra fish. Manonjaya sSnakefruit skin was extracted by maceration using 70% ethanol and then fractionated using n-hexane, ethyl acetate and methanol. The ethyl acetate fraction showed the highest yield and total phenolic content as well as the strongest α-glucosidase inhibition with an inhibition percentage of 64.43%. The ethyl acetate fraction provided the most active spot as an α-glucosidase inhibitor. The profile of active compounds that act as antidiabetic compound is flavonoid from glycosylflavone and flavone groups. Antidiabetic activity using zebra fish was indicates that Manonjaya Snakefruit skin extract from ethanol extract and ethyl acetate fraction can decrease blood sugar level of zebra fish after induced with alloxan. The crude extract group had 54.51% lower sugar content than the induction group and 41.70% lower than the negative control. Meanwhile, the ethyl acetate fraction group had 65.10% lower sugar content than the induction group and 55.27% lower than the negative control.

Abstract

Fungus comb is a structure made by termite colony Macrotermitinae with lots of potential because it contains secondary metabolites such as steroids, terpenoids, and saponins and compounds containing antioxidant and anti-tyrosinase properties. This study aimed to determine fungus comb extracts' fingerprint pattern, antioxidant capacity, and anti-tyrosinase activity. Thin layer chromatography (TLC) was used to determine fingerprint patterns. The resulting TLC pattern was processed with ImageJ and converted into a densitogram. The fungus comb was extracted with n-hexane and ethyl acetate. TLC results showed that fungus comb extracts contain steroids and terpenoids and have antioxidant properties. The antioxidant capacity assay was carried out with 2,2-diphenyl-1-picrylhydrazyl reagent. N-hexane extract had an antioxidant capacity value of 9.07 mg ascorbic acid equivalent (AAE)/g extract, while ethyl acetate extract had 6.04 mg AAE/g extract. Anti-tyrosinase activity assay was carried out with L-tyrosine and L-3,4-dihydroxyphenylalanine (L-DOPA) as substrates. N-hexane extract had a better anti-tyrosinase activity with IC50 of 1804 µg/mL for L-tyrosine and 6784 µg/mL for L-DOPA.

Abstract

A simple, portable visible reflectance spectrophotometer was developed for classifying the quality of Curcuma xanthorrhiza Roxb. The device was assembled using consumer electronic components: an LED as the light source, a DVD optical layer as the diffraction grating, and a webcam as the detector, with web-based software (Spectral Workbench) as the signal processor. The spectrophotometer's performance was evaluated using standard Sudan III samples and powdered C. xanthorrhiza samples from two quality classes (A and B). Spectral data were analysed using principal component analysis (PCA) and partial least squares (PLS) regression.  PCA successfully grouped Sudan III samples by concentration and classified C. xanthorrhiza by curcuminoid content. A PLS regression model was developed for quantifying Sudan III, yielding R² values of 0.9909 for both calibration and cross-validation, with RMSEs of 0.3821% and 0.4152%, respectively. The results closely matched those from a commercial spectrophotometer. A PLS-discriminant analysis (PLS-DA) classification model for C. xanthorrhiza was also developed, achieving sensitivity and specificity values of 1. Additionally, semi-quantitative parameters such as decision limit (26.15% B), detection capability (41.06% B), and unreliability range (26.15–41.06% B) were calculated. The classification model showed strong sensitivity, specificity, and detection capabilities within acceptable limits. These findings suggest that this low-cost reflectance spectrophotometer, combined with chemometric methods, holds promise as a practical tool for the quality control of raw herbal materials.

Abstract

Vegetable oil production of malapari (Pongamia pinnata L. Pierre) fruit remains peels that haven't been utilized. Malaparifruit peel is potential for activated carbon. The purpose of this research is to use malapari fruit peel waste as raw material forproducing activated charcoal. Samples were carbonized at temperature of 400°C and continued activation with phosphoric acid2% and physical activation at temperature of 750°C by flowing steam for 60 minutes. The process resulted in the Optimumcondition. It yields 54% charcoal recovery, moisture content of 8.6%, volatile matter of 11.85%, ash content of 24.73%, fixedcarbon of 63.42%, benzene adsorptivity of 10.15%, metilene blue adsorptivity of 93.89 mg/g, iodine adsorptivity of 648.62mg/g, and specific surface area of 348.11 m2/g. Activated carbon in this study is below Indonesian standards, which is influencedby the activation retention time.

Abstract

Minyak atsiri serai (Cymbopogon citratus) dapur telah dipergunakan secara luas di berbagai industri, diantaranya sebagai agen antibakteri. Pada penelitian ini dievaluasi aktivitas antibakteri dan toksisitas minyak serai dapur yang dipekatkan pada suhu berbeda. Toksisitas dievaluasi melalui uji brine shrimp lethality test (BSLT), sementara uji antibakteri dilakukan terhadap bakteri Staphylococcus aureus dan S. epidermidis dengan metode difusi dan dilusi. Komponen kimia yang terdapat dalam minyak serai dianalisis menggunakan kromatografi gas spektrometri massa (GC-MS). Hasil yang diperoleh menunjukkan berdasarkan uji BSLT diperoleh nilai lethal concentration 50 (LC50) sampel minyak serai dapur semua perlakuan < 1000 ppm dengan LC50 terendah sebesar 16,53 ± 2,82 ppm. Nilai konsentrasi hambat minimun (KHM) dan konsentrasi bunuh minimum (KBM) minyak serai dapur terhadap S. aureus umumnya lebih rendah dibanding terhadap S. epidermidis. Aktivitas antibakteri minyak serai dapur terhadap S. aureus terkategori sangat kuat dengan nilai KHM dan KBM terendah ditunjukkan oleh minyak serai tanpa perlakuan pemekatan yaitu sebesar 1250 dan 2500 ppm terhadap S. aureus. Abstract

Penelitian ini bertujuan memodifikasi screen printed carbon electrode (SPCE)  dengan polimer tercetak molekul (MIP) dan mengaplikasikannya untuk deteksi glutation tereduksi (GSH) secara tidak langsung dengan mediator bromin menggunakan teknik voltammetri siklik. SPCE dimodifikasi dengan MIP polipirola (MIPpy) melalui teknik elektrodeposisi. Pengukuran GSH dilakukan dalam elektrolit NaClO4 0.2 M yang mengandung 1 mM NaBr. Hasil yang diperoleh menunjukkan SPCE-MIPpy telah berhasil diperoleh melalui teknik elektrodeposisi. Pada pengukuran diperoleh puncak oksidasi bromida pada potensial ±0.63 V vs. Ag/AgCl. Puncak ini berkorelasi linear dengan konsentrasi GSH dalam larutan sehingga dapat digunakan sebagai dasar penentuan GSH. Polipirola meningkatkan konduktivitas listrik elektrode sehingga dapat meningkatkan puncak arus oksidasi GSH. Pengukuran glutation dengan SPCE-MIPpy menunjukkan limit deteksi dan limit kuantitasi yang lebih baik dibandingkan SPCE, yaitu sebesar 7.6626 µM dan 25.4092 µM. Keberadaan interference glukosa tidak mengganggu pengukuran glutation. Abstract

In this study, zeolite was synthesized from kaolin which was founded from Bangka Belitung as the starting material. Meanwhile, sodium silicate was used as silica source. Zeolite is material with many benefits, such as become an adsorbent. Kaolin has been used as starting material due to high content of silica and alumina, i.e 35 - 50 %. Zeolite was prepared by hydrothermal synthesis process, by mixing the metakaolin which was produced by calcination of kaolin at 700 °C for 6 hours, with sodium silicate and NaOH. The NaOH was used to activate the major components of Si and Al in the kaolin.The hydrothermal process was conducted at 100 °C for 24 hours with various concentrations of NaOH, i.e 1.5 N and 2.5 N. The NaOH concentration might determine the type of zeolite obtained. To improve the nature as adsorbent, zeolites modified into zeolit/TiO2. The addition of TiO2into the prepared zeolit aimed to produce a material with capability as adsorbent-photocatalyst. The result of research found that the addition of NaOH 1.5 N produced zeolite NaP1. Meanwhile, the addition of NaOH 2.5 N produced the NaP1 zeolite, zeolite X, and faujasite. The composite of zeolit/TiO2has ability on adsorption-photocatalytic as it was proven by methylene blue degradation under ultraviolet light.
