Abstract

Free radicals can react to cause damage when they enter the human body. The use of antioxidants can neutralize free radicals in the body. This study aims to determine the total flavonoid levels and antioxidant activity in premium and original green tea with variations in brewing temperature. Green tea produced by Kulon Progo is brewed with aquadest at 75 ℃ and 100 ℃, and then the filtrate obtained is ground by freeze-drying. Identification of flavonoid compounds using the flavanoid-ammonia vapor color complex. The total flavonoid levels analysis using UV-vis spectrophotometry and antioxidant activity was tested by the DPPH (1,1-diphenyl-2-picrylhydrazil) method. Based on the test results, the highest to lowest total flavonoid levels were premium green tea at 100 ℃ original at 100 ℃, original at 75 ℃, and premium at 75 ℃ (155.095 ± 3.158; 153.333 ± 2.788; 151.7181 ± 4.944; and 148.634 ± 2,095 mgQE/gram extract). The highest to lowest potential antioxidant activity, respectively, is quercetin (flavonoid standard), premium green tea at 100 °C, premium at 75 °C, original at 100 °C, and original at 75 °C, with IC50 values of 3.0692 ± 1.064; 32.359 ± 7.346; 31.502 ± 6.746; 27.527 ± 7.868; and 27.167 ± 1.817 μg/mL. Analysis of the total flavonoids of green tea showed that the highest levels were in premium green tea at a temperature of 100 °C. An antioxidant activity test with the DPPH reagent produced the highest antioxidant potential in premium green tea at 100 °C. So premium green tea at a brewing temperature of 100 °C is more recommended to consumers.

Abstract

Cervical cancer is one of the most prevalent cancer types, making the development of effective anticancer agents critical. Cisplatin (CIS) is a drug that has been used to treat cancer, but it also affects normal cells. Hibiscus sabdariffa L. extract (HSE), which has the potential as an anticancer agent, can be developed as a co-chemotherapy with CIS. This study aimed to determine the potential of HSE as a co-chemotherapy with CIS against HeLa cervical cancer cells and determine specific and non-specific parameters of the studied extract. Hibiscus sabdariffa L. (HS) simplicia powder was macerated with 96% ethanol. An in vitro evaluation was carried out on antiproliferative activity by calculating the doubling time. Additionally, a selectivity test was conducted to calculate combination index (CI) values based on the microtetrazolium (MTT) method. Apoptosis mechanisms were explored based on immunocytochemical methods using p53, caspase, and Bax antibodies, followed by an observation of apoptotic induction using a flow cytometer. The HSE and CIS selectivity index values obtained for Vero cells and HeLa cells were 209 and 278, respectively. The antiproliferation test results showed that the combination of HSE and CIS could better extend the doubling time of cells compared to the negative control. This combination also demonstrated a strong synergistic effect, with a CI value of 0.001. The extract as a co-chemotherapy with CIS was capable of increasing the expression of p53, caspase-3, and Bax. The flow cytometry analysis results indicated that HSE could induce apoptosis. Based on the results on the IC50 and CI value of HSE, as well as on the doubling time and apoptosis induction of HSE-influenced HeLa cells, it is concluded that HSE has the potential as a co-chemotherapy against cervical cancer.

Abstract

Indonesia is rich in aromatic plants, producing approximately 40 types of essential oils used in various industries, including health and aromatherapy. Essential oils have significant potential but face high rates of adulteration. This study aimed to identify kaffir lime leaf oil (Citrus hystrix D.C.), to determine the essential oil profile, and to detect adulteration in commercial oil products. The methods used involved steam distillation and characterization tests of kaffir lime leaf essential oil, including yield percentage and organoleptic, density, refractive index, and solubility in alcohol. An essential oil content test and adulteration detection using FTIR combined with chemometrics were also conducted. The FTIR results showed distinctive peaks for kaffir lime leaf oil, allowing the identification of the main compounds in the oil. Multivariate analysis using the PLS model could predict oil adulteration with RMSEC 1.319, RMSECV 1.913, and RMSEP 2.362, indicating the model’s accuracy. Furthermore, PCA successfully separated and grouped the tested oils. This study concludes that the analyzed oil was kaffir lime leaf oil, based on the characterization results and FTIR spectra showing distinctive peaks of the oil. PLS and PCA analysis showed that the three tested commercial oil products did not contain turpentine adulterants.

Abstract

Cosmetic trends in Indonesia are on the rise, particularly for facial serum products. However, most of these products lack halal certification, posing an important concern for Muslim consumers due to the potential use of haram substances, such as pork gelatin. This study seeks to identify the source of gelatin in serum products using the Fourier Transform Infrared (FTIR) spectroscopy and chemometric analysis. The samples used in this study include a reference facial serum formulated with pure bovine and porcine gelatin concentrations in the ratios of 1:0, 8:2, 5:5, 2:8, and 0:1, three hydrolyzed collagen serums that are not yet labeled halal and available on Shopee, and one serum that is labeled halal. The method used was FTIR combined with PLS chemometrics and PCA. The isolation of facial serum gelatin was performed using acetone at -20oC, followed by analysis via FTIR at wave numbers ranging from 4000 to 400 cm-1. FTIR results indicated the presence of functional groups in gelatin constituents, including C O, N H, C N, and C H aliphatic. The wavelengths employed for PLS and PCA analysis ranged from 1631 to 1430 cm-1. Calibration results showed R2=0.9936 and RMSEC=3.0445%. Internal validation yielded RMSECV=0.1674% and R2=0.9994 whilst external validation yielded RMSEP=0.9981% and R2=0.9910. Lastly, PCA analysis revealed that one halal-labeled serum sample contained bovine gelatin, whereas the three examined commercial serums were free from both pork and bovine gelatin.

Abstract

Paprika merah (Capsicum annum L.) banyak mengandung β–karoten  (provitamin A), likopen, vitamin E serta vitamin C telah diuji aktivitasnya sebagai penangkap radikal 2,2–difenil–1–pikrilhidrazil (DPPH). Sampel diekstraksi dengan cara maserasi menggunakan metanol. Ekstrak yang diperoleh dilakukan analisis aktivitas antioksidannya secara kualitatif dengan larutan DPPH 0,4 mM dan reagen ferri tiosianat. Identifikasi terhadap senyawa yang berpotensi sebagai antioksidan dilakukan dengan spektrofotometer UV–Vis. Aktivitas penangkapan radikal DPPH oleh ekstrak metanol paprika merah ditetapkan secara spektrofotometri pada panjang gelombang 517 nm. Dari hasil analisis diperoleh nilai IC50 paprika merahsebesar0,299 ± 4,98x10-3 mg/mL. Aktivitas paprika merah sebagai penangkap radikal DPPH  kurang poten dibandingkan dengan β–karoten (IC50 0,066 ± 3,24x10-3 mg/mL). Kata kunci : antioksidan, paprika merah, Capsicum annuum, DPPH

Abstract

Piroksikam digunakan sebagai analgetik dan antiinflamasi yang sering dialami oleh kalangan lansia yang mempunyai kesulitan menelan. Salah satu upaya yang dapat dilakukan untuk mengatasi masalah tersebut yaitu dengan sediaan Orally Disintegrating Tablet (ODT). Avicel PH 102 merupakan bahan disintegran yang biasa digunakan sebagai penyusun ODT, karena mempunyai daya disintegran yang sangat baik. Tujuan penelitian ini untuk mengembangkan piroksikam menjadi bentuk sediaan ODT. Metode pembuatan ODT dilakukan dengan metode kempa langsung. Variasi konsentrasi Avicel PH 102 yang digunakan untuk formula 1 sebesar 15% dan formula 2 sebesar 20%. Tablet hasil formulasi diperiksa karakteristik fisik yang meliputi identifikasi tablet, kerapuhan, kekerasan, ketebalan, waktu hancur, bentuk dan ukuran tablet, laju disolusinya serta dilakukan penetapan kadar. Metode analisa kadar piroksikam baik pada penentuan disolusi maupun pada penentuan kadar, dilakukan verifikasi metode spektrofotometri UV-Vis terlebih dahulu meliputi spesifisitas, presisi, dan akurasi. Formula terbaik selanjutnya dilakukan uji hedonik. Hasil verifikasi untuk ketiga uji memenuhi persyaratan sehingga dikatakan metode uji valid. Hasil uji fisik tablet dikatakan memenuhi syarat kecuali untuk uji kerapuhan. Data laju disolusi dan penetapan kadar tablet piroksikam memenuhi syarat FI V yaitu laju disolusi tidak kurang dari 75%(Q + 5%) dan hasil penetapan kadar F1 dan F2 berturut-urut sebesar 95,22 ± 3,09% dan 95,14 ± 4,26%. Dari penelitian ini dapat disimpulkan bahwa piroksikam dapat dibuat dalam bentuk sediaan ODT karena menghasilkan sediaan tablet dengan sifat fisik yang baik kecuali pada uji kerapuhan. Formula yang dipilih yaitu formula 2, karena waktu hancur dan disolusi tablet lebih baik dibandingkan dengan formula 1.

Abstract

Bakpao ayam adalah makanan yang dikenal masyarakat Indonesia. Bahan utama umumnya daging ayam yang kemudian dikukus. Daging yang digunakan, kadang belum tentu terjamin keasliannya. Penelitian ini bertujuan mengetahui dan mendeteksi adanya kandungan lemak anjing dalam produk makanan bakpao ayam. Analisis dengan spektrofometri FTIR yang dikombinasi kemometrika dapat memberikan informasi adanya kandungan lemak anjing dalam bakpao ayam. Model analisisnya dengan membuat variasi konsentrasi campuran daging anjing dengan daging ayam. Bakpao produk di pasaran diambil empat sampel yang dijual di kota Yogyakarta dengan teknik stratified random sampling. Hasil spektra FTIR dianalisis dengan kemometrika. Analisis kuantitatif menggunakan Partial Least Square (PLS), dan analisis kualitatif dengan Principles Component Analysis (PCA). Hasil optimasi bilangan gelombang diperoleh pada 1226-840 cm-1. Analisis dengan PLS diperoleh persamaan y=0,9969x+0,1697 ; nilai R2 0,9969; RMSEC 1,38% ; RMSEP 0,27% ; RMSECV 1,31%.  Analisis dengan PCA menunjukkan bahwa keempat sampel bakpao pasaran pedagang kaki lima di kota Yogyakarta tidak mengandung lemak anjing.

Abstract

Minyak kayu putih merupakan salah satu minyak atsiri yang biasa digunakan dalam sehari-hari. Produksi dalam negeri hanya dapat memproduksi minyak kayu putih 650 ton/tahun dari permintaan kebutuhan dalam negeri dalam setahun sebesar 3.500 ton sehingga memacu pemalsuan. Penelitian ini bertujuan untuk membandingkan profil minyak kayu putih hasil destilasi dengan minyak kayu putih yang ada di pasaran berdasarkan mutu Standar Nasional Indonesia (SNI). Destilasi minyak atsiri dilakukan dengan 5 kg daun kayu putih menggunakan metode destilasi uap air selama 3 hari didapatkan rendemen sebesar 2,32%. Minyak kayu putih hasil destilasi dan minyak produk A, B dan C dilakukan pengujian profil minyak berdasarkan SNI 06-3954-2006 meliputi warna, bau, bobot jenis, indeks bias, kelarutan dalam etanol 70%, dan profil KLT. Hasil pengamatan organoleptis minyak hasil destilasi, produk A, B, dan C memiliki perbedaan warna, bobot jenis, dan kelarutan dalam etanol 70% serta tidak ada perbedaan bau dan indeks bias. Hasil penelitian menunjukkan adanya perbedaan profil minyak produk A, B, dan C yang tidak memenuhi SNI pada pengujian bobot jenis pada produk C. KLT dengan menggunakan fase diam silika gel GF254 dan fase gerak Toluen (100%). Profil KLT pada produk C tidak terlihat noda yang sama dengan standar eucalyptol yang diperlihatkan oleh minyak lainnya. Keywords: Analisis profil, Minyak kayu putih, SNI 06-3954-2006

Abstract

(1) Background: Cervical cancer ranks as the fourth most prevalent cancer globally and in Asia, standing as the second most common in Indonesia. Despite its efficacy, doxorubicin chemotherapy is associated with significant side effects. To mitigate these adverse effects, a promising approach involves combining conventional drugs with curcumin. Both curcumin and doxorubicin have demonstrated cytotoxic effects against cervical cancer (HeLa). This research aims to determine the synergistic effect of the combination of doxorubicin and curcumin on HeLa and safety in normal cells (Vero cells). (2) Methods: This study adopted an experimental design utilizing doxorubicin and curcumin samples with HeLa and Vero cells. The investigation was initiated with cytotoxic and combination tests using the MTT method. The obtained results included IC50 values and combination indices, and the analysis involved a comparative examination of outcomes between HeLa and Vero cells. (3) Results: Cytotoxic tests revealed IC50 values for doxorubicin and curcumin on HeLa cells, measured in 2,17 ± 0,06 and 26,37 ± 2,00 µg/mL, and 17,32 ± 1,99 and 181,88 ± 30,48 µg/mL on Vero cells. Combination test results were represented by combination index .(4) Conclusions: The synergistic effect is observed at the combination of curcumin at a concentration of 9 μg/mL and doxorubicin at a concentration of 0,125 μg/mL, resulting in a combination index of 0,50. These findings suggest a promising avenue for enhancing the therapeutic potential of doxorubicin in cervical cancer treatment while minimizing adverse effects.

Abstract

The halal industry is rapidly growing worldwide, especially in Indonesia, which has the largest Muslim population and abundant natural resources. This study aims to analyze the physicochemical properties and quality of beef tallow and lard oil, as well as to identify differences in FTIR and GC-MS spectra to distinguish between these two animal fats.The results show that lard oil has a higher extraction yield (30%) compared to beef tallow (17%). Lard oil also has a higher iodine value (53 g), indicating a greater proportion of unsaturated fatty acids, making it suitable for culinary applications. Conversely, beef tallow has a lower peroxide value (5 meq O₂/kg), indicating better resistance to oxidation.FTIR analysis reveals differences in functional groups, with lard oil showing a higher degree of unsaturation. GC-MS analysis identifies important compounds such as 9-octadecenoic acid methyl ester in lard oil, while beef tallow is dominated by tetradecanoic acid methyl ester.This study emphasizes the need for the development of Indonesian National Standards (SNI) to ensure product quality and halal compliance. These findings provide important insights for the halal industry and food safety in Indonesia, as well as encouraging further research on the composition of animal fats.
