Abstract

Background: The first-line chemotherapy drug for breast cancer is cisplatin. However, it shows a high incidence of resistance. Therefore, Hibiscus Sabdariffa L (HS), containing strong anticancer compounds, was developed using a self-nano-emulsifying drug delivery system (SNEDDS), potentially as cisplatin co-chemotherapy. This study aimed to develop the HS SNEDDS providing anticancer activity against T47D breast cancer cells. Methods: Development of HS SNEDDS (F1-F7) with various surfactant and co-surfactant concentrations was carried out, followed by characterization of HS SNEDDS. The selected formulation was performed regarding the cytotoxicity on T47D cells and selectivity on Vero cells. Results: F7 as a selected formulation indicated a transmittance, globule size, PI, zeta potential, and emulsification time of 97.80%, 15.68 ± 0.19 nm, 0.12 ± 0.01, -8.05 ± 1.88 mV, and 24.76 ± 0.29 seconds, respectively. Furthermore, the IC50 of HS extract, HS SNEDDS, and Cisplatin on T47D cells were 0.37 ± 0.02, 0.24 ± 0.02, and 0.01 ± 0.001 mg/mL The selectivity index (SI) of HS SNEDDS and cisplatin were 108101.25 and 8351.84, respectively. Conclusion: HS SNEDDS providing the required characteristics could be obtained, and potentially be used as a co-chemotherapy of cisplatin, showing the cytotoxic effect on T47D breast cancer cells.

Abstract

Indonesia has the world's largest Muslim population in 2025 and high demand for halal food products, including sausage. Differences in meat prices increase the risk of adulteration, which is wild boar or pork can adulterate beef or chicken. The adulteration can be detected using FTIR combined with chemometrics. The aim of this study was to investigate the adulteration in sausages by analyzed the fingerprint FTIR spectra combining with chemometrics. This study conducted by physicochemical analysis and scanning the extracted fat at the mid-infrared region using FTIR to collect spectral data. Qualitative and quantitative analyses were performed on the fingerprint region using chemometrics of Partial Least Square (PLS) and Principal Component Analysis (PCA), respectively to provide best modeling. The PLS calibration model for the relationship between actual values and predicted value of three binary mixed meats obtained from 1399 – 1321 cm-1, 1770 – 1681 cm-1, 780 – 650 cm-1. The calibration models demonstrated strong linearity, with R² values ranging from 0.9367 to 0.9884. The chicken–beef model exhibited the lowest RMSEC (5.85), indicating superior predictive accuracy. Although the chicken–pork model showed the highest R² (0.9884), its higher RMSEC (7.32) suggests reduced prediction precision. The chicken–wild boar model displayed the lowest R² and highest RMSEC, indicating greater model uncertainty, likely due to increased spectral similarity and matrix complexity. Fifteen marketed products analyzed. PCA of FTIR data demonstrated clear discrimination between halal-labeled and pork-containing sausages based on lipid profile differences, highlighting its potential as a rapid screening tool for halal authentication.

Abstract

Mangkokan leaves contains flavonoids that are known as natural antioxidant source. The aim of this study is to find the effect of drying temperature variations towards free radical scavenging activity of mangkokan leaves. Mangkokan leaves macerated with 96% ethanol and dried at 40oC, 50oC, and 60oC. Then were macerated with ethanol 96%. Antioxidant activity tested by DPPH method and quercetin was used as positive control. Dry shrinkage with temperatures at 40oC, 50oC, and 60oC were: (9.25 ± 0.276)%; (8.76 ± 0.348)% and (7.97 ± 0.372)%, respectively. Water content at 40oC, 50oC and 60oC were: (5.13 ± 0.575)%; (4.53 ± 0.512)% and (4.04 ± 0.575)%, respectively. The antioxidant potency of 96% ethanolic mangkokan leaves by ES50 value at drying temperatures 40oC, 50oC, 60oC and quercetin were (1.75 ± 0.058); (2.39 ± 0,089); (5.38 ± 0,078) mg/ml and (9.16x10-4 ± 4.36x10-5) mg/ml. Ethanolic extract of mangkokan leaves which dried at 40oC has biggest radical DPPH scavenging activity but much weaker than quercetin.

Abstract

Minyak jagung mengandung protein dan asam lemak yang melimpah, sehingga diperlukan metode dan kondisi ekstraksi yang tepat untuk menghasilka kualitas minyak berkualitas tinggi. Minyak dapat diperoleh dengan cara pemanasan menggunakan alat Soxhlet maupun Reflux dan akan terisolasi dengan pelarut non polar. Penelitian ini bertujuan untuk mengoptimalkan ekstraksi minyak jagung manis, membandingkan kualitas minyak jagung dari dua metode isolasi serta menganalisis komposisi asam lemak menggunakan GC-MS. Minyak jagung manis yang dihasilkan dari dua metode isolasi Reflux dan Soxhlet dibandingkan dengan parameter SNI dan diidentifikasi komposisi asam lemaknya menggunakan instrumen GC-MS. Parameter kualitas minyak biji jagung manis yang dihasilkan dari metode Soxhlet dan Reflux, memiliki karakteristik warna, bau, indeks bias, kadar air, bilangan asam, bilangan peroksida, dan bilangan iod yang memenuhi persyaratan SNI. Hasil analisis GC-MS menunjukkan metode Reflux menghasilkan 3 kandungan senyawa yaitu, asam palmiat (31,37%), linoleoyl chloride (67,15%), dan asam ftalat (1,48%), sedangkan metode Soxhlet menghasilkan 2 kandungan senyawa, yaitu asam palmiat (32,28%) dan asam linoleat (66,89%). Hasil penelitian menunjukkan bahwa variasi metode ekstraksi minyak biji jagung memengaruhi kualitas minyak serta kandungan asam lemak yang dihasilkan.
