Abstract

Cassava (Manihot esculenta crantz) is one of the agricultural commoditythat produce tuber for food source in Indonesia. The research objective was to determine the effect of manure on tuber yield and genetic diversity of the five genotypes of cassava Taluk Kuantan. The research was arranged in the randomized block design (RBD) with two factors. The first factor was five cassava genotypes namely: (IY1) cassava pucuk hitam, (IY 2) cassavabangka, (IY3) cassava pulut, (IY4) cassava roti; and (IY5) cassava pucuk hijau. The second factor was the fertilizer of cow manure consists of five doses, namely: P0 (0 kg/plant), P1 (2 kg/plant), P2 (4 kg/plant), P3 (6 kg/plant); and P4 (8 kg/plant). The parameters observed were morphologicaland agronomiccharacters of cassava, such as plant height,number of tubers/plant, tuber diameter, tuber length, and tuber weight/plant. Pulut cassava produced heaviest tuber in the fertilizer with a dose of 4 kg, bangka and pucuk hijau cassavas produced the highest tuber weight in the fertilizer with 6 kg dose.

Abstract

Mungbean (Vigna radiata L.) is one of important legume in Indonesia. G-SSR and EST-SSR markers had been widely used in mungbean genetic diversity research. DNA isolation and DNA amplification are required to obtain genetic information about mungbean to obtain accurate data on the genetic diversity of mungbean. This study aims to determine the Temperature of annealing (Ta) of the g-SSR and EST-SSR primer pairs. The total DNA was isolated from young leaves mungbean origin Pelalawan and the eight primer pairs of the g-SSR and EST-SSR were optimized. The optimal Ta for G2436, G3598, G2516, G7472, G0483. G1671, G3302, and G3427 were 50,55ºC, 51,15ºC,51,25ºC, 51,2ºC, 51,6ºC, 49,0ºC, 49,8ºC, and 52,8ºC respectively. Meanwhile, the optimal Ta for E51985, E19823, E24080, E22860, E26637, E16266, E11659, and E10675 were 52,2ºC, 54,4ºC, 52,5ºC,51,25ºC, 53,25ºC, 54ºC, 54,35ºC, and 53,6ºC respectively.

Abstract

Determination of annealing temperature of the primer is the first step for genetic diversity analysis using molecular markers such as RAPD (Random Amplified Polymorphic DNA). This study aims to determine annealing temperature (Ta) of RAPD primers on Kampar Mungbean. Methods included total DNA extraction, electrophoresis, and annealing temperature optimization of four RAPD markers namely OPD-20, OPI-06, OPI-13, dan OPX-13. Optimization was conducted by reducing the Tm value (Time melting) of each primer with 3 (Tm-3) and 5 (Tm-5). The results showed that the optimization using OPD-20 and OPX-13 produced bands at Tm-3 and Tm-5. Meanwhile, optimization using OPI-06 and OPI-13 resulted in bands at Tm-3. The next step was to choose the exact Ta based on the clear and bright band. In conclusion, exact Ta for OPD-20, OPI-06, OPI-13, and OPX-13 were 36,1°C, 38,1°C, 35,4°C, and 32,5°C respectively.

Abstract

Mung bean (Vigna radiata L.) is one of the important because it has a high nutritional value, so many Indonesia consumption its crop. The production of mung beans is still relatively low due to the reduced availability of land, so it is necessary to use marginal land such as saline land. Therefore, this study aims to analyze the resistance of 10 mung bean genotypes (Vignaradiata L.) from Riau Province on salinity stress. Mung beans seeds of 10 genotypes were grown in polybags with treatments including Control and Treatment saline 7 g/L NaCl of Dolphin salt with 7 replications for 66 HST. The interval of watering saline treatment was five days on time the plant begins to bloom. All genotypes showed different morphological responses to salinity stress. Characteristic of 10 genotypes of mung beans from Riau Province based on salinity stress treatment was obtained a significantly different result on the character of productive branch number, plant height, flowering period, number of pods, pod length, seed number, 100 seed weight, and root length. The tolerant genotype of the salinity of Rohul origin 1 (RU1) was superior to plant height, flowering period, number of pods, number of seeds, and root volume. Genotypes from Pelalawan (P) for branch character, Inhu (IU) genotype was for long pods and 100 seeds, and Rohul 2 (RU2) genotype was for long root characters.

Abstract

ABSTRACT Genes expression information involved in Pandanus sp. adaptation in Kajuik Lake sp. to environmental stress is unknown. The genes expression analysis requires the internal control gene such as actin. Actin gene is one of the genes that can be expressed continuously at all stages of plant development. Before obtaining genetic information, it is necessary to isolate and amplify DNA. The purpose of this study was to determine the annealing temperature for actin gene amplification in Pandan (Pandanus sp.). The DNA amplification uses two primer pairs with 20 annealing temperatures. The annealing temperatures were calculated based on the mean values of Tm which were then reduced and added with 5, 4, 3, 2 and 1. The annealing temperature of 53.4 ºC (Tm +1) yielded single thick clear using. The annealing temperature of 53.4 ºC (Tm +1) using P_act_F / P_act_R1 primer pair yielded single, thick and firm DNA band. It was concluded that the annealing temperature for actinin gene amplification in Pandanussp was 53.4 ºC with primer P_act_F / P_act_R1. Keywords: Actin gene, Annealing temperatures, PCR

Abstract

Maman is a traditional plant frequently used as food by people in Melayu Rokan of Riau Province. Morphological detection indicates that the scientific name of maman is Cleome gynandra L.  The objective of this study is to confirm the taxonomic status of maman from Riau basedon matK and trnL-trnL-trnF intergenic spacer (IGS) sequences. Methods utilized in this study included sampling, DNA extraction, PCR, electrophoresis, sequencing, and data analysis. The fresh leaves were picked up from Rokan Hulu Regency, Riau Province. In this study, the matK and trnL-trnL-trnF IGS sequences of maman with a length of 754 bp and 938 bp, respectively, had been obtained. The BLASTn analysis based on both sequences showed that maman had 100% similarity to Gynandropsis gynandra. Cleome gynandra was synonymous with G. gynandra. There were 49 nucleotide variations, 16 critical nucleotides, and there were no indels in matK. Meanwhile, in trnL-trnL-trnF IGS there were 181 nucleotide variations, 13 critical nucleotides, and 61 indels. In conclusion, this study succeeded in confirming the taxonomic status of maman from Riau as C. gynandra syn. G. gynandra. The result enriched the abundance of DNA barcode sequences of this plant in GenBank which is useful for the molecular identification.

Abstract

The productivity of palm trees grown is generally measured in monthly and yearly periods, but on a shorter time scale, it is particularly measured by the plant maintenance of palm trees. Consideration of the general productivity in time is how to produce good fruit fertilizer (chemical aspect), healthy plants, fast growing and evolving (agricultural aspect), and genetic type of palm that grows (biological aspect). However, the growth and production on the physical aspect is still less attention. Through physical treatment, the growth of palm trees can be optimized to support aspects of the previous aspects, as it has been proven in advanced countries on the plant's aloe vera, avocado, and pine trees. In this paper, the methodology proposes a direct current voltage treatment to the palm tree that can accelerate the flow of ions to grow and develop nutrients. The palm tree samples are aged 1 to 5 years corresponding to the provision of 10, 16, 25, 35, and 50 V for each of the ages of the palm tree respectively. Identifying samples before and after treatment by electricity is based on the geometry of leaves, midribs, and ions effect. There have been reduced and increased levels of nutrients. This identification can optimize the productivity of palm trees.

Abstract

A taxonomic status of Pucuk Seminyak plant is still unclear. This plant has a fruit like Melinjo (Gnetum gnemon) fruit and is often classified as a member of Gnetaceae family. This study aims to determine the taxonomic status of Pucuk Seminyak from Riau using trnL-trnL-trnF intergenic spacer (IGS) and matK sequences. Methods included leaf samples from three individuals collected from the Kampar Regency, Riau Province, total DNA extraction, electrophoresis, polymerase chain reaction (PCR), and data analysis. The results showed that the trnL-trnL-trnF IGS DNA sequences from the three samples of Pucuk Seminyak were obtained 937 bp length, while the matK had a size of 775 bp. These sequences have been registered in GenBank with registration numbers OQ174512, OQ174513, and OQ174514 for trnL-trnL-trnF IGS, and OQ174525, OQ174526, and OQ174527 for matK. The highest similarity was found between Pucuk Seminyak and Champereia manillana with an identity value of 99.67% based on trnL-trnL-trnF IGS and 99.74% based on matK. BLASTn analysis by default settings and organism Champereia (taxid: 50116) resulted in hits on the Champereia genus of the Opiliaceae. Further phylogenetic analysis placed the study objects in the Champereia clade (bootstrap support 49 for trnL-trnL-trnF, 68 for matK, and 71 for concatenated sequences). Lastly, identification by morphological characters also confirmed that the specimen agreed with the description and identification key for C. manillana. The species is classified into two varieties: var. longistaminea and var. manillana. The former was distributed in Southwest China, and the latter was distributed in Malesia to India. Furthermore, the phylogenetic analysis separated the two varieties into different clades. Therefore, it can be concluded that our specimen of Pucuk Seminyak is identified as C. manillana var. manillana Merr. (Opiliaceae).

Abstract

Species within the Pandanaceae family, particularly those in the Pandanus genus, exhibit substantial morphological variation, often leading to challenges in accurate species identification. This study conducted a multilocus DNA barcode analysis using sequences of matK, atpB-rbcL Intergenic Spacer (IGS), and trnL-trnF Intergenic Spacer (IGS) obtained from GenBank. Sequence alignment was performed using MAFFT v.7, and the aligned sequences were concatenated into multilocus datasets. Further analyses included frequency data evaluation and phylogenetic tree construction using the Neighbor Joining method in MEGA11. Results revealed that matK had the highest number of conserved sites and the greatest genetic stability, making it particularly effective for identifying broader taxonomic groups. In contrast, atpB-rbcL IGS and trnL-trnF IGS showed greater genetic variation, useful for distinguishing species with similar morphological features. Phylogenetic trees based on single- or two-locus data often failed to group species accurately, while multilocus combinations yielded well-resolved and more reliable tree structures. Therefore, the combined use of atpB-rbcL IGS + matK + trnL-trnF IGS significantly improves species resolution and reduces taxonomic misidentification within Pandanaceae. This study underscores the importance of multilocus barcoding in addressing limitations of morphological classification and enhancing molecular taxonomy in morphologically complex plant groups.

Abstract

Maman plant (Cleome gynandra L.) is an underutilised leafy vegetable that has a high nutritive value and contains phenolic compounds that are essential in reducing or preventing the occurrence of chronic and infectious diseases. In this study, we investigate the effect of drying methods on the total phenolic content of maman plant from Riau. The drying treatment process for maman leafs was tried using three methods: room air-dried without sunlight exposure at 21-24oC for seven days, freeze-dried at -40oC for four days, and oven-drying at 40oC for three days. Freeze-dried maman leafs exhibited significantly (p < 0.05) higher total phenolics (5,90 mgGAE/g) than those dried using other methods, followed by oven-dried (5,14 mgGAE/g) and room-air dried (4,50 mgGAE/g). Drying maman leaves for further research is recommended using the freeze-dried method to obtain optimal phytochemical content.
