Abstract

Latar Belakang: Sekitar 30-50% karsinoma payudara positif terhadap virus Epstein-Barr (EBV). Studi melaporkan bahwa karsinoma payudara positif EBV cenderung lebih agresif. Infeksi EBV dapat dibuktikan salah satunya melalui pemeriksaan LMP1. Sejauh ini studi tentang infeksi EBV terhadap karakteristik klinikopatologis karsinoma payudara di negara-negara Asia termasuk Indonesia masih terbatas. Penelitian ini bertujuan menganalisis hubungan ekspresi LMP1 dengan karakteristik klinikopatologis karsinoma payudara invasif.Metode: Penelitian cross-sectional menggunakan 64 sampel blok parafin dari pasien karsinoma payudara invasif di RSUP Dr. Kariadi periode Juni-September 2019. Ekspresi LMP1 dinilai dengan immunostaining. Data klinikopatologis berupa usia, ukuran tumor, grade, ekspresi ER, PR, HER2, Ki67, subtipe molekuler dan keterlibatan KGB aksila. Analisis menggunakan uji korelasi dan SPSS 17.Hasil: Dari 64 kasus karsinoma payudara invasif, sebagian besar berusia >50 tahun (59,4%), ukuran T2 (45,3%), grade 2 (70,3%), ekspresi ER positif (62,5%), ekspresi PR negatif (56,3%), ekspresi HER2 negatif (64,1%), indeks Ki67 > 20% (75%), subtipe molekuler Luminal B-like (HER2 negative) (31,3%) dan metastasis KGB N1 (35,9%). Ekspresi hanya berkorelasi dengan metastasis KGB (p=0.01), bermakna karsinoma payudara invasif dengan LMP1 positif menunjukkan semakin tinggi risiko keterlibatan KGB.Kesimpulan: Ekspresi LMP1 berkorelasi dengan tingkat metastasis KGB.

Abstract

Tujuan dari penelitian ini untuk mengetahui pengaruh ekstrak daun kari terhadap kadar glukosa darah, malondialdehid (MDA), dan gambaran histopatologi hepar dan ginjal. Penelitian ini menggunakan metode eksperimen laboratorium dengan desain kelompok kontrol posttest acak. Subjek penelitian adalah 16 tikus Wistar (Ratus novergicus). Dalam penelitian ini pembuatan ekstrak dengan proses maserasi menggunakan 96% etanol (1:3). Analisis kandungan ekstrak ekstrak daun kari seperti flavonoid menggunakan metode reduksi dengan standar quercetin, kandungan tanin dan saponin. Intervensi akan dilakukan selama 21 hari. K- 1 ml aquades / hari; Metformin K + 45mg / kgBB; P1 160 mg / kgBB ekstrak daun kari; P2 400 mg / kbBB ekstrak daun kari. Induksi DM menggunakan intraperitoneal aloksan dosis tunggal 120 mg / kgBB. Pengukuran glukosa darah pada hari ke-21 setelah induksi pengobatan menggunakan metode GOD-PAP. Hasil dari penelitian ini yaitu Flavonoid total dalam daun kari adalah 599 mg / 100 g ekstrak  dan mengandung tanin dan saponin. Pada uji ANOVA menunjukkan ada perbedaan kadar glukosa darah dan kadar MDA yang signifikan antara kelompok 0,00 (p <0,05). Uji post hoc menunjukkan bahwa dosis 320 mg / kgBB ekstrak daun kari dapat menurunkan kadar glukosa hingga 75 mg/dl p= 0,00(p<0,05), dan menurunkan kadar MDA hingga 31.46 nmol/ml. Mann-withney menunjukkan ada perbedaan yang signifikan antara kelompok p = 0,00 (p <0,05) untuk derajat kerusakan hepar dan degenerasi tubulus ginjal. Sehingga dapat disimpulkan bahwa terdapat perbedaan pada pemberian ekstrak daun kari 320 mg/kgBB dengan kelompok kontrol negatif terhadap kadar glukosa darah, MDA, dan gambaran histopatologi hepar dan ginjal.

Abstract

Background: Diabetes mellitus (DM) is a chronic metabolic disease that can cause microvascular and macrovascular complications. The formation of superoxide (O2-) by dysfunctional mitochondria in diabetes has been recognised as a major driver of diabetic complications. Consequently, the utilisation of antioxidants may prove advantageous. The plant Andredera cordifolia contains flavonoids, which act as antioxidants.Objective : to investigate the effect of binahong leaf extract on the development of atherosclerosis in the carotid artery.Methods: The research employed a laboratory experimental method with a randomised post-test control group design. The research subjects were 15 Wistar rats (Rattus norvegicus). In this research, the extraction of the active compounds was achieved through a maceration process using 96% ethanol (1:3) and emulsification by the addition of sunflower oil, CMC, Span80 and Tween 20. The analysis of Anredera cordifolia extract content included the determination of flavonoid content and IC50. The intervention will be conducted for a period of 14 days. The rats were administered 1 ml of aquades per day, 45 mg/kg BW of metformin (K+), 200 mg/kg BW of EAC (P1), and 400 mg/kg BW of EAC (P2). The diabetic model was induced using a single dose of 50 mg/kgBW streptozotocin intraperitoneally. On the fourteenth day following the induction of treatment, blood glucose measurements were taken using the GOD-PAP method, and a histopathological examination of the common carotid artery was conducted using eosin haematoxylin staining at a magnification of 100-400x.Results: The total flavonoid content of binahong leaves is 1525 mg/100 g extract, while the IC50 value is 1.9 ppm. The results of the ANOVA indicate a statistically significant difference between the fasting blood glucose levels of the two groups (p < 0.05). The post hoc test demonstrated that a 400 mg/kgBW dose can result in a reduction in glucose levels to 305.75 mg/dl, with a p-value of 0.001 (p < 0.05). The Mann-Whitney test revealed a statistically significant difference among the groups (p < 0.05) for the inhibition of atherosclerosis in the common carotid artery. Conclusion: A significant difference was observed in the administration of 400 mg/kgBB binahong leaf emulsion extract in comparison to a negative control group, with regard to the histopathological picture of atherosclerosis of the common carotid artery

Abstract

Extranodal NK/T-cell lymphoma, nasal type (ENKL) is a lymphoid neoplasm, which in the past has been grouped as a type of granulomatous diseases, is an aggressive non-Hodgkin’s lymphoma. The aggressiveness of ENKL is characterized clinically by nonrelenting destruction of the midline structures of the palate and nasal fossa. The histopathological diagnosis of this disease is difficult to enforced despite the malignant clinical course recognizably because of extensive tissue necrosis and multiple biopsies are often required. ENKL has an ominous prognosis, as the average survival rate is between 6 and 25 months. This case report describes the difficulties in making histopathological diagnosis of a 52-year-old male patient who presented with nasal obstruction and foul smelling, destructive and ulseratif lesion over for 3 months duration, and failed to treat with antibiotics and anti-inflammatories drugs. The diiferential diagnosis of the multiple biopsies was nasal cavity granuloma, NK/T cell lymphoma and Wegener granulomatosis. After immunohistochemical panel analysis of Pansitokeratin (AE1/3), LCA, CD3 and CD56, the patient be confirmed had an ENKL, nasal type which in the past also known as angiocentric T-cell lymphoma.

Abstract

Ovarian cancer is the 7th leading cancer in incidence (3,4%) after breast, colorectal, lung, cervix uteri, thyroid, corpus uteri and stomach cancer among women worldwide according to Globocan, 2018. Ovarian cancer in Indonesia is the 3rdmost leading cause of cancer death, after cervix and breast cancer. The most common type of ovarian cancer is epithelial ovarian carcinoma (EOC), which most prevalent in older women. This study was aimed to describe clinicopathological profile of EOC in kariadi general hospital semarang from January 2016 to December 2018. This is the observational analytical studyusing data of ovarian cancer patient medical records at the laboratory of pathological anatomy in kariadi general hospital semarang. Data collection include : age of diagnosis, location, size tumor and histological subtype. 330 EOC patient were included in this study. Most of EOC patient (52%) were diagnosed more than 40 years old. 81% of EOC patient had unilateral location. 60% of patient had size tumor more than 14 cm in diameter. Histological subtype of patient was mostly mucinous carcinoma (40%). EOC patient in kariadi general hospital semarang was mostly diagnosed at more than 40 y.o, had unilateral location and had mucinous carcinoma subtype.

Abstract

Background: Elevated intraocular pressure (IOP) induces inflammation in the trabecular meshwork (TM) through increases in tumor necrosis factor-alpha (TNF-α) expressions by activating nuclear factor kappa B (NF-κB). Mechanical injury to the TM, such as increased IOP, can cause extracellular matrix (ECM) remodeling, impacting the permanent outflow of aqueous humor. Moringa oleifera (MO) leaf extract is an anti- inflammatory agent that inhibits NF-κB activation and has been studied to have antioxidant effects.Objective: to analyze the effects of MO leaf extract on TNF- α expression and the thickness of the trabecular meshwork ECM in a Wistar rat glaucoma model. Methods: Fourteen male Wistar rats were divided into two groups (n=7): the glaucoma group received oral MO leaf extract at 300 mg/kg body weight for four weeks, and the control group. TNF-α expressions were measured by immunohistochemical stain, and the thickness of the ECM was examined with hematoxylin eosin staining. P-values of less than 0.05 were considered to represent statistical significance.Results: The mean of TNF-α expressions in the treatment group was 4.29 ± 0.76, and the control group was 5.29 ± 0.49. There was a significant difference in TNF-α expressions between the two groups (p=0.015). The mean thickness of the ECM TM experimental group and control group were 155.65 ± 28.72μm and 218.63 ± 40.85μm respectively. The extracellular matrix TM experimental group was statistically significantly thinner than the control group (p=0.006).Conclusion: MO could protect TM from damage caused by high IOP by suppressed TNF- α expression and ECM thickness in the TM Wistar glaucoma model.

Abstract

The wound healing process of traumatic injuries in the oral cavity involves four stage: hemostasis, inflammation, proliferation, and remodeling. Fibroblast cells are essential for the wound healing process, appearing on the 3rd day and reaching their peak on the 7th day after injury. This study reveals that bay leaf extract can accelerate wound healing process by mitigating inflammation and enhancing fibroblast cell activity. This research utilized a post-test only control group design. Fifteen male Wistar rats divided in 3 groups equally:  control group and 2 groups were given 5% and 10% of the bay leaf extract gel was applied twice a day until the 5th  day. The results indicated that the group treated with 10% bay leaf extract gel had significantly higher value compared to the control group and the group treated with 5% bay leaf extract gel. In this study, we can learn that bay leaf extract gel can increase of fibroblast cells in the ulcer healing process.

Abstract

Introduction & ObjectivesGlaucoma is a disorder characterized by the presence of optic neuropathy. Visual impairment inglaucoma is caused by retinal ganglion cell (RGC) apoptosis associated with N-methyl-D-aspartate(NMDA)-mediated excitotoxicity. Excitotoxicity causes an increase in calcium in cells as well asactivation of caspase-3. Glaucoma therapy is currently given to reduce intraocular pressure (IOP),but the damage is still progressing progressively. Hesperidin is known as a neuroprotector that cansuppress calpain overactivation in NMDA-treated retina, thereby reducing the upregulation of TNF-,an inflammatory cytokine, inhibiting calpain activation by suppressing oxidative stress, andattenuating caspase-3 activation. The aim of this study to proving the effect of oral Hesperidin on theexpression of caspase-3 Retinal Ganglion Cell in Wistar rats model of NMDA-induced glaucoma. MethodsExperimental research with post test design only randomized control trial Wistar rats with NMDAinduction were divided into 2 treatment and control groups. The treatment group was given oralHesperidin at a dose of 100mg/kg/day while the control group was given Na-CMC 182 mg/kg/day for3 weeks. Retinal caspase-3 expression was examined by immunohistochemical staining. Data werecollected and processed and then the Mann- Whitney test was performed. (significant p<0.05) ResultsThe mean percentage and intensity of RGC caspase-3 expression in the treatment group were 4.29 ±0.49 it was significantly lower than the control group 6.29 ± 0.95 (p value = 0.003). ConclusionHesperidin provides neuroprotection for RGC againts NMDA- induced exitotoxicity by inhibition ofcaspase-3.

Abstract

Introduction & ObjectivesRetinal ganglion cell (RGC) damage can affect the thinning of the GCC layers, which is the nervefiber layer, the ganglion cell layer and the inner plexiform layer. The loss of RGC and inner plexiformlayer (IPL) is strongly correlated with the loss of visual field defect. RGC and inner plexiform layerare potential targets to evaluate the progression of glaucoma. Glaucoma treatment not only focus onreducing IOP. Decreasing IOP according to the target pressure often can’t stop the process of RGCdamage and make it permanently and irreversible. Vitamin D3 is known as neuroprotector which hasthe ability to protect RGC and prevent layer thinning of GC-IPL in retina. The aim of this study is todetermine the effect of oral Vitamin D3 on the GC- IPL layer thickness in retinal of Wistar rats withNMDA-induced glaucoma model. MethodsGlaucoma Wistar rats were divided into 2 groups. The treatment group was given oral Vitamin D3 ata dose of 500 IU/kgBW/day for 10 weeks. GC-IPL layer thickness in retinal was examined byHematoxylin Eosin staining. Data were collected and processed then used with Independent T Test.(significant p<0.05) ResultsThe mean GC-IPL layer Thickness in the treatment group was 77,59 ± 15,15 ?m and 35,32 ± 4,04?m in the control group. There was a significant different between two group ( p = <0,001 ) ConclusionVitamin D3 has a neuroprotective effect on ganglion cell-inner plexiform layer Wistar rats in NMDAinducedexcitotoxicity model of glaucoma

Abstract

The use of synthetic insecticides in treating myiasis is associated with adverse side effects and potential disruption of metabolic systems, prompting interest in natural alternatives. This study investigated the in vivo larvicidal efficacy of Gigantochloa apus bamboo shoot extract, formulated as a spray gel. Twenty-five Wistar rats were randomly divided into 5 groups: a negative control group, a positive control (ivermectin), and 3 treatment groups receiving of 1%, 3%, and 5% G. apus bamboo shoot extract. Myiasis was induced by introducing fly larvae into standardized wounds. Treatments were applied topically twice daily for 32 hr, and larval mortality was assessed every 8 hr. Phytochemical screening and GC-MS analysis revealed the presence of bioactive compounds, including alkaloids, flavonoids, tannins, saponins, and hydrogen cyanide, all of which are known for their larvicidal, neurotoxic, and antiproliferative properties. The 5% extract group showed the highest mortality rate (100%) at 32 hr. The LC50 and LC95 values were determined at 1.43% and 6.01%, respectively. Compared to the standard ivermectin treatment, the 5% extract demonstrated a shorter lethal time and more rapid larval death. Morphological examination revealed darker abdominal segments in the dead larvae, indicating a potential interaction with the digestive tract. These findings indicate that G. apus bamboo shoot extract has potential larvicidal activity and can be an effective natural alternative for treating myiasis.
