Abstract

Background: Breast cancer is the second largest cancer in the world. The gold standard for breast cancer diagnosis and prognosis is grade assessment (mitotic, nuclear pleiomorphism, tubular formation), based on breast tissue histopathology. However, this assessment has limitation in that it is subjective and semi quantitative. A genetic biomarker known as  microRNA (miRNA) may regulate human gene expression by targetting mRNA. One of the miRNA, i.e. miR-21, is able to suppress tumor suppressor mRNA, resulting in increased cellular proliferation andneoplastic transformation, and decreased apoptosis. This study aimed to investigate the expression of miR-21in breast cancer with histopathologic grade I, II, and III.Subjects and Method: This was a descriptive study with cross-sectional design involving paraffin block (FFPE) samples of ductal-type breast cancer tissue. Sixty four samples were obtained from the department of Anatomical Pathology, Haji Adam Malik Hospital, Medan. Patients history were obtained through medical records followed by sample processing (Total RNA Isolation, cDNA Synthesis, and Examination of miR-21 Expression using Real-Time qPCR). MiR-21 expression from every hystopathologic grade was obtained using Fold Change Calculation. Results: Breast cancer occurred more frequently in age group of 30-49 years old. Based on the histopathologic grade, most breast cancer was found in grade II. Analysis of miR-21 expression showed that miR-21 expression was higher in grade III compared to other grade. Conclusion: Breast cancer occured more frequently in the productive age group. Most breast cancer were found in grade II. Increased miR-21 expression in grade III may suggest that miR-21 is an oncomiR, which plays an important role in the development of malignant breast cancer. Keywords: breast cancer, histopathology grade, miRNA, miR-21.Correspondence: Juwita. Abstract

Background: MicroRNA (miRNA) is noncoding-RNA that comprises 18-22 nucleotides. miRNA binds its gene targets in the 3-untranslated region (3-UTR), causing direct degradation of mRNA and repressed translation of RNA. MicroRNA regulates various biological processes, including cell proliferation, apoptosis, cell growth, cell differentiation, and metabolism. Various changes have been identified miRNA expression in different types of cancers. This study aimed to describe the expression of miR-155 in patients with breast cancer tissue in Haji Adam Malik Hospital in Medan.Subjects and Method: This was a descriptive study. A total sample was 64 paraffin tissue blocks was taken from female patients with breast cancer at the surgical oncology unit, Haji Adam Malik Hospital in 2013-2014. RNA sample were isolated and checked using a spectrophotometer followed by cDNA synthesis. Then this sample was analyzed for miR-155 expression by using Real Time qPCR. Histopathological grade data were obtained from the medical record. Results: The miR-155 expression in the breast cancer patients is higher than that in the UniSp6 RNA Spike-in (internal controls). miR-155 expression was 29.64, 28.44, 29.22, in grade-I, grade-II, grade-III breast cancer, respectively. UniSp6 RNA Spike-in was 22.30, 22.89, 23.10, respectively in the control groups. Thus, increased of miR-155 expression was the highest in grade I breast cancer patients. Conclusion: Increased miR-155 expression can be used as a biomarker for early diagnosis and prognosis of breast cancer. Keywords: miRNA, miR-155, breast cancer.Correspondence: Henny Erina Saurmauli Ompusunggu. Abstract

Cardiac Arrest is the leading cause of death in the world including Indonesia, which can occur anytime and anywhere. The causes vary, ranging from fatigue, and underlying health conditions, to traffic accidents. Basic Life Support (BLS) is the initial step that can be taken to restore respiratory and circulatory function in individuals experiencing respiratory or cardiac arrest. It is important for online motorcycle taxi drivers to possess these skills, as they often spend time on the road and interact with various people in different situations. Perhimpunan Dokter Spesialis Anesthesiology dan Terapi Intensif (PERDATIN) North Sumatra Branch concluded a series of Kursus Penyegar dan Penambah Ilmu Anestesia (KPPIA) by providing basic life support training to 521 public transport or online motorcycle taxi drivers in the city of Medan on September 29, 2024. Held at the Gedung Serba Guna of the North Sumatra Provincial Government, the participants were divided into 52 groups guided directly by anesthesiologist specialists as instructors. By providing this education and training, they can be better prepared to face emergency situations on the road, thereby enhancing safety and service for passengers

Abstract

RTI (Respiratory Tract Infection) is an acute upper and lower respiratory tract inflammation caused by infectious agents, namely infections of microorganisms or bacteria, viruses, without or accompanied by lung parenchyma. RTI disease in Indonesia is on the 10th list of most common diseases in hospitals. PKM FK-UMI aims to bring the SPA (RTI-Free School/Sekolah Bebas ISPA) program, increase knowledge about RTI, and prevent and reduce the occurrence of RTI to Galilea Hosana Kids Elementary School. The method used is to deliver material about the definition, symptoms, how to get RTI, and how to prevent it. The results of the activity show that Galilea Hosana Kids Elementary School students still do not understand the meaning of RTI, the signs and symptoms of decreased body immunity, how to increase body immunity, and how to prevent the spread of RTI. Counseling for elementary school students at Galilea Hosana Kids Elementary School was very effective in educating students about RTI.

Abstract

Aging is a complex biological process characterized by a decline in cellular function and increased susceptibility to degenerative diseases. In recent decades, research has shown that epigenetic modifications—such as DNA methylation, histone modification, and regulation by non-coding RNAs—play a key role in the aging process. These changes influence gene expression without altering the DNA sequence and contribute to cellular damage, chronic inflammation, and tissue dysfunction. As scientific understanding advances, various epigenetic-based molecular interventions have emerged, including epigenetic reprogramming, senolytic therapies, and pharmacological and lifestyle approaches targeting epigenetic mechanisms. Although challenges remain in terms of safety, specificity, and clinical applicability, these approaches offer promising potential for extending healthspan and improving quality of life in later years.

Abstract

Background: Type 2 diabetes mellitus (T2DM) is a complex metabolic disorder associated with chronic inflammation and male reproductive dysfunction. The ethanol extract of turmeric (Curcuma longa) has demonstrated potential as an alternative therapeutic agent due to its anti-inflammatory and reproductive restorative properties. This study aims to analyze the anti-inflammatory and anti-infertility effects of the ethanol extract of Curcuma longa in male Wistar rats with T2DM induced by a streptozotocin-nicotinamide-high fat diet (STZ-NA-HFD). Subjects and Method: Thirty male Wistar rats were randomized into six groups (n=5): normal control (K1), diabetic control (K2), positive control (T2DM + metformin 45 mg/kgBW, K3), and three treatment groups receiving Curcuma longa extract at doses of 50 mg/kgBW (K4), 100 mg/kgBW (K5), and 150 mg/kgBW (K6). Diabetes was induced with STZ (55 mg/kgBW) and nicotinamide (120 mg/kgBW). Blood glucose, IL-6, testosterone levels, sperm count, and sperm motility were evaluated after 21 days of treatment. Results: The turmeric extract significantly reduced blood glucose levels (p<0.001) and IL-6 concentrations from 574.06±68.99 to 214.06±78.71 ng/mL at a dose of 150 mg/kgBW (p<0.001). Normal sperm motility increased significantly at 150 mg/kgBW (20.0±2.55%) compared to the diabetic control (8.40±2.07%) (p=0.008). Conclusion: The ethanol extract of Curcuma longa demonstrated significant anti-inflammatory activity by reducing IL-6 levels and improving glycemic control. Its anti-infertility effect was reflected by enhanced sperm motility, particularly at the 150 mg/kgBW dose. These findings indicate that Curcuma longa possesses potential as an adjuvant therapy for the management of T2DM and its associated complications.
