Abstract

Kaimana and Fakfak are two regions in West Papua which have high biodiversity. The total fish production in Kaimana and Fakfak are 10,039 tons.y-1 and 17,806 tons.y-1, respectively. The snapper fish (Lutjanus spp.) is one of the important economic commodities in Kaimana and Fakfak regions. There has been a decrease in the number of exports of snapper fish in 2018 to 2019 from 4,742 tons to 4,290 tons due to overfishing and environmental pollution. This study employed DNA barcoding technology to identify the species of snapper fish collected from Kaimana and Fakfak. The DNA isolation was conducted by using genomic DNA mini kit (tissue) and the amplification of COI gene with Go Taq green master mix. Agarose gel electrophoresis was used to visualize the PCR product.  A total of 16 sequences with length 654 base pairs of COI gene were identified as five species of Lutjanidae, which were Lutjanus decussatus, L. gibbus, L. quinquelineatus, L. malabaricus, and L. johnii. Homology analysis with BLAST NCBI and BOLD System showed that all samples have similarity of 99.08-100% and query cover of 93-100%. Relationship analysis using phylogenetic tree and genetic distances showed results of intraspecific close relatives (0.001-0.016) and interspecific distant relatives (>0.1000). The phylogenetic tree illustrated that all species of Lutjanidae are separated into monophyletic clades. DNA barcoding technology successfully identified the snapper fish collected from Kaimana and Fakfak.

Abstract

Sebagai upaya memulihkan kondisi perikanan dan ekosistem pesisir,  masyarakat Kampung Asai melaksanakan tasamu rawanang (peleh lautan) yang berlangsung sejak Agustus 2022.  Penelitian ini dilaksanakan pada April hingga Mei 2023, bertujuan untuk mengetahui apakah restorasi mangrove secara alami dapat terjadi tanpa adanya penanaman mangrove, melalui pengkajian struktur vegetasi mangrove dan indeks ekologi mangrove. Untuk memperoleh data vegetasi digunakan metode transek kuadran di 3 stasiun. Struktur komunitas mangrove yang dianalisi adalah Indeks Nilai Penting (INP) dan indeks ekologi yang dianalisis antara lain indeks keanekaragaman, indeks kemerataan, dan indeks dominansi. Hasil penelitian menginformasikan bahwa terdapat 11 jenis mangrove sejati. INP tingkat pohon tertinggi adalah R. apiculata (154,743%) diikuti B. gymnorrhiza (59,696%) dan R. mucronata (26,655%). Pada tingkat anakan dan semai, R.apiculata juga memiliki INP tertinggi. Nilai rata-rata keanekaragaman pada tingkat pohon adalah 1,263, tingkat anakan 1,165 dan tingkat semai 1,051, yang masuk dalam kelas keanekaragaman sedang. Indeks kemerataan jenis pada tingkatan pohon berkisar antara 0,595-0,625. Untuk tingkat anakan berkisar antara 0,626-0,786. Sedangkan untuk tingkatan semai berkisar antara 0,768-0,772. Hal ini menunjukkan bahwa penyebaran individu mangrove di lokasi penelitian cenderung merata atau memiliki tingkat kemerataan yang tinggi. Hasil penelitian ini mengindikasikan bahwa pelaksanaan tasamu rawanang selama 9 bulan telah berkontribusi positif terhadap restorasi mangrove secara alami. Meskipun demikian,  kondisi ekosistem masih mengalami tekanan sehingga upaya pengelolaan berkelanjutan yang inklusif harus terus diupayakan.  The Asai village community has been conducting tasamu rawanang since August 2022 to restore the condition of fisheries and coastal ecosystems. This research was conducted from April to May 2023 to determine whether natural mangrove restoration can occur without mangrove planting by studying mangrove vegetation structure and the mangrove ecological index. To obtain vegetation data, the quadrant transect method was used at three stations. The mangrove community structure analysed was the Important Value Index (INP), and the ecological indices analysed were the Diversity Index, Uniformity Index, and Dominance Index. The study's results indicate that there are 11 true mangrove species. The highest INP at the tree level was R. apiculata (154.743%), followed by B. gymnorrhiza (59.696%) and R. mucronata (26.655%). At the sapling and seedling level, R. apiculata also had the highest INP.  The species uniformity index at the tree level ranged from 0.595 to 0.625. For the sapling level, it ranged from 0.626 to 0.786. At the seedling level, it ranged from 0.768 to 0.772. This shows that the distribution of mangrove individuals in the study area tends to be even, or at least highly uniform. Furthermore, this study's findings demonstrate that, even within a relatively short 9-month implementation period, the Tasamu approach has contributed to natural mangrove restoration. Nevertheless, the ecosystem remains under pressure, and inclusive, sustainable management efforts need to continue.

Abstract

Nusmapi Island, located in Doreri Bay, Manokwari Regency, West Papua Province, is exposed to various anthropogenic activities that may introduce pollutants transported from the mainland. Fish are commonly used as bioindicators of aquatic pollution due to their ability to accumulate contaminants through food webs. This study was conducted on Nusmapi Island from June to August 2020 to examine differences in lead (Pb) concentrations among fish species and trophic groups. Fish samples represented two trophic groups: carnivores and herbivores that feed on algae and seagrass. Pb concentrations in fish muscle were analyzed using a Shimadzu AA-6880 atomic absorption spectrophotometer (AAS). The results showed that the average Pb concentration in the carnivorous group was 0.88 mg/kg, while the herbivorous group exhibited a higher average concentration of 1.13 mg/kg. The highest Pb concentration was recorded in Siganus spinus (1.34 mg/kg), whereas the lowest concentration was found in Neoniphon sammara (0.27 mg/kg). Elevated Pb levels in sediments and seagrass around Nusmapi Island may exert continuous environmental pressure on marine biota. As Pb concentrations in these environmental compartments exceed threshold levels, they may serve as major sources of contamination entering the food chain, increasing the toxicological risk for fish occupying higher trophic levels. Keywords: heavy metal Pb, bioaccumulation, Doreri Bay   Abstrak Pulau Nusmapi merupakan daerah yang berada di kawasan Teluk Doreri Kabupaten Manokwari yang merupakan daerah yang terdampak akibat padatnya berbagai aktivitas dari daratan yang ditransport secara langsung maupun tidak langsung. Ikan merupakan organisme yang sering digunakan sebagai bioindikator untuk menilai tingkat pencemaran suatu perairan. Penelitian dilakukan pada bulan Juni - Agustus Tahun 2020. Sampel ikan yang diambil terdiri dari dua kelompok trofik, yaitu karnivora dan herbivora yang memakan alga dan lamun. Tujuan utama penelitian ini adalah menguji perbedaan konsentrasi Pb berdasarkan spesies dan kelompok trofiknya guna memahami distribusi polutan di wilayah tersebut. Daging ikan sebanyak 30 g hingga 50 g diambil pada bagian punggung, selanjutnya daging ditimbang lalu dibungkus dengan aluminium foil. Daging ikan di destruksi dan dianalisis dengan atomic absorbtion spectrophotometer (AAS). Konsentrasi Pb pada kelompok karnivora adalah sebesar 0,88 mg/kg, sedangkan pada kelompok herbivora sebesar 1,13 mg/kg. Konsentrasi tertinggi ditemukan pada Siganus spinus (1,34 mg/kg), sementara konsentrasi terendah pada Neoniphon sammara (0,27 mg/kg). Mengingat logam Pb telah melampaui ambang batas pada sedimen dan lamun akan menjadi sumber utama polusi yang masuk ke dalam rantai makanan dan pada akhirnya menempatkan ikan sebagai tingkat trofik yang lebih tinggi pada posisi rentan secara toksikologis.
