Abstract

Abstrak: Pengabdian masyarakat ini dilaksanakan dengan tujuan memberikan edukasi pemanfaatan limbah sisik ikan yang dimanfaatkan sebagai bahan utama gelatin halal menjadi kembang gula jeli untuk meningkatkan perekonomian pada warga Margahayu, Bekasi. Adapun warga yang mengikuti kegiatan ini berjumlah 40 orang. Metode pelaksanaan kegiatan berupa penyampaian materi, pelatihan, dan pendampingan. Metode pelaksanaan tersebut dirangkum dalam tiga hari pelaksanaan workshop yang meliputi, workshop pembuatan gelatin halal, kemudian workshop pembuatan gummy, dan terakhir workshop packaging dan online marketing. Hasil evaluasi berdasarkan pre-test dan post-test yang diberikan, menunjukkan pengetahuan dan pemahaman terkait materi hari pertama tentang pembuatan gelatin halal meningkat dari 45% menjadi 88%, kemudian setelah penyampaian materi pembuatan kembang gula jeli (gummy) pemahaman peserta juga meningkat dari sebelumnya hanya 65% menjadi 92%, dan terakhir untuk penyampaian materi akan packaging dan online marketing pemahaman para peserta pun meningkat dari sebelumnya hanya 73% menjadi 95%. Diharapkan peserta mampu mengimplementasikan seluruh rangkaian materi workshop dengan baik dalam memanfaatkan limbah sisik ikan menjadi sebuah hasil akhir berupa produk yang bernilai jual ekonomi.Abstract: This community service is carried out with the aim of providing education on the utilization of fish scale waste which is used as the main ingredient of halal gelatin into jelly candy to improve the economy of the residents of Margahayu, Bekasi. The residents who participated in this activity amounted to 40 people. The method of implementing the activities is in the form of material delivery, training, and mentoring. The implementation method is summarized in a three-day workshop which includes a halal gelatin making workshop, then a gummy making workshop, and finally a packaging and online marketing workshop. The evaluation results based on the pre-test and post-test given, showed that knowledge and understanding of the material on the first day about making halal gelatin increased from 45% to 88%, then after the delivery of the material for making candy jelly (gummy) participants' understanding also increased from before. only 65% to 92%, and finally for the delivery of materials on packaging and online marketing the understanding of the participants also increased from previously only 73% to 95%. Participants are expected to be able to implement the entire series of workshop materials properly in utilizing fish scale waste into a final product in the form of products that have economic selling value.

Abstract

Erythrina variegata is one of the plants that have efficacy as thrombolytic agent. Isoflavonoid compound has the potential to accelerate the changes of plasminogen which plays important role in the forming of fibrin, that causes the formation of fibrin clots that can clog the arteries thus causing myocardial infarction. This study aimed to look at the activity of isoflavonoid compounds from Erythrina variegata bark which are expected to be candidates for thrombolytic drugs. This research aims at looking for the interaction of isoflavonoid compounds of bark  which is expected can be thrombolytic drug candidates. The method used in this research was the molecular docking with Autodock Vina and PyMOL software. The results showed that the value of ΔG binding affinity as ligands standard streptokinase -8.4 kcal/mol and isoflavonoid derivatives that have ΔG binding affinity value was lowest for the folitenol -11.5 kcal / mol and orientanol C -11.4 kcal/mol. Can be concluded that isoflavonoid derivatives in Erythrina variegata have better binding potential compared to streptokinase as standard ligands.

Abstract

Chewable lozenges atau gummy merupakan sediaan berbentuk kenyal yang dapat melepaskan zat aktifnya langsung di dalam mulut atau tenggorokan. Bahan yang berpengaruh dalam konsistensi gummy tersebut berasal dari basis. Dalam sediaan gummy, gelatin digunakan sebagai basis yang sebagian besar bersumber dari babi dan sapi. Identifikasi sumber bahan dapat dilakukan dengan teknik PCR dan sekuensing DNA. Penelitian ini bertujuan mengetahui sumber gelatin sediaan gummy impor tanpa logo halal. Isolasi DNA genom daging babi dan sediaan gummy dilakukan dengan GeneJet Kit. Isolat DNA kemudian diamplifikasi menggunakan primer spesifik DNA mitokondria sitokrom b. Selanjutnya, amplikon dianalisis dengan elektroforesis dan dilakukan sekuensing DNA. Hasil elektroforesis amplikon daging babi dan sediaan gummy A menghasilkan pita DNA dengan ukuran 553 bp. Analisis sekuensing DNA menunjukkan homologi dengan Sus scrofa breed long lin. Berdasarkan hasil tersebut dapat disimpulkan gelatin yang digunakan pada sediaan gummy A mengandung gelatin yang berasal dari babi Sus scrofa breed long lin.

Abstract

Penyakit menular atau penyakit infeksi adalah penyakit yang disebabkan oleh sebuah agen biologi (seperti virus, bakteria atau parasit). Salah satu lingkungan yang rentan penularan penyakit terutama pada anak-anak adalah lingkungan sekolah. Oleh karena itu perlu dibiasakan melakukan kebiasaan hidup sehat dengan kebiasaan mencuci tangan dan menggunakan antiseptik. Salah satu yang umum dipakai di kegiatan sehari-hari adalah hand sanitizer. Tujuan pelaksanaan kegiatan ini adalah memberikan informasi tentang kesehatan salah satunya tentang kebiasaan hidup sehat pada anak-anak sejak dini dengan kebiasaan mencuci tangan agar dapat mencegah penularan penyakit dan workshop pembuatan Hand sanitizer bahan alami. Peningkatan pemahaman tentang penularan penyakit dan cara pembuatan hand sanitizer sebagai salah satu solusi permasalahan tersebut. Metode pelaksanaan kegiatan Pengabdian masyarakat pada Mitra dilakukan dengan beberapa tahap meliputi penyampaian dalam bentuk edukasi tentang penyakit menular, tata cara mencuci tangan yang benar, pelatihan dan workshop pembuatan “Hand sanitizer” dari bahan alami. Berdasarkan hasil pengabdian yang dilakukan di TK Aisyiyah diketahui tingkat pengetahuan peserta tentang Perilaku Hidup Bersih dan Sehat (PHBS) masih rendah (35,1%) sedangkan hasil post test menunjukkan tingkat pengetahuan peserta tentang PHBS menjadi tinggi yaitu 81,1 %. Hasil positif tersebut menunjukkan bahwa kegiatan pengabdian masyarakat yang telah dilakukan pada mitra tersebut cukup efektif dalam meningkatkan pengetahuan peserta pengabdian mengenai penyakit menular, tata cara mencuci tangan yang benar dan pengetahuan tentang handsanitizer.

Abstract

The novel compound of 2-hydroxy-N-phenylnicotinamide was synthesized from modification of the UK-3A compound, which was known biologically active to inhibit bacterial and cancer cells growth. Synthesis of this compound was carried out in one-step reaction. Analog UK-3A compound namely 2-hydroxy-N-phenylnicotinamide was synthesized from 2-hydroxynicotinic acid as the starting material using amidation reaction with aniline in presence of DCC/DMAP in pyridine. Analog UK-3A was analyzed and identified using TLC, FT-IR, LC-MS and FT-NMR spectrofotometer. The yield after purification was 42.97%. The IC50 value 85 μg/mL was obtained from cytotoxicity test against P388 Murine Leukemia cells. It indicated that the synthesis products were sufficiently potential for leukemia P388 treatment.

Abstract

The estrogen receptor alpha (ERα) plays an important role in breast development and pro-proliferation signal activation in the normal and cancerous breast. The ERα inhibitors were potentially active as cytotoxic agents against breast cancer. This study was conducted in order to find Asymmetrical Hexahydro-2H-Indazole Analogs of Curcumin (AIACs) as hits of ERα inhibitor. A training set of 17 selected ERα inhibitors was used to create 10 pharmacophore models using LigandScout 4.2. The pharmacophore models were validated using 383 active compounds as positive data and 20674 decoys as negative data obtained from DUD.E. Model 2 was found as the best pharmacophore model and consisted of three types of pharmacophore features, viz. one hydrophobic, one hydrogen bond acceptor, and aromatic interactions. Model 2 was utilized for ligand-based virtual screening 186 of AIACs, AMACs, intermediates, and Mannich base derivative compounds. The hits obtained were further screened using molecular docking, analyzed using drug scan, and tested for its synthesis accessibility. Fourteen compounds were fulfilled as hits in pharmacophore modeling, in which 10 hits were selected by molecular docking, but only seven hits met Lipinski’s rule of five and had medium synthesis accessibility. In conclusion, seven compounds were suggested to be potentially active as ERα inhibitors and deserve to be synthesized and further investigated.

Abstract

ABSTRAK Manggis (Garcinia mangostana L.) merupakan buah yang prospektif untuk di kembangkan. Secara empiris kulit buah manggis digunakan untuk pengobatan berbagai penyakit. Penelitian sebelumnya menunjukkan bahwa fraksi butanol-air ekstrak kulit buah manggis secara in vitro mempunyai aktivitas sebagai imunomodulator. Pada penelitian ini, uji imunomodulator dilakukan secara in vitro dengan parameter aktivitas dan kapasitas fagositosis sel makrofag yang diambil dari peritoneum mencit, menggunakan fraksi etanol 70% dalam konsentrasi 0,1; 1,0; 10,0; 100,0; dan 1000,0 ppm. Stimuno sebagai kontrol positif. Dari hasil uji ANOVA satu arah diketahui bahwa hasil percobaan uji efek imunomodulator berdasarkan peningkatan aktivitas dan kapasitas fraksi etanol dari eksrak etanol 70% menunjukkan adanya perbedaan bermakna dari masing-masing perlakuan. Uji Tukey menunjukkan adanya perbedaan bermakna dari konsentrasi 0,1; 1,0; 10,0; 100,0; dan 1000,0 ppm dibandingkan terhadap kontrol negatif. Aktivitas dari fraksi etanol pada konsentrasi 0,1; 1,0; 10,0; dan 100,0 ppm lebih kecil dari kontrol positif, tetapi pada konsentrasi 1000,0 ppm mempunyai aktivitas yang sama atau tidak ada perbedaan yang bermakna dengan kontrol positif. Untuk kapasitas makrofag, fraksi etanol pada konsentrasi 0,1; 1,0; 10,0; 100,0; dan 1000,0 ppm memiliki nilai kapasitas lebih tinggi dibandingkan dengan kontrol negatif, tetapi masih dibawah kontrol positif. Dari data tersebut disimpulkan bahwa fraksi etanol eksrak etanol 70% pada dosis 1000,0 ppm memiliki aktivitas sebagai imunomodulator. Kata kunci: fagositosis, fraksi etanol, Garcinia, immunomodulator. ABSTRACT The immunomodulator assay of ethanol fraction of 70% ethanolic extract of Mangosteen pericarp based on increased number of intraperitonial macrophage phagocytocide latex after infected by S. epidermidis. Five concentrations of ethanol fraction of of 70% ethanolic extract Mangosteen pericarp was used, they were 0.1; 1.0; 10.0; 100.0; and 1000.0 ppm, respectively. Stimuno was used as the positive control, while distilled water was used as the negative control. Phagocyte activity was measured by in vitro ability of mice peritonial macrophages to phagocytize latex particles. The result of this research indicated ethanol fractions at the concentrations of 0.1; 1.0; 10.0; 100.0 ppm possessed a lower activity than that of positive control. Somehow, machrophage activity at the dose of 1000.0 ppm was not significantly different to that of the positive control. The result indicated that ethanol fraction of 70% ethanolic extract of mangosteen pericarp induced immune respone at the concentration of 1000 ppm. Abstract

Vannamei shrimps (Litopenaeus vannamei), also known as white leg shrimps are widely cultivated and consumed by many people. However, most consumers have removed the shrimp’s head which integrates with its digestive tract. The digestive tract of white leg shrimp contains digestive enzymes, including amylase. This study aimed to determine the protein content and amylase activity from Vannamei shrimps’ digestive tract using the size exclusion chromatography method. The protein isolation of size exclusion chromatography was prepared in three steps, namely; centrifugation, precipitation, and dialysis. The protein from the dialysis step was purified by using gel filtration chromatography. Each obtained fraction was determined the protein content and amylase activity by using spectrophotometer UV-Vis method. Abstract

The application of nuclear technology in the health sector is increasing. One example is the use of irradiation in production of wound dressings. Research activities have been conducted to study whether polyvinyl alcohol (PVA)-gelatin-based hydrogel from white snapper scales can be processed using gamma irradiation into wound dressings. A series of PVA (10 %) solutions containing gelatin in various concentrations (0-4 %) were treated with three freeze-thaw cycles and then irradiated at doses of 10 and 20 kGy. They were subsequently characterized using Fourier transform infrared (FTIR) spectroscopy and scanning electron microscope (SEM). Gel fraction, water absorption, and percentage of hydrogel water evaporation rate were tested gravimetrically, while the elongation at break and the tensile strength of the hydrogels were tested with a universal testing meter. The evaluation showed that the hydrogel gel fraction decreased with increasing gelatin concentration from 0 % to 4 % for both irradiation doses (10 and 20 kGy). The rising gelatin concentration demonstrated that increasing gamma radiation dose improved the hydrogel's water absorption, evaporation rate, tensile strength, and elongation at break. PVA-gelatin hydrogel with irregular pore structure was observed from SEM test results. The FTIR measurement results confirmed the formation of crosslinks in the hydrogel matrix. The PVA-gelatin hydrogel produced through gamma irradiation could be used for wound dressings.

Abstract

The development of analog curcumin compounds by modifying the structure of monocarbonyl into an analog indazole of curcumin (AIC) is recognized to have a great potential. Still, only a few reports have been available. Rarely occurring in nature, indazole molecules are typically created through chemical synthesis. Therefore, this study aimed to synthesize six new AIC compounds with a particular focus on testing in vitro antioxidant activity using the DPPH and FRAP methods, as well as anti-inflammatory activity using the protein denaturation method. The results showed that the compounds formed had high anti-inflammatory activity but low antioxidant activity. All synthesis products produced higher anti-inflammatory activity than standard diclofenac sodium and curcumin compounds. Specifically, compound 3a showed the highest anti-inflammatory activity with an IC50 = 0.548 ± 0.062 μM. Therefore, it was concluded that compound 3a has the potential to be further studied for anti-inflammatory activity.
