Abstract

Indo-Pacific bottlenose dolphin (Tursiops aduncus) is a marine mammal that lives in relatively small populations. The geographic ranges of this species are susceptible to the effects of human activities, thereby necessitating conservation efforts to prevent extinction. Therefore, this study aimed to evaluate the daily quality of dolphin sperm after several days of refrigeration. The sperm of two male dolphins were stored at 4oC for 4 days, and the quality was observed daily to determine the motility, viability, membrane integrity, and sperm abnormalities. Sperm samples were divided into four groups, consisting of two centrifuged followed by the removal of seminal plasma, and two groups without centrifugation, containing 100x106 and 200x106 sperm/ml each. After liquid storage, the motility of sperm was 63-75% with no significant reduction in the first 3 days. Sperm viability following storage was 65-75% and the percentage with abnormal morphology ranged from 2-6%. Furthermore, there was no significant increase in abnormal morphology of sperm on any day of storage for 3 days. Sperm membrane integrity was 36-49%, with no significant reduction in the membrane integrity in the first 2 days. There was no significant difference in sperm quality, although centrifugation and removal of seminal plasm had a slight effect. The results of this study showed that Indo-Pacific bottlenose dolphin sperm could be stored for a short period as liquid storage while maintaining a quality that allows for future use.

Abstract

One factor that affects frozen semen characteristics is bull age. Increasing age induces changes in sperm that reduce frozen semen quality. This study aimed to evaluate the frozen semen characteristics of Bali bulls aged 3, 8, and 13 years. Frozen semen was derived from fresh semen with sperm motility > 70%. The frozen semen characteristics tested included motility, viability, plasma membrane integrity, acrosome integrity, and sperm abnormalities. Sperm motility was analyzed by computer-assisted sperm analysis (CASA). Viability was assessed using eosin-nigrosin staining; plasma membrane integrity was assessed using the hypoosmotic swelling test (HOST); acrosome integrity was assessed using Giemsa staining; and sperm abnormalities were assessed using Williams staining. The plasma membrane integrity of the 13-year-old group was significantly lower (P < 0.05) than that of the younger age groups. However, there were no significant differences (P > 0.05) in the other parameters. We conclude that age affects the integrity of the sperm plasma membrane in frozen semen from Bali bulls.

Abstract

Glutamine is a non-essential amino acid that can reduce free radicals and act as an antioxidant. This study aims to determine the effect of adding glutamine in egg yolk tris diluent at concentrations of 0 mM (K), 2,5 mM (G2,5), 5 mM (G5), and 7,5 mM (G7,5) on the quality of liquid semen and kept in a refrigerator at 4 °C. Microscopic evaluation was performed immediately after dilution (H0) and every 24 hours until day 5 (H5). The results showed that the G 7,5 mM group could maintain better motility and viability (P < 0,05) than the K, G 2,5, and G 5 groups. The addition of glutamine did not have a significant effect (P> 0.05) in maintaining the MPU of spermatozoa. Glutamine with a concentration of 7.5 mM is the best concentration to maintain the quality of sheep's liquid semen.

Abstract

Excessive free radicals can induce oxidative stress, leading to the inhibition of phosphorylation processes that supply energy for sperm capacitation and the acrosome reaction in spermatozoa during fertilization. This study investigated the impact of incorporating the antioxidant melatonin into the tris-fructose egg yolk diluent on the quality of ram liquid semen preserved at 4°C. Fresh semen with ≥70% motility was diluted in the tris-fructose egg yolk group (control), tris-fructose egg yolk with 0.5 mM melatonin (TFEY + M0.5), tris-fructose egg yolk with 1 mM melatonin (TFEY + M1), tris-fructose egg yolk with 2 mM melatonin (TFEY + M2). Semen was stored in the refrigerator at 4°C for 5 days (D0−D5). The motility, viability, abnormality, and plasma membrane integrity of the semen were evaluated every 24 hours. The results indicated no significant difference (P>0.05) between the control group and the liquid semen treatment at D0. Liquid semen quality was significantly improved with melatonin treatment compared to the control group. The results of this study indicate that TKTF + M0.5 had the highest motility and viability values ​​and can maintain abnormality values. The TKTF + M2 group had a higher plasma membrane integrity value than the other treatment groups.

Abstract

Improvement of canine reproduction has been developed by artificial insemination (AI) for effective breeding. -tocopherol widely recognized as an antioxidant that has been used to improve the quality of the spermatozoa. This study evaluated the effect of -tocopherol supplementation in dog chilled semen quality. Beagle dog breed semen samples were obtained from male Beagles using massage technique. The semen was then diluted with tris-skim milk extender supplemented with -tocopherol of various concentration, 0% as control group (T-0), 0.1% (T-0.1), 0.2% (T-0.2), and 0.3% (T-0.3), then were stored at 4 C and evaluated daily for four days, particularly for variables namely sperm motility, sperm viability, and plasma membrane integrity. Results showed that the quality of spermatozoa was higher when spermatozoa preserved in extender containing of 0.1% or 0.2% -tocopherol compared to those of control and 0.3% -tocopherol group (P0.05). The highest quality found in group with 0.2% -tocopherol. In conclusion, supplementation -tocopherol 0.2% in dog chilled semen extender is effective for improving and maintaining chilled dog sperm quality for 4 days of storage.

Abstract

Age is among the factors affecting reproductive organs, particularly semen characteristics. Semen characteristics are also influenced by composition of seminal plasma that is ejaculated with the sperm. Therefore, this study aimed to investigate characteristics of fresh semen and seminal plasma composition from Balinese cattle aged 3, 8, and 13 years. Semen characteristics observed were concentration, progressive motility, viability, plasma membrane integrity, acrosome integrity, and sperm abnormalities. Biochemical composition of seminal plasma analyzed was fructose, malondialdehyde (MDA), total antioxidant capacity (TAC), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), total protein (TP), albumin, cholesterol, and triglycerides. Mineral compositions observed were Na, K, Ca, Mg, Zn, Cu, Fe, and Se. The results showed that there were no significant differences (p > 0.05) in semen characteristics across age groups, meaning that semen of Balinese cattle can still be processed into frozen semen even at an older age, provided it meets the SNI 4869-1:2024 standards. Furthermore, the concentrations of each seminal plasma biochemical component were as follows: fructose (377.5–615.4 mg/dL), MDA (9.7–10.36 nmol/mL), TAC (66.22–67.22%), GOT (203–265 U/L), GPT (19–35 U/L), TP (4.3–5.5 g/dL), albumin (1.7–1.9 g/dL), cholesterol (11–35 mg/dL), and triglycerides (822–1.078 mg/dL). Surprisingly, the concentration of triglycerides in semen plasma in this study was very high compared to those reported in previous studies on several cattle breeds. Seminal plasma mineral concentrations included Na (1.500–1.900 mg/kg), K (600–700 mg/kg), Ca (600–800 mg/kg), Mg (38.85–45.16 mg/kg), Zn (5.33–9.64 mg/kg), Cu (0.06–0.12 mg/kg), Fe (11.85–15.88 mg/kg), and Se (0.92–1.48 mg/kg). These findings indicate that age did not affect semen characteristics, biochemical, and mineral composition of seminal plasma in Balinese cattle.

Abstract

Collagen is one of the determinant factors of beef tenderness. This study analyzed the expression and content of type III collagen connective tissue in two muscle types of Aceh cattle through immunohistochemical staining. Meat samples were obtained from the gluteobiceps and psoas major muscles of three Aceh cattle bulls weighing 250-300 kg, with a body condition score of three. Histology preparations were created from samples and subjected to immunohistochemical staining utilizing specific antibodies targeting type III collagen. Qualitative data were analyzed descriptively, while quantitative data were analyzed using IMAGE J software. The results showed that the expression of type III collagen connective tissue was found in the endomysium and perimysium layers of both muscles. The expression of type III collagen connective tissue varied significantly among the muscles (P<0.05). The gluteobiceps muscle had an average percentage of the area containing type III collagen connective tissue of 4.47±2.41%, while the psoas major muscle was lower at 2.51±1.69%. This study concludes that the gluteobiceps muscle of Aceh cattle has a higher amount of type III collagen connective tissue than the psoas major muscle.

Abstract

Trichinellosis merupakan penyakit zoonotik yang disebarkan oleh konsumsi daging mentah atau kurang matang yang mengandung larva cacing Trichinella sp. Keberadaan Trichinella spiralis di Indonesia telah teridentifikasi sejak masa kolonial Belanda, tetapi informasi mengenai kejadian trichinellosis pada manusia dan hewan di negara ini masih sangat terbatas. Sejumlah penelitian terkini mengindikasikan terjadinya peningkatan kasus trichinellosis pada hewan, khususnya babi dan babi hutan. Hal ini tidak hanya berpotensi mengancam kesehatan manusia, tetapi juga satwa liar karnivora yang mengonsumsi daging tersebut, termasuk satwa yang berada dalam penangkaran ex situ. Penelitian ini dilakukan untuk mendeteksi keberadaan larva Trichinella sp. pada daging babi hutan hasil buruan di Provinsi Bengkulu yang dimanfaatkan sebagai sumber pakan satwa liar karnivora di Taman Margasatwa Ragunan, Jakarta. Sebanyak 44 sampel daging babi hutan asal Bengkulu serta serum dari empat Harimau Bengala, dua Jaguar, dan satu Singa Afrika asal Taman Margasatwa Ragunan diperiksa menggunakan teknik Indirect Enzyme Linked Immunosorbent Assay (ELISA) untuk mendeteksi keberadaan antibodi terhadap Trichinella sp. Dari seluruh sampel daging yang diuji, tujuh (15,9%) menunjukkan hasil positif, 35 (79,5%) negatif, dan dua (4,5%) dubius. Pengujian serum Harimau Bengala menunjukkan satu (25%) seropositif, sedangkan sisanya dubious. Adapun pengujian pada serum Jaguar dan Singa Afrika menunjukkan hasil seronegatif. Harimau Bengala yang diuji merupakan individu yang lahir di Taman Margasatwa Ragunan dan hanya mendapat pasokan pakan berupa babi hutan asal Bengkulu. Oleh sebab itu, terdapat indikasi kuat bahwa infeksi Trichinella sp. pada babi hutan di Bengkulu dapat berpotensi memengaruhi kesehatan satwa liar karnivora yang ditangkarkan di Taman Margasatwa Ragunan, Jakarta.

Abstract

Semen was collected using an artificial vagina, and then examined macroscopically and microscopically. Semen with motility of 70% was diluted into different extenders: tris egg yolk-glucose (TEYG), tris egg yolk-glucose extender with 0.5 mM melatonin (TEYG + M0.5), tris egg yolk-glucose extender with 1 mM melatonin (TEYG + M1), tris egg yolk-glucose extender with 2 mM melatonin (TEYG + M2). All semen samples were stored at 4 C for 5 days (H0-H5) and examined every 24 hours. Semen was examined for spermatozoa motility, viability, abnormality, and membrane plasma integrity. The results showed that there were no significant changes of semen quality on H0 (P0.05). In general, the melatonin supplemented groups had a higher semen quality value than the control. Melatonin supplementation can protect and maintain the quality of chilled semen. In addition, the result showed that TEYG + M1 extender was the best extender treatment, which was indicated by higher values than other treatments.

Abstract

The aim of this study was to evaluate the ability of rats Leydig cells collected with Nycodenz gradient in producing testosterone in vitro. Leydig cells were collected using 5 column of Nycodenz gradient (4, 8, 10, 12, and 15%) and cells were evaluated regarding its concentration, viability, and purity of Leydig cells. Media used to cultured Leydig cells were Dulbeccos Modified Eagles Medium (DMEM)+10% newborn calf serum (NBCS); DMEM+10% NBCS+2,5 IU/mL human chorionic gonadotrophin (hCG); DMEM+10% NBCS+ 5 g/mL insulin, 10 g/mL transferrin, and 5 g/mL Se (ITS); DMEM+10% NBCS+hCG+ITS at 5% CO2 incubator with temperature of 37.5 C for 3 days. Culture medium was collected every day for testosterone analysis with enzyme-linked immunosorbent assay (ELISA). By adding ITS to the medium, Leydig cells concentration was significantly increased (8.92x106 cells/mL) compared to medium with serum (7.74x106 cells/mL) or hCG (7.68x106 cells/mL) (P0.05). ITS and hCG in medium significantly increased Leydig cells concentration (10.40x106 cells/mL) at day 3 of culture (P0.05). The result of parallelism test showed that the assay obtained good validity to measure testosterone concentration in culture medium. Testosterone in medium was detected at 1.80-2.60 ng/mL at day 1 of culture. In conclusion, Leydig cells collected with Nycodenz gradient had no effect to testosterone secretion from Leydig cells in vitro.
